背景技术
当今社会随着人们生活节奏的加快以及工作压力的加大,常常会造成饮食不规律,引发胃肠炎、胃溃疡、胃胀、食欲不振、消化不良等胃肠疾病。据WHO统计,仅中国就有1.2亿胃肠疾病患者,其中,中老年人占到70%以上,若不及时治疗,反复发作容易转化成胃癌,因此研发一种治疗肠胃的药物具有中药意义。
刺梨,为蔷薇科多年生落叶灌木缫丝花的果实,具有很高的药用价值,有健胃、消食、滋补、止泻、清热生津、美容养颜的功效。特别是刺梨富含超氧化物歧化酶(SOD),具有抗衰老、防癌、抗病毒和抗辐射的作用。因此,以刺梨为原料,并配伍其他组分制成一种组合物,不仅具有健胃消食的功效,同时保健效果显著。
发明内容
本发明的目的在于针对现有技术不足,提供一种刺梨健胃消食药物组合物及其制备方法。本发明的药物组合物具有健胃消食、滋补止泻、清热生津、补气安神和消肿解毒的功效。各组分相互配合,发挥协同作用,起到显著的养胃、健胃功效,其效果已经远超过各组分单独作用时所起效果的总和。
为实现上述目的,本发明采用如下技术方案:
一种刺梨健胃消食药物组合物,其原料组成按重量份数计为:刺梨提取物150~250份、苹婆种仁80~100份、楮实子提取物60~90份、红豆蔻提取物100~135份和刺嫩芽根提取物100~150份。
优选的组合物的原料组成按重量份数计为:刺梨提取物180~220份、苹婆种仁90~100份、楮实子提取物70~80份、红豆蔻提取物110~130份和刺嫩芽根提取物110~140份。
优选的组合物的原料组成按重量份数计为:刺梨提取物200~215份、苹婆种仁80~90份、楮实子提取物75~85份、红豆蔻提取物105~120份和刺嫩芽根提取物120~130份。
优选的组合物的原料组成按重量份数计为:刺梨提取物200份、苹婆种仁90份、楮实子提取物75份、红豆蔻提取物115份和刺嫩芽根提取物125份。
所述刺梨提取物的制备方法为:取刺梨洗净后进行压榨后过滤得刺梨果汁和刺梨渣,往所得刺梨渣中加入4倍重量的水,85℃下提取1小时,过滤得刺梨渣提取液;将所得刺梨果汁和刺梨渣提取液混合后真空浓缩至稠膏,冷冻干燥并粉碎后得刺梨提取物。
所述楮实子提取物的制备方法为:取楮实子先用6倍重量85wt%的乙醇回流提取2次,每次1.5小时,收集醇提有效成分;将醇提后的药渣用6倍重量的水煎煮2小时,过滤收集水提有效成分;合并醇提液和水提液并真空浓缩得浸膏,将所得浸膏喷雾干燥得楮实子提取物。
所述红豆蔻提取物的制备方法为:取红豆蔻先用6倍重量85wt%的乙醇回流提取2次,每次1.5小时,收集醇提有效成分;将醇提后的药渣用6倍重量的水煎煮2小时,过滤收集水提有效成分;合并醇提液和水提液并真空浓缩得浸膏,将所得浸膏喷雾干燥得红豆蔻提取物。

刺梨能提高胃液量、总酸排出量及胃蛋白酶活性,起到健胃消食的作用,而且其所含有的刺梨多糖能显著提高抗应激性,所含的SOD能调节人体免疫系统,起到延缓衰老的作用。苹婆,以种入药,性味:甘,温;功用主治:温胃,杀虫。治虫积腹痛,翻胃吐食,疝痛。红豆蔻,性味:味辛;性温;功用主治:温中燥湿、醒脾消食、利水除湿、和血排脓、消肿解毒。刺嫩芽根中总皂甙为人参根总皂甙的3倍左右,具有健胃收敛、免疫防癌的作用。楮实子,性寒,味甘;功能主治:补肾清肝、明目、利尿;用于腰膝酸软、虚劳骨蒸、砂晕目昏、目生翳膜、水肿胀满。《药性通考》:“楮实子,阴痿能强,水肿可退,充肌肤,助腰膝,益气力,补虚劳,悦颜色,壮筋骨,明目。久服滑肠。补阴妙品,益髓神膏。世人弃而不用者,因久服滑肠之语也,楮实滑肠者,因其润泽之故,非嫌其下行之速也,防其滑而以茯苓、薏仁、山药同施,何惧其滑乎”。
本发明的有益效果在于:本发明的药物组合物由刺梨提取物、苹婆种仁、楮实子提取物、红豆蔻提取物和刺嫩芽根提取物复配而成,含有多种有效成分,具有健胃消食、滋补止泻、清热生津、补气安神和消肿解毒的功效。各组分相互配合,发挥协同作用,起到显著的养胃、健胃功效,其效果已经远超过各组分单独作用时所起效果的总和。
具体实施方式
以下结合具体实施例对本发明做进一步说明,但本发明不仅仅限于这些实施例。
实施例1
一种刺梨健胃消食药物组合物,其原料组成按重量份数计为:刺梨提取物150mg、苹婆种仁80mg、楮实子提取物60mg、红豆蔻提取物100mg和刺嫩芽根提取物100mg份。将苹婆种仁粉碎后过200目筛得到苹婆种仁粉,并与其他组分按照重量配比混合制得。
实施例2
一种刺梨健胃消食药物组合物,其原料组成按重量份数计为:刺梨提取物250mg、苹婆种仁100mg、楮实子提取物90mg、红豆蔻提取物135mg和刺嫩芽根提取物150mg。将苹婆种仁粉碎后过200目筛得到苹婆种仁粉,并与其他组分按照重量配比混合制得。
实施例3
一种刺梨健胃消食药物组合物,其原料组成按重量份数计为:刺梨提取物180mg、苹婆种仁90mg、楮实子提取物70mg、红豆蔻提取物110mg和刺嫩芽根提取物110mg。将苹婆种仁粉碎后过200目筛得到苹婆种仁粉,并与其他组分按照重量配比混合制得。
实施例4
一种刺梨健胃消食药物组合物,其原料组成按重量份数计为:刺梨提取物200 mg、苹婆种仁90 mg、楮实子提取物75 mg、红豆蔻提取物115 mg和刺嫩芽根提取物125 mg。将苹婆种仁粉碎后过200目筛得到苹婆种仁粉,并与其他组分按照重量配比混合制得。
健胃消食效果测试
本发明研究了刺梨提取物、苹婆种仁、楮实子提取物、红豆蔻提取物、刺嫩芽根提取物及其配伍对对大鼠溃疡指数、胃粘膜血流量、胃液酸度、胃酸分泌速度和胃壁结合液的影响。结果发现,该组合物起到协同增强对肠胃疾病的疗效。
其中实验性大鼠胃溃疡模型采用幽门结扎型溃疡,结扎大鼠幽门造成胃液在胃中滞留,致溃疡因素增强,胃壁防御能力减弱,导致溃疡形成。
溃疡指数的测定:选取体重在180~200g健康大鼠,禁食自由饮水72h后,用水合氯醛麻醉,打开腹腔,用缝合线结扎幽门十二指肠结合部。结扎时小心避开血管,缝合腹壁。术后18h结扎贲门取出全胃,将胃浸入1%甲醛溶液中,10min后,在胃大弯处剪一小口收集胃液。然后沿胃大弯将胃剪开,用自来水轻轻冲洗胃黏膜面,将胃平铺于白纸上,观察溃疡发生情况并测量溃疡斑大小(mm)即得溃疡指数。
胃粘膜血流量的测定:大鼠给药1小时后结扎幽门和贲门,从右股静脉缓慢注射负荷量中性红 (1200μg/kg),再以中性红 (1200μg/kg·h) 维持。1小时后采血,收集胃液,分别测定中性红浓度,计算清除率,即胃粘膜血流量(mL/min)。
胃液酸度的测量:幽门结扎收集大鼠胃液,以酚酞为指示剂,以 0.01mol/L NaOH滴定直至溶液变红不退为止,即为胃酸浓度(mEq/L)。
胃壁结合粘液:取幽门结扎大鼠将胃剖开,浸入阿利新兰液孵育2小时 (25℃),染液离心后取上清液在751型分光光度计上比色。
胃蛋白酶的测定:胃蛋白酶是胃溃疡形成的重要攻击因子之一,它可与盐酸(胃酸)一起导致溃疡的形成。胃蛋白酶分泌增加或胃酸分泌增加均可导致溃疡;而当胃粘膜的防御机能减弱时,胃蛋白酶的攻击力量相对处于优势,也可导致溃疡的形成。采用麦特氏毛细玻管测定法。将内径1~2mm粗细均匀的毛细玻管裁成10cm长,洗净烤干待用。用鸡蛋一个,取其蛋清充分打匀后用滤布过滤,将备用毛细玻管利用虹吸作用,灌满蛋清(注意管内无气泡)。然后放在85℃热水中使蛋白凝固,贮冰箱中备用。试验时取胃液1ml放入50ml的三角烧瓶中,加0.05mol/L盐酸溶液15ml摇匀,放进蛋白管二根。塞好瓶口,在37℃恒温孵育24h,取出蛋白管,用尺测量蛋白管两端透明部分的长度 (mm),以四端之值求其平均值。胃蛋白酶活性单位=平均值2×16。
空白组为正常无溃疡大鼠组,对照组为幽门结扎型溃疡大鼠,该组无任何药物治疗,其他实验组为幽门结扎型溃疡大鼠胃部灌注对应药物或组合,灌注量为2mg/次,每日3次。分别测量各项指标。其结果见表 1。

从表1中可以看出,对照组中溃疡指数、胃液酸度和胃蛋白酶活力均显著高于空白组,而胃粘膜血流量和胃壁结合粘液显著低于空白组,说明大鼠溃疡模型建立成功。刺梨提取物、苹婆种仁、楮实子提取物、红豆蔻提取物和刺嫩芽根提取物组中溃疡指数、胃液酸度和胃蛋白酶活力要比对照组低,而胃粘膜血流量和胃壁结合粘液要比对照组高,说明刺梨提取物、苹婆种仁、楮实子提取物、红豆蔻提取物和刺嫩芽根提取物对大鼠胃溃疡均有一定功效。而实施例1~4组中各项数据均显著优于刺梨提取物、苹婆种仁、楮实子提取物、红豆蔻提取物和刺嫩芽根提取物单独组别,也优于各单独组别作用时的效果之和,说明刺梨提取物、苹婆种仁、楮实子提取物、红豆蔻提取物和刺嫩芽根提取物之间发挥协同作用,起到显著的健胃、养胃、消食的效果。
以上所述仅为本发明的较佳实施例,凡依本发明申请专利范围所做的均等变化与修饰,皆应属本发明的涵盖范围。
Claims (6)
1.一种刺梨健胃消食药物组合物,其特征在于:所述组合物的原料组成按重量份数计为:刺梨提取物150~250份、苹婆种仁80~100份、楮实子提取物60~90份、红豆蔻提取物100~135份和刺嫩芽根提取物100~150份。
2.根据权利要求1所述的刺梨健胃消食药物组合物,其特征在于:所述组合物的原料组成按重量份数计为:刺梨提取物180~220份、苹婆种仁90~100份、楮实子提取物70~80份、红豆蔻提取物110~130份和刺嫩芽根提取物110~140份。
3.根据权利要求1所述的刺梨健胃消食药物组合物,其特征在于:所述组合物的原料组成按重量份数计为:刺梨提取物200~215份、苹婆种仁80~90份、楮实子提取物75~85份、红豆蔻提取物105~120份和刺嫩芽根提取物120~130份。
4.根据权利要求1所述的刺梨健胃消食药物组合物,其特征在于:所述组合物的原料组成按重量份数计为:刺梨提取物200份、苹婆种仁90份、楮实子提取物75份、红豆蔻提取物115份和刺嫩芽根提取物125份。
5.根据权利要求1~4中任一项权利要求所述的刺梨健胃消食药物组合物,其特征在于:所述刺梨提取物的制备方法为:取刺梨洗净后进行压榨后过滤得刺梨果汁和刺梨渣,往所得刺梨渣中加入4倍重量的水,85℃下提取1小时,过滤得刺梨渣提取液;将所得刺梨果汁和刺梨渣提取液混合后真空浓缩至稠膏,冷冻干燥并粉碎后得刺梨提取物。
6.一种制备如权利要求1~4中任一项权利要求所述的刺梨健胃消食药物组合物的方法,其特征在于:具体包括以下步骤:将苹婆种仁粉碎后过200目筛得到苹婆种仁粉,并与刺梨提取物、楮实子提取物、红豆蔻提取物和刺嫩芽根提取物按照重量配比混合。
