本发明涉及电泳转移一种大分子核酸的方法,其主要是首先制做支持胶,然后将硝基纤维素纸,电泳后切下的含DNA酶解谱带的琼脂条先后垂直贴在一块支持胶的断面上,再用另一块支持胶一将硝基纤维素纸条和电泳胶条夹紧,给两块支持胶搭上纸桥,接通电源即可,使用该方法可防止杂质离子进入,同时它还适用于任何一种电极设备,时间短、转移完全,该方法制做简单、使用方便,安全可靠。

电泳转移一种大分子核酸的方法
本发明涉及采用电泳转移一种大分子核酸的方法
在进行核酸的各种杂交实验前,都要先做核酸的硝基纤维素纸转移,即把经琼脂糖凝胶电泳分离的DNA片段转移到硝基纤维素纸上,然后进行DNA-DNA,DNA-RNA杂交实验,根据国同内外现通常采用核酸转移方法为吸印法,电转移方法,吸印法对于大于15Kb的DNA分子,需要很长的转移时间(至少20小时以上),而且转移常常不彻底,只是利用了吸水纸对水的吸力,而使DNA分子随水向上移动,由于吸水纸的作用力很有限,转移速度自然缓慢,电转移方法采用不锈钢电极专用设备,在不锈钢板上需人为打眼,其孔径及孔的均匀程度不能保证,很容易引进杂质离子,同时它必须是专用的电极设备。本发明经过大量的试验研究探索,研制出采用电泳转移一种大分子核酸的方法。
本发明的目的在于,研制电泳转移一种大分子核酸的方法,其主要是首先制做支持胶,然后将硝基纤维素纸,电泳后切下的含DNA酶解谱带的琼脂条先后垂直贴在一块支持胶的断面上,再用另一块支持胶将硝基纤维纸条和电泳胶条夹紧,给两块支持胶搭上纸桥,接通电源即可,该方法是利用琼脂糖中自身的孔径转移,可防止杂质离子进入,同时它还适用于任何一种水平电泳设备,时间短,转移完全,该方法制做简单,使用方便,安全可靠。
本发明所述的电泳转移一种大分子核酸的方法,首先制做支持胶,然后将硝基纤维素纸、电泳后切下的含DNA酶解谱带的琼脂条先后垂直贴在一块支持胶的断面上,再用另一块支持胶将硝基纤维素纸条和电泳胶条夹紧,给两块支持胶搭上纸桥,接通电源,电流50-100MA,电泳1-2小时。支持胶是用电泳缓冲液配制1-2%琼脂糖溶液,倒入玻璃盒内凝固,凝固后除去玻璃盒切成胶条即可。本发明所述的电泳转移方法还可推广用于聚丙烯酰胺凝胶中DNA片段的转移,聚丙烯酰胺凝胶电泳,但聚丙烯酰胺凝胶的孔径小,一般难于进行通常的转移,而改用电泳转移,则可得到好的结果。
实施例1:
首先制做支持胶,用电泳缓冲液配制定1%琼脂糖溶液,倒入玻璃盒内凝固,凝固后除去玻璃盒内切成胶条即成支持胶,然后将硝基纤维素纸,电泳后切下的含DNA敏解谱带的琼脂条,先后垂直贴在一块支持胶的垂直断面上排尽气泡,再用另一块支持胶将硝基纤维素纸条和电泳胶条夹紧,给两块支持胶搭上纸桥,接通电源、电流50MA,电泳1小时即可得到转移完全的硝基纤维素纸条,用于杂交实验。
实施例2:
首先制做支持胶,用电泳缓冲液配制2%琼脂糖溶液,倒入玻璃盒内凝固,凝固后除去玻璃盒切成胶条即成支持胶,然后将硝基纤维素纸、电泳后切下的含DNA酶解谱带的琼脂条,先后垂直贴在一块支持胶的垂直断面上排尽汽泡,再用另一块支持胶将硝基纤维素纸条和电泳胶条夹紧,给两块支持胶搭上纸桥,接通电源,电流100MA,电泳2小时,即可得到转移完全的硝基纤维纸条,用于杂交实验。

Claims (2)

1、一种电泳转移一种大分子核酸的方法.其特征在于,首先制做支持胶。然后将硝基纤维素纸、电泳后切下的含DNA酶解谱带的琼脂条先后垂直贴在一块支持胶的断面上,再用另一块支持胶将硝基纤维素纸条和电泳胶条夹紧,给两块支持胶搭上纸桥,接通电源,电流50-100MA,电泳1-2小时。
2、根据权利要求1所述的电泳转移一种大分子核酸的方法,其特征在于支持胶是用电泳缓冲液配制1-2%琼脂糖溶液,倒入玻璃盒内凝固,凝固后除去玻璃盒切成胶条即可。
