Abstract
一种用于恢复剖腹产新生儿共生微生物抹布的制备方法,是将待产孕妇阴道分泌物用干净脱脂棉吸收,之后擦拭于剖腹产新生儿的嘴部、面部及身体的其他部位,弥补剖腹产新生儿细菌匮乏导致的后期免疫缺陷类疾病和抵抗力低下。本发明直接将待产孕妇的阴道分泌物取出,用于擦拭剖腹产新生儿嘴部、面部、肛门等身体的其他部位,恢复其共生微生物,具有简便、安全、有效、可操作性等优点。

技术领域
本发明涉及一种用于恢复剖腹产新生儿共生微生物抹布的制备方法,属于微生物生物技术与生物医药领域。
背景技术
现代医学技术的发展为人类创造了剖腹产手术, 这种新的分娩方式挽救了无数产妇和婴儿的生命, 为保障人类的健康和人口再生长做出了巨大贡献。在阴道分娩无法达成, 或经阴道分娩可能对产妇或新生儿造成威胁时, 才需要剖腹产。然而近年来,我国剖腹产的孕妇数量逐年增多,如今我国的剖宫产率已经高达40%-50%,某些地区甚至更高。
分娩方式的选择直接影响新生儿共生菌群的组成。在顺产的婴儿中,观察到微生物组成往往与母体产道和肠道相似,而在剖腹产的婴儿中,微生物组成往往更接近于母亲的皮肤以及医院环境包括医护人员和其它新生儿。阴道分娩儿的菌群定植主要受母亲阴道和会阴区域菌群的影响,剖腹产儿则主要受医院环境的影响。以肠道菌群为例,前者最初的肠道菌群主要是母亲阴道和外阴周围的细菌,多为乳酸杆菌、普雷沃氏菌、双歧杆菌等;而后者主要以链球菌、葡萄球菌、厌氧球菌、类杆菌、丙酸杆菌为主。其中金黄色葡萄球菌是导致新生儿败血症的主要原因,双歧杆菌数量则很低甚至没有,而双岐杆菌对机体健康及免疫功能有重要作用。
正常阴道分娩过程中婴儿经过产道时会被产道挤压,在分娩过程中及出生后不久,母亲身体里的细菌就会通过胎儿呼吸道及消化道最早进入胎儿体内定植,这个细菌定植对婴儿一生的健康非常重要,它为一个基本无菌的新生儿肠道内建立起新的微生物生态系统。这些定植的细菌主要是在分娩过程,产道环境微生物起作用的结果。而新生儿的这一微生物生态系统的构成成分直接影响婴儿的免疫功能。剖腹产新生儿因出生时没有经产道挤压、不能及时建立共生菌群。最终造成剖腹产儿的微生物生态系统的构成与顺产儿有很大的不同。
大量的研究发现,剖腹产新生儿中观察到的微生物组成改变与远期多种疾病发生风险升高有着密切的关系,包括过敏、湿疹、哮喘、肥胖、1型糖尿病、慢性相关炎症性疾病等。英国研究人员警告说,剖腹产的孩子比顺产的孩子更易患哮喘,剖腹产孩子患哮喘的概率比正常顺产的孩子高80%。剖腹产剥夺了孩子最先感觉统合锻炼的权利,使得剖腹产儿容易患感统失调。而且剖腹产令“湿肺症”增多。总的来说,与自然分娩相比,剖腹产新生儿的免疫力及抗感染能力比自然分娩的孩子要差。
发明内容
本发明的目的是针对现有技术不足提供一种用于恢复剖腹产新生儿共生微生物抹布的制备方法,该方法具有简单方便、安全有效、可操作等优点,有助于更好地增强和提高剖腹产新生儿的抗感染能力和免疫功能。
本发明所述方法包括下列步骤:
1、阴道分泌物的收集和保存:在进行剖腹产手术之前1小时,将一块大小适宜的无菌纱布放置于待产母亲的阴道当中,手术开始时取出并保存在无菌容器中。
2、阴道分泌物的擦拭:在新生儿产下的1—3分钟内,将浸有阴道分泌物的纱布依次擦拭新生儿的嘴部、面部、肛门等身体的其他部位。
3、纱布微生物的鉴定:为验证纱布所浸的微生物与阴道分泌物的差异性,鉴定纱布以及母亲阴道分泌物样本中的菌群情况。
4、新生儿共生微生物的鉴定:为验证通过这种阴道菌群移植的方法是否可以恢复新生儿共生微生物,在新生儿出生的1、3、7、14、21及30天这六个时间点,分别在受试母婴的肛门、口腔及皮肤等部位共采集了样本,采用16S rRNA测序的方法鉴定其群落组成。
本发明的有益效果:本发明直接将待产孕妇的阴道分泌物取出,用于擦拭剖腹产新生儿嘴部、面部、肛门等身体的其他部位,恢复其共生微生物,该方法具有简单方便、安全有效、可操作等优点,有助于更好地增强和提高剖腹产新生儿的抗感染能力和免疫功能。
具体实施方式
本发明所述方法包括下列步骤:

1.一种用于恢复剖腹产新生儿共生微生物抹布的制备方法,其特征在于:所述方法包括下列步骤:
A、阴道分泌物的收集和保存:在进行剖腹产手术之前1小时,将一块大小适宜的无菌纱布放置于待产母亲的阴道当中,手术开始时取出并保存在无菌容器中;
B、阴道分泌物的擦拭:在新生儿产下的1—3分钟内,将浸有阴道分泌物的纱布依次擦拭新生儿的嘴部、面部、肛门身体的其他部位;
C、纱布微生物的鉴定:为验证纱布所浸的微生物与阴道分泌物的差异性,鉴定纱布以及母亲阴道分泌物样本中的菌群情况;
D、新生儿共生微生物的鉴定:为验证通过这种阴道菌群移植的方法是否可以恢复新生儿共生微生物,在新生儿出生的1、3、7、14、21及30天这六个时间点,分别在受试母婴的肛门、口腔及皮肤部位共采集了样本,采用16S rRNA测序的方法鉴定其群落组成。
