Abstract

发明领域
本发明关于复合纳米颗粒用于靶向癌症干细胞和治疗癌症的方法。
发明背景
人们提出不同的方法来治疗不同类型的癌症。有的建议是用标靶癌症细胞的方法来治疗癌症。然而,在表面标靶癌细胞一直是个挑战,因为实现高效特异性靶向在目前来说仍然是困难的。如果一种治疗方法不能有效地标靶问题细胞,那么治疗的效力会被削弱,更糟糕的是,会引起不良副作用。
本发明旨在解决上述问题,或至少向公众提供一种有用的替代方案。
发明内容
根据该本发明的第一方面,这里提供复合纳米颗粒包含核心部分,其含有作为热源的磁性纳米颗粒和用于治疗癌症组织的化疗剂;外壳部分,其包覆着所述核心部分;抗体,其连接在所述外壳表面并用于靶向癌症干细胞。在具体的实施方式中,该复合纳米颗粒可同时包含用于在活的有机体内定位的荧光染料。
所述外壳部分由二氧化硅或二氧化硅基材料制成。所述复合纳米颗粒的直径或宽度基本上为5至500纳米的范围。所述外壳部分厚度可为10至100纳米。所述磁性纳米颗粒的直径或宽度可为1至50纳米。
该磁性纳米颗粒是磁响应的,根据不同应用的需求,可以为铁磁性或超顺磁性纳米颗粒。该磁性纳米颗粒可对交变磁场产生响应。该磁性纳米颗粒的化学组成为Fe3O4。
化疗药物包含但不限于热休克蛋白抑制剂。在此实施方式中,该热休克蛋白抑制剂是临床批准的药物,尽管在其他的实施方式中,其他化疗剂也可被使用。该抗体可被连接在所述外壳的外表面上。该抗体可能够靶向到差异化集群分子或癌症干细胞的其他表面分子。
根据本发明的第二方面,这里提供利用靶向癌症干细胞来处理癌症的方法,包含施用如上述描述的复合纳米颗粒。
该方法可包括制备该纳米颗粒与靶向癌症干细胞的步骤。

根据该本发明的第四方面,这里提供在生物体中处理癌症的方法,包含对生物体至少每周一次施用热疗和化疗的组合处理的步骤。该方法可包含在接受热疗/化疗组合时利用荧光成像或磁性共振成像识别标靶组织的步骤。该方法可包含使用处理器以控制交替磁性场的功率和频率来调节标靶组织的温度上升的步骤。

该发明的具体实施方式
本发明关于通过同时使用化疗和热疗来协同地靶向癌症干细胞(CSCs)来处理癌症的手段和方法。
在特定的实施方式中,该手段包括使用平均颗粒尺寸为5至500纳米范围的二氧化硅基材料的纳米颗粒,包裹磁性核心和化疗药物,并在纳米粒子表面连接特定的抗体来识别出癌症细胞的表面标志物并对抗肿瘤组织尤其是肿瘤组织中的CSCs。使用CSCs作为靶向目标治疗的策略主要是干预CSCs的生长和繁殖。本发明使用纳米颗粒基材料的组合热疗和化疗是新颖的,如下所述,于癌症处理中展示了显著的效果。以CSCs作为靶向目标是特别理想的,因为它对肿瘤细胞的存活,增殖复发,和转移有着重要作用。通过利用抗体识别特定抗原将药物运送到肿瘤位置,结合加热,可以在不破坏该周围正常组织的情况下进一步增强治疗效果。

材料和方法
由Fe3O4@SiNPs释放的HSPI的体外分析
药物释放研究是在玻璃装置中,于37℃下,以AMF进行。该药物被称为如上所述的复合纳米颗粒。请参阅图1A所示该纳米颗粒组合物的结构。该组合物可以被认为是一个由热敏磁性颗粒和抗癌药物组成的核心,包覆抗体修饰的硅基外壳而成的复合纳米颗粒。
首先,加载HSPI的Fe3O4@SiNPs分散在1mL培养基并置于10kDa分子量的透析袋。然后将该透析袋浸于9ml的PBS,利用搅拌器保持恒定温度和搅拌,由AMF设备提供高频交变磁场。定期从透析袋外部溶液中收集300mL溶液作为测试样品,并补充与该体积相同的新鲜PBS。通过紫外-可见分光光度法(PerkinElmer,PE Lamda 750,USA),以及浓度-吸光度的标准方程分析HSPI的释放量。对每个该样品进行三次该药物释放研究。
LCSCs摄取的多功能的纳米颗粒
LCSCs(第3代)在24孔平板以1×104细胞/孔的密度接种于盖玻片上,在37℃培养24小时,然后与浓度为100μg/mL的PE-CD20标记的Fe3O4@SiNPs(CD20-Fe3O4@SiNPs)和Fe3O4@SiNPs在37℃下分别培养1小时和24小时。用DAPI(1mg/mL)核染色5分钟后,该细胞被洗涤,利用共聚焦显微镜(SPE,Leica,Germany)观察。
体外标靶的内化
LCSCs(第3代)以1×104细胞/孔的密度被接种在24孔板上。培养24小时后,分别用100mg/ml的CD20-Fe3O4@SiNPs和Fe3O4@SiNPs处理细胞1小时,并用PBS洗涤两次。接着,在冷的0.1M的二甲胂酸钠缓冲液中用2%戊二醛来收集并固定细胞至少2小时。该细胞随后在0.2M二甲胂酸钠缓冲液中用1%的四氧化锇固定1小时,然后在室温温度下用2%水铀盐染色30分钟,接着通过浓度逐渐升高的乙醇溶液分级脱水。该超薄切片的样本用乙酸铀酰和柠檬酸铅染色后在透射电子观察显微镜(TEM)(FEI/Philips Tecnai 12BioTWIN)下观察。
在交变磁场(AMF)下的体外热疗和化疗
该交变磁场(AMF)由3千瓦的交变磁场发生器供电给直径5厘米8匝感应线圈产生。LCSCs以5×104细胞/孔的密度被接种在6孔板上。孵育24小时后,细胞分别用100mg/ml的CD20-Fe3O4@SiNPs,CD20-HSPI&Fe3O4@SiNPs,Fe3O4@SiNPs,HSPI&Fe3O4@SiNPs,SiNPs,和HSPI处理1小时。未经处理的细胞作为对照。用PBS洗涤两次后,细胞被置于该线圈内,加热到一个限定温度(37和50℃之间)30分钟。保持恒定频率在350kHz,使用温度计浸在含有2mL溶液的试管中监测温度。使用传统的加热方法(水浴加热器)与AMF加热比较。细胞存活率利用MTT法评估。
流式细胞仪分析
为了检测在AMF热疗和水浴加热后的LCSCs的细胞凋亡情况,LCSCs用CD20-HSPI&Fe3O4@SiNPs进行处理后用PBS洗涤,根据细胞凋亡检测试剂盒说明书(YO-PRO-1/7-AAD,Invitrogen)对细胞凋亡情况进行测试。简单地说,将待处理的细胞在暗黑中利用YO-PRO-1和7-AAD溶液染色30分钟,和然后通过流式细胞仪分析(BD FACSCanto II system,BDBiosciences)。
建立人肺癌细胞的异种移植
BAL B/c裸鼠(5-6周龄和重15-20g)由伊利沙伯医院(中国香港)提供,所有的动物的培养和使用都遵从护理和使用实验动物规定。为建立该肿瘤模型,在裸鼠的背部皮下注射LCSCs(3×104细胞/200μL)。每4天密切观察每只小鼠的肿瘤生长情况。肿瘤体积可以利用本公式计算:长×宽×深×π/6。
溶血试验

为了进一步研究纳米颗粒对老鼠免疫系统的副作用,在后注射的第1、2、3、4、5、6、7、和40天从NPs处理的老鼠体内收集全血并加入抗凝剂。使用流式细胞仪的正向和侧向散射分析使白血细胞种群进入淋巴细胞,单核细胞和巨噬细胞,和嗜中性粒细胞。使用对抗典型B-细胞抗原(CD20)的抗体来分析淋巴细胞中的B-细胞数量。没有注射NPs的老鼠作为对照实验。
活体内骨髓衍生的间充质干细胞(MSCs)对NPs的摄取情况
在活体内骨髓衍生的间充质干细胞(MSCs)摄取NPs,根据前期工作,在注射纳米粒子后的第40天,从NPs处理的老鼠体内提取MSCs。使用FACSCalibur流式细胞仪系统分析该纯化MSCs。没有注射NPs的老鼠作为对照实验。
多功能的纳米颗粒在裸鼠体内的分布
长有肺癌肿瘤的老鼠经眼眶后静脉窦或尾静脉注射CD20-HSPI&Fe3O4@SiNPs或HSPI&Fe3O4@SiNPs。利用活体成像系统(XenogenSpectrum)分别在注射后的0.5,1,2,和24小时拍摄影像。该裸鼠在24小时被处死并解剖,拍摄包括心,肝,脾,肺,肾的活体外器官影像。
热疗和化疗组合在动物模型的功效
在大约第10天,当肿瘤体积达到约100mm3,该老鼠被随机分为五组(n=10):CD20-Fe3O4@SiNPs,CD20-HSPI&Fe3O4@SiNPs,HSPI&Fe3O4@SiNPs,CD20-HSPI@SiNPs,和PBS。每周一次通过眼眶后静脉窦或尾静脉注射样品(50mg/kg)至裸鼠。在注射后的一天,该老鼠暴露于AMF(3千瓦的交变磁场发生器供电予直径10cm、12匝感应线圈)30分钟(每星期3次)。每4天测定所有老鼠体重和瘤体积。
肿瘤异种移植和器官组织的染色
为了进一步研究通过由眼眶后静脉窦或尾静脉注射多功能的NPs对老鼠体内肿瘤的治疗效果,在注射后的第40天切除该肿瘤进行免疫组化分析。同时,收集器官利用免疫组化分析以研究多功能的NPs对老鼠的副作用。用10%中性缓冲的福尔马林固定组织,用石蜡包埋,以5μm厚切片,用苏木和曙红(H&E)染色。然后通过数码影像系统(Axioplan2,Zeiss)观察切片。
对肿瘤异种移植切片进行荧光染色以确认多功能的NPs对LCSCs的显著疗效。在血清被阻塞后,在37℃下对组织切片用PE-共轭的CD20抗体孵育1小时。利用共聚焦激光扫描显微镜观察该染色组织。
统计分析
所有数据均表示为平均值±标准偏差(SD)。使用t-检验,当P<0.05时,测定的差异被认为是显著的(P<0.05)。
结果
多功能的纳米颗粒特征
TEM影像显示Fe3O4@SiNPs和CD20-Fe3O4@SiNPs能够良好分散在PBS缓冲液中,保存几个星期后无明显聚集。颗粒尺寸大多在35nm到40nm之间,粒径分布较窄(图1C和1F)。与PE-CD20抗体共轭后颗粒尺寸稍微改变(图1D)。如在图1E中所示,该二氧化硅厚度可以在15nm至20nm的范围内进行调节。Fe3O4NPs的核心(深颜色)直径为大约30纳米。zeta电位结果(图1G)表明Fe3O4@SiNPs和CD20-Fe3O4@SiNPs的表面电荷分别为-42.86和-22.04mV。此外,PE-CD20抗体在Fe3O4@SiNP的表面上的共轭可以通过荧光光谱(FluoroMax-4)得到证实。如图1H所示,该PE-CD20标记的NPs的荧光信号峰在580nm,与PE-CD20抗体溶液相同,说明PE-CD20抗体被成功连接在HSPI&Fe3O4@SiNPs的表面上。
磁性超高温特性研究
从振动样品磁强计(VSM)获得的磁滞回线显示Fe3O4NPs和CD20-Fe3O4@SiNPs的该饱和磁化强度值(Ms)。该曲线通过原点表明被测Fe3O4NPs和Fe3O4@SiNPs是超顺磁性的。如图2A和2B所示,该Fe3O4NPs和CD20-Fe3O4@SiNPs的Ms分别为26emu/g和2.6emu/g。在相同强度的应用磁场下,Fe3O4@SiO2NPs比裸的Fe3O4有较弱的磁化值,因为相同质量的样品中,前者所含磁性材料量相对较少。
高Ms值对于提高NPs在AMF下的加热速度至关重要。图2C显示在该暴露时间下NPs混悬液的温度上升情况比较。Fe3O4@SiNPs悬浮液实现的最高温度为50.5℃,PBS溶液作为对照。因此,在中性培养基分散该NPs可实现有效加热,Fe3O4@SiNPs是一个用磁致热引发药物输送及其他生物医学应用的候选物。
该数据在图2A至2D表示为n=3的平均值±SD。
体外药物释放研究
可控和持续的药物释放在药物输送系统中非常重要。图2D示出了以1mg/mL的Fe3O4@SiNPs中HSPI的累计释放情况。该体外释放研究表明,Fe3O4@SiNPs在AMF下,HSPI的持续释放长达72小时(70%释放),从而可以在动物身上实现药物的可控释放。然而,无AMF的触发下,药物释放率在72小时的观察中只有21.5%。
体外细胞摄取和内化
LCSCs细胞(高表达CD20)对Fe3O4@SiNPs和CD20-Fe3O4@SiNPs的摄取通过激光共聚焦扫描显微镜进行研究。该LCSCs(第3代)利用浓度在100μg/mL的Fe3O4@SiNPs和CD20-Fe3O4@SiNPs分布在37℃下培养1小时和24小时。图3中,包括A至H,表明在1小时培养后LCSCs对CD20-Fe3O4@SiNPs的摄取高于Fe3O4@SiNPs。这结果还表明CD20标记的Fe3O4@SiNPs进入细胞的速度比自由的Fe3O4@SiNPs快得多,这可能是由于该受体介导的内噬途径。除此之外,细胞摄取纳米粒子量随培养时间从1小时至24小时而增加。图3显示细胞使用CD20-Fe3O4@SiNPs(图3-A和图3-B)和Fe3O4@SiNPs(图3C和图3-D)处理1小时和24小时的共聚焦细胞影像。图3还显示LCSCs内化CD20-Fe3O4@SiNPs(图3-E和图3-F)和Fe3O4@SiNPs(G和H)后的TEM影像。
基于LCSCs的细胞摄取的结果,NPs的内化通过TEM进行进一步的研究。正如图3E和3F所示,CD20-Fe3O4@SiNPs在与细胞共同培养1小时后被发现在该细胞膜附近聚集和内化,继而定位在溶酶体和在细胞质中,但是,即使在培养时间延长至24小时后,被定位在溶酶体和在细胞质中的Fe3O4@SiNPs(图3-G和3-H)仍然较少,这表明CD20能够促进该纳米材料靶向受体并被细胞摄取。
多功能的NPs在LCSCs上的体外热疗和化疗效果
为了评估CD20-Fe3O4@SiNPs的热治疗效果,试通过MTT方法检测LCSCs在AMF下处理30分钟或在特定的温度下水浴中的存活率。正如图4A所示,当它们与CD20-Fe3O4@SiNPs处理后可以得出细胞存活率为76%,68%,和63%和水浴加热到42℃,45℃,和50℃(该温度分别地是通过水浴或AMF在37℃,40℃,42℃,45℃和50℃来控制)。此结果表明由于该热休克蛋白(HSPs)家族成员的高表达,LCSC有热耐受性。相反地,只有约12%的LCSCs在AMF加热到42℃可存活。此外,当温度在AMF处理下保持50℃,只有约8%的LCSCs才能生存。这些结果说明LCSCs对AMF控制的CD20-Fe3O4@SiNPs介导的热疗敏感的。但是,高温度不仅可以杀死癌细胞,但它们也可以伤害或杀死正常细胞和组织。为了实现在较低的温度下选择性杀死LCSCs的目的,HSP90抑制剂17-(甲基氨基乙基氨基)-17-去甲氧基格尔德霉素(17-DMAG)被包裹在CD20-Fe3O4@SiNPs内以抑制HSP90的表达和克服LCSCs的热耐受性。
为了测试CD20-HSPI&Fe3O4@SiNPs的组合热疗和化疗效果,LCSCs与NPs孵育并在37℃的AMF下加热30分钟。结果显示,与该对照实验(只是培养基)相比,LCSCs在CD20-HSPI&Fe3O4@SiNPs的存活率有显着下降(约12%)。请参阅图4B(该温度是通过水浴或AMF在37℃处理30分钟。数据中显示为平均值±SD,*p<0.05和**p<0.01表明显着差,n=5)。在该其他方面,细胞存活率在AMF处理下的HSPI,SiNPs,Fe3O4@SiNPs,和HSPI&Fe3O4@SiNPs分别地降低到77%,88%,81%,和73%(图4B)。实验结果显示在AMF处理下CD20-HSPI&Fe3O4@SiNPs组合热疗和化疗具有选择性靶向治疗肿瘤干细胞的功效。
多功能的NP介导的热疗和化疗引起的坏疽
为了解多功能的NPs介导的热疗和化疗造成细胞死亡的该机制,LCSCs通过水浴或AMF 37℃下30分钟处理和使用YO-PRO1标签(细胞凋亡的标志物)和7-AAD渗透性(质膜完整性的指示物)量度。与上述发现一致的是,水浴超高温没有导致的细胞死亡。在水浴加热过后未观察到7-AAD和YO-PRO1阳性细胞(图4C)。与此相反,经过CD20-HSPI&Fe3O4@SiNPs处理LCSCs 7-AAD和YO-PRO1的阳性达到83.9%。然而,凋亡细胞(YO-PRO1阳性,7-AAD阴性)在AMF处理后未有观察到,表明坏疽是LCSCs观察到的细胞死亡的主要形式。纳米颗粒介导的组合热疗和化疗可引起细胞的膜破坏并导致坏死的细胞死亡方式。
靶向肿瘤的纳米颗粒在体内的积累及全身分布
在评估小鼠对纳米颗粒的肿瘤标靶和治疗功效前,以溶血试验和全血分析来评估CD20-HSPI&Fe3O4@SiNPs的血兼容性。对于溶血分析,如果红细胞裂解,血红蛋白将被释放,上清液会现红并可以在吸光度540nm被测定。如在图10A中所示,在1mg/mL的高浓度纳米颗粒处理后没有观察到可见的血红蛋白,这表明多功能的NPs具有良好的血液兼容性(<4%溶血)。未检测NPs对白血细胞的影响,注射NPs到老鼠并在AMF下处理30分钟。取小鼠眼缘静脉血并利用流式细胞仪正向和侧向散射分析白细胞中淋巴细胞,单核细胞,和嗜中性粒细胞。结果在图10B表明,在对照实验和NPs处理之间的免疫细胞数目没有显着差异。这些结果表明多功能的NPs具有良好的血兼容性且可用于小鼠活体实验。
然后通过活体成像系统,研究CD20-HSPI&Fe3O4@SiNPs的肿瘤标靶功效及其在异体种植瘤小鼠体内的分布情况。
图5示出了CD20-Fe3O4@SiNPs和Fe3O4@SiNPs的该荧光信号,这两个囊化荧光染料Ru(bppy)3,均在注射后30分钟位于该肝。该结果示出了在0.5,1,2,和24小时的CD20-Fe3O4@SiNPs和Fe3O4@SiNPs(眼眶后静脉窦或尾静脉注射)处理。大部分Fe3O4@SiNP聚集在肝,而CD20-Fe3O4@SiNP主要集中在肿瘤区。(C:对照实验;1:注射Fe3O4@SiNPs;2:注射CD20-Fe3O4@SiNPs)
随着时间的过去,使用CD20-Fe3O4@SiNPs处理的小鼠,在肿瘤区域检测到明显的荧光信号,在注射后24小时的时间点,CD20-Fe3O4@SiNPs的荧光信号差不多在瘤的位置附近被找出且有少量的积累在肝内。然而,没有记录到Fe3O4@SiNPs在肿瘤区域聚集的荧光信号。CD20-Fe3O4@SiNPs能够特异的靶向肿瘤,其比Fe3O4@SiNPs有更高的效率。相比于Fe3O4@SiNPs,CD20-Fe3O4@SiNPs的特异靶向效率和肿瘤积累进一步以活体外(ex vivo)成像(图5)证实。在脾,肺,心,肾没有观察到明显的荧光信号,仅在肝检测到微弱信号,结果显示在24小时后,存在于其他器官纳米颗粒被排出体内。
多功能的NPs介导的热化疗法在体内抑制肿瘤生长
为了确定CD20-HSPI&Fe3O4@SiNPs结合热疗和化疗的抗肿瘤功效,LCSCs在几个实验组中(n=10)异种移植在裸鼠背部。本模型是一种高度恶性瘤模型,肿瘤体积可在14天之内增加至约1500mm3。根据兽医的咨询,肿瘤的总体积超过2000mm3视为垂死或死亡。CD20-HSPI&Fe3O4@SiNPs分散在正常的PBS由该眼眶后静脉窦尾静脉注入肿瘤老鼠。该鼠被置于直径为10cm的水冷磁性感应线圈(AMF)。之后对瘤体积达进行36天监控。如在图6A和6B所示,得到对照实验组的一个快速瘤生长曲线(在图6A中,示出了裸老鼠在AMF处理前异种移植LCSCs和AMF处理后的36天;图6B是纳米颗粒处理后的瘤体积(V/V开始)相对日数的曲线图;图6C是裸老鼠注入NPs的累计存活率;图6D是该老鼠处理各种纳米颗粒后显示相对体重的图与;图6E显示皮下瘤注射NPs(2:CD20-HSPI&Fe3O4@SiNPs;3:HSPI&Fe3O4@SiNPs;4:CD20-Fe3O4@SiNPs;5:CD20-HSPI@SiNPs)后的照片影像。数据表示为平均值±SD,(n=10)。)
结果显示,接收CD20-HSPI&Fe3O4@SiNPs的组合热疗和化疗处理的肿瘤生长明显被抑制且几乎没有明显的生长。相反地,未修饰的HSPI&Fe3O4@SiNPs,CD20-Fe3O4@SiNPs或CD20-HSPI@SiNPs没有明显影响瘤的生长。相比对照实验组中,CD20-HSPI&Fe3O4@SiNPs处理的老鼠平均存活期由12天延长到36天(图6C)。在观察期中每组老鼠的体重按比例增加(图6D)。与其他群体的老鼠比较,用PBS处理的老鼠体重最低(图6D)。
为了进一步评估多功能的NPs的抗肿瘤效果,对肿瘤组织进行体外切片病理分析。对照实验组的肿瘤组织的细胞形态维持正常。然而,在NPs处理组的肿瘤切片染色中发现有明显的坏疽发生。坏疽细胞表现为深色胞浆嗜酸性和密集的紫色核的圆形(图7A)。为了更好地确定CD20-HSPI&Fe3O4@SiNPs的治疗功效,使用PE连接CD20抗体通过免疫组织化学染色来检查肿瘤标本中CD20的表达(AMF处理36天后)。相比于未处理瘤(图7B),CD20-HSPI&Fe3O4@SiNPs可杀死大部分LCSCs,如所示的CD20的荧光信号显着减弱。另外,利用透射电子显微镜成像显示在CD20-HSPI&Fe3O4@SiNPs处理的肿瘤组织内观察到纳米颗粒的堆积,表明该多功能的纳米颗粒靶向肿瘤能力(图7C-F)。图7C-7E显示纳米颗粒积聚在肿瘤组织,在AMF处理36天后肿瘤组织发生坏死。
多功能的NPs未有在活的有机体内引起任何毒性的迹象
该多功能的NPs在活的有机体内的毒性在AMF处理36天后,通过组织病理学切片观察纳米颗粒对各器官如心,肺,肝和肾的毒性效果。如在图8所示,与正常组作为对照实验进行比较,处理组未观察到组织病理学的变化。此外,没有发现NPs在这些组织内的积累。从病理组织学分析,可以确认该CD20-HSPI&Fe3O4@SiNPs没有严重损害那些裸鼠的器官。图8显示了多功能的NPs在36天后无诱导的毒性迹象。在那些器官中没有观察到异常病理变比。该影像在20×放大倍数下拍摄。
根据白血细胞(WBC)计数,包括淋巴细胞,单核细胞,和嗜中性粒细胞(图9A-D),对由CD20-HSPI&Fe3O4@SiNPs处理诱导的免疫细胞损伤和恢复进行了评估。AMF处理后3天的淋巴细胞在WBC内明显减少和第6天数目回复到该正常的水平(图9A)。此外,通过使用CD20抗体对B-细胞进行详细分析。虽然以CD20-HSPI&Fe3O4@SiNPs处理的B-细胞于第3天降到最低点,但是实验观察到B-细胞于第4天快速恢复,并最早于第6天回复到基础水平(图9B)。值得注意的是,B细胞数目同时在约第4天开始增加和该WBCs于第6天恢复。该结果展示出通过造血功能的激活,损坏的B细胞在AMF处理后的约第4天开始恢复。除此以外,从CD20-HSPI@Fe3O4@SiNPs处理的老鼠骨髓中提取的骨髓间充质干细胞(MSCs)没有观察摄取CD20-HSPI@Fe3O4@SiNPs的证据(图9E)。图9A显示WBC计数和图9B显示老鼠以CD20-HSPI&Fe3O4@SiNP介导的AMF处理的B-细胞的变化。图9C和图9D示出老鼠7天痊愈后有或没有CD20-HSPI&Fe3O4@SiNPs的白细胞和B-细胞的百分比;图9E显示通过流式细胞仪监测该骨髓内老鼠MSCs的CD20-HSPI&Fe3O4@SiNPs摄取。
肿瘤内异质性是癌症治疗过程中的一个重大障碍。越来越多的证据体表明,肿瘤可由一小群被称为癌症干细胞来驱动,它们具有自我更新,对抗传统治疗的能力,且具有分化成形成不同的癌症细胞种群的能力。最新研究结果显示,靶向癌症干细胞可有效提高癌症治疗的效率。然而,目前并没有关于CSCs的最佳标志物的共识,大量研究使用表面抗原作为CSCs的标志物。在这个发明中,肺癌干细胞(LCSCs)是从人肺肿瘤细胞种群中分离并利用细胞表面标志物和干细胞标志物来表征,例如,CD20,CD15,ABCG2,和Oct4。请参阅图11。研究结果显示,相比CD20阴性细胞,CD20阳性肿瘤干细胞有更强的肿瘤形成,迁移和侵袭的能力。请参阅图12和图13。利用裸鼠体内研究结果表明在注射相同的细胞数量后,LCSCs比分化的LCSCs能够更快的形成肿瘤,表明LCSC的肿瘤起始能力。请参阅图14。由于这些LCSCs细胞是高度成瘤的,在常规治疗中有效的消灭LCSC为成功治疗肺癌的关键。因此,CSCs标靶疗法的发展提供了一个治疗方法来彻底消除癌症细胞以达到治愈肺癌患者的目标。
临床结果表明基于纳米颗粒的药物输送系统可以在癌症治疗过程中增强功效,同时降低了副作用。目前基于纳米颗粒的同时热疗和化疗(热化疗)的CSCs的癌症治疗则不幸甚少研究。在本专利中,我们合成和表征该生物兼容性多功能的二氧化硅为基础的纳米颗粒;该纳米颗粒包裹了磁性核心(Fe3O4NPs)和化疗药物(包括热休克蛋白抑制剂)并在表面连有特定的抗体(CD20);在交变磁场(AMF)下以肺癌症干细胞的表面标志物为标靶对其进行组合的热疗和化疗。为了确定CD20-Fe3O4@SiNPs的磁性和发热性,测试了磁化饱和值并绘制了滞回曲线。该曲线通过该原点显示这两个Fe3O4NPs和CD20-Fe3O4@SiNPs是超顺磁性。同时评估了CD20-Fe3O4@SiNPs在AMF下的发热性。正如图2C中所示,该NPs因磁性滞回具有AMF引起的加热能力和在AMF下发热。接下来,响应AMF而释放相应的药物。相比于HSPI,该NPs复合物可以延长了HSPI在体外的释放。
虽然一直有以热疗治疗癌症的建议,但热疗只能杀死大部分癌细胞而不是癌症干细胞(CSCs),结果癌症干细胞形成对热疗的耐受性,从而引起复发。此外,癌细胞热休克蛋白的高表达可以触发防御机制从而保护癌细胞免受高温的破坏。在这方面研究中使用热休克蛋白抑制剂(使用17-甲基氨基乙基氨基-17-去甲氧基格尔德霉素(17-DMAG)的为例,其靶向HSP90,FDA已批准用于临床试验)来结合磁性纳米颗粒同时同时进行热疗和化疗。此外,该多功能的纳米颗粒可以通过CD20抗体的修饰靶向LCSC。使用该CD20-HSPI&Fe3O4@SiNPs体内应用于异种移植小鼠瘤模型来靶向LCSCs的能力被进一步证实。经过1小时孵育,与非修饰NPs比较,体外细胞实验表明CD20抗体修饰的NPs促进纳米颗粒靶向到LCSCs。然而,LCSCs对该Fe3O4@SiNPs的摄取率相对于该孵育时间的增加而略有增加,这表明长孵育时间可以提高非特定的摄取和降低标靶和非标靶纳米颗粒的该差异,这与其他研究一致。CD20-Fe3O4@SiNPs在体外可选择性靶向到LCSCs这表明CD20抗体的修饰可以促进LCSCs对纳米颗粒的内吞过程,促进纳米颗粒能够更迅速分布于细胞质中。有研究指出受体泛素化可能会引发膜网格蛋白涂覆坑断裂和完成程序内吞。预泛素化表皮生长因子受体(EGFR)和ErbB2的可组成型内吞进入细胞。因此,靶向份子(CD20抗体)和受体(CD20)间的相互作用诱导受体的该泛素化,从而导致抗体修饰的纳米颗粒迅速内吞。同时,体内分布数据有力地证明了CD20抗体修饰的纳米颗粒可于很短的时间内聚集在肿瘤组织内,这与许多早先的报告一致。为了确认体内成像结果,解剖各器官用于生物体外(ex vivo)成像。结果显示,注入CD20-Fe3O4@SiNPs的裸鼠的异植瘤内纳米颗粒的荧光信号清晰可见,而其他器官无该信号,仅有肝脏观察到弱信号。该肾显示清晰影像,可解释为在注射该NPs的24小时内纳米颗粒被肾脏迅速消除。为了进一步评价这些多功能的纳米颗粒的血兼容性,我们进行了溶血和全血分析。对于溶血分析,以1mg/mL高浓度纳米颗粒处理红细胞,结果显示没有观察到可见的血红蛋白,这表明该多功能的纳米颗粒具有良好的血液兼容性。经过静脉CD20-HSPI&Fe3O4@SiNPs处理的裸鼠,血液中淋巴细胞,单核细胞和巨噬细胞和嗜中性粒细胞与正常的不经过处理对照裸鼠相比似乎没有任何改变。此外,获得的数据也显示CD20修饰的纳米颗粒是针对CSCs而不和一般的体内“干细胞”结合。来自骨髓和血的MSCs未被检测到与CD20-HSPI&Fe3O4@SiNPs的结合。因此,该纳米颗粒上的CD20抗体部分有利于CD20-HSPI&Fe3O4@SiNPs全身用药时排除正常的干细胞非特异性的毒性,防止有害的和有潜在危险的副作用。本专利发明的纳米颗粒潜在的临床应用的纳米输送系统有显着的优点。多功能的NP的其他优点是CD20-HSPI&Fe3O4@SiNP介导的LCSC标靶的组合热疗和化疗。
研究显示本发明表明热疗或超高温治疗在组合疗法起着重要的作用,在AMF中的四氧化三铁纳米颗粒产生的40℃-50℃被认为是最佳超高温。在本发明过程中,CD20-Fe3O4@SiNPs的热疗效果在体外被评估。除了预期的LCSCs细胞死亡,AMF控制的CD20-Fe3O4@SiNPs介导的热疗也诱导意想不到的生物反应,例如因热休克蛋白的表达引起的肿瘤特定免疫反应。这些结果表明超高温不仅能杀死暴露在热处理中的LCSCs,也能在40℃-50℃的温度杀死正常的细胞。为实现选择性在较低的温度(37℃)消除LCSCs的目标,HSP90抑制剂17-DMAG被包裹在CD20-Fe3O4@SiNPs中以抑制HSP90的表达从而克服了LCSCs的热耐受性。CD20-HSPI&Fe3O4@SiNPs的组合热疗和化疗在AMF下30分钟对LCSCs的存活影响进行了研究。与其他组别比较,CD20-HSPI&Fe3O4@SiNPs可特异性靶向到LCSCs并降低其存活率。此外,通过流式细胞仪的对细胞凋亡和坏死进行分析,实验证实多功能的NPs通过引起临界膜损伤和随后的坏死而杀死LCSCs。LCSCs内的温度增加到42℃以上,造成细胞膜损坏细胞和随后的坏死的细胞死亡,这表明经过NPs介导的AMF处理后,观察到坏疽是LCSCs细胞死亡的主要形式。

Claims (19)
1.复合纳米颗粒包括:中心部分,其含有作为热源的磁性纳米颗粒和用于治疗癌症的化疗剂;外壳部分,其包覆着中心部分且表面连接功能性基团,用于改善生物相容性及连接;抗体,其连接在所述外壳表面用于靶向癌症干细胞。
2.如权利要求1所述的复合纳米颗粒,进一步包含用于体内定位的荧光染料。
3.如权利要求1所述的复合纳米颗粒,其中所述外壳部分是由二氧化硅或二氧化硅基材料制成。
4.如权利要求1所述的复合纳米颗粒,其中所述纳米颗粒的直径或宽度大约为5~500纳米。
5.如权利要求1所述的复合纳米颗粒,其中所述外壳部分厚度为10至100纳米。
6.如权利要求1所述的复合纳米颗粒,其中所述磁性纳米颗粒的直径或宽度为1至50纳米。
7.如权利要求1所述的复合纳米颗粒,其中所述磁性纳米颗粒是磁响应的,并且是超顺磁性纳米颗粒。
8.如权利要求1所述的复合纳米颗粒,其中所述磁性纳米颗粒被应用于交变磁场并产生响应。
9.如权利要求1所述的复合纳米颗粒,其中所述磁性纳米颗粒的化学成分为Fe3O4。
10.如权利要求1所述的复合纳米颗粒,其中所述化疗剂为热休克蛋白抑制剂。
11.如权利要求1所述的复合纳米颗粒,其中所述抗体是连接在所述外壳部分的表面。
12.如权利要求1所述的复合纳米颗粒,其中所述抗体是针对癌症干细胞的表面分子。
13.通过靶向癌症干细胞来治疗癌症的方法,包括施用如权利要求1所述的复合纳米颗粒。
14.如权利要求13所述的方法,包括制备该复合纳米颗粒与靶向癌症干细胞的步骤。
15.如权利要求13所述的方法,包括利用表面的抗体使纳米粒子靶向癌细胞,通过施加交变磁场,中心部分在磁热效应作用下释放热量,进而抑制或杀死癌细胞;同时,中心部分温度上升促使化疗药物从纳米粒子内部释放出来,增强癌症治疗效果。此外,通过控制交变磁场的物理参数,可以控制纳米颗粒温度上升的程度和化疗药物的释放。
16.如权利要求15所述的方法,该标靶位置的温度可以提升至至40℃-52℃。
17.如权利要求13所述的方法,,以每公斤体重静脉注射10微克至500毫克的所述复合纳米颗粒的剂量。
18.如权利要求13所述的方法,每周施用一次所述复合纳米颗粒。
19.使用如权利要求1所述的复合纳米颗粒用于治疗癌症。
