组成的,试剂是由淋病免疫血清,稳定菌液,防腐剂组
成的;本试剂盒能简单快速的对淋病进行诊断,3分
钟可出结果,诊断急性淋病阳性率可达100%,慢性
淋病阳性率可达98%,非常适于基层医疗单位的使
用。
Description
本发明属于医疗诊断新技术。
本发明的技术背景:性病在我国六十年代初期基本消灭,但是,八十年代中期又开始由国外传入,并且正在蔓延。性病中发病率最高的是淋病。国内现有的实验室诊断主要是用直接涂片,革兰氏染色,镜检病原菌,细菌培养,生化鉴定等方法。以上各种方法对急性淋病、症状明显,分泌物多的患者阳性率高,容易诊断。而对慢性淋病,症状不明显,迁延时间较久的,分泌物不多的病例,检测的阳性率不高,诊断比较困难。国外采用淋酶法诊断淋病,是一种可靠而且准确的方法,但是需要较高的实验室条件,操作也比较复杂,在我国的具体条件下,不适于广泛使用。
葡萄球菌A蛋白协同凝集试验,国外用于细菌及病毒性疾病的诊断,国内有用于鼠疫病原菌抗原检测的。但是用于检测淋球菌抗原快速诊断淋病方面没有报导。
本发明的目的在于寻找建立性能可靠、简便、敏感度高的淋病诊断试剂盒,提高对淋病的诊断率。
本发明的技术构思是:利用特异性抗体致敏A蛋白葡萄球菌进行协同凝集试验。检测淋球菌抗原。本试剂盒的试剂是由淋球菌免疫血清,稳定菌液和防腐剂(硫柳汞磷酸盐缓冲液)组成。
1.制备淋球菌免疫血清:
首先取患者分泌物为标本,经分离培养鉴定为淋球菌,纯培养于兔血琼脂斜面30~40℃,在5%~10%的CO2环境,用生理盐水洗下菌落,每斜面10~20ml,离心、沉淀加0。5福尔马淋生理盐水 ~8ml,加入等量佐剂,混均为免疫源,取数只家兔分别注射免疫原2次。一周后,再注射活菌2次,一周后采血,进行玻片凝集试验阳性者,取血分离血清。即制得淋球菌免疫血清备用。
2.制备稳定菌液:
用金黄色葡萄球菌孔埃I株(COwanI),在30~40℃条件下,培养基内悬浮菌液,经20~40小时后,离心得菌膏,配成10%的磷酸盐菌悬液,制得稳定菌液。
3.试剂盒制作:
以淋球菌免疫血清与稳定菌之溶积比为1∶8~15,用0。1%的硫柳汞做防腐剂,配成淋球菌诊断试剂。每盒以5ml眼药水瓶(或其他容器)分装即为本试剂盒。
本发明的积极效果:
本发明是用特异性抗体检测淋球菌抗原,并且是凝集法,因此不受实验条件的限制,不需要任何设备,可以避免形态学的误差和假阳性结果出现。诊断快速,3分钟可出结果,诊断急性淋病阳性率100%,慢性淋病阳性率98%,优于革兰氏染色和细菌培养诊断方法,有利于基层医疗单位使用。

1、一种由盒底,盒盖及试剂组成的淋病诊断试剂盒,其特征在于:试剂是由淋病免疫血清、稳定菌液和防腐剂组成;
2、如权利要求1所述的试剂盒,其特征在于:淋病免疫血清与稳定菌液之容积比为1∶8~15。
