Abstract

发明内容
前述需求通过本发明得以满足。
本发明提供了纳米颗粒,其包含具有八个三角双锥臂(trigonal bipyramidalarm)的八角(octapod)铁氧化物。
本发明还提供了包封纳米颗粒,其包含纳米颗粒和包封剂 (encapsulatingagent),所述纳米颗粒包含具有八个三角双锥臂的八角铁氧化物。
本发明还提供制备包含具有八个三角双锥臂的八角铁氧化物的纳米颗粒的方法,其所述方法包括加热羧酸铁、羧酸、氯盐和溶剂的混合物。

具体实施方式
本发明提供了纳米颗粒,其包含具有八个三角双锥臂的八角铁氧化物。
在某些实施方案中,八角铁氧化物包含Fe3O4单元。在某些优选实施方案中,八角铁氧化物包含磁铁矿。在某些优选实施方案中,八角铁氧化物具有反尖晶石晶体结构。
如本领域中已知的,尖晶石是式A2+B2 3+O4 2-的矿石。磁铁矿是式 Fe2+Fe2 3+O4 2-的尖晶石。
在任意上述实施方案中,纳米颗粒包含八角铁氧化物和氯离子。在某些实施方案中,氯离子与八角铁氧化物表面上的Fe(III)离子螯合。在某些实施方案中,氯离子与暴露在八角铁氧化物表面的[311]面上的Fe(III) 离子螯合。
在任意上述实施方案中,八角铁氧化物包含由[311]高指数面界定并且具有14个面和24条棱的凹多面体几何结构。在上述某些实施方案中,八角铁氧化物的平均棱长为约15nm至约40nm,例如约15nm、约20 nm、约25nm、30nm、35nm或40nm。
本发明还提供了制备包含具有八个三角双锥臂的八角铁氧化物的纳米颗粒的方法,所述方法包括加热羧酸铁、羧酸、氯盐和溶剂的混合物。在某些实施方案中,所述方法还包括从混合物中分离纳米颗粒的步骤。可以使用任何合适的技术分离纳米颗粒。例如,可以将混合物冷却到室温并且通过向反应混合物中添加合适的非溶剂如异丙醇来从反应混合物中沉淀出纳米颗粒。
羧酸铁是羧酸的铁盐,优选长链脂肪酸,例如C8-C20羧酸。在某些优选实施方案中,羧酸铁是油酸铁。在某些优选实施方案中,羧酸是油酸。
在某些优选实施方案中,氯盐是有机或无机氯盐,例如金属氯盐如氯化钠或氯化钾,或者有机氯盐如四烷基氯化铵。当氯盐是四烷基氯化铵时,所述四烷基氯化铵可以是任意合适的四烷基氯化铵。四烷基基团可以是相同的或不同的,例如,烷基可以独立地为C1-C20烷基。在一个优选实施方案中,四烷基氯化铵是十六烷基三甲基氯化铵。
用于进行反应的溶剂可以是任何合适的溶剂。在某些实施方案中,非极性溶剂可以选自烯烃、烷基醚、芳基醚、烷基芳基醚、胺和卤代芳烃。在某些优选实施方案中,溶剂是非极性溶剂,所述非极性溶剂选自:1- 十八烯、1-十六烯、1-二十烯、苯基醚、苄基醚、三辛胺、辛基醚和邻二氯苯。在一个优选实施方案中,溶剂是1-十八烯。
通常,相对于1当量的羧酸铁,反应混合物包含约0.1当量或更多、约0.2当量或更多、约0.3当量或更多、约0.4当量或更多、或者约0.5 当量或更多的氯盐。或者或另外,反应混合物包含约1当量或更少、约 0.9当量或更少、约0.8当量或更少、约0.7当量或更少、或者约0.6当量或更少(的氯盐)。因此,反应混合物可以包含由上述端点中的任意两个界定的量的氯盐。例如,反应混合物可以包含约0.1至约1当量、约0.1 至约0.9当量、约0.1至约0.8当量、约0.1至约0.7当量、或者约0.2至约0.7当量的氯盐。本文提到的当量是指摩尔当量。
可以将反应混合物加热任意合适量的时间。例如,可以将反应混合物加热约0.5小时至约3小时或更久。通常将混合物加热到约300℃至约 350℃,例如约310℃至约340℃或者约320℃至约330℃的温度。在一些实施方案中,所形成的八角铁氧化物的平均棱长可至少部分地取决于加热反应混合物的时间长度,棱长度通常随着反应混合物加热时间的延长而增加。
本文使用的术语“约”在结合温度数值使用时是指值±5%、±4%、± 3%、±2%、或±1%。例如,“约300℃”意指300℃±15℃、300℃±12℃、 300℃±9℃、300℃±6℃、或300℃±3℃。
在某些实施方案中,本发明提供了包封纳米颗粒,其包含具有八个三角双锥臂的八角铁氧化物和包封剂。包封剂可以是任意合适的包封剂。优选地,包封剂有助于纳米颗粒分散在合适的溶剂(例如,水)中。在一些优选实施方案中,包封剂是HDA-G2。HDA-G2是通过1-十六烷基胺、丙烯酸甲酯和乙二胺缩合获得的树枝状聚合物,其制备描述在Zhou,Z.J.等,Adv.Mater.24:6223-6228(2012)中。
在某些实施方案中,包封剂包含游离胺基。在这些实施方案中的某些中,包封剂与靶向配体缀合。合适的靶向配体的非限制性实例包括肽、抗体和叶酸。理想地,靶向配体通过表面受体特异性识别病理性细胞。
本文使用的术语“靶向配体”是指在向对象施用后相对特异性地与存在于特定器官或组织中的分子结合的特定分子。通常,选择性靶向至少部分地通过检测与对照器官或组织相比对器官或组织以至少2倍高的选择性结合的分子来表征。在某些实施方案中,选择性结合为与对照器官或组织相比为至少三倍或至少四倍高。
在肿瘤靶向分子的情况下,这样的分子与在特定癌组织中选择性过表达的受体结合。过表达意指与正常组织相比在肿瘤组织中的表达为至少 1.5倍高。在某些实施方案中,与非肿瘤相比,肿瘤中的表达为至少5倍高。

本发明还提供了用于磁共振成像的造影剂,其包含前述纳米颗粒或者前述包封纳米颗粒。在一些优选实施方案中,造影剂是T2造影剂。
本发明的造影剂可用作用于MRI(磁共振成像)、X射线CT(计算机断层扫描)、超声成像和闪烁照相的造影剂。特别适合用作用于MRI 的造影剂。造影剂可以胃肠外施用或经口施用。当造影剂胃肠外施用时,造影剂还可以包含用于生产注射产品的已知添加剂,例如溶剂、助悬剂等。添加剂的实例包括水、丙二醇、聚乙二醇、苯甲醇、油酸乙酯、卵磷脂等。这些添加剂可以单独使用,或者两种或更多种组合使用。另外,当造影剂经口施用时,造影剂单独施用或者与可药用载体一起施用。特别地,造影剂经口以例如颗粒剂、细颗粒剂、粉剂、片剂、硬糖浆剂(hard syrup)、软胶囊剂、糖浆剂、乳剂、混悬剂、脂质体、溶液等的形式施用。当形成颗粒剂、细颗粒剂、粉剂和片剂时,可以使用赋形剂。赋形剂的实例包括乳糖、蔗糖、淀粉、滑石、纤维素、糊精、高岭土、碳酸钙等。这些赋形剂可以单独使用,或者两种或更多种组合使用。当形成乳剂、糖浆剂、混悬剂和溶液时,可以使用常用非活性稀释剂。稀释剂的实例包括植物油等。造影剂还可以包含已知添加剂。添加剂的实例包括润湿剂、助悬剂、甜味剂、芳香剂、着色剂、防腐剂等。这些添加剂可以单独使用或者两种或更多种组合使用。此外,乳剂等中形成的造影剂可以放置在由可吸收物质如明胶制成的胶囊中。本发明造影剂的施用剂量没有特别限制,在一次诊断中每个成人为0.1mg至10g,优选1mg至5g。
化学
在氯化钠(NaCl)的存在下通过油酸铁的分解制备八角铁氧化物纳米颗粒。在一个实施方案中,在含有油酸作为表面活性剂以及NaCl作为封端剂的1-十八烯中,使油酸铁在320℃分解2小时。透射电子显微镜 (TEM)图像(图1A和1B)表明以高产率(>95%)所获得的产物由均一的四臂星形铁氧化物颗粒组成。两个临近的臂端点之间的平均棱长为约30nm。在高分辨率TEM(HRTEM)图像(图1C)中观察到了横越整个纳米颗粒的均匀晶格条纹,其间隔对应于Fe3O4(220)。在仔细考察这些独特纳米颗粒后,发现了一些可能属于星形颗粒的四条臂的阴影,表明颗粒中可能存在凹陷特征。为了更好的显现纳米颗粒的三维结构,将样品从垂直于电子束的方向倾斜。随着倾斜,纳米颗粒从四臂星形变成了细长的六臂星(图1D),其适合于具有八条三角锥臂的八角纳米颗粒的特征。在这些观察的基础上,对这种独特铁氧化物纳米结构提出了凹多面体模型 (图1A,插图)。为了确认这种提出的结构,通过HRTEM和相关选定区电子衍射(SEAD)测量对单个八角纳米颗粒进行了表征。单个纳米颗粒的轮廓及棱之间的角度二者均与通过[311]高指数面界定的凹面几何模型一致(图2)。八角纳米颗粒的X射线粉末衍射(XRD)图与不具有任何铁相的磁铁矿Fe3O4参考值(JCPDS No.82-1533)良好匹配,表明八角铁氧化物纳米颗粒是具有反尖晶石晶体结构(图3)的纯磁铁矿。XRD 图在约31、36、43、57和63度的2θ值处示出有峰。
根据本发明的一个实施方案,氯盐(例如,NaCl)在八角铁氧化物纳米颗粒的合成中可能是必不可少的。通过向10mL反应溶液中提供一定量的NaCl(0.17毫摩尔比0.86毫摩尔油酸铁),获得了均一的八角铁氧化物纳米颗粒。当NaCl的量减少到0.085毫摩尔时,八角铁氧化物纳米颗粒的产率显著降低。当NaCl的量进一步降低到0.034毫摩尔时,只有很少的八角铁氧化物纳米颗粒(图4)。这些结果表明NaCl在八角铁氧化物纳米颗粒的形成中具有重要作用。为了理解氯阴离子和钠阳离子在八角铁氧化物纳米颗粒的形成中的作用,进行了许多对照试验。当使用 NaOH和油酸钠代替NaCl时,产物主要是球形和立方体铁氧化物纳米颗粒的混合物。另外,当使用NaF或KBr代替NaCl时未获得八角结构(图 5),表明在八角铁氧化物纳米颗粒的形成中氯离子比钠离子更关键。此外,使用十六烷基三甲基氯化铵(CTAC)、KCl、十六烷基三甲基溴化铵 (CTAB)和KBr代替NaCl以研究最终产物的结构(图6)。通过添加 CTAC或KCl获得了类似的八角产物,而在CTAB或KBr的存在下没有获得期望的八角结构。这些结果进一步证实氯离子是形成八角铁氧化物纳米颗粒的关键诱导剂。
还可以通过在NaCl的存在下改变反应时间来控制八角纳米颗粒的尺寸。分别通过反应0.5、1、2和2.5小时,形成了平均棱长为14、20、30 和36nm的八角纳米颗粒(图7),表明氯离子可能在整个颗粒生长过程影响八角纳米颗粒的形成。形成八角铁氧化物纳米颗粒的一种可能的机制是在颗粒生长过程中氯离子选择性地与铁氧化物的暴露在高指数面(可能[311])的铁离子结合。这种氯离子辅助形成机制得到了使用能量色散X 射线光谱(EDS)和X射线光电子光谱(XPS,图8中所示)观察到的八角铁氧化物纳米颗粒上痕量氯的存在的支持。图9描绘了一种模型,其在纳米颗粒的表面上示出了氯离子的存在。
八角铁氧化物纳米颗粒的结构和性质。不希望受到任何理论或机制的约束,认为本发明实施方案的纳米结构的复杂形貌(sophisticated morphology)可以改变颗粒核心的有效半径。根据量子力学外球理论 (quantum mechanical outer sphere theory),模拟出覆盖整个八角铁氧化物纳米颗粒的球体作为模型,以代表在外磁场B0下八角纳米颗粒的客观存在(图10)。因此,模型的直径表示八角铁氧化物纳米颗粒的有效直径。发现八角铁氧化物纳米颗粒的有效半径为具有相同几何核心体积的球形纳米颗粒的有效半径的约2.4倍,证明八角形貌可以显著提高纳米颗粒的有效半径,并且指示八角铁氧化物纳米颗粒可比具有类似几何体积的球形纳米颗粒具有更高的T2弛豫率。为了研究具有相同几何体积的八角铁氧化物纳米颗粒和球形铁氧化物纳米颗粒的MRI对比能力,选择平均棱长为30nm的八角铁氧化物纳米颗粒(表示为八角-30)和平均棱长为20nm 的八角铁氧化物纳米颗粒(表示为八角-20)作为两个代表性实例。因此,将平均直径为16nm的球形纳米颗粒(表示为球形-16)和平均直径为10 nm的球形纳米颗粒(表示为球形-10)用于比较,这是因为体积类似(即,八角-30与球形-16以及八角-20与球形-10)。然后通过超导量子干涉器件 (SQUID)测试八角铁氧化物纳米颗粒和球形铁氧化物纳米颗粒的磁性质。在环境温度下八角-30、八角-20、球形-16和球形-10均表现出光滑 M-H曲线而没有滞后(图11)。八角-30和八角-20的阻隔温度(blocking temperature)分别为290K和240K,这进一步证实八角-30和八角-20在室温下表现出超顺磁行为,使这些纳米颗粒能够用于许多生物医学应用 (例如,生物学分离和MRI对比增强)。八角-30、八角-20、球形-16和球形-10的Ms值分别为约71emu/g、51emu/g、67emu/g和55emu/g。八角-30比球形-16稍高的Ms值可能是由于与球形颗粒相比八角形貌中降低的自旋倾斜效应引起。尽管具有类似的Ms值,这些尖的凹面磁铁矿纳米结构(spiked concave magnetite nanostructure)中的形状各向异性和磁核心显著增加的有效半径可能是明显高的T2弛豫率的原因。

由于所制备的纳米颗粒是疏水的,使用树枝状分子和1-十六烷基胺的缀合物(表示为HDA-G2)通过疏水-疏水相互作用将纳米颗粒转移到水性介质中(Zhou等,同上)。包封纳米颗粒在水溶液中表现出优异的胶体稳定性。在储存超过一个月后,没有观察到聚集或形貌变化。通过动态光散射(DLS)测量所有样品的流体动力学直径(HD)。球形-10、球形-16、八角-20和八角-30的HD分别为22±2nm、30±3nm、49±5nm和58± 2nm,表明铁氧化物纳米颗粒在水中是单分散的而没有任何显著的群集和聚集。此外,在这些包封纳米颗粒中,在水分散性八角纳米颗粒的表面上有大量可用的游离胺基,允许进一步改性和官能化。应注意,八角-30 的HD为球形-16的约2倍,与提出的模型一致(图10)。此外,由于独特的八角结构,八角-30在水介质中的物理表面与体积的比率大于模拟的球形模型。因此,预期,八角-30对于水分子扩散的有效表面积可能超过球形-16的4倍。
八角铁氧化物纳米颗粒的横向弛豫率。在7T MR扫描仪上测试上述 4种样品的横向弛豫率(r2)。如所预期的,随着Fe浓度的增加,T2加权模型图像的信号强度降低(图13),表明所有样品在T2加权序列下具有产生MRI对比增强的潜力。显著地,八角铁氧化物纳米颗粒比球形铁氧化物纳米颗粒表现出更强的T2对比效果,表明八角铁氧化物纳米颗粒可以充当高灵敏T2造影剂。八角-30、八角-20、球形-16和球形-10的r2值分别为约679.25±30mM-1S-1、209.03±15mM-1S-1、125.86±9mM-1S-1和 59.91±6mM-1S-1。由于八角铁氧化物核心增加的有效半径,八角-30的 r2值为球形-16的约5.4倍。同时,八角-20也具有比球形-10大(约3.5倍)的r2值。需要提到的是,两种八角铁氧化物纳米颗粒与相应球形铁氧化物纳米颗粒(即,八角-30与球形-16以及八角-20值球形-10)的Ms值和几何体积非常接近。这些结果证明,通过形貌控制来在结构上增加铁氧化物的有效半径是对增加铁氧化物纳米颗粒的T2弛豫率的现有策略如金属掺杂和颗粒群集的有吸引力的替代选择。
使用八角铁氧化物纳米颗粒进行肝MR成像。在进行动物研究之前,首先使用HepG2细胞系作为模型测试水分散性八角铁氧化物纳米颗粒的细胞毒性。3-(4,5-二甲基噻唑-2-基)-2,5-二苯基四唑溴化物(MTT)测定表明,即使在高达100μg Fe/mL的浓度下,八角铁氧化物纳米颗粒在24 小时没有明显的细胞毒性,表明八角铁氧化物纳米颗粒是生物相容的(图 14)。为了证明在体内八角铁氧化物纳米颗粒比常规球形铁氧化物纳米颗粒具有更好的对比效果,选择八角-30和球形-16作为代表性样品并且使用BALB/c小鼠作为模型进行肝的T2加权的MRI。在以1mg Fe/kg小鼠体重的剂量静脉内注射八角-30和球形-16样品后,对于两种纳米颗粒,在注射后(post-injection,p.i.)0.5小时在肝脏区域确实观察到了显著的信号衰减(图15)。为了量化对比度,肝被认为是目的区域(region of interest,ROI),计算每只动物的信噪比(SNR)和SNR后/SNR前值并且列在下表中。
八角-30(注射后0.5小时和1小时,分别为63.9±3.2%和67.3±1.3%) 表现出比球形-16(注射后0.5小时和1小时,分别为39.8±1.5%和53.5± 1.6%)更高的对比度,表明在体内肝的T2成像中具有更高r2值的八角-30 比球形-16更灵敏。在施用八角-30和球形-16后,普鲁士蓝染色检测到了遍布肝脏切片的蓝色斑点(图16),证实肝中的信号衰减是由铁氧化物纳米颗粒积累造成的。电感耦合等离子体质谱(ICP-MS)分析(图17)表明肝脏以可比的方式摄取八角-30和球形-16,证明了好得多的对比是由于八角-30更高的r2值造成的。在MR成像中,更低剂量的造影剂可能意味着更低的成本和更少的副作用,这保证了在临床诊断中更好的前景。八角 -30更高的对比度暗示了以更低剂量进行肝MR成像。通过将八角-30的注射剂量减少到0.5mg Fe/kg,对比(0.5小时39.9±2.5%,1小时56.2± 1.9%)依然略高于1mg/kg剂量的球形-16(图18)。
使用八角铁氧化物纳米颗粒通过MRI检测肝癌。为了进一步评估八角-30用于肝癌成像的能力,在原位HepG2肿瘤模型上进行T2加权MRI。通过向裸鼠肝脏中接种小的皮下HepG2肿瘤碎片来建立原位肝肿瘤模型。当肝癌直径达到3-5mm时,将八角-30和球形-16静脉内注射到该裸鼠中(2mg Fe/kg),并且在7T microMRI扫描仪上对动物进行扫描。由于肝肿瘤包含少得多的活跃枯否细胞和巨噬细胞,其不能像正常肝组织那样有效地积累铁氧化物纳米颗粒。因此与正常肝组织相比,肝肿瘤将表现出假阳性对比。在静脉内施用后,两种颗粒均在肿瘤部位造成明显的对比增强(图19)。但是,八角-30的注射在肿瘤部位导致比球形-16更高的 MR对比,使得容易区分MR图像中的肝病灶和正常肝组织。肿瘤与肝脏的对比随着时间而增加,并且在八角-30和球形-16注射后4小时分别高达136.9±8.5%和64.5±2.7%,表明八角-30比球形-16在肝肿瘤成像中表现出更高的信号变化和检测限。因此,使用具有超高T2弛豫率的八角 -30作为造影剂可以显著改善T2成像的灵敏度,这对于癌症的准确检测和早期诊断极其重要。
一般程序
FeCl3、NaCl、KC1、KBr、NaF、己烷、油酸钠、异丙醇和乙醇购自国药集团化学试剂有限公司(中国上海)。1-十八烯(90%)、油酸(90%)、十六烷基三甲基氯化铵和十六烷基三甲基溴化铵购自Alfa Aesar(Ward Hill,MA)。所有试剂以接收状态使用而无进一步纯化。
铁氧化物纳米颗粒的MR弛豫率的测量。为了测量T2弛豫率,使具有不同铁浓度的八角-30、八角-20、球形-16和球形-10分散在1%琼脂糖溶液中。使用多重回波T2加权的快速自旋回波成像序列(TR/TE=2000/20、 40、60、80、100ms,切片厚度=2mm)在7T MRI扫描仪(Varian 7T micro MRI System)上扫描样品。
细胞培养。HepG2细胞购自中国科学院细胞库(中国上海)并且培养在补充有10%胎牛血清(FBS,Hyclone)和抗生素(100mg/mL链霉素和100U/mL青霉素)的Dulbecco改良的Eagle培养基(DMEM培养基)中。所有细胞保持在37℃、5%CO2的潮湿气氛下。
体外细胞毒性评估。将HepG2细胞(1×104)接种在96孔平板上并且在DMEM(含10%FBS)中孵育12小时。在将细胞用PBS洗涤两次后,以不同浓度(Fe当量浓度为100、67、44、30、20、13、9、6、4和 0μg Fe/mL)添加含八角铁氧化物纳米颗粒的新鲜培养基并且孵育细胞 24小时。相同浓度下的每个实验在5个孔中进行。将生长培养基替换成含0.5mg/mL 3-(4,5-二甲基噻唑-2-基)-2,5-二苯基四唑溴化物(MTT)的 DMEM并且再次孵育细胞4小时。弃掉培养基并且添加100μL DMSO以溶解沉淀物,使用MultiSkan FC微量板读取器(ThermoScientific)测量所得溶液在492nm的吸光度。
体内肝MR成像。根据厦门大学实验动物管理和使用委员会 (InstitutionalAnimal Care and Use Committee of Xiamen University)批准的方案进行动物实验。使用BALB/c小鼠作为模型进行体内肝的MR成像。在以1mg Fe/kg小鼠体重的剂量静脉内注射铁氧化物纳米颗粒后,在Varian 7T microMRI扫描仪上使用fSEMS序列(TR/TE=3000/40ms, 256×256矩阵,平均值=1)扫描冠状和横向平面MR图像。获得注射前、注射后0.5小时、和注射后1小时(n=3/组)的MR图像。为了量化信号增强,通过等式SNR肝脏=SI肝脏/SD噪声计算信噪比(SNR),其中SI表示信号强度,SD表示表标准偏差。
体内肝肿瘤MR成像。通过向裸鼠肝脏中接种小的皮下HepG2肿瘤碎片来建立原位肝肿瘤模型。当肿瘤直径达到3-5mm时,以2mg Fe/kg 的剂量向小鼠静脉内注射八角-30和球形-16。在7T MRI扫描仪上使用 fSEMS序列(TR/TE=3000/40ms,256×256矩阵,平均值=1)获得冠状和横向平面MR图像。在注射后0、0.5、1、2和4小时(n=3/组)连续获得MR图像。为了量化对比增强效力,引入对比度-噪声比(CNR),其通过CNR=(SNR肿瘤-SNR肝脏)/SNR肿瘤给出。
普鲁士蓝染色。在MR成像后,处死小鼠并且将小鼠肝脏保持在最佳切片温度(optimal-cutting-temperature,O.C.T)化合物中并且储存在 -80℃。当混合物冰冻时,将样品切成10μm厚切片并且立即用冰冷丙酮固定5分钟。在室温干燥5分钟后,将切片放到染色溶液(20%盐酸和 10%亚铁氰化钾溶液混合物,1:1体积比)中30分钟,并且用曙红复染5分钟。然后,使用90%、95%和纯水清洗切片3次。
以下实施例进一步描述了本发明,但是当然,其不应解释为以任何方式限制本发明的范围。
实施例1
本实施例说明了油酸铁的合成
将4.56g油酸钠(15毫摩尔)和0.81g FeCl3(5毫摩尔)溶解在20 mL蒸馏水和10mL乙醇的混合物中。将所得溶液在氩气氛下加热到70℃并且在该温度下保持4小时。当反应结束时,添加己烷,并且将己烷层在分液漏斗中用蒸馏水洗涤三次。蒸发掉己烷后,获得蜡状固体形式的油酸铁配合物。
实施例2
本实施例展示了16nm尺寸的对比球形铁氧化物纳米颗粒的合成。
通过别处建立的程序进行球形铁氧化物纳米颗粒的合成(Park等,Nat.Mater.2004,3:891-895)。在室温下,将0.93g如实施例1所述合成的油酸铁(1毫摩尔)和160μL油酸(0.5毫摩尔)溶解在15mL 1-十八烯中。在室温下将混合物在真空中脱气30分钟,并利用氩气回填以除去任何低挥发性杂质和氧气。然后以3.3℃min-1的恒定加热速率将反应溶液加热到320℃,并且在该温度下保持1小时。反应溶液由初始红棕色变成棕黑色。然后将所得溶液冷却到室温并且与30mL异丙醇混合以沉淀纳米颗粒。通过离心来分离纳米颗粒并且用乙醇洗涤3次。洗涤后,将纳米颗粒溶解在己烷中以在4℃下长期储存。
实施例3
本实施例展示了对比合成25nm尺寸的球形铁氧化物纳米颗粒的合成
在室温下将0.93g油酸铁(1毫摩尔)和160μL油酸(0.5毫摩尔) 溶解在10mL三辛胺中。在室温下将混合物在真空中脱气30分钟,并利用氩气回填以除去任何低挥发性杂质和氧气。以3.3℃min-1的恒定加热速率将反应溶液加热到350℃,并且在该温度下保持5小时。反应溶液由初始红棕色变成棕黑色。然后将所得溶液冷却到室温并且与30mL乙醇混合以沉淀纳米颗粒。通过离心来分离纳米颗粒并且用乙醇洗涤3次。洗涤后,将纳米颗粒溶解在己烷中以在4℃下长期储存。
实施例4
本实施例展示了根据本发明一个实施方案的棱长为30nm的八角铁氧化物纳米颗粒的合成。
将油酸铁(0.8g,0.86毫摩尔)、NaCl(10mg,0.17毫摩尔)、油酸 (110μL,0.35毫摩尔)和蒸馏水(60μL)与10mL 1-十八烯混合在一起。在室温下将所得溶液在真空中脱气30分钟,并利用氩气回填以除去任何低挥发性杂质和氧气。以3.3℃min-1的恒定加热速率将反应溶液加热到320℃,并且在该温度下保持2小时。溶液颜色从红棕色转变为透明橙色并最终变为棕黑色。将所得溶液冷却到室温并且与30mL异丙醇混合以沉淀纳米颗粒。通过离心来分离纳米颗粒并且用乙醇洗涤3次。将所得产物溶解在己烷中以在4℃下长期储存。
实施例5
本实施例展示了根据本发明一个实施方案的棱长为20nm的八角铁氧化物纳米颗粒的合成。
如实施例4中所述进行棱长为20nm的八角铁氧化物纳米颗粒的合成,只是在加热到320℃后,将溶液在该温度保持1小时。通过离心来分离纳米颗粒并且用乙醇洗涤3次。将纳米颗粒溶解在己烷中以在4℃下长期储存。
实施例6
本实施例展示了水溶性HDA-G2包封纳米颗粒的制备
通过将纳米颗粒与HDA-G2共沉淀来获得具有HDA-G2的八角铁氧化物纳米颗粒复合物。将含有20mg HDA-G2的1mL氯仿添加到含有10 mg八角铁氧化物纳米颗粒的1mL氯仿中,然后容器打开放在通风厨中以在室温下缓慢蒸发溶剂。通过泵完全除去残余氯仿,并且通过超声处理使干燥样品重新分散在水中。通过尺寸排阻色谱(PD-10柱,GE HealthcareLife Science)对水分散性样品进行进一步纯化。将最终水溶液储存在4℃下用于进一步使用。
