背景技术
目前手术器械清洗液一般为戊二醛或者新洁尔灭,戊二醛属高效消毒剂。因具有杀菌谱广、杀细菌芽孢效果可靠、杀菌作用受有机物影响较小、对金属腐蚀性小等特点,适用于不耐热的医疗器械和精密仪器的消毒与灭菌,尤其是内窥镜的消毒灭菌。但由于戊二醛对人体具有潜在的毒性、对皮肤黏膜具有刺激性,目前有些人正在积极寻找其替代品用于内窥镜和其他医疗器械的灭菌。新洁尔灭兼有杀菌和去垢效力,作用强而快,对金属无腐蚀作用,不污染衣服,性质稳定,易于保存,属消毒防腐类药.稀释液可用于外科手术前洗手(0.05~0.1%,浸泡5分钟),皮肤消毒和霉菌感染(0.1%),黏膜消毒(0.01~0.05%),器械消毒(置于0.1%的溶液中煮沸15分钟后再浸泡30分钟).忌与肥皂,盐类或其他合成洗涤剂同时使用,避免使用铝制容器,消毒金属器械需加0.5%亚硝酸钠防锈,不宜用于膀胱镜,眼科器械及合成橡胶的消毒.对革兰氏阴性杆菌及肠道病毒作用弱,对结核杆菌及芽孢无效。目前缺少有效的手术器械清洗液。

所述的手术器械清洗液,制备方法包括如下步骤:取上述中药野席草、黄囊杓兰、狭叶蓬莱葛、定草根、黄花岩松、树扁竹,加药材10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,减压浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置,过滤,滤液减压浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,加入次氯酸盐、十二烷基磺酸钠,即手术器械清洗液。

黄花岩松为景天科景天属植物黄花岩松SedumleucocarpumFranch.,以全草入药。秋季采,晒干或鲜用,清热消炎,用于肝炎,外用治中耳炎,烧烫伤。
树扁竹为兰科莪白兰属植物鸢尾扁竹兰OberoniairidifoliaLindl.,以全草入药。四季可采,开水烫后,晒干,理气消食,清热利尿,止咳止痛。用于消化不良,胃痛,肠炎,尿路感染,咳嗽,支气管炎,哮喘,跌打损伤,骨折,毒蛇咬伤。其余为药典品种。
有益效果:本发明中野席草、黄囊杓兰、狭叶蓬莱葛、定草根、黄花岩松、树扁竹清热解毒,诸药相合,大量减少化学消毒剂的用量,具有抗菌抗病毒作用,用于消毒手术器械。本发明处方设计合理,配伍严谨,经长期的验证,疗效确切。本发明组成为中药材,无毒副作用,价格便宜。

以下通过实施例形式,对本发明的上述内容再作进一步的详细说明,但不应将此理解为本发明上述主题的范围仅限于以下的实例,凡基于本发明上述内容所实现的技术均属于本发明的范围。
实施例1:取野席草22份、黄囊杓兰10份、狭叶蓬莱葛12份、定草根12份、黄花岩松19份、树扁竹12份,加药材10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,减压浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置,过滤,滤液减压浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,加入次氯酸盐9份、十二烷基磺酸钠2份,即得手术器械清洗液,使用方法为将手术器械放于清洗液中煎煮30分钟。
实施例2:取野席草28份、黄囊杓兰10份、狭叶蓬莱葛12份、定草根12份、黄花岩松19份、树扁竹12份,加药材10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,减压浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置,过滤,滤液减压浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,加入次氯酸盐5份、十二烷基磺酸钠4份,即得手术器械清洗液,使用方法为将手术器械放于清洗液中煎煮30分钟。
实施例3:取野席草26份、黄囊杓兰15份、狭叶蓬莱葛10份、定草根14份、黄花岩松17份、树扁竹14份,加药材10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,减压浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置,过滤,滤液减压浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,加入次氯酸盐7份、十二烷基磺酸钠3份,即得手术器械清洗液,使用方法为将手术器械放于清洗液中煎煮30分钟。
实施例4:本发明抑菌试验
药品及试剂:本发明按实施例3制备方法制备;对照组含有次氯酸盐7份、十二烷基磺酸钠3份,加50份去离子水,配制成消毒液;青霉素钠:华北制药股份有限公司规格:80万单位/瓶批号:Y1105656,氯化钠,上海久亿化学试剂有限公司,批号:00090130。营养肉汤,上海中科昆虫生物技术开发有限公司,批号:120240。营养琼脂,江苏省疾病预防控制中心微生物试剂厂,批号:130026。小牛血清,杭州四季青生物工程材料有限公司,批号:140403;脱纤绵羊血,购自南京绵羊血供应站,批号:14090:7
菌株:临床分离菌株:金黄色葡萄球菌、大肠杆菌、乙型化脓性链球菌、白色念珠菌,由江苏省人民医院微生物室分离鉴定提供。
仪器设备:电子天平,北京塞多利斯天平有限公司生产;全自动高压灭菌锅,日本SANYO;超净工作台,苏州净化设备厂;隔水式恒温培养箱,上海一恒科技有限公司
增菌培养:取临床分离菌株金黄色葡萄球菌、大肠杆菌、乙型化脓性链球菌、白色念珠菌菌株少许,分别接种于肉汤培养基中,37℃培养18h。(乙型化脓性链球菌肉汤培养基中加20%小牛血清)取18h培养的各菌株营养肉汤培养物,用营养肉汤作10-3稀释用于实验。
二倍稀释法(试管法):取灭菌试管11支,第1管加入营养肉汤液体培养基1.6mL,其余每管1mL,分别取本发明护理液加入第一管中,混匀后取1mL至第2管,依次稀释,第10管吸出1mL弃去,第11管不加药液作为对照。每管加入稀释菌液0.1mL,37℃培养24h,取出观察细菌生长情况。如药液管混浊,即表示细菌生长,供试药物无抑菌作用;如药液管清亮则表示细菌生长受到抑制,其能抑制细菌生长的最大稀释度的药液,即为该药物的最小抑菌浓度(MIC)。将清亮的药液肉汤各管转种肉汤琼脂平板,培养24h观察,仍无细菌生长的最小药物浓度即为该药物的杀菌浓度。
取青霉素钠(配成16万单位/mL)按上述方法,依次稀释,培养,作阳性对照。
实验结果显示:本发明对临床分离菌金黄色葡萄球菌、大肠杆菌、乙型化脓性链球菌、白色念珠菌均具有较强的抑菌作用,MIC在5mg~500mg/mL,表明本发明具有体外抗菌作用。见表1。

实验结论:本发明对临床分离菌金黄色葡萄球菌、大肠杆菌、乙型化脓性链球菌、白色念珠菌均具有较强的抑菌作用,MIC在5mg~200mg/mL,表明本发明具有体外抗菌作用,而且比单纯含有次氯酸盐、十二烷基磺酸钠的消毒液效果好。
Claims (4)
1.一种手术器械清洗液,其特征在于,它由如下重量份数的原料药制备得到:野席草22-28份、黄囊杓兰10-20份、狭叶蓬莱葛8-12份、定草根12-16份、黄花岩松15-19份、树扁竹12-16份、次氯酸盐5-9份、十二烷基磺酸钠2-4份。
2.根据权利要求1所述的手术器械清洗液,其特征在于,它由如下重量份数的原料药制备得到:野席草26份、黄囊杓兰15份、狭叶蓬莱葛10份、定草根14份、黄花岩松17份、树扁竹14份、次氯酸盐7份、十二烷基磺酸钠3份。
3.权利要求1所述的手术器械清洗液,其特征在于,制备方法包括如下步骤:取上述中药野席草、黄囊杓兰、狭叶蓬莱葛、定草根、黄花岩松、树扁竹,加药材10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,减压浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置,过滤,滤液减压浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,加入次氯酸盐、十二烷基磺酸钠即手术器械清洗液。
4.权利要求1所述手术器械清洗液在手术器械清洗液中的应用,使用方法为将手术器械放于清洗液中煎煮30分钟。
