Abstract
本发明公开了一种高浓度NAA与甘油凝胶的沾根方法,属于植物生物技术领域。首先配制1mg/mL的NAA母液;将该母液稀释成500mg/L的高浓度NAA工作液;以上述工作液配制质量浓度为1%的羧甲基纤维素钠、质量浓度为8%海藻酸钠的高浓度NAA与甘油凝胶封闭液;将健康粗壮的枝条下端沾在上述凝胶封闭液中3-5秒后拿出;将上述枝条插入栽培基质中生根。

技术领域
一种高浓度NAA与甘油凝胶的沾根方法,属于植物生物技术领域。
背景技术
扦插繁殖是植物、尤其是树木无性繁殖的常用方法之一,该方法具有成本低、见效快、周期短、所用枝条来源丰富且能够保持亲本优良性状等诸多优点。扦插繁殖时常常会使用到不同类型的激素,如NAA、IBA、NAA、2,4-D、GA3、6-BA、KT、TDZ、TIBA、乙烯利、B9、PP333、MH等,上述激素的添加会使植物在体或离体条件下能够提早或延迟生根、开花、坐果等,增加根数、诱导腋芽萌发、提高座果率等,在农业生产上发挥着重要的用途。然而,在离体繁殖或生根过程中难以分化的枝条由于长时间的水分或营养物质的亏损,极其容易导致死亡。而现有的采用催根诱芽开花的方法具有技术步骤较多、成本高、不稳定等缺点,在生产上效果并不理想。因此,亟需一种快速简便的方法用于植物腋芽诱导、生根及成花等。
发明内容
一种高浓度NAA与甘油凝胶的沾根方法,其特征在于,所述方法中包括有激素母液配制、工作液配制、激素凝胶配制、沾根、扦插,具体操作步骤如下:
①激素母液配制:准确称量重量为0.1g的NAA用少许95%的乙醇溶解后,用蒸馏水定容至100mL,即为1mg/mL的NAA母液;
②工作液配制:将上述母液按照体积比1:2的比率稀释成浓度为500mg/L的高浓度激素工作液,同时按照90ml激素工作液加入10ml甘油;
③激素凝胶配制:准确称取1g羧甲基纤维素钠和8g海藻酸钠,在50℃水浴条件下用100mL的浓度为500mg/L的激素与甘油工作液溶解备用;
沾根:将健康粗壮的枝条下端沾在上述凝胶封闭液中3-5秒后拿出;
扦插:将上述枝条插入栽培基质中生根。
本发明的优点在于从保水、促根、诱芽、开花、坐果等方面考虑,以激素工作液为溶剂,羧甲基纤维素钠和海藻酸钠为凝胶载体,配制成含有不同类型、浓度激素的凝胶溶液。使用时将枝条、花朵稍稍浸泡于上述溶液或者将上述溶液滴加在腋芽处,凝胶干燥后会迅速形成薄膜,达到减少水分蒸发、持续施加激素、操作简便等特点。

实施例
一种高浓度NAA与甘油凝胶的沾根方法,其特征在于,所述方法中包括有激素母液配制、工作液配制、激素凝胶配制、沾根、扦插,具体操作步骤如下:
①激素母液配制:准确称量重量为0.1g的NAA用少许95%的乙醇溶解后,用蒸馏水定容至100mL,即为1mg/mL的NAA母液;

沾根:将8-10cm长、健康粗壮的黄杨枝条下端沾在上述凝胶封闭液中3-5秒后拿出;
扦插:将上述枝条插入栽培基质中生根。

1.一种高浓度NAA与甘油凝胶的沾根方法,其特征在于,所述方法中包括有激素母液配制、工作液配制、激素凝胶配制、沾根、扦插,具体操作步骤如下:
