Abstract
本发明涉及小杆线虫在防治核桃云斑天牛中的应用。小杆线虫R.(O.)chongmingensis Tumian(简称Tumian)可防治蛀干害虫。本发明人将小杆线虫Tumian制成制剂施用于核桃园中,结果表明小杆线虫Tumian防治核桃云斑天牛的效果显著,防治幼虫效果达90%(校正有效率91.4%)以上,阻止卵孵化效果近70%(68.4%)。
Description
技术领域
本发明涉及一种生物防治技术领域,特别是涉及利用一种小杆线虫Rhabditis(Oscheius)chongmingensis Tumian防治核桃云斑天牛的技术。
背景技术
云斑天牛(Batocera horsfieldi)又称云斑白条天牛,属鞘翅目(Coleoptera)天牛科(Cerambycidae),是我国重大林木蛀干害虫,在核桃树上危害尤为严重。主要以幼虫在树干木质部取食钻蛀危害,并以幼虫和成虫在蛀道内和蛹室中越冬,形成大量蛀道,严重时造成树木死亡或风折,从而使之失去经济价值和生态价值。
长期以来对于此类害虫的防治主要靠人工钩杀和蛀道内注入高浓度、高毒性化学农药。但这些方法效果并不理想,而且对施药人员和生态环境毒性较大。
小杆属线虫资源丰富,分布广泛,易于获得,易于培养。传统上认为小杆属Rhabditis(Oscheius)线虫是食细菌类线虫,国外尚未见小杆线虫用于防治害虫的报道,国内仅有本发明人用小杆线虫Rhabditis spp防治地下害虫蛴螬的报道,迄今为止国内外尚未见用该小杆线虫Rhabditis(Oscheius)chongmingensis Tumian防治核桃云斑天牛的报道。
发明内容
本发明针对上述领域的空白,提供小杆线虫R.(Oscheius)chongmingensisTumian在防治核桃云斑天牛中的应用,以解决生产上的难题,拓宽生物防治途径。
小杆线虫R.(O.)chongmingensis Tumian(简称Tumian)在防治蛀干害虫中的应用。
所述蛀干害虫为天牛。
所述天牛为核桃云斑天牛。
所述应用是将所述小杆线虫制剂在植物生长发育期施于被天牛危害的树干、树枝刻槽部和排粪孔内。
所述植物为核桃树,所述天牛为核桃云斑天牛。
所述防治是抑制云斑天牛卵孵化或杀死云斑天牛幼虫或成虫。
所述小杆线虫制剂为小杆线虫的悬浮水溶液,浓度为250-8000条/孔,施用量为2-4mL/蛀孔。
所述小杆线虫制剂的浓度为2000-4000条/mL。
本发明提供了一种新的防治云斑天牛的方法,采用本发明人从内蒙棉田土壤中采集到的小杆线虫Tumian制成制剂施用于核桃园中,结果表明小杆线虫Tumian防治核桃云斑天牛的效果显著,防治幼虫效果达90%(校正有效率91.4%)以上,阻止卵孵化效果近70%(68.4%)。

具体实施方式
现结合实施例对本发明作进一步的详述。
本发明实施例虽然只是应用于核桃云斑天牛的防治,但对危害其它林木果树的及其它蛀干害虫(如吉丁虫、小蠹虫等)来说,基于同样的原理,小杆线虫Tumian同样适用于其它天牛危害和蛀干害虫危害的果园和林地。唯有不同的是实用技术,既不同蛀干害虫在不同地区、不同寄主上的危害时期和部位深浅不同,需根据生物学特性而调整。
实施例一、田间防治核桃云斑天牛卵的效果案例
1材料与方法
1.1试验材料
供试药剂:小杆线虫Rhabditis(Oscheius)chongmingensis Tumian(Liu Qizhi,Zdenˇek M.Zhang Lijuan,Vladimír P.and Dong Lamei.2012.Re-description ofOscheius chongmingensis(Zhang et al.,2008)(Nematoda:Rhabditidae)and itsentomopathogenicity.Nematology,14(2):139-149.),本申请第一发明人从内蒙棉田土壤中采集、分离、纯化、扩繁所得,可对外公开发放。收集后的线虫悬浮在水溶液中,在4℃保存,20天内施用。线虫施用浓度如下述试验方法。
1.2试验方法
2014年7-9月在贵州省赫章县野马川镇苏噶村随机选取被云斑天牛严重危害的核桃园,不进行其它防治。在核桃树上标记有天牛卵的新刻槽。
将线虫R.chongmingensis Tumian分别配成250IJs/mL、500IJs/mL、1000IJs/mL、2000IJs/mL、4000IJs/mL五个浓度梯度。每个处理设置3个重复(即3棵核桃树),每个重复(每棵核桃树上)的刻槽数20~100个不等,标记所有刻槽。每个刻槽表层涂施1-2mL某一种浓度的线虫粘稠液。对照施用等量清水粘稠液。每7天观察并记录一次有粪排除的刻槽数后清除虫粪,共观察7次,至第49天。
1.3计算方法
按下列公式计算:
卵的有效率%=无虫粪率%=[1-(排粪刻槽数/总刻槽数)]×100%
校正有效率%=(处理无虫粪率-对照无虫粪率)/(100%-对照无虫粪率)×100%
2结果与分析
2.1防治核桃云斑天牛卵的有效率
从图1看出:供试线虫O.chongmingensis Tumian对核桃云斑天牛卵的抑制效果在4000IJs/mL线虫剂量下达54.7~68.4%(7天——49天)。从整体趋势来看,线虫侵染时间越长,孵化抑制率越高。
实施例二、田间防治核桃云斑天牛幼虫的效果案例
1材料与方法
1.1试验材料
供试药剂:小杆线虫Rhabditis(Oscheius)chongmingensis Tumian,本第一发明人从内蒙棉田土壤中采集、分离、纯化、扩繁所得,可对外公开发放。收集后的线虫悬浮在水溶液中,在4℃保存,20天内施用。线虫施用浓度如下述试验方法。

校正有效率%=(处理无虫粪率-对照无虫粪率)/(100%-对照无虫粪率)×100%
2结果与分析
2.1防治核桃云斑天牛幼虫的有效率
从表1看出:供试线虫Tumian对核桃云斑天牛幼虫具有很高的侵染力,防治效果达75.3~93.7%。施用该线虫浓度为250IJs/mL时,7-28天的有效率达75.3~80.1%;施用500IJs/mL时,有效率达78.4~84.4%;施用1000IJs/mL时,有效率达81.8~88.5%;施用2000IJs/mL时,有效率达83.5~90.3%;线虫施用为4000IJs/mL时,有效率达84.5~93.7%。

2.2防治核桃云斑天牛幼虫的校正有效率
从图2看出:供试线虫Tumian对核桃云斑天牛幼虫的校正有效率在4000IJs/mL线虫剂量下第28天达91.4%。
从整体趋势来看,线虫侵染剂量越大、侵染时间越长,对幼虫的防治有效率越高。

1.小杆线虫R.(O.)chongmingensis Tumian在防治蛀干害虫中的应用,所述蛀干害虫为核桃云斑天牛;
所述应用是将小杆线虫制剂在植物生长发育期施于被天牛危害的树干、树枝刻槽部和排粪孔内,所述植物为核桃树。
2.根据权利要求1所述的应用,所述防治是抑制云斑天牛卵孵化或杀死云斑天牛幼虫。
3.根据权利要求1所述的应用,所述小杆线虫制剂为小杆线虫的悬浮水溶液,浓度为250-8000条/mL,施用量为2-4mL/蛀孔。
4.根据权利要求3所述的应用,所述小杆线虫制剂的浓度为2000-4000条/mL。
