Abstract

Description
发明领域
本发明涉及一种液体酶组合物。本发明还涉及一种从培养液回收和/或提纯酶的方法。
发明背景
液体酶产品在商业上被大量地用作不同生产过程中(例如在从淀粉或(半)纤维素原料生产生物乙醇中)的加工助剂。通常,通过微生物培养,然后是培养液回收、提纯、标定和配制来生产酶产品。
在回收、提纯、储存以及运输过程中液体酶组合物中的微生物生长是一个问题。细菌、酵母菌以及霉菌均可引发问题,并且特别地乳酸杆菌的生长倾向于是有问题的。为了限制此类微生物的生长,在终产品中通常保持较高固体含量,并且例如通过添加抗微生物剂诸如亚硫酸盐、山梨酸盐以及苯甲酸盐可以进一步抑制微生物的生长。
发明内容
诸位发明人已经发现在液体酶组合物中添加C6-C12脂肪酸或其盐对乳酸杆菌具有抗微生物作用。
因此,本发明提供了一种液体组合物,该液体组合物包含一种酶和一种对乳酸杆菌具有抗微生物作用的量的C6-C12脂肪酸或其盐。
本发明还提供了一种从培养液回收、提纯和/或配制酶的方法,该方法包括将一种C6-C12脂肪酸或其盐以一个对乳酸杆菌具有抗微生物作用的量添加至该培养液中。最后,本发明提供了C6-C12脂肪酸或其盐在液体酶组合物的回收、提纯、储存或运输过程中杀死或抑制乳酸杆菌生长的用途。
发明详细说明
抗微生物作用
该液体组合物包含一种酶和一种对乳酸杆菌具有抗微生物作用的量的C6-C12脂肪酸或其盐,即该C6-C12脂肪酸或其盐的量对于杀死或抑制乳酸杆菌的生长是有效的。
该抗微生物作用可以通过分析用乳酸杆菌接种并且在20℃-25℃孵育1-2周后液体配制品的活细胞(CFU)并且与没有该脂肪酸或盐的相似的液体组合物比较来确定。
液体组合物
通常,该液体组合物具有一个10%w/w以上、15%w/w以上、20%w/w以上、25%w/w以上、30%w/w以上、35%w/w以上或45%w/w以上的干物质固体总含量。这可以通过在105℃干燥直到恒重来确定。
除了酶,这些固体还可以包含一种或多种多元醇和/或无机盐。适合的多元醇包括丙三醇、丙二醇(MPG)、山梨糖醇以及单、二和三糖,诸如葡萄糖、果糖、蔗糖和海藻糖。无机盐可以是碱金属卤化物盐,诸如NaCl或KCl。多元醇和/或无机盐的含量可以是10%w/w以上、15%w/w以上、20%w/w以上、25%w/w以上、30%w/w以上、35%w/w以上或45%w/w以上。
该液体组合物包含一种或多种酶。它具有一个至少1%w/w,特别是至少2%w/w、至少5%w/w或至少10%w/w的酶蛋白含量。该酶含量可以是25%w/w以下、20%w/w以下、15%w/w以下或10%w/w以下。
该液体组合物典型地具有一个至少3.5、至少4.0或至少4.5的pH值。该pH值典型地是7以下,特别是6.5以下、6以下、5.5以下或5以下。特别地,该pH可以在4-5.5的范围内。
该液体组合物典型地本质上不含表面活性剂(阴离子和非离子的两者)。
酶
本发明对于包含一种或多种酶的液体配制品是可适用的。实例包括用于在从淀粉或(半)纤维原料生产生物乙醇(第一和第二代生物乙醇)中使用的酶或用于在纺织品工业中使用的酶。该酶可以是水解酶、碳水化合物分解酶、糖苷酶(EC 3.2)、α-淀粉酶(EC 3.2.1.1)、葡聚糖1,4-α-葡糖苷酶(葡糖淀粉酶;淀粉转葡糖苷酶,EC 3.2.1.3)、纤维素酶、半纤维素酶、植酸酶、蛋白酶或支链淀粉酶。
用于从淀粉原料生产生物乙醇(1G)的酶
可以根据本发明配制以下酶产品:
芽孢杆菌属α-淀粉酶(WO 1996/023873,WO 1999/019467)。
原料淀粉水解(RSH)α-淀粉酶和RSH葡糖淀粉酶的共混物(WO2006/069289,WO 2006/069290)。
葡糖淀粉酶(WO 1999/028448,WO 2006/069289,WO 2006/069290)。
商用产品的实例是LiquozymeTM、TermamylTM SC和Novozyme BPX、SpirizymeTM(诺维信公司(Novozymes A/S)的产品)。
用于从(半)纤维原料生产生物乙醇(2G)的酶
可以根据本发明配制以下酶产品:
含有米曲霉β-葡糖苷酶融合蛋白(WO 2008/057637)和橙色嗜热子囊菌GH61A多肽(WO 2005/074656)的里氏木霉纤维素酶制剂。
棘孢曲霉GH10木聚糖酶(WO 94/021785)。
棘孢曲霉GH10木聚糖酶(WO 94/021785)与含有烟曲霉β-葡糖苷酶(WO2005/047499)和橙色嗜热子囊菌GH61A多肽(WO 2005/074656)的里氏木霉纤维素酶制剂的共混物。
含有棘孢曲霉GH10木聚糖酶(WO 94/021785)的里氏木霉纤维素酶制剂。
烟曲霉GH10木聚糖酶(WO 2006/078256)和烟曲霉β-木糖苷酶(WO2011/057140)与含有烟曲霉纤维二糖水解酶I(WO 2011/057140)、烟曲霉纤维二糖水解酶II(WO 2011/057140)、烟曲霉β-葡糖苷酶变体(PCT/US2011/054185)和青霉属(Penicillium sp.)(埃默森青霉(emersonii))GH61多肽(WO 2011/041397)的里氏木霉纤维素酶制剂的共混物。
含有烟曲霉GH10木聚糖酶(WO 2006/078256)和烟曲霉β-木糖苷酶(WO2011/057140)的里氏木霉纤维素酶制剂。
商用产品的实例是CTec、Cellic HTec、Cellic CTec2、Cellic HTec2、Cellic CTec3、Cellic HTec3(诺维信公司的产品)。
α-淀粉酶
该α-淀粉酶可以是微生物来源的,例如细菌或真菌。该细菌α-淀粉酶可以来源于芽孢杆菌属,例如来源于地衣芽孢杆菌、解淀粉芽孢杆菌、枯草芽孢杆菌或嗜热脂肪芽孢杆菌的菌株。
真菌α-淀粉酶包括来源于曲霉属的菌株的α-淀粉酶,诸如米曲霉、黑曲霉和白曲霉(Aspergillus kawachii)α-淀粉酶。其他考虑到的野生型α-淀粉酶包括那些来源于根毛霉属和多孔菌属(Meripilus)的菌株,优选微小根毛霉属或大型多孔菌(Meripilus giganteus)的菌株。
该真菌α-淀粉酶可以是一种包含一个淀粉结合结构域(SBM)和一个α-淀粉酶催化结构域的野生型酶(即,非杂交种)或其变体。在一个实施例中,该野生型α-淀粉酶来源于一种白曲霉菌株。
葡糖淀粉酶
优选的葡糖淀粉酶是真菌或细菌来源的,例如,选自由曲霉属葡糖淀粉酶组成的组,特别是黑曲霉、泡盛曲霉、米曲霉
其他的葡糖淀粉酶包括罗氏阿太菌(Athelia rolfsii)(以前命名为罗尔伏革菌(Corticium rolfsii))葡糖淀粉酶、踝节菌属葡糖淀粉酶,特别是来源于爱默生踝节菌、Talaromyces leycettanus、Talaromyces duponti、嗜热踝节菌。
考虑到的细菌葡糖淀粉酶包括来自梭状芽孢杆菌属,特别是嗜热解淀粉梭状芽孢杆菌(C.thermoamylolyticum)和嗜热硫化氢梭状芽孢杆菌(C.thermohydrosulfuricum)以及瓣环栓菌(Trametes cingulata)、纸质厚孢孔菌(Pachykytospora papyracea);以及大白桩菇(Leucopaxillus giganteus)或红边隔孢伏革菌(Peniphora rufomarginata)的葡糖淀粉酶。
纤维素酶
纤维素酶可以包括纤维二糖水解酶(EC 3.2.1.91),例如纤维二糖水解酶I和纤维二糖水解酶II,以及内切葡聚糖酶(EC 3.2.1.4)和β-葡糖苷酶(EC3.2.1.21),特别是内切葡聚糖酶I和/或II(EG-I,EG-II),纤维二糖水解酶I和/或II(CBH-I CBH-II)和/或β-葡糖苷酶。
纤维素酶可以来源于真菌来源,诸如木霉属菌株,优选里氏木霉菌株;腐质霉属菌株,诸如特异腐质酶菌株;或金孢子菌属菌株,优选拉克淖金孢子菌(Chrysosporium lucknowense)的菌株。纤维素酶制剂可以包含一种具有纤维素水解增强活性的多肽(GH61A)。
半纤维素
优选的半纤维素包括木聚糖酶、阿拉伯呋喃糖苷酶、乙酰木聚糖酯酶、阿魏酸酯酶、葡萄糖醛酸酶、内切半乳聚糖酶、甘露聚糖酶、内切或外切阿拉伯糖酶、外切半乳聚糖酶以及β-木糖苷酶。
木聚糖酶可以是微生物来源的,诸如真菌来源的(例如,木霉属、多孔菌属、腐质霉属、曲霉属、镰刀菌属)或来自一种细菌(例如,芽孢杆菌)。在一个优选的实施例中,木聚糖酶来源于丝状真菌,优选地来源于曲霉属菌株,诸如棘孢曲霉;或腐质霉属菌株,优选柔毛腐质霉。
C6-C12脂肪酸或其盐
该C6-C12脂肪酸可以是C6(己酸(hexanoic acid)、己酸(caproic acid))、C8(辛酸(octanoic acid)、辛酸(caprylic acid))或C10(癸酸(decanoic acid)、癸酸(capric acid)),优选C6或C8。还可以使用相应的盐,特别是Na或K盐。
该液体组合物中的C6-C12脂肪酸或盐的含量可以是至少0.01%w/w,特别是至少0.02%w/w、至少0.05%w/w、至少0.1%w/w、至少0.25%w/w或至少0.5%w/w。该含量可以是1.5%w/w以下、1.0%w/w以下或0.75%w/w以下。
可任选的另外的抗微生物剂
可任选地,除C6-C12脂肪酸或盐之外,该液体组合物还可以包含一种或多种抗微生物剂。这可以用来改进乳酸杆菌生长的抑制作用和/或抑制其他微生物的生长。实例是例如0.05%w/w-0.8%w/w的量的甲酸、山梨酸、苯甲酸的Na或K盐以及亚硫酸钠。
杀死或抑制乳酸杆菌的生长
根据本发明,C6-C12脂肪酸或盐被用于在制备、回收、提纯、储存以及运输液体酶组合物过程中杀死或抑制乳酸杆菌的生长。
典型地,通过适合的微生物的培养(发酵),可任选地接着是酶的下游处理(回收和/或提纯)以及配制成一种液体组合物来制备该酶。该酶还可以被用作一种粗制品;例如,该产品可以从发酵液直接获得。
C6-C12脂肪酸或盐可以被用于抑制发酵液下游处理中的乳酸杆菌的生长。发酵过程结束后,使用针对所讨论的酶研发的标准技术可以从该发酵液回收该酶。
从发酵液回收该酶的方法将典型地(但不局限于)包括下列步骤的一步或多步:
1)发酵液的预处理(例如,pH处理和/或絮凝)
2)将细胞和其他固体材料从发酵液中除去(初级分离)
3)过滤
4)浓缩
5)过滤
6)稳定化并且标准化。
除以上所列的单元操作之外,可以应用多个其他的回收流程和步骤,例如温度变化、结晶、用活性碳处理包含感兴趣的化合物的溶液、以及不同吸附剂的使用。
可以将C6-C12脂肪酸或盐加入至这个过程的任何阶段,以便在随后的阶段中抑制乳酸杆菌的生长。
实例
实例1:具有己酸盐的液体葡糖淀粉酶配制品
如下配制根据本发明的两种液体酶组合物:
·酶:来自踝节菌属的葡糖淀粉酶
·酶含量:约19%w/w的酶蛋白
·总固体含量:43%w/w,通过添加葡萄糖
·脂肪酸:0.5%w/w的己酸钠
·pH:5.0
乳酸杆菌的抑制作用
通过用105-106CFU/ml乳酸杆菌(布氏乳酸杆菌(L.buchneri)和副干酪乳酸杆菌(L.para paracasei)接种,20℃-25℃培养两周,并且分析在接种前后以及1周和2周后活细胞(CFU)来进行激发(challenge)筛选。
结果显示为CFU/ml(检出限100):

1周和2周后,没有己酸钠的空白试验(有和没有甲酸钠两者)都显示了增加的CFU。
因此,结果证明该液体酶配制品中的己酸钠具有针对乳酸杆菌的抗微生物(杀菌)作用。
实例2:含有己酸盐的液体木聚糖酶和纤维素酶配制品
如下配制具有两种酶混合物的液体组合物:
·酶:木聚糖酶和纤维素酶
·酶含量:约18%w/w的酶蛋白
·总固体含量:56.5%w/w,通过添加22%w/w的蔗糖
·脂肪酸:0.5%w/w的己酸钠
·另外的抑制剂:0.45%w/w苯甲酸钠+0.13w/w山梨酸钾
·pH:4.6
如实例1中测试该样品。包括一个没有己酸钠的空白,用于比较。结果:

·总固体含量:52%w/w,通过添加25%w/w蔗糖
·脂肪酸:0.5%w/w的己酸钠
·另外的抑制剂:0.4%w/w苯甲酸钠+0.12%山梨酸钾
·pH:4.55
如实例1中测试该样品。结果:

·酶含量:约19%w/w的酶蛋白
·总固体含量:43%w/w,通过添加葡萄糖
·脂肪酸:0.5%w/w的辛酸钠(辛酸钠)
·另外的抑制剂:0.5%的甲酸钠
·pH:pH 5.0
如实例1中测试该样品。包括一个含有0.75%w/w甲酸钠+0.1%w/w山梨酸钾+0.63%w/w丙酸钠的常规配制品,用于比较。结果:

·酶葡糖淀粉酶(两种变体的混合物)
·酶含量:约23%w/w的酶蛋白
·总固体含量:48%w/w,通过添加葡萄糖
·脂肪酸:0.33%w/w辛酸钠(Na-Caprylate)(辛酸钠(Na-octanoate))
·另外的抑制剂:有或没有0.3%w/w三水醋酸钠
·pH:pH 4.7
如实例1中测试该样品。包括含有苯甲酸钠而不是辛酸钠的常规配制品,用于比较。结果:
结果证明将0.3w/w辛酸钠添加至液体酶配制品中足以提供针对乳酸杆菌的强的抗微生物(杀菌)作用。无需添加醋酸盐作为另外的抑制剂。常规抑制剂(苯甲酸盐、醋酸盐)的添加不是有效的。
Claims (12)
1.一种液体组合物,包含至少1%w/w的量的一种酶和对乳酸杆菌具有抗微生物作用的量的一种C6-C12脂肪酸或其盐。
2.如前一项权利要求所述的组合物,其中该脂肪酸或盐是己酸或辛酸或其盐。
3.如前述权利要求中任一项所述的组合物,该组合物具有一个7以下的pH值。
4.如前述权利要求中任一项所述的组合物,该组合物具有一个至少3.5,特别是4-5.5的pH值。
5.如前述权利要求中任一项所述的组合物,该组合物本质上不含表面活性剂。
6.如前述权利要求中任一项所述的组合物,该组合物包含总量至少10%w/w的一种或多种多元醇和/或有机盐。
7.如前述权利要求中任一所述的组合物,该组合物包含至少2%w/w量的该酶。
8.如权利要求1或2所述的组合物,其中该脂肪酸或其盐以至少0.01%w/w的量存在。
9.如权利要求1所述的组合物,该组合物包含至少10%w/w的干物质固体。
10.如前述权利要求中任一项所述的组合物,该组合物包含一种另外的抗微生物剂,特别是一种甲酸盐、苯甲酸盐、山梨酸盐或亚硫酸盐。
11.一种从培养液回收、提纯和/或配制酶的方法,该方法包括将一种C6-C12脂肪酸或其盐以对乳酸杆菌具有抗微生物作用的量的加入至该培养液中。
12.一种C6-C12脂肪酸或其盐在液体酶组合物的回收、提纯、储存或运输过程中杀死或抑制乳酸杆菌生长的用途。
