本专利涉及对A型流感病毒PB基因有干扰作用的siRNA序列的设计及应用,属于生物技术领域。
背景技术
流行性感冒(Influenza),在流行性感冒病毒,是一种造成人类及动物患流行性感冒的RNA病毒,在分类学上,流感病毒属于正黏液病毒科,它会造成急性上呼吸道感染,并借由空气迅速的传播,在世界各地常会有周期性的大流行。目前在全球甲型流感的不同亚型对人类及陆地和水生动物(禽,鸭,鹅)均造成大规模的感染和瘟疫。
流行性感冒是由甲型流感病毒的一种亚型引起的传染性疾病综合征,广泛在人类和动物中传播。在禽类引起的禽流感被国际兽疫局定为A类传染病。禽流感可分为高致病性、低致病性和非致病性三大类。非致病性禽流感不会引起明显症状,仅使染病的禽鸟体内产生病毒抗体。低致病性禽流感可使禽类出现轻度呼吸道症状,食量减少、产蛋量下降,出现零星死亡。高致病性禽流感最为严重,发病率和死亡率高,感染的鸡群常常“全军覆没”。最早的禽流感记录在1887年,意大利发生鸡群大量死亡,当时被称为鸡瘟。到1955年,科学家证实其致病病毒为甲型流感病毒。此后,这种流行性疾病被更名为禽流感。就最近5年的记录,1999年3月至11月,英国伦巴第地区暴发禽流感,到次年3月1300万只病禽被捕杀。2002年10月,美国加州暴发禽流感,当年12月疫情扩散到内华达州和亚利桑那州。到2003年3月,仅加州就销毁326多万只鸡;4月,荷兰发生禽流感,人类感染者达80人,并出现死亡病例。自2003年12月以来,禽流感在亚洲十多个国家和地区肆虐,造成数千万只家禽被宰杀销毁,染病死亡者已达数十人。我国近几年连续发生禽流感疫情。造成禽类大面积死亡,经济损失严重。禽流感被发现100多年来,人类并没有掌握有效的预防和治疗方法,仅能以消毒、隔 离、大量宰杀禽畜的方法防止其蔓延。因此,提高有效的预防和治疗方法和措施是非常重要的。
我国富源辽阔,农业占我国的总产值的70%,养殖业是我国农民的经济来源的重要部分。伴随着禽流行性感冒的发生和造成禽类的大面积死亡,我国的预防疫苗的生产远远不能满足现有的养殖业的市场需要,我国目前使用的禽流感疫苗对于禽病的疫情发作预防和隔离性很小,疫苗效果不好。因此,加大力度进行疫苗及预防性生物制剂的研制,是当前疫情的需要,是发展我国国民经济的需要。
流感病毒[1]属正粘液病毒科(Orthomyxoviridae),分甲、乙、丙三型,多为球形,直径80~120nm,有时呈丝状。三型病毒具有相似的生化和生物学特征。病毒由三层构成,内层为病毒核衣壳,含核蛋白(NP)、P蛋白和病毒RNA。NP是可溶性抗原(S抗原),具有型特异性,抗原性稳定。P蛋白(PA、PB1、PB2)可能是RNA转录和复制所需的多聚酶。流感病毒RNA分节段,甲型和乙型由8个节段、丙型由7个节段构成。中层为病毒包膜,由两层组成,内层为基质蛋白M1,抗原性稳定,也具有型特异性;包膜外层来源于宿主细胞膜的脂质双层膜,M2为嵌于包膜中的膜蛋白,形成膜通道,利于脱壳及HA的产生。外层为两种不同糖蛋白构成的辐射状突起,即血凝素(hemagglutinin,HA)和神经氨酸酶(neuraminidase,NA)。HA能引起红细胞凝集,是病毒吸咐于敏感细胞表面的工具,NA则能水解粘液蛋白,水解细胞表面受体特异性糖蛋白末端的N-乙酰神经氨酸,是病毒复制完成后脱离细胞表面的工具。HA和NA均有变异特性,故只有株特异的抗原性,其抗体具有保护作用。
根据NP和MP的不同,将流感病毒分为甲(A)、乙(B)、丙(C)三型。A型流感病毒按HA和NA抗原不同,可分为若干亚型。目前已发现HA有15种(H1~H15)、NA有9种(N1~N9)抗原,所构成的亚型均可从禽类中分离到。至今人群中有甲型H1~2的组合和N1~2的组合,但已有新出现的H5N1亚型和H9N2亚型禽流感病毒可感染人和动物的报道。因此,研究预防和资料流感的药物具有重要意义。
RNA干扰(RNAinterference,RNAi)既目的RNA与同源的靶目标mRNA结合,阻断病毒翻译蛋白质是解决流感病毒流行的重要手段和方法,其作用原理为:iRNA形成阶段。RNAi的诱导物在细胞质内被RNaseIII Dicer切割成为21-23nt 的小分子干扰RNA,;②RNA诱导的沉默复合物(RNA-induced Silencing Complex,RISC)的形成阶段。siRNA与RNAi特异性酶(如A go-2)结合,形成RISC,具有特异性的核酸内切酶活性能特异性地降解与siRNA同源的mRNA;③效应阶段。RISC中siRNA变性,双链解开,卸下正义RNA,反义RNA仍结合在复合物上,并引导RISC与同源的靶目标mRNA结合,在核酸内切酶的作用下,将靶目标mRNA切断(切割位置在siRNA的中央,距离5′端10-nt),从而阻断了其翻译成蛋白质,表现为基因沉默。

发明内容
本发明四条对A型流感病毒复制有抑制作用的siRNA编码序列。所述的四条对A型流感病毒复制有抑制作用的siRNA的序列分别如序列表400<1>,400<2>,400<3>,400<4>所示。
本发明所述的四条siRNA的动物水平的实验结果表明,对流感病毒H1N1,H9N2,H3N2的病毒复制有抑制作用。
具体实施方式
实施例1:鸡胚实验验证重组质粒干扰H9N2流感病毒增殖作用
H9N2病毒毒力ED50108/0.1ml,将H9N2病毒稀释含有100个ED50病毒液,体积0.1ml,注射到9日龄SPF鸡胚尿囊腔中,同时注射SiRNA载体质粒6微克,33℃培养72小时,取尿囊液,测血凝效价。
实施例2:鸡胚实验验证重组质粒干扰H1N1流感病毒增殖作用
H1N1病毒毒力ED50108.6/0.1ml,将H1N1病毒稀释含有100个ED50病毒液,体积0.1ml,注射到9日龄SPF鸡胚尿囊腔中,同时注射SiRNA载体质粒6微克,33℃培养72小时,取尿囊液,测血凝效价。
实施例3:鸡胚实验验证重组质粒干扰H3N2流感病毒增殖作用
H3N2病毒毒力ED50107.6/0.1ml,将H1N1病毒稀释含有100个ED50病毒液,体积0.1ml,注射到9日龄SPF鸡胚尿囊腔中,同时注射SiRNA载体质粒6微克,33℃培养72小时,取尿囊液,测血凝效价。
实施例4:尿囊液病毒滴度检测
1.微量血凝板的每孔中滴加PH7.2PBS100μL,(2)吸取被检尿囊液样品100μL,倍比稀释12孔,第12孔弃掉100μL。每孔中加入0.1%红细胞悬液100μL。
2.设阴性对照和流感病毒阳性对照。
(4)置微型振荡器上振荡1min混匀。
(5)放室温下(18~20℃)30min,观察结果。
经三次重复实验,序列表所述的四条序列<400>1,400<2>,400<3>,400<4>对

2.权利要求1、对所述的siRNA作为抑制流感病毒PB基因沉默的靶向基因的应用。
