技术领域
本发明属于成分提取技术领域,尤其涉及一种蝴蝶戏珠花的化学成分提取方法。
背景技术
荚蒾属(Viburnum)为忍冬科之大属,全世界大约有230余种,分布于温带和亚热带地区。我国约有108 种(含亚种、变种、变型),广布于全国各地,尤以西南地区最多。该属植物在我国民间具有悠久的药用历史,首见于唐代《新修本草》:“荚蒾,味甘苦平,无毒。主三虫下气、消谷......;煮树枝叶和米作粥甘美,以伺小儿杀蛔......”。此后,历代本草中均有记载,其多种植物的根、茎、叶、果实等均可入药,具有清热解毒、解痉止痛、通经活络、活血利气、养生抗衰等功效。然而,对荚蒾属药用植物的研究报道并不多,大大限制了这一资源丰富的药用植物的开发利用。
蝴蝶戏珠花(Viburnum plicatum Thunb. var. tomentosum Miq.)在我国分布较为广泛,陕西、安徽、浙江、江西等地均有分布,各地也常有栽培。其根及茎入药,具有清热解毒、健脾消积、祛风止痛等功效,主治疮毒、淋巴结炎、小儿疳积、风热感冒、风湿痹痛。
发明内容
本发明就是针对上述问题,提供一种效率高、操作简单的蝴蝶戏珠花的化学成分提取方法。
为了实现上述目的,本发明采用如下技术方案。
(1)取蝴蝶戏珠花干燥茎,切成3~5cm长的小段,用5~15倍量95%乙醇回流提取3次,每次2小时,合并提取液,减压回收得总浸膏。将总浸膏混悬于水中,依次用二氯甲烷、乙酸乙酯、水饱和正丁醇萃取,分别得到二氯甲烷部分浸膏,乙酸乙酯部分浸膏,正丁醇部分浸膏。
(2)二氯甲烷部分浸膏经硅胶柱层析,石油醚-丙酮梯度洗脱,各流分再经硅胶、柱层析、重结晶,分离得到β-谷甾醇、胡萝卜苷。
(3)乙酸乙酯部分浸膏经硅胶柱层析,三氯甲烷-甲醇梯度洗脱,得到绿原酸。
(4)正丁醇部分浸膏经硅胶柱层析,三氯甲烷-甲醇梯度洗脱,得到蔗糖。
作为一种优选方案,本发明所述步骤(1)小段的长度为4cm。
作为另一种优选方案,本发明所述步骤(1)乙醇的倍量为10。
本发明有益效果。
本发明从蝴蝶戏珠花茎中提取4个化合物。酚酸类化合物均具有较为广泛的生物活性,大多具有抗炎、抗氧化、保肝、抗肿瘤、镇静、止痛等活性。其为蝴蝶戏珠花清热解毒、祛风止痛等传统功效的主要物质基础,这将为开发利用蝴蝶戏珠花等荚蒾属这一民间药用植物资源提供科学依据。
具体实施方式
本发明采用如下技术方案。
(1)取蝴蝶戏珠花干燥茎,切成3~5cm长的小段,用5~15倍量95%乙醇回流提取3次,每次2小时,合并提取液,减压回收得总浸膏。将总浸膏混悬于水中,依次用二氯甲烷、乙酸乙酯、水饱和正丁醇萃取,分别得到二氯甲烷部分浸膏,乙酸乙酯部分浸膏,正丁醇部分浸膏。
(2)二氯甲烷部分浸膏经硅胶柱层析,石油醚-丙酮梯度洗脱,各流分再经硅胶、柱层析、重结晶,分离得到β-谷甾醇、胡萝卜苷。
(3)乙酸乙酯部分浸膏经硅胶柱层析,三氯甲烷-甲醇梯度洗脱,得到绿原酸。
(4)正丁醇部分浸膏经硅胶柱层析,三氯甲烷-甲醇梯度洗脱,得到蔗糖。
所述步骤(1)小段的长度为4cm。
所述步骤(1)乙醇的倍量为10。
本发明可采用以下设备。
核磁共振谱用Bruker AV-500 45 型核磁共振仪测定(TMS为内标);ESI-MS 谱用Agilent1100-G1946 MSD 液相色谱-质谱仪测定。高效薄层板及柱层析硅胶(青岛海洋化工厂),Sephadex LH-20 (GE Healthcare 公司)。其它试剂均为分析纯。
可以理解的是,以上关于本发明的具体描述,仅用于说明本发明而并非受限于本发明实施例所描述的技术方案,本领域的普通技术人员应当理解,仍然可以对本发明进行修改或等同替换,以达到相同的技术效果;只要满足使用需要,都在本发明的保护范围之内。

Claims (3)

1.蝴蝶戏珠花的化学成分提取方法,其特征在于包括以下步骤:
(1)取蝴蝶戏珠花干燥茎,切成3~5cm长的小段,用5~15倍量95%乙醇回流提取3次,每次2小时,合并提取液,减压回收得总浸膏;将总浸膏混悬于水中,依次用二氯甲烷、乙酸乙酯、水饱和正丁醇萃取,分别得到二氯甲烷部分浸膏,乙酸乙酯部分浸膏,正丁醇部分浸膏;
(2)二氯甲烷部分浸膏经硅胶柱层析,石油醚-丙酮梯度洗脱,各流分再经硅胶、柱层析、重结晶,分离得到β-谷甾醇、胡萝卜苷;
(3)乙酸乙酯部分浸膏经硅胶柱层析,三氯甲烷-甲醇梯度洗脱,得到绿原酸;
(4)正丁醇部分浸膏经硅胶柱层析,三氯甲烷-甲醇梯度洗脱,得到蔗糖。
2.根据权利要求1所述蝴蝶戏珠花的化学成分提取方法,其特征在于所述步骤(1)小段的长度为4cm。
