本发明涉及化学分离纯化领域,尤其涉及一种层析柱装置。
背景技术
层析柱作为分离纯化的常用设备应用非常普遍,现有技术中,层析柱多为直筒柱状结构,在用洗脱液洗脱时,特别是干法装柱的层析柱,由于力学效应,与层析柱内表面接触的部分填料总是先于中间的填料浸润,导致层析柱色谱带扭曲变宽,降低了分离度。
发明内容
鉴于上述现有技术存在的缺陷,本发明的目的是提出一种层析柱装置,使得干法装柱的填料能均匀渗透,提高分离度。
本发明的目的将通过以下技术方案得以实现:
一种层析柱装置,包括带标准磨砂口的柱颈,柱肩和柱体,所述柱颈直径是所述柱体直径的25%~50%,所述柱肩为倒漏斗状,上窄下宽,连接柱颈和柱体。在装柱时,填料装至领接柱颈处,柱颈内填装待分离样品,在洗脱时即可利用力学原理,使洗脱液带动色谱带扩散,使色带均匀一致。
本发明的突出效果为:
利用力学原理,使得色谱填料浸润均匀,色谱带变窄,变整齐,提高了色谱分离度。

实施例:
本实施例一种层析柱装置,如图1所示,包括带标准磨砂口的柱颈1,柱肩2和柱体3,柱颈1直径是柱体3直径的25%~50%,柱肩2为倒漏斗状,上窄下宽,连接柱颈1和柱体3。在装柱时,填料装至领接柱颈1处,柱颈1内填装待分离样品,在洗脱时即可利用力学原理,使洗脱液带动色谱带扩散,使色带均匀一致。
本实施例利用力学原理,使得色谱填料浸润均匀,色谱带变窄,变整齐,提高了色谱分离度。
本发明尚有多种实施方式,凡采用等同变换或者等效变换而形成的所有技术方案,均落在本发明的保护范围之内。
