Abstract

发明领域
本发明涉及聚醚共聚物和微水溶性活性物质与能影响配制剂稳定性和/或生物活性物质释放的其它聚合物组合的固体药物制剂,所述聚醚共聚物通过乙酸乙烯酯和N-乙烯基内酰胺在聚醚的存在下聚合而得到。此外,本发明涉及用于制备这些制剂的方法及其用途。
相应的聚醚共聚物用作微水溶性生物活性物质的增溶剂。
在均质制剂,特别是生物活性物质的制备中,疏水性,即微水溶性物质的增溶在实践中变得非常重要。
增溶应当理解意指通过表面活性化合物,增溶剂赋予微溶或不溶于某些溶剂,特别是水中的物质以溶解性。这类增溶剂能将弱水溶或水不溶性物质转化成透明,至多乳白色的水溶液而不使这些物质的化学结构经受因此的改变(参见Chemie Lexikon,第9版,第5卷,第4203页,Thieme Verlag,Stuttgart,1992)。
在所制备的增溶物中,弱水溶或水不溶性物质作为胶状溶液存在于形成于水溶液中的表面活性化合物的分子联系如疏水性域或胶束中。所得溶液为显示出光学透明至乳白色的稳定或亚稳定单相体系。
在药物制剂的情况下,药物的生物药效率和因此功效可通过使用增溶剂而提高。
关于增溶剂的另一理想要求是用微溶性物质形成所谓“固溶体”的能力。术语“固溶体”指物质以胶状形式分散或理想地以分子形式分散在固体基体如聚合物基体中的一种状态。这类固溶体,例如当用于微溶性活性物质的固体药物给药形式中时导致改善的活性物质释放。关于所述固溶体的重要要求是它们甚至在相对长时间储存时为稳定的,即活性物质不结晶。此外,固溶体的能力,换言之形成具有尽可能高的活性物质含量的稳定固溶体的能力也是重要的。
在固溶体的形成中,除增溶剂形成固溶体的基本能力外,增溶剂的吸湿性也起到重要作用。由周围空气中吸收太多水的增溶剂导致固溶体的潮解和不理想的活性物质结晶。过高的吸湿性还可导致加工以得到给药形式期间的问题。
许多已知的聚合增溶剂的缺点是它们不形成稳定的固溶体,特别是当活性物质以高于饱和浓度存在于聚合物中时。因此,动态地控制该体系且活性物质在储存过程中结晶。这代表主要问题。
另外,已知的增溶剂和配制剂仍留有改进的空间,这涉及含水体系,特别生物体系中的增溶。口服生物药效率通常未能提高至理想的程度以无副作用地实现均衡的再现作用。
关于加工性,一些已知的增溶剂也由于它们倾向于变粘而具有缺点,因为它们不是充分可流动的粉末。
在微水溶性活性物质的药物制剂的情况下,另一问题是释放的控制。通常,在这类形式下控制释放的不是赋形剂,而是活性物质的晶体性能。这意指活性物质粒度的变化、晶体改性的差别和粒子形状的差别对溶解具有相当的影响。由于这些参数难以精确地建立,产生不同的释放率。在水溶性活性物质的情况下,不存在该问题,因为活性物质的溶解过程比通过延缓聚合物控制的扩散过程相当地更快。
关于释放,速释(快速释放)型、肠溶释放(耐胃酸)型和缓释(延时、缓释)型之间应存在区别。在1小时以后释放至少75%的剂型成为速释型。在肠溶释放型的情况下,在胃液中进行轻微释放(通常<10%),但在肠液中快速释放。缓释型在胃液和肠液中均具有缓慢的释放。当在3小时以后释放慢于75%时使用术语缓释型。
微水溶性聚合物应当理解意指在整个pH范围,或至少在某一pH范围内微溶的聚合物。这些包括所谓的缓释聚合物(在pH1-14下通常不溶)、酸性耐胃酸聚合物(在酸性pH范围内微溶)和碱性逆转肠溶聚合物(在中性至碱性pH范围内微溶)。进一步参见下文关于溶解度的定义。
EP-A 876 819描述了至少60重量%N-乙烯基吡咯烷酮和具有长链烷基的酰胺或酯的共聚物的用途。
EP-A 948 957描述了单烯属不饱和羧酸如丙烯酸,和疏水改性共聚单体如具有C8-C30烷基的不饱和羧酸的N-烷基-或N,N-二烷基酰胺的共聚物的用途。
DE-A 199 350 63公开了基于乙烯基内酰胺和乙酸乙烯酯的含聚氧化烯接枝聚合物及其作为气体水合物抑制剂的用途。
EP-A 953 347公开了含聚氧化烯接枝聚合物作为增溶剂的用途。此处所述且包含乙酸乙烯酯和聚氧化烯的接枝聚合物通常不是粉末,而是粘性粘液,这是使用中的缺点。
WO 2007/051743公开了N-乙烯基内酰胺、乙酸乙烯酯和聚醚的水溶性或水分散性共聚物在药物、化妆品、食品、农业技术和其它技术应用中作为增溶剂的用途。其中非常一般地规定相应的接枝聚合物还可以与活性物质一起以熔体加工。
WO 2009/013202公开了可将N-乙烯基内酰胺、乙酸乙烯酯和聚醚的这种接枝聚合物在挤出机中熔融并与粉状或液体活性物质混合,描述了基本在活性物质熔点以下的温度下挤出。
然而,令人满意的高且同时稳定的活性物质负载不可通过将熔融接枝聚合物与粉状或液体活性物质混合而实现。特别地,活性物质的稳定X射线无定形状态的实现不总是达到令人满意的程度。
本发明的目的是提供微水溶性活性物质的改进制剂,所述制剂容许有目标的调整释放以及良好的生物药效率。
因此,发现包含在聚醚共聚物和至少一种微水溶性聚合物的聚合物基体中的微水溶性活性物质的制剂的剂型,所述聚醚共聚物通过30-80重量%N-乙烯基内酰胺、10-50重量%乙酸乙烯酯和10-50重量%聚醚的混合物自由基聚合得到,其中微水溶性活性物质以无定形形式存在于聚合物基体中。
此外,发现一种制备微水溶性活性物质的制剂的方法,其中活性物质以无定形形式嵌入基于30-80重量%N-乙烯基内酰胺、10-50重量%乙酸乙烯酯和10-50重量%聚醚的聚醚共聚物的聚合物基体中,其中除聚醚共聚物外,将至少一种微水溶性聚合物掺入聚合物基体中并将聚合物与微水溶性活性物质彻底混合。
存在于聚合物基体中的聚醚共聚物通过以下组分的混合物自由基聚合得到:
i)30-80重量%N-乙烯基内酰胺,
ii)10-50重量%乙酸乙烯酯,和
iii)10-50重量%聚醚。
条件是i)、ii)和iii)之和等于100重量%。
根据一个工艺变化方案,将聚醚共聚物与微水溶性聚合物和微水溶性活性物质彻底混合并将混合物加热至共聚物的玻璃化转变温度以上。
根据另一工艺变化方案,在有机溶液中制备聚合物和活性物质的混合物,然后干燥。
聚醚共聚物自由地溶于水中,这意指在20℃下,1份共聚物溶于1-10份水中。
根据本发明一个实施方案,使用由如下组分得到的优选的聚醚共聚物:
i)30-70重量%N-乙烯基内酰胺,
ii)15-35重量%乙酸乙烯酯,和
iii)10-35重量%聚醚。
特别优选使用的聚醚共聚物可由如下组分得到:
i)40-60重量%N-乙烯基内酰胺,
ii)15-35重量%乙酸乙烯酯,和
iii)10-30重量%聚醚。
非常特别优选使用的聚醚共聚物可由如下组分得到:
i)50-60重量%N-乙烯基内酰胺,
ii)25-35重量%乙酸乙烯酯,和
iii)10-20重量%聚醚。
组分i)、ii)和iii)之和等于100重量%的条件也适用于优选和特别优选的组合物。
合适的N-乙烯基内酰胺为N-乙烯基己内酰胺或N-乙烯基吡咯烷酮或其混合物。优选使用N-乙烯基己内酰胺。
聚醚用作接枝基础。优选的聚醚为聚亚烷基二醇。聚亚烷基二醇的分子量可以为1000-100000D[道尔顿],优选1500-35000D,特别优选1500-10000D。分子量起始于根据DIN 53240测得的OH值测定。
特别优选的聚亚烷基二醇为聚乙二醇。此外,聚丙二醇、聚四氢呋喃或可由2-乙基环氧乙烷或2,3-二甲基环氧乙烷得到的聚丁二醇也是合适的。
其它合适的聚醚为由氧化乙烯、氧化丙烯和氧化丁烯得到的聚亚烷基二醇的无规或嵌段共聚物,例如聚乙二醇-聚丙二醇嵌段共聚物。嵌段共聚物可以为AB型或ABA型。
优选的聚亚烷基二醇还包括在一个末端OH基团上或在两个末端OH基团上烷基化的那些。合适的烷基为支化或直链C1-C22烷基,优选C1-C18烷基,例如甲基、乙基、正丁基、异丁基、戊基、己基、辛基、壬基、癸基、十二烷基、十三烷基或十八烷基。
用于制备本发明聚醚共聚物的一般方法本身是已知的。制备通过自由基聚合,优选在溶液中、在非水有机溶剂中或在混合非水/含水溶剂中进行。合适的制备方法例如描述于WO 2007/051743和WO 2009/013202中,通过引用将其关于制备方法的公开内容结合到本文中。
就释放的目标控制而言,具有增溶效果的共聚物与适于影响生物活性物质释放的微水溶性聚合物组合使用。影响释放的程度在这里通常取决于微水溶性聚合物的浓度。
聚醚共聚物与微水溶性聚合物之比可以为99∶1-10∶90。优选它为90∶10-30∶70,特别优选80∶20-40∶60。
关于聚合物的溶解性,术语“微水溶”应当理解如下:根据本发明术语“微水溶”包含微水溶以及实际上不溶的物质,且意指对于在20℃下聚合物在水中的溶液,每g聚合物需要至少100-1000g水。在实际上不溶物质的情况下,每g物质需要至少10000g水。
在微水溶性聚合物的以下描述中,术语“微水溶”简写为“微溶”。
微水溶性聚合物
在本发明上下文中,微水溶性聚合物为中性微溶性聚合物(缓释聚合物)、阴离子微溶性聚合物(耐胃酸聚合物)或碱性微溶性聚合物。
中性微溶性聚合物
微溶性聚合物应当理解意指为在1-14的总pH范围内微溶于水或在水中仅溶胀的那些聚合物。通常,药物组合物仅包含一种水不溶性聚合物。然而,两种或多种水不溶性聚合物也可随着彼此或作为混合物存在。
合适微溶性聚合物的实例为:
中性或基本中性的甲基丙烯酸酯共聚物。这些可特别包含至少95重量%,特别是至少98重量%,优选至少99重量%,尤其是至少99重量%,特别优选100重量%经受自由基聚合且具有中性基团,特别是C1-C4烷基的(甲基)丙烯酸酯单体。
合适的具有中性基团的(甲基)丙烯酸酯单体例如为甲基丙烯酸甲酯、甲基丙烯酸乙酯、甲基丙烯酸丁酯、丙烯酸甲酯、丙烯酸乙酯、丙烯酸丁酯。甲基丙烯酸甲酯、丙烯酸乙酯和丙烯酸甲酯是优选的。具有阴离子基团的甲基丙烯酸酯单体如丙烯酸和/或甲基丙烯酸可以以小于5重量%,优选不大于2重量%,特别优选不大于1重量%或0.05-1重量%的小含量存在。
例如20-40重量%丙烯酸乙酯、60-80重量%甲基丙烯酸甲酯和0至小于5重量%,优选0-2重量%或0.05-1重量%的中性或基本中性的(甲基)丙烯酸甲酯共聚物(NE型)是合适的。Eudragit NE为30重量%丙烯酸乙酯和70重量%甲基丙烯酸甲酯的共聚物。
其它合适的微溶性(甲基)丙烯酸酯共聚物例如为不依赖pH而溶解或溶胀且适于药物涂层的聚合物。
微溶性聚合物可以为98-85重量%丙烯酸或甲基丙烯酸C1-C4烷基酯和2-15重量%具有季铵基团的(甲基)丙烯酸酯单体的聚合物或多种这类物质的聚合物的混合物。
优选的丙烯酸或甲基丙烯酸C1-C4烷基酯为丙烯酸甲酯、丙烯酸乙酯、丙烯酸丁酯、甲基丙烯酸丁酯和甲基丙烯酸甲酯。
甲基丙烯酸2-三甲基铵乙基酯氯化物特别优选作为具有季氨基的(甲基)丙烯酸酯单体。合适的共聚物例如包含65重量%甲基丙烯酸甲酯、30重量%丙烯酸乙酯和5重量%甲基丙烯酸2-三甲基铵乙基酯氯化物(Eudragit RS)。
微溶性聚合物可以为93-88重量%丙烯酸或甲基丙烯酸C1-C4烷基酯和7-12重量%具有季铵基团的(甲基)丙烯酸酯单体的聚合物(Eudragit RL型)。
水不溶性聚合物可以为比值为20∶1-1∶20的Eudragit RS型和EudragitRL型聚合物的混合物。
比值为20∶1-1∶20重量份的Eudragit RS和Eudragit RL的混合物也特别合适。
药物组合物还可包含聚乙酸乙烯酯作为微溶性聚合物。合适的聚乙酸乙烯酯例如为乙酸乙烯酯的均聚物。此外,微溶性聚乙酸乙烯酯共聚物是合适的,例如乙酸乙烯酯和N-乙烯基吡咯烷酮的水不溶性共聚物。市售的合适聚乙酸乙烯酯例如为SR 30D或SR。
其它合适的微溶性聚合物为烷基纤维素如乙基纤维素。
阴离子微溶性聚合物
此外,也可使用阴离子微溶性聚合物。阴离子聚合物应当理解优选意指具有至少5%,特别优选5-75%具有阴离子基团的单体基团的聚合物。阴离子(甲基)丙烯酸酯共聚物是优选的。合适的市售具有阴离子基团的(甲基)丙烯酸酯共聚物例如为型L、L100-55、S和FS。
合适的阴离子(甲基)丙烯酸酯共聚物例如为25-95重量%丙烯酸或甲基丙烯酸C1-C4烷基酯和5-75重量%具有阴离子基团的(甲基)丙烯酸酯单体的聚合物。取决于阴离子基团的含量和其它单体在大于pH5.0的pH下的性质,合适聚合物为水溶性的,因此还可溶于肠液中。通常,所述比例之和为100重量%。
具有阴离子基团的(甲基)丙烯酸酯单体可例如为丙烯酸,但优选为甲基丙烯酸。
此外,40-60重量%甲基丙烯酸和60-40重量%甲基丙烯酸酯或60-40重量%丙烯酸乙酯的阴离子(甲基)丙烯酸酯共聚物是合适的(Eudragit L或Eudragit L1 00-55型)。
Eudragit L为50重量%甲基丙烯酸酯和50重量%甲基丙烯酸的共聚物。
Eudragit L1 00-55为50重量%丙烯酸乙酯和50重量%甲基丙烯酸的共聚物。Eudragit L 30D-55为包含30重量%Eudragit L 100-55的分散体。
合适的还有20-40重量%甲基丙烯酸和80-60重量%甲基丙烯酸甲酯的阴离子(甲基)丙烯酸酯共聚物(S型)。
共聚物优选基本至仅包含上述比例的单体甲基丙烯酸、丙烯酸甲酯和丙烯酸乙酯。
然而,可任选存在0-10重量%,例如1-5重量%的少量其它烯属可共聚单体如甲基丙烯酸甲酯、甲基丙烯酸丁酯、丙烯酸丁酯或甲基丙烯酸羟乙酯,同时这不导致基本性能的削弱。
共聚物可通过常规连续或分批自由基本体、溶液、成珠或乳液聚合方法在自由基引发剂和如果合适的话用来调整分子量的链转移剂存在下制备。平均分子量Mw(重均,例如通过测量溶液粘度测定)可例如为80000-1000000(克/摩尔)。在水溶引发剂和(优选阴离子)乳化剂的存在下在水相中乳液聚合是优选的。在本体聚合的情况下,可将共聚物以固体形式通过压碎、挤出、粒化或热端面车削加工。
适用作阴离子聚合物的还有乙酸羟丙基甲基纤维素琥珀酸酯,一种在碱性条件下可溶的耐胃酸聚合物。
碱性微溶性聚合物
固体制剂可通过本身已知的方法制备。
根据一个实施方案,首先将制剂的所有成分一起在合适溶剂中产生溶液,然后除去溶剂。可使用的溶剂为常用于药物技术中的所有溶剂,例如乙醇、异丙醇、正丁醇、异丁醇、乙酸乙酯、丙酮或二甲基甲酰胺。溶剂的除去可通过所有可能类型的干燥方法,例如通过喷雾干燥、流化床干燥、转鼓式干燥、使用超临界气体干燥、冷冻干燥、蒸发而进行。
根据优选程序,固体制剂通过挤出制备。
可将聚合物以粉状形式和以溶液或分散体形式供入挤出机中。
可将聚合物的分散体或溶液通过除去挤出机中的分散介质或溶剂而转化成熔融态,和通过熔体冷却转化成固体形式。
然后可将因此所得熔体冷却和粒化。为此,进行在空气或惰性气体下,例如在特氟隆皮带或链带上进行所谓的热端面车削或冷却,随后将冷却的熔体条粒化。然而,冷却也可在聚合物明显不溶于其中的溶剂中进行。
原则上使用以下方法A-E:
原则上,本领域技术人员已知的常规挤出机类型适于本发明方法。这些通常包含外壳、具有传动的驱动单元和由一个或多个装配有螺杆元件的挤出机轴组成的加工单元,在这种情况下呈现出标准设计。
挤出机由多个段组成,所述段每种情况下与某些加工单元配对。这些段各自由一个或多个桶(桶块)作为最小的独立单元和具有对应于方法目的的螺杆元件的相关螺杆段组成。
各个桶应为可加热的。此外,桶还可设计用于冷却,例如用水冷却。各个桶块优选为相互独立地可加热和可冷却的,使得沿着挤出方向还可建立不同的温度区域。
挤出机有利地为具有共轴旋转螺杆的双螺杆挤出机。螺杆构型可取决于产物提供不同的剪切度。捏合元件必须特别用于熔融区中。还可使用相反的捏合元件。
合适双螺杆挤出机可具有16-70mm的螺杆直径和25-40D的长度。
总挤出机由可单独自动调温的桶块组成。最初两个桶可就更好的材料进入而自动调温。自第三桶向前,优选建立恒温,选择所述恒温以便为材料特异性的并特别取决于所用活性物质的熔点和聚合物的玻璃化转变温度。然而,所得产物温度通常取决于所用螺杆元件的剪切度且在一些情况下可以比设置的桶温度高20-30℃。
有利地在环境压力下操作的液化区可存在于熔融区下游。
所用圆模可具有0.5-5mm的直径。同样可使用其它压模形状如狭缝压模,尤其是当需要较大的材料生产率时。
设计两个共轴旋转螺杆使得在由输送元件组成的进料区下游,具有下游流量限制器的捏合块已用于第三加热区中。在包含输送元件的短减压区之后,将现熔融的材料在捏合区中再次彻底混合。这之后是具有下游捏合元件的输送元件区。具有下游捏合区的输送元件区随后。最后通过输送元件区确保材料的排出。
可将所得挤出物通过制粒机加工以得到团粒,又可将这些进一步粉碎(研磨)以得到粉末。可随着常规制片助剂的使用将团粒或粉末引入胶囊中或压制以得到片剂。在这种情况下,也可使用控制释放的其它赋形剂。
此外,在挤出期间可使用水、有机溶剂、缓冲物质或增塑剂。特别地,水或挥发性醇适于该目的。该方法容许在相对低温下反应。溶剂或增塑剂的量通常为可挤出材料的0-30%。可将水或溶剂在大气压力下或通过施加降低的压力通过挤出机中的液化点除去。作为选择,当挤出物离开挤出机且压力降至大气压力时,这些组分蒸发。在略挥发的组分的情况下,可随后将挤出物以合适方式干燥。
在制备方法的特别变化方案中,可在挤出以后直接将热塑性材料砑光,以得到类片剂压块,其构成最终的给药形式。在该变化方案中,可有利地在挤出以前或期间加入其它组分,例如用于调整玻璃化转变温度和熔体粘度的聚合物、崩解剂、增溶剂、增塑剂、染料、香料、增甜剂等。原则上,当首先将挤出物粉碎,然后压制以得到片剂时也可使用这些物质。
具有高玻璃化转变温度的水溶性聚合物如K值为17-120的聚乙烯吡咯烷酮、羟烷基纤维素或羟烷基淀粉可用于调整配制剂的玻璃化转变温度。配制剂的过高玻璃化转变温度可通过加入增塑剂而降低。也用于药物涂层的所有增塑剂如柠檬酸三乙酯、柠檬酸三丁酯、柠檬酸乙酰三丁酯、醋精、丙二醇、聚乙二醇400、癸二酸二丁酯、单硬脂酸甘油酯、月桂酸、鲸蜡基硬脂醇原则上适于该目的。
此外,可另外将降低熔体粘度和因此挤出温度的表面活性剂掺入制剂中。这些物质也可正面影响可能的结晶并产生更好的配制剂润湿和更快的溶解。合适的物质为离子和非离子表面活性剂,例如HS 15(聚乙二醇硬脂酸酯15)、80、聚氧乙基化脂肪酸衍生物如RH40(聚氧乙烯醚(40)氢化蓖麻油,USP)、Cremophor EL(聚氧乙烯醚(35)蓖麻油,USP)、泊洛沙姆、多库酯钠或月桂基硫酸钠。
优选通过压模将仍塑性的混合物挤出,冷却并粉碎。原则上,用于该目的的所有已知常规技术如热端面车削或冷端面切削适于粉碎。
将挤出物例如通过转动叶片或通过空气喷射端面车削,然后用空气或在惰性气体下冷却。
也可将挤出物作为熔体带放在冷却皮带(不锈钢、特氟隆、链带)上并在固化以后将它粒化。
然后可任选将挤出物研磨。制剂作为自由流动和可流动水溶性粉末得到。优选建立20-250μm的粒度。
此外,也可通过注塑加工包含聚合物和活性物质的塑性混合物。
令人惊讶的是,本发明配制剂具有相当改善的稳定性,即活性物质以分子分散或胶状分散无定形状态保留在配制剂中且不结晶。因此,释放性能也不随时间过去而改变。如果挤出物的粒度为300μm以上,则证明微水溶性聚合物对释放的影响。在这些情况下,这意指释放取决于添加是缓释或耐胃酸的。然而,如果将挤出物粉碎使得粒度为100μm以下,则在耐胃酸或缓释聚合物的使用中令人惊讶地实现快速释放。
本发明制剂具有较高的活性物质生物药效率。
通过本发明方法得到的制剂原则上可用于所有其中仅微水溶性或不溶性物质用于含水制剂中或在含水介质中显示出其效果的领域。
在本发明上下文中,微水溶性物质优选应当理解意指生物活性物质,例如用于人类和动物的药物活性物质、化妆品或农用化学活性物质或食品增补剂或饮食活性物质。
此外,染料如无机或有机颜料也适用作待溶解的微溶性物质。
挤出温度通常为50-260℃,优选70-200℃。温度下限取决于每种情况下待挤出混合物的组成和待加工的微溶性物质。至少一部分混合物必须可在所选择的温度下塑化。温度上限由活性物质或聚合物的分解限定。
所用药物活性物质为根据DAB 9中的定义不溶或略溶于水的物质。
根据DAB 9(German Pharmacopeia),物质的溶解性分类如下:略溶(溶于30-100份溶剂中);微溶(溶于100-1000份溶剂中);几乎不溶(溶于大于10000份溶剂中),每种情况下基于在20℃下一份待溶解物质。
活性物质可源自任何指示领域。
这里作为实例可提到苯并二氮杂、抗高血压药、维生素、细胞抑制剂-特别是紫杉醇、麻醉药、神经安定药、抗抑郁药、抗病毒剂如抗HIV剂、抗生素、抗真菌剂、抗老年痴呆药、杀真菌剂、化疗药物、泌尿系统用药、血小板凝集抑制剂、磺酰胺类、解痉药、激素、免疫球蛋白、血清、甲状腺治疗药、精神药物、抗帕金森药和其它抗运动机能亢进药、眼科用药、神经病用制剂、钙代谢调节剂、肌肉松弛药、类脂降低药、肝治疗药、冠状动脉药、心脏药、免疫治疗药、调节性肽及其抑制剂、催眠药、镇静药、妇科用药、抗痛风药、纤维蛋白溶解药、酶制剂和运输蛋白、酶抑制剂、催吐药、血流刺激药、利尿药、诊断助剂、皮质激素类、胆碱药、胆用治疗药、平喘药、支气管解痉药、β受体阻断剂、钙拮抗剂、ACE抑制剂、抗动脉硬化药、抗炎药、抗凝血药、抗低血压药、抗低血糖药、抗高血压药、抗纤维蛋白溶解药、抗癫痫药、抗呕吐药、解毒药、抗糖尿病药、抗心律不齐药、抗贫血药、抗过敏药、驱肠虫药、止痛药、兴奋剂、醛固酮拮抗药、减肥药。
在上述药物制剂中,可口服给药的配制剂的那些是特别优选的。
药物制剂中聚醚共聚物和微溶性聚合物的混合物的含量取决于活性物质为1-99重量%,优选5-80重量%,特别优选10-70重量%。
对于药物给药形式的制剂如片剂,可将常规药物赋形剂加入挤出物中。
这些为选自填料、增塑剂、增溶剂、粘合剂、硅酸盐和崩解剂和吸附剂、润滑剂、流动改进剂、染料、稳定剂如抗氧化剂、湿润剂、防腐剂、脱模剂、芳香剂或增甜剂的物质,优选填料、增塑剂和增溶剂。
可加入的填料例如为无机填料如镁、铝、硅的氧化物、碳酸钛或碳酸钙、磷酸钙或磷酸镁,或有机填料如乳糖、蔗糖、山梨醇、甘露醇。
合适的增塑剂例如为醋精、柠檬酸三乙酯、单硬脂酸甘油酯、低分子量聚乙二醇或泊洛沙姆。
HLB值(亲水-亲脂平衡)为大于11的表面活性物质例如用40个氧化乙烯单元乙氧基化的氢化蓖麻油(RH 40)、用35个氧化乙烯单元乙氧基化的蓖麻油(Cremophor EL)、聚山梨醇酯80、泊洛沙姆、多库酯钠或月桂基硫酸钠适用作另外的增溶剂。
铝、钙、镁和锡的硬脂酸盐和硅酸镁、聚硅氧烷等可用作润滑剂。
例如,滑石或胶状二氧化硅可用作流动改进剂。
例如,微晶纤维素适用作粘合剂。
为另外调整延时释放,还可将其它微水溶性聚合物加入制片混合物中。
崩解剂可以为交联聚乙烯吡咯烷酮或交联羧甲基淀粉钠。稳定剂可以为抗坏血酸或生育酚。
染料例如就将给药形式着色而言,为铁氧化物、二氧化钛、三苯基甲烷染料、偶氮染料、喹啉染料、靛青染料、类胡萝卜素,就提高透光率和降低染料的使用而言,乳浊剂如二氧化钛或滑石。
除用于化妆品和药物中外,根据本发明制备的制剂还适用于食品领域,例如用于微水溶性或水不溶性营养、助剂或添加剂如脂溶性维生素或类胡萝卜素的掺入。可作为实例提到用类胡萝卜素着色的饮料。
根据本发明得到的制剂在农用化学中的用途可尤其包括包含农药、除草剂、杀真菌剂或杀虫剂的配制剂,尤其是待用作用于喷雾或洒水的配制剂的作物保护剂的那些制剂。
微溶物质的所谓固溶体可借助本发明方法得到。根据本发明,其中观察不到微溶物质的结晶部分的体系称为固溶体。
当视觉评估稳定的固溶体时,证明没有无定形组分。视觉评估可使用光学显微镜,用或不用滤波器地在40放大倍数下进行。
此外,还可借助XRD(X射线衍射法)和DSC(差示扫描量热法)研究制剂的结晶度或无定形性能。
如所述,通过本发明方法得到的制剂以无定形形式存在,这意指生物活性物质的结晶部分为小于5重量%。优选可通过DSC或XRD检查无定形状态。这种无定形状态也可称为X射线无定形状态。
本发明方法容许制备具有高活性物质负载和在微溶性物质的无定形状态方面良好稳定性的稳定制剂。

实施例
聚合物1的制备
在搅拌设备中,将最初取得的混合物在N2气氛下加热至77℃而无一部分进料2。当达到77℃的内部温度时,加入一部分进料2并进行预聚合15分钟。其后,在5小时内计量加入进料1,并在2小时内计量加入进料2。在已计量加入所有进料以后,随后使反应混合物聚合另外3小时。在随后的聚合后,将溶液调整至50重量%的固体含量。
最初取得的混合物:25g乙酸乙烯酯
104.0g PEG 6000
1.0g进料2
进料1:240g乙酸乙烯酯
进料2:456g乙烯基己内酰胺
240g乙酸乙酯
进料3:10.44g过氧化新戊酸叔丁酯(在脂族化合物混合物中75重量%浓度)
67.90g乙酸乙酯。
其后,通过喷雾方法除去溶剂并得到粉状产物。如作为1重量%浓度乙醇溶液测量,K值为16。
用于制备以下实施例所述配制剂的双螺杆挤出机具有16mm的螺杆直径和40D的长度。整个挤出机由8个独立的可自动调温的桶块组成。为更好地材料引入,最初两个桶分别自动调温在20℃和70℃下。自第三个桶向前,建立恒温。
使用以下设备和条件通过XRD(X射线衍射法)和DSC(差示扫描量热法)研究所制备的固溶体的结晶度和无定形性能:

测量方法:θ-θ反射几何

取得重量约8.5mg
加热速率:20K/min
活性物质释放根据USP设备(划桨法2),37℃,50rpm(BTWS 600,Pharmatest)进行。通过制粒机将挤出物粉碎至3mm的长度并引入硬明胶胶囊中。所释放活性物质的检测通过UV光谱(Lamda-2,Perkin Elmer)进行。
实施例1
将1200g聚合物1、400g Eudragit E PO和400g塞来昔布(Celecoxib)(熔点162℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第三桶向前的区域温度:140℃
·螺杆速度200rpm
·生产率:500克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在0.1标称HCl中1小时以后,释放出95%活性物质。
在30℃/70%相对湿度下储存6个月以后,制剂仍为无定形的。
实施例2
将800g聚合物1、800g Eudragit E PO和400g Naproxen(熔点157℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第三桶向前的区域温度:120℃
·螺杆速度200rpm
·生产率:600克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在0.1标称HCl中1小时以后,释放出89%活性物质。
实施例3
将1200g聚合物1、400g Eudragit E PO、40g月桂基硫酸钠和400g伊曲康唑(itraconazole)(熔点166℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第三桶的区域温度:140℃
·螺杆速度200rpm
·生产率:800克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在0.1标称HCl中1小时以后,释放出99%活性物质。
在30℃/70%相对湿度下储存6个月以后,制剂仍为无定形的。
实施例4
将600g聚合物1、1000g Kollidon SR和400g非诺贝特(fenofibrate)(熔点81℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第三桶的区域温度:110℃
·螺杆速度200rpm
·生产率:1000克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在0.1标称HCl中2小时以后,活性物质的释放为小于20%。在再缓冲至pH6.8以后,这是另外10小时以后,其后一起释放出80%活性物质。
在30℃/70%相对湿度下储存6个月以后,制剂仍为无定形的。
实施例5
将600g聚合物1、1000g Eudragit S 100和400g桂利嗪(cinnarizine)(熔点122℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。将柠檬酸三乙酯借助往复式活塞泵供入挤出机中。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第三桶的区域温度:130℃
·螺杆速度100rpm
·生产率:800克/小时
·液体计量:80克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在0.1标称HCl中2小时以后,活性物质的释放为小于10%;在再缓冲至pH6.8以后,释放出100%。
在30℃/70%相对湿度下储存6个月以后,制剂仍为无定形的。
实施例6
将400g聚合物1、1200g Eudragit E PO和400g酰胺咪嗪(carbamazepine)(熔点192℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第3桶的区域温度:160℃
·螺杆速度200rpm
·生产率:600克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在0.1标称HCl中1小时以后,释放出95%活性物质。
在30℃/70%相对湿度下储存6个月以后,制剂仍为无定形的。
实施例7
将600g聚合物1、1000g Eudragit L 100-55和400g洛哌丁胺(loperamide)(熔点223℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第3桶的区域温度:170℃
·螺杆速度200rpm
·生产率:1000克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在0.1标称HCl中2小时以后,活性物质的释放为小于10%;在再缓冲至pH6.8以后,释放出98%。
在30℃/70%相对湿度下储存6个月以后,制剂仍为无定形的。
实施例8
将600g聚合物1、500g Eudragit RS PO、500gEudragit RL PO和400g克霉唑(clotrimazole)(熔点148℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第3桶的区域温度:150℃
·螺杆速度100rpm
·生产率:700克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在pH6.8的磷酸盐缓冲液中2小时以后,活性物质的释放为20%;在10小时以后,释放出84%的起初所用活性物质。
在30℃下储存6个月以后,制剂仍为无定形的。
实施例9
将600g聚合物1、1000g HPMCAS(乙酸羟丙基甲基纤维素琥珀酸酯)和400g桂利嗪(熔点122℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第3桶的区域温度:140℃
·螺杆速度100rpm
·生产率:800克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在0.1标称HCl中2小时以后,活性物质的释放为小于10%;在再缓冲至pH6.8以后,释放出90%。

实施例10
将600g聚合物1、500g Eudragit RS PO、500g Eudragit RL PO和400g吡罗昔康(piroxicam)(熔点199℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第3桶的区域温度:170℃
·螺杆速度100rpm
·生产率:700克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在pH6.8的磷酸盐缓冲液中2小时以后,活性物质的释放为20%;在10小时以后,释放出93%的起初所用活性物质。
在30℃/70%相对湿度下储存6个月以后,制剂仍为无定形的。
实施例11
将600g聚合物1、1000g乙基纤维素和400g非洛地平(felodipine)(熔点145℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。将Eudragit NE 40D借助往复式活塞泵供入挤出机中。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第3桶的区域温度:140℃
·螺杆速度100rpm
·生产率:800克/小时
·液体计量:60克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在pH6.8的磷酸盐缓冲液中2小时以后,活性物质的释放为31%;在10小时以后,释放出79%的起初所用活性物质。
在30℃/70%相对湿度下储存6个月以后,制剂仍为无定形的。
实施例12
将600g聚合物1、1000g Eudragit RS PO和400g伊曲康唑(熔点166℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第3桶的区域温度:150℃
·螺杆速度100rpm
·生产率:800克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在pH6.8的磷酸盐缓冲液中2小时以后,活性物质的释放为27%;在10小时以后,释放出82%的起初所用活性物质。
在30℃/70%相对湿度下储存6个月以后,制剂仍为无定形的。
实施例13
将400g聚合物1、1200g Eudragit RL PO和400g酰胺咪嗪(熔点192℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第3桶的区域温度:160℃
·螺杆速度100rpm
·生产率:800克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在HCl中2小时以后,活性物质的释放为小于20%;在再缓冲至pH6.8以后,在10小时以后释放出75%。
在30℃/70%相对湿度下储存6个月以后,制剂仍为无定形的。
实施例14
将1200g聚合物1、400g Eudragit E PO、20g多库酯钠和400g非诺贝特(熔点81℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第3桶的区域温度:120℃
·螺杆速度100rpm
·生产率:800克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在HCl中2小时以后,活性物质的释放为83%。
在30℃下储存6个月以后,制剂仍为无定形的。
实施例15
将600g聚合物1、500g Kollidon SR,500g乙基纤维素、20g CremophorRH40和400g克霉唑(熔点148℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
Kollidon SR:80重量%聚乙酸乙烯酯、19重量%聚乙烯吡咯烷酮K30、0.8重量%月桂基硫酸钠、0.2重量%二氧化硅的物理混合物。
将混合物在以下条件下挤出:
·第一桶的区域温度:20℃;第二桶:40℃
·自第3桶的区域温度:160℃
·螺杆速度100rpm
·生产率:700克/小时
·压模直径3mm
通过XRD和DSC研究固溶体并发现为无定形的。在0.1标称HCl中2小时以后,活性物质的释放为小于20%。在再缓冲至pH6.8以后,在10小时以后释放出87%。
在30℃/70%相对湿度下储存6个月以后,制剂仍为无定形的。
实施例16
将600g聚合物1、1000g HPMCAS和400g非诺贝特(熔点81℃)称重加入Turbula混合容器中并在Turbula混合机T10B中混合10分钟。
将混合物在以下条件下挤出:

·自第3桶的区域温度:120℃
·螺杆速度100rpm

实施例17
将来自实施例7的挤出物通过空气喷射磨机粉碎至小于25μm的粒度。在0.1标称HCl中1小时以后,活性物质的释放为82%。
实施例18
将来自实施例15的挤出物通过空气喷射磨机粉碎至小于20μm的粒度。在0.1标称HCl中1小时以后,活性物质的释放为79%。
Claims (13)
1.一种包含在聚醚共聚物和至少一种微水溶性聚合物的聚合物基体中的微水溶性活性物质的制剂的剂型,所述聚醚共聚物通过30-80重量%N-乙烯基内酰胺、10-50重量%乙酸乙烯酯和10-50重量%聚醚的混合物自由基聚合得到,其中所述微水溶性活性物质以无定形形式存在于聚合物基体中,其中聚醚共聚物与微水溶性聚合物之比为99:1-10:90,其中微水溶性聚合物选自基于丙烯酸或甲基丙烯酸或其酯或所述单体的混合物的微水溶性聚合物、乙酸乙烯酯的均聚物和共聚物、乙基纤维素、乙酸羟丙基甲基纤维素琥珀酸酯和碱性甲基丙烯酸酯共聚物。
2.根据权利要求1的剂型,其包含在水中溶解度为pH依赖的那些聚合物作为微水溶性聚合物。
3.根据权利要求1或2的剂型,其包含基于丙烯酸或甲基丙烯酸或其酯或所述单体的混合物的微水溶性聚合物。
4.根据权利要求1的剂型,其包含乙酸乙烯酯的均聚物和共聚物作为微水溶性聚合物。
5.根据权利要求1的剂型,其包含乙基纤维素作为微水溶性聚合物。
6.根据权利要求1-5中任一项的剂型,其中聚醚共聚物与微水溶性聚合物之比为90:10-30:70。
7.根据权利要求1-6中任一项的剂型,其中聚醚共聚物与微水溶性聚合物之比为80:20-40:60。
8.根据权利要求1-7中任一项的剂型,其中所述聚合物基体包含其它增溶剂。
9.一种制备如权利要求1-8中任一项所限定的微水溶性活性物质的制剂的方法,其中所述活性物质以无定形形式嵌入基于30-80重量%N-乙烯基内酰胺、10-50重量%乙酸乙烯酯和10-50重量%聚醚的聚醚共聚物的聚合物基体中,其中除聚醚共聚物外,将至少一种微水溶性聚合物掺入聚合物基体中并将聚合物与微水溶性活性物质彻底混合。
10.根据权利要求9的方法,其中将聚合物和活性物质的混合物加热至聚醚共聚物的玻璃化转变温度以上。
11.根据权利要求9或10的方法,其中聚合物和活性物质的混合在挤出机中进行。
12.根据权利要求9的方法,其中聚合物和活性物质的混合在有机溶液中进行,然后除去有机溶剂。
13.根据权利要求12的方法,其中通过喷雾干燥除去有机溶剂。
