Abstract

本发明涉及一种应用于卵巢癌病人手术后预后的评判的方法。针对卵巢癌病人死亡率高、现有的预后评价体系繁琐且不够准确,本发明的特征在于在卵巢癌病人手术标本上,采用免疫组织化学的方法,联合检测TRPC各蛋白亚型的表达强度,采用友好的人机对话软件对数值进行处理,得出较为准确的卵巢癌预后评价。

TRPC蛋白亚型联合检测判断卵巢癌预后的方法
1.技术领域
本检测方法可应用于卵巢癌病人手术后预后的评判。对于选择手术后的辅助性治疗有提示作用。
2.背景技术
卵巢癌是一种严重威胁女性健康的杀手,因为早期缺乏典型症状,直至出现肿瘤转移出现相关症状才被发现,病死率高。大部分患者需要进行手术治疗,手术标本经常规病理诊断得到组织学类型和分级指标,再结合临床分期指标进行预后的判断,操作上复杂,而且准确性不高。为了更加准确的评判卵巢癌病人的危险度,利于合理使用辅助治疗和随诊周期的确定,需要新的预后评判指标。
TRPC是一类细胞膜表面的离子通道,它的表达量与功能与细胞的活性状态有较强的相关性。与多种肿瘤的发生和发展有关,例如:Sydorenko V发现淋巴结转移的前列腺癌细胞株中TRPC1和TRPC3表达增高并与肿瘤的增殖有关。El Hiani Y等发现乳腺癌细胞株中TRPC1和TRPC6表达增高。Aydar E等研究乳腺癌结果显示,癌组织表达TRPC3和TRPC6高于正常对照组织,转移率低的细胞株只表达TRPC3而高转移率的细胞株表达TRPC3和TRPC6,两者在细胞内结合在一起,TRPC6的激活剂可以抑制肿瘤细胞增殖并杀灭细胞,而抑制TRPC6则只抑制细胞增殖而不能杀灭细胞。Cai R等研究胃癌显示TRPC6表达增高,使用TRPC的抑制剂SKF96365则可以抑制细胞周期于G2/M期并抑制体内成瘤性。同样在食管鳞癌的研究也提示了TRPC6参与细胞周期的G2期跨越。Beck B等发现TRPC1和TRPC4与鳞状上皮的分化有关,这与基底细胞癌的发生有关。Yang SL在卵巢癌细胞株中发现TRPC3表达增高,抑制TRPC3的表达可以抑制细胞增殖,阻止细胞周期的M期通过;并抑制体内成瘤。TRPC在肿瘤局部神经系统的分布还与肿瘤性疼痛有关。目前申请者也发现TRPC1、3、6、7等在不同级别的卵巢癌中表达不同(见附图1-4)。
就目前的肿瘤研究发现,肿瘤细胞与正常细胞相比,往往出现多种基因和蛋白呈系统性的改变。正如TRPC离子通道蛋白各种亚型是相互关联起作用的,因此,单一的TRPC亚型检测不能全面反映肿瘤细胞的状态。因此,本发明是将TRPC各亚型联合检测,并采用数学建模,在模型的基础上,判断卵巢癌患者的预后。
3.发明内容
本发明目的在于用联合检测卵巢癌组织中TRPC各亚型的表达强度,配合数学模型进行卵巢癌病人预后的较准确的评价。

5.具体实施方式
本发明是这样实现的:
1)征得病人或病人家属同意,采集不同地区、不同医院的卵巢癌组织标本和临床病史资料。标本进行免疫组织化学染色,标记TRPC1、3、4、5、6、7六种亚型。显微镜观察表达强度,进行评级。所得评级与临床指标、病理指标以及病人预后进行多重线性回归模型建立数学模型,并制作相关计算软件;
2)在此基础上,手术病人的标本送检病理科化验时,除进行常规的病理检查,再进行TRPC1、3、4、5、6、7六种TRPC亚型的免疫组织化学染色(采用指定的公司抗体、指定的浓度和一定的操作流程);
3)染色完毕的切片在显微镜高倍镜下随机选择5个肿瘤组织视野进行染色强度的评判,判断结果取平均值;
4)六种TRPC亚型的相关值输入软件(附图5),得出与预后相关的提示。

Claims (7)

1.一种对上皮性卵巢癌病人预后和后续治疗效果评判的有效方法。
2.如权利要求1的方法,对于卵巢癌病人手术切取的手术标本,采用免疫组织化学方法对标本进行TRPC1,TRPC3,TRPC4,TRPC5,TRPC6,TRPC7六种TRPC亚型蛋白的染色。染色结果采用显微镜高倍镜随机选择5个肿瘤组织部分进行染色强度的评判,按0、1、2、3和4进行评分,取平均值。
3.将上述染色值代入特定公式,计算得出与预后相关的参数,对病人的术后情况进行评估。
4.如权利要求2的方法,TRPC各亚型的检测抗体选自特定公司的产品,并按照特定的浓度进行稀释使用。
5.如权利要求3的方法,计算公式来源于对已知病人预后的卵巢癌组织标本进行联合检测的数据采用多重线性回归模型等统计学处理后得到数学模型。
6.如权利要求5的方法,将数学模型编程,该软件具有良好的人机对话界面,方便医师操作。
7.如权利要求5的方法,卵巢癌病人的标本采集自上海、河南以及广东多家医院,从而保证该方法地域局限小。
