solide, liquide, poudre, lyophilisât, crème, pommade, par exemple, permettant la fabrication de toutes les formes galéniques pharmaceutiquement acceptables, sans aucune restriction: comprimés, sachets, gélules, globules, granules, sirops, liquides, par exemple, pour usage par voie orale, sublinguale, nasale, oculaire, et cutanées, par exemple, pouvant être préparés selon différents types de fabrication.
[0002] On pourra aussi utiliser le lait de tous les mammifères, dont le lait humain, les laits végétaux et les laits de toutes autres provenances, par exemple.
[0003] La formulation qui sera proposée pour une gélule par exemple, pourra aussi servir à la fabrication de compléments alimentaires à usage humain et animal, par exemple, de produits cosmétologiques, et pour d'autres produits, par exemple, cette formulation étant proposée à titre purement indicatif et n'est pas restrictive.
[0004] La présente invention associe les qualités du lait d'ânesse, par exemple, à l'activité des inhibiteurs naturels des enzymes protéasiques humaines (inhibiteurs extraits du blanc d'oeuf de caille japonaise de l'espèce Coturnix coturnix japonica de souche B. Mina), comme l'ovomucoïde et l'ovoinhibiteur, association qui potentialise l'efficacité de chacun des principes actifs et maximise leurs activités anti-inflammatoires et anti-allergiques, par exemple, innovation qui bénéficie de l'effet technique surprenant obtenu par la lyophilisation et par le mélange intime desdits principes actifs, ce qui induit une excellente synergie d'action.

[0008] Le lait d'ânesse frais contient principalement des protéines, des glucides, des lipides, des vitamines, des sels minéraux et des oligo-éléments; la composition du lait d'ânesse et très proche de celle du lait maternel, et peut donc être un substitut idéal en cas d'intolérance au lait de vache:
les protéines sont essentiellement représentées par une forte teneur en lysozyme et une faible teneur en caséine ce qui explique, d'une part, sa faible contamination microbienne et d'autre part, une digestion facilitée,
les glucides sont essentiellement le lactose, qui intervient comme stimulant dans l'absorption intestinale du calcium,
les lipides comportent des acides gras essentiels non synthétisés par l'organisme, les omégas 3 et les omégas 6,
les vitamines sont les vitamines A, B1, B2, B6, D, C et E,
les sels minéraux et les oligo-éléments comprennent le calcium, 'e magnésium, le phosphore, le sodium, le fer et le zinc.
[0009] Par "inhibiteurs naturels des enzymes protéasiques humaines", on entend des glycoprotéines contenues dans le blanc de l'oeuf de caille japonaise de l'espèce Coturnix coturnix japonica de souche B. Mina (l'oeuf de caille étant considéré comme un produit naturel), dotées d'une puissante activité anti-protéasique humaine (protéases du type trypsine et ses homologues), ce qui leur confère une très forte activité anti-inflammatoire;
deux fractions ont été isolées du blanc de l'oeuf de caille:
l'ovomucoïde, d'un poids moléculaire de 28 kDA, inhibe la trypsine humaine et ses homologues comme l'élastase, la tryptase et la carboxypeptidase, par exemple.
l'ovoinhibiteur, d'un poids moléculaire de 57 kDA, n'inhibe pas la trypsine humaine, mais la protéinase, serine protéase d'une moisissure nommée aspergillus orizae, la subtilisine, serine protéase du bacille subtilis, et enfin l'&-chymotrypsine.
[0010] L'association de ces deux inhibiteurs naturels des serines protéases humaines dans une proportion, par exemple, de trois fractions d'ovomucoïde pour une fraction d'ovoinhiteur, confère à l'innovation une activité anti-protéasique plus large et plus puissante par rapport à chacun de ces inhibiteurs, proposés séparément.
[0011] L'innovation se compose, par exemple, in fine, d'un mélange intime d'un lyophilisât de lait d'ânesse et d'un lyophilisât d'ovomucoïde et d'ovoinhibiteur intimement mélangés et incorporés dans une gélule à délitement rapide pour la voie orale, par exemple, mais d'autres voies d'administration sont possibles avec des formes galéniques différentes, les comprimés pour la voie sublinguale et, des crèmes, des pommades pour la voie percutanée, par exemple.
[0012] Selon l'état antérieur, on connaît le brevet FR 2 901 137, dans lequel on décrit une composition médicamenteuse à base d'inhibiteurs naturels des protéases humaines, associés à des vitamines et ou à des sels minéraux et ou à des acides aminés, incorporés dans un excipient, composition administrée par voie orale et sublinguale, sous la forme d'un comprimé à délitement rapide.
[0013] Il n'était donc pas évident, même pour l'homme de l'art, d'associer du lait d'ânesse ou une autre variété de lait provenant d'un mammifère, d'un végétai ou de toutes autres provenances, à un ou plusieurs inhibiteurs naturels des enzymes protéasiques humaines.
[0014] Le mélange intime des lyophilisats incorporés dans une gélule, par exemple, est une composition médicamenteuse constituée par un lyophilisât de lait d'ânesse mélangé à un lyophilisât d'inhibiteurs naturels des protéases humaines extraits du blanc des oeufs de caille de l'espèce Coturnix coturnix japonica de souche B. Mina, adapté à un délitement rapide par voie orale, ce qui favorise la libération des principes actifs dans l'organisme.
[0015] Cette gélule comprend des principes pharmacologiquement actifs intimement mélangés: un lyophilisât de lait d'ânesse et un lyophilisât d'inhibiteurs naturels des protéases humaines (ovomucoïde et ovoinhibiteur).
[0016] Cette gélule possède des activités qui lui sont propres:
une forte valeur nutritionnelle grâce à l'apport combiné des protéines, des glucides, des lipides, des vitamines, des sels minéraux et des oligo-éléments contenus dans le lait d'ânesse, un exemple de formulation sera décrit ci-après,
une activité anti-inflammatoire globale ciblée sur tous les tissus et organes de l'organisme et,
une activité anti-inflammatoire non spécifique liée aux inhibiteurs des protéases humaines, activité intervenant dans la prévention et le traitement des processus infectieux de l'adulte et de l'enfant, par exemple,
une activité anti-inflammatoire, anti-allergique spécifique,
grâce à l'inhibition de la libération des médiateurs chimiques excrétés dans le sang par les cellules basophiles et les mastocytes lors d'un conflit allergènes-anticorps IgE (allergènes d'acariens, de moisissures, de pollens, par exemple), médiateurs responsables des symptômes cliniques de l'allergie,
une activité anti-inflammatoire dermatologique, grâce à son action inhibitrice sur les enzymes de l'inflammation contenues dans les cellules immunitaires du derme telles que l'élastase sécrétée par les cellules neutrophiles en cas d'inflammation cutanée par exemple,
une activité anti-inflammatoire sur tout le système oculo-naso-respiratoire,
une forte action stimulante du système immunitaire par l'intermédiaire du zinc et de la vitamine C contenus dans le lait d'ânesse.
1/Obtention des Iyophilisats du lait d'ânesse et des inhibiteurs naturels des protéases humaines
[0017] On utilise un lyophilisateur; la lyophilisation ou séchage à froid, est un procédé qui consiste à retirer l'eau d'un aliment ou de tout autre produit solide ou liquide, afin de le rendre stable à la température ambiante et à faciliter sa conservation; la lyophilisation comprend trois étapes: la congélation, la dessiccation primaire et la dessiccation secondaire ou séchage final:
la congélation consiste à ramener la température du produit à lyophiliser à une température comprise entre -20[deg.] et -80[deg.]C de façon à bloquer l'eau sous forme de glace, cela évite l'altération des principes actifs,
la dessiccation primaire ou sublimation est le passage d'une substance de l'état solide ou liquide à l'état gazeux; le produit ou l'aliment est desséché en
le mettant sous vide, la glace devient alors de la vapeur;
la déshydratation du produit entraîne une perte d'eau de 80 à 90[deg.],
la dessiccation secondaire commence quand toute la glace est sublimée ou supprimée, les aliments ou les produits sont séchés, la température du lyophilisateur s'élève spontanément et brièvement une fois que toute l'eau est sublimée ce qui permet d'obtenir un produit sec à 95% voire plus, sans la moindre altération de sa composition initiale; la lyophilisation permet le mélange des produits ainsi obtenus, leur conservation et leur stabilité sont garanties pour plusieurs années;
l'excellente action pharmacologique du produit fini (lait d'ânesse et inhibiteurs de protéases) repose sur l'effet surprenant induit par la technique de lyophilisation, l'association des produits potentialise et maximise leur efficacité grâce à une excellente synergie d'action.
2/Obtention d'un ou de plusieurs inhibiteurs naturels anti-protéases
[0018] Les inhibiteurs naturels des protéases humaines extraits du blanc de l'oeuf de caille comprennent:
l'ovomucoïde, qui est contenu dans le blanc, est isolé par précipitation à l'aide de l'acide trichloracétique et de l'acétone; la fraction protéique isolée est ensuite purifiée par chromatographie séquentielle sur DEAE-cellulose; une fois purifié, l'ovomucoïde est lyophilisé; l'activité anti-protéases humaines (trypsine et homologues) est testée sur un substrat de caséine; on fait réagir 2 p.g d'ovomucoïde sur 1 jag de trypsine humaine: l'ovomucoïde du blanc de l'oeuf de caille n'est détruit ni par la digestion ni par un pH gastrique acide ce qui permet son administration par voie orale, par exemple,
l'ovoinhibiteur qui est obtenu par précipitation à l'aide de l'acide trichloracétique et de l'acétone;
on obtient un précipité qui est dialyse et lyophilisé; on mesure la capacité d'inhibition des inhibiteurs protéasiques en faisant réagir la fraction obtenue sur la protéinase d'aspergillus orizae en utilisant un substrat de caséine, puis la subtilisine du bacillus subtilis, l'a-chymotrypsine et la trypsine humaine: l'ovoinhibiteur n'inhibe pas l'activité protéasique de la trypsine humaine, mais inhibe la protéinase, la subtilisine et l'[alpha]-chymotrypsine.
