Gegenstand der Erfindung ist ein Verfahren zur Durchführung biotechnologischer Prozesse unter Kultivierung aerober Organismen in hoher Zelldichte im Durchlaufverfahren in einem Zyklonreaktor mit einem Zyklondurchmesser : Einlaufdurchmesser-Verhältnis </= 10, dem am oberen Ende tangential flüssiges Medium mit einer Beschleunigungskennziffer (Zentrifugalbeschleunigung/Erdbeschleunigung) bis zu 25 zugeführt wird, in das sauerstoffhaltiges Gas gemischt ist und/oder dem sauerstoffhaltiges Gas über die Mantelfläche des Reaktors zugeführt wird und das vom unteren Ende des Reaktors entnommen und über einen äusseren Rezyklierungsumlauf auf Prozessbedingungen gebracht wird, während vom oberen Ende des Reaktors akkumuliertes Gas entfernt wird, wobei mit einem Leistungseintrag im Kreislauf von 4-40 kW/m<3> gearbeitet wird, sowie eine dafür geeignete Anordnung.
Biotechnologische Prozesse, insbesondere Fermentationsprozesse, die unter Kultivierung von Mikroorganismen oder Zellen ablaufen, bedürfen insbesondere bei Arbeit mit hohen Zelldichten einer intensiven Sauerstoffzufuhr.
Es sind daher bereits unterschiedliche Techniken zur Steigerung des Sauerstoffeintragsvermögens für Fermentationsprozesse bekannt, die einzeln oder in Kombination angewandt werden können:
1. Eine Vergrösserung des treibenden Konzentrationsgefälles durch Erhöhung des Sauerstoffpartialdrucks im Reaktor mit möglichst grossem hydrostatischen Druck ("Turmbiologie"), durch Reaktionen unter höheren Drücken (Druckfermentation) und durch Begasung mit sauerstoffangereicherter Luft bis hin zum Einsatz von reinem Sauerstoff.
2. Eine Vergrösserung der volumenspezifischen Stoffaustauschfläche a durch möglichst feine Dispergierung der Gasphase im gesamten Reaktor unter Vermeidung von Koaleszenz ("Spaltinjektoren", Radialstromdüsen" mit Leitblechen).
3.
Eine Verringerung des Stofftransportwiderstands auf der Flüssigkeitsseite 1/kL durch Erhöhung des Energieeintrags (z.B. "Strahlschlaufenreaktoren").
4. Höchste Sauerstoffeintragsraten werden im Zentrifugalreaktor erreicht: Hierbei wird eine dampfsterilisierbare Siebzentrifuge als Reaktor eingesetzt (H. Voit, A. Mersmann, Chem. Ing. Techn. 61, 5 (1989) S. 416-417). Bei 50facher Erdbeschleunigung lassen sich im Zentrifugalbioreaktor bei der Begasung mit Luft Sauerstoffeintragsraten von bis zu 100 g/(l*h) verwirklichen (Newtonsche Medien, kLa = 10 h<-><1>). Dies ist neben der Intensivierung des Phasenkontakts (kL-Erhöhung) und der Feindispergierung der Gasblasen (a-Erhöhung) auf die lokale Erhöhung des Sauerstoffpartialdrucks (Erhöhung des treibenden Konzentrationsgefälles) im Zentrifugalfeld zurückzuführen.
Bekannt ist auch bereits ein Zyklon-Säulenreaktor (P.S.S. Dawson, Biotechnol. & Bioeng. Symp. No. 4 (1974) 809-819), bei dem eine Zyklon-Säule in einen Kreislauf für das flüssige Medium eingeschaltet ist, das am Säulenkopf tangential zugeführt wird und sich schraubenartig als bewegter Film längs der Säulenwand abwärts bewegt unter lebhaftem Gasaustausch mit dem am Reaktorboden zugeführten Gas.
Diese bereits längere Zeit bekannte Zyklonsäulentechnik wurde von J.D. Sheppard u.a. (J. Chem. Tech. Biotechnol. 59 (1994) 83-89) aufgegriffen und in einer Scaleup Version von 75 l Kapazität des Reaktors untersucht. Bei der Scale-up Einheit wurde die Säule verkürzt und dafür mit Luftinjektion in einem stark vergrösserten Kreislauf an zumindest zwei Stellen unter Erhöhung der Rotationsgeschwindigkeit der Förderpumpe gesorgt. Auf diese Weise wurde eine mit dem Zyklonsäulenreaktor vergleichbare Fermentationsleistung erreicht, die als ähnlich zum herkömmlichen Rührkesselreaktor bezeichnet wird.
Eine höhere Effizienz scheint der von H.Voit et al. (s.o.) beschriebene Zentrifugalreaktor zu besitzen, der in der DE 3 905 609 A1 in Verbindung mit einem Zyklonentschäumer näher beschrieben wird.
Eine für biotechnologische Prozesse brauchbare Verfahrensweise mit einem im Kreislauf betriebenen Zyklonreaktor der eingangs spezifizierten erfindungsgemässen Art ist mithin bislang nicht in Betracht gezogen worden.
Gemäss der Erfindung wird durch intensiven Tangentialeintrag in den Zyklonreaktor bei relativ niedrigem Durchmesserverhältnis von Zyklon zu Zykloneinlauf mit Beschleunigungskennziffern bis zu 25 (insbesondere 2-25) gearbeitet unter Verwendung von Verdrängerpumpen wie Drehkolbenpumpen, Exzenterschneckenpumpen, Sine-Pumpen oder Taumelscheibenpumpen oder auch Peristaltikpumpen und Doppelmembranpumpen unter Erzielung von Sauerstofftransportkoeffizienten kLa bis zu 3000 h<-><1> (bei Gasinjektion im Umlauf) bzw. bis 25000 h<-><1> (bei Gaszufuhr über den Zyklonmantel).
Durch die Anordnung eines Prallkörpers im unteren Bereich wird die Ausbildung des Sekundärwirbels gefördert, so dass insgesamt die volumenspezifische Begasungsmenge verringert werden kann, was zur Einsparung von Energie und Milderung von Schaum- und Abgasproblemen führt.
Besonders zweckmässig ist ein Prallkörper in Form eines Tellers, der ggf. gleichzeitig - allein oder ergänzend - als Gasverteiler bzw. Begasungseinrichtung wirken kann oder in Form eines mit der Spitze nach unten weisenden Kegels, der auch ggf. - mit poröser Basis - dem Gaseintrag dienen kann.
Der erfindungsgemässe Zyklonreaktor kann jedoch auch ohne Prallkörper im unteren Bereich vorgesehen werden, wobei dann erhöhte Gasanteile in den Rezyklierungskreis gelangen können, was bisweilen für die ausreichende Sauerstoffversorgung empfindlicher Zellsysteme auch im Rezyklierungskreis erwünscht sein kann.
Auch bei erhöhtem Caseintrag über den Prallkörper oder über einem perforierten Zylindermantel des Zyklons kann ein Teil des zugeführten Cases in den Kreislauf gedrückt werden. Hierdurch wird auch bei hohen Zelldichten eine Sauerstoffversorgung im Kreislauf sichergestellt, da die Speicherkapazität der Flüssigphase für Sauerstoff sehr begrenzt ist. Eine genaue Quantifizierung ist nicht möglich, da die Stoffdaten gas/flüssig den Umlauf-Gasanteil wesentlich beeinflussen.
Besonders zweckmässig ist die Einbeziehung einer Mikrofiltration in den Rezyklierungskreis zur Steigerung der Zelldichten, wobei z.B. mit Röhrenmodulen gearbeitet werden kann und die Überströmgeschwindigkeit dem Tangentialeinlauf in den Zyklonreaktor angepasst ist.
Alle Messsonden für Temperatur, pH-Wert, pO2, Trübung etc. sowie alle Zu- und Abläufe der Flüssigphasen (Substrate, Korrekturmittel, Produkt) werden vorzugsweise im Umlauf installiert, um das Strömungsfeld im Zyklonreaktor nicht zu stören.
Eine Anordnung zur Durchführung biotechnologischer Prozesse gemäss der Erfindung unter Kultivierung aerober Organismen in hoher Zelldichte ist gekennzeichnet durch einen Kulturflüssigkeitskreislauf mit Elementen zur Regelung der Prozessbedingungen für einen als Zyklon ausgebildeten, in den Kreislauf integrierten Reaktor mit Gasentnahme am Kopf und Flüssigkeitsentnahme am Boden, dessen oberer Tangential-Einlauf mit einer im Kreislauf vorgesehenen Pumpe für einen Leistungseintrag für Beschleunigungskennziffern (ZEinlauf) bis zu 25 (Zentrifugalbeschleunigung/Erdbeschleunigung) in Verbindung steht und vor dessen unterem Ende ein Prallkörper zur Unterstützung des Sekundärwirbels angeordnet sein kann und durch Mittel zur Begasung im Kreislauf vor dem Tangentialeinlauf zum Zyklonreaktor und/oder über perforierte Gasverteilerflächen im Reaktor.
Anwendungsgebiete für die erfindungsgemässe Technik sind:
die Bioverfahrenstechnik zur effektiven Herstellung mikrobieller niedermolekularer Metabolite (z.B. organische Säuren, Aminosäuren, Vitamine) sowie Exoenzymen (z.B. Proteasen, Amylasen) und zur effektiven Herstellung rekombinanter Proteine in Hochzelldichte-Fermentationen;
die biotechnologische Forschung zur Kultivierung hoher Zelldichten unter definierten, nicht sauerstofflimitierten Bedingungen.
Weitere Besonderheiten der Erfindung ergeben sich aus den Patentansprüchen sowie aus der nachfolgenden Beschreibung von Ausführungsbeispielen unter Bezugnahme auf die angefügten Zeichnungen, die schematisch vier Varianten einer erfindungsgemässen Anordnung wiedergeben.
Gemäss Fig. 1 ist ein Zyklonreaktor 1 mit unterem (hohlen) Prallkegel 2 in einen Kulturflüssigkeitsumlauf einbezogen, der vom unteren Ende 3 des Zyklons abzweigend über eine Pumpe 4 geleitet wird, mit einem Auslass für zellhaltige Flüssigkeit 5 sowie einer Mikrofiltration 6, insbesondere in Form von Rohrmodulen zur Abgabe von Filtrat über 7, während bei 8 insbesondere ergänzendes Kulturmedium eingespeist werden kann. Mit 9 ist eine Gasinjektion angedeutet für die Gaszufuhr über 10. Das daraufhin erzielte Flüssigkeitsgasgemisch tritt bei 11 tangential in den Zyklonreaktor ein, in dem sich ein Flüssigkeitsspiegel, etwa wie angedeutet, ausbildet. Das oberhalb desselben akkumulierte Gas tritt bei 12 über ein Tauchrohr 13 aus dem Reaktor aus.
Dieses Tauchrohr kann mit (nicht dargestellten) Schaumzerstörerelementen unterhalb des Gasauslasses versehen sein, etwa in Form von Dreiecken, die in den Gasraum hineinragen. Die Gasinjektion kann auch zwischen Pumpe und Filter vorgesehen sein.
Fig. 2 und 3 zeigen Varianten zu der soeben erläuterten Ausführungsform, bei denen die Begasung, wie angedeutet, über die porös ausgebildete Zyklonwand bzw. die poröse Basis des Prallkegels erfolgt.
Gemäss Fig. 4 ist die Flüssigkeitsentnahme aus dem Zyklonreaktor abgewandelt, und zwar wird flüssiges Kulturmedium aus dem Zyklonreaktor über ein Tauchrohr 14 vom geschlossenen Boden des Zyklonreaktors 1 entnommen, das durch den Prallkegel 2 und das Tauchrohr 13 zur Gasentnahme hindurchgeführt ist.
Beispiel
Zyklon Reaktor für Labor-Fermentationen gemäss Fig. 4
Dieser Zyklon-Reaktor besitzt einen Innendurchmesser von 70 mm und eine zylindrische Länge von 190 mm. Der Einlauf-Durchmesser beträgt 9 mm. Der Prallkegel (Winkel in der Kegelspitze 130 DEG ) vor dem Unterlauf hat einen Durchmesser von 62 mm, so dass über einen Ringspalt von 4 mm der gasfreie Unterlauf in das Axialrohr nach oben abgesaugt werden kann (Fig. 4). Die Durchmesserreduzierung vom zylindrischen Teil des Zyklon-Reaktors zum Zentralrohr (Innendurchmesser 9 mm) erfolgt über eine Länge von 10 mm. Das Tauchrohr am Kopf des Zyklon-Reaktors zur Entnahme der Gasphase ist 5 mm eingetaucht und hat einen Innendurchmesser von 30 mm. Die Gasentnahme aus dem nach oben abgeschlossenen Tauchrohr erfolgt seitlich mit einem Rohrdurchmesser von 9 mm. Durch den Deckel des Tauchrohrs ist zentral das Saugrohr des Unterlaufs durchgeführt. Der gesamte Zyklon-Reaktor ist aus Glas gefertigt.
Direkt neben dem Tauchrohr ist eine SCHOTT GL Verschraubung angebracht, die zur Aufnahme einer kontinuierlichen Niveausonde (potentiometrisches Messprinzip) dient, um das Flüssigphasen-Volumen im Zyklon-Reaktor einzustellen (Arbeitsvolumen 800 ml).
Alle anderen Messsonden (Temperatur pH pO2, Trübung) sowie alle Zu- und Abläufe der Flüssigphasen (Substrat, Korrekturmittel, zellhaltiger Ablauf) sind im Umlauf integriert.
Auf der Druckseite der Umlaufpumpe (Filtron-Sinus-Pumpe) ist ein Mikrofiltrationsmodul (keramisches Rohrmodul mit 19 Kanälen, Kanaldurchmesser 2,7 mm, Austauschfläche 0,14 m<2>) installiert. Danach erfolgt der Lufteintrag über ein Sinterrohr, bevor der Umlaufstrom über einen Wärmetauscher wieder tangential in den Zyklon-Reaktor eingespeist wird. Damit ergibt sich ein Gesamt-Arbeitsvolumen von 1,5 l.
Bestimmung des Sauerstoff-Transportkoeffizienten (kLa):
Der kLa-Wert wurde nach der stationären Methode mit Medium ohne Zellen bestimmt, indem der in den Zyklon-Reaktor eingetragene Sauerstoff extern in einem Stripper mit Stickstoff wieder entfernt wurde (Stickstoffdesorptionsmethode).
Bei verschiedenen Beschleunigungskennziffern z und unterschiedlichen volumetrischen Gaseintragsraten wurde der kLa-Wert bestimmt. Die Flüssigvolumenströme wurden zwischen 400 und 700 I/h variiert, die Luftvolumenströme zwischen 50 und 300 I/h. Die Ergebnisse lassen sich mit einem Potenzansatz korrelieren:

Die Korrelationskoeffizienten wurden wie folgt bestimmt (Medium, siehe Tab. 1):
c = 1.3865, a = 0.5995, b = 0.4387
Es sind kLa-Werte bis 3000 1/h bzw. Sauerstoffeintragsraten bis 21 g/(I*h) gemessen worden.
Kultivierung des aeroben Bakteriums Corynebacterium glutamicum im Zyklon-Reaktor:
Zur Vorkultivierung werden 1000 ml Schüttelkolben mit 4 Schikanen und einem Arbeitsvolumen von 200 ml verwendet. Die Medienbestandteile werden 30 min bei 121 DEG C autoklaviert (Zusammensetzung des Nährmediums: Tabelle 1). Nach Abkühlung auf Umgebungstemperatur wird mit 1 ml Organismensuspension aus der Stammhaltung angeimpft. Es folgt eine 24stündige Inkubierung bei 30 DEG C.
Das Zyklon-Reaktor-System wird chemisch sterilisiert. Dazu wird Dimethyldicarbonat 1:1000 mit Wasser verdünnt und anschliessend in das Reaktorsystem gepumpt. Bei 20 DEG C ist nach ca. 20 Minuten der Wirkstoff in Kohlendioxid und Methanol zerfallen. Nach der chemischen Sterilisierung des Reaktorsystems wird das übriggebliebene Wasser durch sterilisiertes Nährmedium verdrängt. Die Zusammensetzung des Nährmediums zur kontinuierlichen Kultivierung ist Tabelle 1 zu entnehmen. Salze und Glucose sind getrennt zu autoklavieren (30 min, 121 DEG C). Vitamine und Aminosäure werden im Anschluss sterilfiltriert zugegeben.
Zum Starten der Reaktion werden 200 ml inkubierte Vorkultur in den Zyklon-Reaktor mit einem Gesamtvolumen von 1.51 gepumpt. Zur Adaption der Mikroorganismen an das synthetische Hauptkulturmedium folgt ein 12stündiger Batchbetrieb des Zyklon-Reaktors. In dieser Zeit erhöht sich die Zellmassekonzentration von 3 auf 15 g Biotrockenmasse/l.
Der Volumenstrom der Umlaufpumpe wird konstant auf 500 I/h eingestellt. Das entspricht einer Strömungsgeschwindigkeit v der Flüssigkeit am tangentialen Einlauf des Zyklon-Reaktors von 2m/s.

Die Beschleunigungskennziffer ZEinlauf (Zentrifugalbeschleunigung/Erdbeschleunigung) wird wie folgt berechnet:
ZEinlauf = 2 * v<2> / (D * g)
wobei
v = Leerrohrgeschwindigkeit am Einlauf (m/s)
D = Innendurchmesser des Zyklons (m)
g = Erdbeschleunigung (9.81 m/s<2>).
Damit ist die Beschleunigungskennziffer ZEinlauf = 11.7.
Die Zuluftmenge wird auf 400 I/h eingestellt. Damit ergibt sich ein volumenspezifischer Energieeintrag (Leistungseintrag der Umlaufpumpe und kinetische Energie des Gaseintrags) von knapp 10 kW/m<3>. Über den Wärmetauscher im Umlauf wird durch einen Thermostaten mit externer Temperaturegelung die Temperatur im Zyklon-Reaktor-System auf 30 DEG C eingestellt. Der pH-Wert wird durch Zugabe des Korrekturmittels (4N NaOH) auf 7,0 geregelt.
Bei einer Biotrockenmassekonzentration von 15 g/l kann auf kontinuierlichen Betrieb umgestellt werden. Der Zulauf wird auf 300 ml/h eingestellt. Bei einem Gesamtvolumen des Reaktorsystems von 1,5 l entspricht das einer mittleren Verweilzeit des Nährmediums von 5 h im Reaktorsystem. Durch die im Umlauf integrierte Mikrofiltrationseinheit kann dem System ein zellfreier Flüssigkeitsstrom entnommen werden. Damit kann die mittlere Verweilzeit der Mikroorganismen im System von der Verweilzeit des Nährmediums entkoppelt werden.
Es wird ein Verweilzeitfaktor F von 5 eingestellt (Der Verweilzeitfaktor F stellt das Verhältnis zwischen der Verweilzeit der Mikroorganismen und der Verweilzeit des Nährmediums dar). Ein Verweilzeitfaktor F von 5 bedeutet, das sich die Zellmasse (der Biokatalysator) 5 mal länger im Zyklon-Reaktor-System befindet als das Nährmedium. Bei einem Zulaufvolumenstrom von 300 ml/h ist dies durch die Entnahme eines zellhaltigen Bleedstroms von 60 ml/h und eines zellfreien Filtratstroms von 240 ml/h zu erreichen. Der Filtratstrom stellt sich bei fester Vorgabe des Bleedstroms über die Füllstandsregelung ein.
Ergebnisse:
Nach 3 Zellmasse-Verweilzeiten (75 h) wird im Fliessgleichgewicht eine Zellmassekonzentration von 60 g BTM/I im Zyklon-Reaktor erreicht. Als pO2 wird bei dieser Zelldichte ein Wert von 6% Luftsättigung an der Messstelle im Umlauf gemessen.
Zur biochemischen Kontrolle einer im gesamten Zyklon-Reaktor-System ausreichenden Sauerstoffversorgung wurde das Fermentationsmedium auf das Gärendprodukt Laktat untersucht, das nur gebildet wird, wenn nicht genügend Sauerstoff vorhanden ist. Im untersuchten Betriebsbereich konnte keine Laktatbildung festgestellt werden.
Gegenüber dem herkömmlichen Rührkesselverfahren kann durch den Einsatz des Zyklon-Reaktors die Zellmassekonzentration im Reaktor um den Faktor 3 gesteigert werden, ohne dass es zu unerwünschten Sauerstofflimitierungen kommt. Bei unveränderter zellmassenspezifischer Reaktionsgeschwindigkeit der Aminosäurebildung lässt sich damit die volumetrische Produktivität ("Raum-Zeit-Ausbeute") ebenfalls gegenüber dem kontinuierlichen Rührkesselverfahren um den Faktor 3 steigern.

Biotechnological processes, especially fermentation processes that take place with the cultivation of microorganisms or cells, require an intensive supply of oxygen, especially when working with high cell densities.
Different techniques for increasing the oxygen input capacity for fermentation processes are therefore already known, which can be used individually or in combination:
1. An enlargement of the driving concentration gradient by increasing the oxygen partial pressure in the reactor with the greatest possible hydrostatic pressure ("tower biology"), by reactions under higher pressures (pressure fermentation) and by gassing with oxygen-enriched air up to the use of pure oxygen.
2. An enlargement of the volume-specific mass transfer area a by finely dispersing the gas phase in the entire reactor while avoiding coalescence ("gap injectors", radial flow nozzles "with baffles).

This cyclone column technique, which has been known for a long time, was developed by J.D. Sheppard et al. (J. Chem. Tech. Biotechnol. 59 (1994) 83-89) and examined in a scale-up version of 75 l capacity of the reactor. In the scale-up unit, the column was shortened and air injection in a greatly enlarged circuit ensured it at at least two points while increasing the speed of rotation of the feed pump. In this way, a fermentation performance comparable to the cyclone column reactor was achieved, which is referred to as similar to the conventional stirred tank reactor.
A higher efficiency seems to be that of H.Voit et al. (see above) to have the described centrifugal reactor, which is described in more detail in DE 3 905 609 A1 in connection with a cyclone foamer.
A procedure which can be used for biotechnological processes and has a cyclone reactor of the type according to the invention specified at the outset, which has been operated in a circuit, has therefore not hitherto been considered.
According to the invention, intensive tangential entry into the cyclone reactor with a relatively low diameter ratio of cyclone to cyclone inlet with acceleration ratios up to 25 (in particular 2-25) is carried out using displacement pumps such as rotary lobe pumps, eccentric screw pumps, sine pumps or swash plate pumps or also peristaltic pumps and double diaphragm pumps Achievement of oxygen transport coefficients kLa up to 3000 h <-> <1> (with gas injection in circulation) or up to 25000 h <-> <1> (with gas supply via the cyclone jacket).
The arrangement of a baffle in the lower region promotes the formation of the secondary vortex, so that overall the volume-specific fumigation quantity can be reduced, which leads to savings in energy and mitigation of foam and exhaust gas problems.
A baffle in the form of a plate is particularly expedient, which can optionally act as a gas distributor or fumigation device at the same time, alone or in addition, or in the form of a cone with the tip pointing downwards, which may also serve to introduce gas with a porous base can.
However, the cyclone reactor according to the invention can also be provided in the lower region without a baffle, in which case increased gas fractions can get into the recycling circuit, which can sometimes be desirable in the recycling circuit for the adequate oxygen supply to sensitive cell systems.
Even if the casein is raised above the impact body or over a perforated cylinder jacket of the cyclone, part of the supplied case can be pressed into the circuit. This ensures an oxygen supply in the circuit even at high cell densities, since the storage capacity of the liquid phase for oxygen is very limited. An exact quantification is not possible because the material data gas / liquid have a significant influence on the circulating gas content.
It is particularly expedient to include microfiltration in the recycling circuit to increase cell densities, e.g. can be used with tube modules and the overflow speed is adapted to the tangential inlet into the cyclone reactor.
All measuring probes for temperature, pH value, pO2, turbidity etc. as well as all inflows and outflows of the liquid phases (substrates, correction agent, product) are preferably installed in circulation so as not to disturb the flow field in the cyclone reactor.
An arrangement for carrying out biotechnological processes according to the invention with the cultivation of aerobic organisms in high cell density is characterized by a culture liquid circuit with elements for regulating the process conditions for a cyclone-type reactor integrated in the circuit with gas removal from the head and liquid removal from the bottom, its upper tangent -Inlet is connected to a pump provided in the circuit for a power input for acceleration indicators (ZInlet) up to 25 (centrifugal acceleration / gravitational acceleration) and an impact body can be arranged in front of the lower end to support the secondary vertebra and by means of gassing in the circuit in front of the Tangential inlet to the cyclone reactor and / or via perforated gas distributor surfaces in the reactor.
Areas of application for the technology according to the invention are:
bioprocess engineering for the effective production of microbial low-molecular metabolites (e.g. organic acids, amino acids, vitamins) and exoenzymes (e.g. proteases, amylases) and for the effective production of recombinant proteins in high-cell density fermentations;
biotechnological research for the cultivation of high cell densities under defined, non-oxygen-limited conditions.

This cyclone reactor has an inside diameter of 70 mm and a cylindrical length of 190 mm. The inlet diameter is 9 mm. The impact cone (angle in the cone tip 130 °) in front of the underflow has a diameter of 62 mm, so that the gas-free underflow can be sucked upwards into the axial tube via an annular gap of 4 mm (Fig. 4). The diameter reduction from the cylindrical part of the cyclone reactor to the central tube (inner diameter 9 mm) takes place over a length of 10 mm. The immersion tube at the head of the cyclone reactor for withdrawing the gas phase is immersed 5 mm and has an inner diameter of 30 mm. The gas is withdrawn from the top of the immersion tube with a tube diameter of 9 mm. The suction pipe of the underflow is carried out centrally through the cover of the immersion pipe. The entire cyclone reactor is made of glass.
A SCHOTT GL screw connection is attached directly next to the immersion tube, which is used to hold a continuous level probe (potentiometric measuring principle) in order to adjust the liquid phase volume in the cyclone reactor (working volume 800 ml).
All other measuring probes (temperature pH pO2, turbidity) as well as all inflows and outflows of the liquid phases (substrate, correction agent, outflow containing cells) are integrated in the circulation.
A microfiltration module (ceramic tube module with 19 channels, channel diameter 2.7 mm, exchange area 0.14 m <2>) is installed on the pressure side of the circulation pump (Filtron sine pump). The air is then introduced via a sintered tube before the recycle stream is again tangentially fed into the cyclone reactor via a heat exchanger. This results in a total working volume of 1.5 l.
Determination of the oxygen transport coefficient (kLa):
The kLa value was determined using the stationary method with medium without cells, by removing the oxygen introduced into the cyclone reactor externally in a stripper using nitrogen (nitrogen desorption method).
The kLa value was determined at different acceleration indicators z and different volumetric gas entry rates. The liquid volume flows were varied between 400 and 700 I / h, the air volume flows between 50 and 300 I / h. The results can be correlated with a potency approach:

Cultivation of the aerobic bacterium Corynebacterium glutamicum in the cyclone reactor:
For pre-cultivation, 1000 ml shake flasks with 4 baffles and a working volume of 200 ml are used. The media components are autoclaved at 121 ° C. for 30 min (composition of the nutrient medium: Table 1). After cooling to ambient temperature, inoculation is carried out with 1 ml organism suspension from the stock. A 24-hour incubation at 30 ° C. follows.
The cyclone reactor system is chemically sterilized. For this purpose, dimethyl dicarbonate is diluted 1: 1000 with water and then pumped into the reactor system. At 20 ° C., the active ingredient has broken down into carbon dioxide and methanol after about 20 minutes. After the reactor system has been chemically sterilized, the remaining water is displaced by sterilized nutrient medium. The composition of the nutrient medium for continuous cultivation is shown in Table 1. Salts and glucose are to be autoclaved separately (30 min, 121 ° C). Vitamins and amino acids are then added under sterile filtration.
To start the reaction, 200 ml of incubated preculture are pumped into the cyclone reactor with a total volume of 1.51. A batch operation of the cyclone reactor for 12 hours follows to adapt the microorganisms to the synthetic main culture medium. During this time, the cell mass concentration increases from 3 to 15 g dry biomass / l.
The volume flow of the circulation pump is constantly set to 500 I / h. This corresponds to a flow velocity v of the liquid at the tangential inlet of the cyclone reactor of 2 m / s.

The supply air volume is set to 400 I / h. This results in a volume-specific energy input (power input of the circulation pump and kinetic energy of the gas input) of just under 10 kW / m <3>. The temperature in the cyclone reactor system is set to 30 ° C. via the heat exchanger in the circulation by means of a thermostat with external temperature control. The pH is regulated to 7.0 by adding the correcting agent (4N NaOH).
With a dry biomass concentration of 15 g / l you can switch to continuous operation. The feed is adjusted to 300 ml / h. With a total volume of the reactor system of 1.5 l, this corresponds to an average residence time of the nutrient medium of 5 h in the reactor system. A cell-free liquid flow can be removed from the system by the microfiltration unit integrated in the circulation. The mean residence time of the microorganisms in the system can thus be decoupled from the residence time of the nutrient medium.
A residence time factor F of 5 is set (the residence time factor F represents the relationship between the residence time of the microorganisms and the residence time of the nutrient medium). A residence time factor F of 5 means that the cell mass (the biocatalyst) is in the cyclone reactor system 5 times longer than the nutrient medium. With an inlet volume flow of 300 ml / h this can be achieved by removing a cell-containing bleed flow of 60 ml / h and a cell-free filtrate flow of 240 ml / h. The filtrate flow is set via the level control if the bleed flow is fixed.
Results:
After 3 cell mass dwell times (75 h) a cell mass concentration of 60 g BTM / I is reached in the cyclone reactor in steady state. At this cell density, a value of 6% air saturation at the measuring point in circulation is measured as pO2.
For the biochemical control of an adequate oxygen supply in the entire cyclone reactor system, the fermentation medium was examined for the final fermentation product lactate, which is only formed when insufficient oxygen is available. No lactate formation was found in the examined operating area.
Compared to the conventional stirred tank process, the use of the cyclone reactor allows the cell mass concentration in the reactor to be increased by a factor of 3 without undesired oxygen limitations. With the cell mass-specific reaction rate of the amino acid formation unchanged, the volumetric productivity ("space-time yield") can also be increased by a factor of 3 compared to the continuous stirred tank process.
