BESCHREIBUNG
Die Erfindung betrifft die neuen 2-Difluormethyl-2-cyanethylmalonsäureniedrigalkylester, ein Verfahren zur Herstellung dieser Verbindungen und die Verwendung dieser Verbindungen zur Herstellung von racemischem Difluormethylornithin.
Es ist bekannt, Difluormethylornithin ausgehend von Ornithin herzustellen (US-PS-4 309442 und US-PS-4 413141). Ornithin ist ein teures Ausgangsprodukt, das infolge der niedrigen Ausbeuten, die nach diesem Verfahren erzieltwerden, zu einem erheblichen Teil verloren geht.
Bei der Synthese müssen weiter beide Aminogruppen des Ornithins, vorzugsweise durch Überführung von Ornithin mit Benzaldehyd in den Dibenzylidenornithinester, geschützt und später unter Verlust der Schutzgruppe wieder freigesetzt werden.
Das Ziel vorliegender Erfindung war es, einen neuen, von leicht zugänglichen Ausgangsprodukten ausgehenden Weg zur Verfügung zu stellen.
Erfindungsgemäss wird dies durch die neuen Zwischenprodukte 2-Difluormethyl-2-cyanethylmalonsäureniedrigalkylester der Formel

worin R ein Niedrigalkylrest bedeutet, erreicht.
Bevorzugte Vertreter dieser Zwischenprodukte sind der 2 Difluormethyl-2-cyanethylmalonsäuremethylester und der 2-Difluormethyl-2-cyanethylmalonsäureethylester.
Die neuen Zwischenprodukte werden ausgehend von Malon säureniedrigalkylestem erhalten. Das Verfahren ist dadurch gekennzeichnet, dass man Malonsäureniedrigalkylester der Formel

in welcher Rein Niedrigalkylrest ist, mit Acrylnitril in Gegenwart eines Alkalimetalles, eines Alkalimetallhydriils, eines Alka- lialkoholats oder eines Alkalihydroxids zum 2-Cyanethylmalonsäureniedrigalkylester umsetzt und diesen in Gegenwart von einem Alkalimetallhydrid oder einem Alkalialkoholat und in einem polaren organischen Lösungsmittel mit Difluorchlormethan umsetzt und den gebildeten 2-Difluormethyl-2-cyanethylmalonsäureniedrigalkylester isoliert.
Unter einem Niedrigalkylrest wird vorzugsweise der Methylsowie der Ethylrest verstanden. Selbstverständlich sind nach dem erfindungsgemässen Verfahren auch längerkettige Esterreste herstellbar. Für den weiteren Verwendungszweck der Zwischenprodukte zur Herstellung von Difluormethylornithin haben solche Verbindungen aber keine grosse Bedeutung.
Als Alkalimetall können Kalium, Lithium oder Natrium eingesetzt werden, vorzugsweise wird Natrium verwendet.
Mit entsprechendem Erfolg können auch Alkalimetallhydride wie Natriumhydrid, Alkalialkoholate wie Natriummethylat oder auch Alkalihydroxide wie Natriumhydroxid verwendet werden.
Bei der Umsetzung mit Acrylnitril geht man zweckmässig so vor, dass das Alkalimetallbzw. das Alkalialkoholat, das Alkalimetallhydrid oder das Alkalihydroxid bei erhöhter Temperatur im Malonsäureniedrigalkylester aufgelöst und anschliessend bei Temperaturen zwischen 10 bis 90 "C das Acrylflitril zugetropft wird. Der entstehende 2-Cyanethylmalonsäureniedrigalkylester wird vorteilhaft durch Destillation isoliert.
Die Alkylierung mit Difluorchlormethan wird in Gegenwart eines Alkalimetallhydrids oder Alkalialkoholats durchgeführt.
Als Alkalimetallhydrid können Kalium- oder Natriumhydrid verwendet werden, vorzugsweise wird Natriumhydrid verwen- det. Als Alkalialkoholat setzt man zweckmässig Alkoholate von niederen aliphatischen Alkoholen wie Natriummethylat, Natriumethylat oder Kalium-tert.-butylat ein.
Geeignete Vertreter eines polaren organischen Lösungsmittels sind N,N-Dimethylformamid, Tetrahydrofuran oder 1,2 Dimethoxyethan, bevorzugtwird aber N,N-Dimethylformamid eingesetzt.
Das gasförmige Difluorchlormethan wird zweckmässig in einem Überschuss von 1 bis 5 Mol, bezogen auf 1 Mol eingesetzten Cyanethylmalonester, eingesetzt.
Die Alkylierung wird zweckmässig bei Temperaturen zwischen 10 bis 80 "C, vorzugsweise zwischen 20 und 40 "C, und bei Drücken zwischen Normaldruck bis 3 bar, vorzugsweise zwischen 2 und 3 bar, durchgeführt.
Das gewünschte Reaktionsprodukt, der 2-Difluormethyl-2- cyanethylmalonsäureniedrigalkylester, wird vorteilhaft durch Destillation isoliert.
Der 2-Difluormethyl-2-cyanethylmalonsäureniedrigalkylester kann erfindungsgemäss weiter zum Difluormethylornithin umgesetzt werden.
Dazu wird zunächst eine Hydrierung mit Wasserstoff in Gegenwart eines Hydrierungskatalysators durchgeführt. Als Katalysatoren können Raney-Nickel-, Raney-Eisen-, Raney Kupfer- oder Raney-Kobaltkatalysatoren oder übliche Edelmetallkatalysatoren, wie Palladium- oder Platinkatalysatoren, in der Regel aufgebracht auf einem Trägermaterial, wie Kohle oder Aluminiumoxid, eingesetzt werden. Vorzugsweise wird ein Raney-Nickelkatalysator verwendet. Die Katalysatormenge bewegt sich zweckmässig zwischen 5 und 15 Mol-%, bezogen auf den eingesetzten 2-Difluormethyl-2-cyanethylmalonsäureester.
Als Lösungsmittel werden zweckmässig Alkohole, wie Methanol oder Ethanol, aber auch Toluol oder Gemische von Alkoholen mit niedersiedenden Erdölfraktionen, wie Benzine eingesetzt.
Vorzugsweise werden Alkohole, die dem Esterrest des Malonesters entsprechen, verwendet.
Die Hydrierungstemperatur liegt vorzugsweise bei 60 bis 110 "C und der Druck bei 5 bis 10 bar.
Das so erhaltene 3-Difluormethyl-3-niedrigalkoxycarbonyl2-piperidon kann auf übliche Weise isoliert werden und darauf nach zwei verschiedenen Wegen weiter in das Difluormethylornithin übergeführt werden.
Der eine Wegführt über das 3-Difluormethyl-3-aminocarbonyl-2-piperidon, das durch Behandeln von 3-Difluormethyl-3 niedrigalkoxycarbonyl-2-piperidon mit Ammoniak erhalten wird. Zweckmässig wird unter einem Ammoniakdruck zwischen Normaldruck und 10 bar vorzugsweise zwischen 5 und 8 bar, und Temperaturen zwischen Raumtemperatur und 70 "C gearbeitet.
Um ein zusätzliches Lösungsmittel zu vermeiden wird vorteilhaft mitkonzentrierterwässriger Ammoniaklösung gearbeitet. Das 3-Difluormethyl-3-aminocarbonyl-2-piperidon wird anschliessend durch Behandeln mit einem Alkalihypochlorit, vorzugsweise mit Natriumhypochlorit, und einem Alkalihydroxid, vorzugsweise mit Natriumhydroxid, bei Temperaturen von 25 bis 60 "C umgesetzt und weiter über das intermediär entstehende 3 Difluormethyl-3-amino-2-piperidon bei Temperaturen von 60 bis 90 "C mit wässriger Salzsäure oder Natronlauge zum Difluormethylornithin hydrolisiert und als Monohydrochloridmonohydrat isoliert.
Zweckmässig wird das Alkalihypochlorit in einem Überschuss von 5 bis 20% und das Alkalihydroxid in einem Überschuss von 1 bis 20% eingesetzt.
Der zweite Weg führt über das 3-Difluormet'nyl-3-hydrazinocarbonyl-2-piperidon, das durch Behandeln des 3-Difluorme- thyl-3-niedrigalkoxycarbonyl-2-piperidons mit Hydrazinhydrat erhalten wird.
Die Reaktion wird zweckmässig in einem organischen Lösungsmittel, vorzugsweise in Alkoholen wie Methanol oder Ethanol, und bei Temperaturen von 30 bis 60 "C durchgeführt.
Das Hydrazinhydrat wird zweckmässig in einem beträchtlichen Überschuss von 5 bis 15 Mol, bezogen auf das eingesetzte Piperidon, verwendet.
Das entstandene 3-Difluormethyl-3-hydrazinocarbonyl-2- piperidon kann, gegebenenfalls nach einer Reinigung durch Umkristallisation, mit einem Alkalinitrit in das 3-Difluormethyl3-azidocarbonyl-2-piperidon überführt werden.
Zweckmässig wird die Umsetzung mit Natriumnitrit in einer verdünnten Mineralsäure, vorzugsweise mit Salzsäure, in Gegenwart eines chlorierten Kohlenwasserstoffes, vorzugsweise Methylenchlorid, durchgeführt. Die Reaktion kann aber auch in einer mit Wasser verdünnten organischen Säure, wie Essigsäure, durchgeführt werden. Das Produkt kann schliesslich mit einem mit Wasser nicht mischbaren organischen Lösungsmittel, vorzugsweise Methylenchlorid oder Ethylacetat, extrahiert werden.
Die Umsetzungstemperaturen bewegen sich vorteilhaft zwi schen 0 und 10 "C.
Nach der Umsetzung kann die die Azidoverbindung enthaltende organische Phase abgetrennt, auf übliche Weise getrocknet und gegebenenfalls mit einem zusätzlichen höhersiedenden Lösungsmittel, wie z. B. Toluol, vermischt, dann einer Tempera turbehandlung von 60 bis 110 "C unterzogen werden, wobei das niedersiedende Lösungsmittel entfernt wird.
Das so erhaltene 3-Difluormethyl-3-isocyanato-2-piperidon kann schliesslich durch Behandeln mit einer Säure, vorzugsweise mit Salzsäure, bei Temperaturen von 60 bis 120 "C in das gewünschte Difluormethylornithin-monohydrochlorid-monohydrat übergeführt werden.
Beispiel 1 a) Herstellung von Cyanethylmalonsäurediethylester
160,2 g (1,0 mol) Malonsäurediethylester und 1,0 g Natriummetall wurden bei 60 "C 2 h gerührt. Die so entstandene gelbe Lösung wurde auf 10 0gekühlt. Dann wurden innert ca. 1 h 34,5 g (0,65 mol) Acrylnitril so zugetropft, dass die Temperatur zwischen 10 und 20 "C gehalten werden konnte. Nach dem Zutropfen wurde das Reaktionsgemisch 1 h bei 10 bis 20 "C gerührt. Man erhielt 195,7ggelbliches Rohprodukt. Dieses wurde bei 107 bis 110 C/1 bis 1,3.102 Pa destilliert. Man erhielt 105,1 g = 49,1% Reinprodukt.
Unter Berücksichtigung von 61,5 gunverbrauchtem und zurückgeführtem Malonsäurediethylester beträgt die Ausbeute 80,6%, bezogen auf eingesetzten Malonsäurediethylester.
b) Herstellung von 2-Difluormetlvl-2-cyanel hylmalonsäuredi ethyl er
Zu einer Suspension von 13,88 g Natriumhydrid (79%ig in Weissöl, 460 mmol) inN,N-Dimethylformamid gab man 86,58 g (400 mmol) Cyanethylmalonsäurediethylester sowie 7,40 g (100 mmol) tert.-Butanol. Im Autoklav wurden in diese Lösung bei 30 "C 173,0 g (2,00 mol) Difluormethan innert 10 min eingeleitet.
Nach 6 h bei 30 "C und einem Druck von 2 bis 3 bar wurde der Autoklav entspannt und das Reaktionsgemisch fil triert. Vom Filtrat wurde vorerst das Lösungsmittel, dann das Produkt selbst abdestilliert (1 mbar/100 "C). Man erhielt 89,88 g 2-Difluormethyl-2-cyanethylmalonsäurediethylester, entsprechend einer Ausbeute von 81,6%, bezogen auf eingesetzten Cyanethylmalonsäurediethylester.

Entsprechend Beispiel la) und lb) nur mit Malonsäuredimethylester als Ausgangsprodukt wurde mit gleicher Ausbeute der 2-Difluormethyl-2-cyanethylmalonsäure-dimethylester hergestellt.

Beispiel 3
Herstellung von 3-Difluormethyl-3-ethoxycarbonyl-2-piperidon
15,0 g (53,5 mmol) 2-Difluormethyl-2-cyanethylmalonsäure- diethylester wurden in 235 g Ethanol gelöst und bei 90 "C/6 bar über 1,46 g (10 Gew.-%) Raney-Nickel hydriert. Nach 3 h wurde das Reaktionsgemisch abgekühlt, durch Filtration vom Katalyt abgetrennt und am Rotationsverdampfer eingeengt.
Durch Urncristailisation aus 50 ml Ethanolin-Hexan 1:4 erhielt man
9,06 g 3-Difluormethyl-3-ethoxycarbonyl-2-piperidon in einer Ausbeute von 75,3% bezogen auf eingesetzten 2-Difluormethyl2-cyanethylmalonsäurediethylester.

Beispiel 4 a) Herstellung von 3-Aminocarbonyl-3-difluormethyl-2-pipe ridon
8,01 g (34,1 mmol) 3-Difluormethyl-3-ethoxycarbonyl-2 piperidon wurden im Autoklavin 80 g 23% Ammoniumhydro xidlösung gelöst. Unter einem Ammoniakdruck von 7 bar wurde das Gemisch 10 h bei 40 0C gerährt. Nach Abkühlen und
Entspannen des Autoklaven engte man die Reaktionslösung am
Rotationsverdampfer auf ein Gewicht von ca. 20 g ein und filtrierte das ausgefallene Produkt ab.
Man erhielt 5,40 g 3 Aminocarbonyl-3-difluormethyl-2-piperidon in einer Ausbeute von 81,2% bezogen auf eingesetztes Edukt.

2,93 g (14,5 mmol) 3-Aminocarbonyl-3-difluormethyl-2-piperidon wurden in 10 g Wasser suspendiert, auf 50 "C erhitzt und mit einer Lösung von 1,22 g Natriumhypochlorit (16,4 mmol) und 1,74 g Natriumhydroxid (43,5 mmol) in 50 g Wasser versetzt.
Die klare Lösung wurde 30 min bei 50 "C und 2 h bei 80 "C gerührt, auf 25 "C abgekühlt, mit 38 g 7%iger Salzsäure (76 mmol) versetzt und zur Trockene eingeengt. Den Rückstand extrahierte man mit 117 g heissem Ethanol in 3 Portionen. Das Ethanolfiltrat wurde zur Trockene eingeengt. Das Rohprodukt wurde in 3,2 g Wasser gelöst und mit 1,10 g (11 mmol)Triethylaminversetzt, wodurch ein pH-Wert zwischen 3,5 und 4,0 errericht wurde. Nach Zugabe von 23 g Ethanol liess man das Gemisch 12 h bei -5 "C stehen, filtrierte das ausgefallene Produkt ab, wusch es mit 5g Ethanol und trocknete es am Hochvakuum.
Man erhielt 2,30 gD,L-a- Difluormethylornithin-monohydrochlorid-monohydrat entsprechend einer Ausbeute von 64,4%, bezogen auf das Edukt.

Beispiel 5 a) Herstellung von 3-Difluormethyl-3-hydrazinocarbonyl-2-pi peridon
12,50 g (55,8 mmol) 3-Difluormethyl-3-ethoxycarbonyl-2piperidon wurden in 100 ml Ethanol gelöst, mit 27,95 g Hydrazinhydrat (100%ig, 558 mmol) versetzt und 23 h auf 50 "C erhitzt.
Überschüssiges Hydrazin und Ethanol wurden abdestilliert und der Rückstand aus 160 ml Isopropanol umkristallisiert. Man erhielt 10,53 g 3-Difluormethyl-3-hydrazinocarbonyl-2-piperidon entsprechend einer Ausbeute von 90,1% bezogen auf eingesetztes Edukt.

5,09 g (24,1 mmol) 3-Difluormethyl-3-hydrazinocarbonyl-2piperidon,
40 ml 1M- Salzsäure und 50 ml Methylenchlorid wurden bei 0 "C vorgelegt und innert 10 min mit einer Lösung von
2,32gNatriumnitrit (34mmol) in 10 ml Wasser versetzt. Nach einer Stunde bei 0 C wurden die beiden Phasen getrennt, die organische Phase mit Natriumbikarbonatlösung und Wasser gewaschen, über Natriumsulfat getrocknet und mit 100 ml Toluol vermischt. Diese Lösung wurde langsam auf 80 "C erhitzt, wobei man das Methylenchlorid abdestillieren liess. Nach 30 min bei
80 C wurde das Gemisch abgekühlt, mit 100 ml 20%iger Salz säure vermischt, unter gutem Rühren während 2 h bei Rückflus stemperatur gehalten, wieder abgekühlt und am Rotationsver dampfer zur Trockene eingeengt.
Man löste das Rohprodukt in 5 g Wasser, gab 1,5 g Triethylamin (15 mmol) zu, verdünnte mit 50 ml Ethanol und liess das Gemisch 2 h bei 0 C stehen. Das ausgefallene Produkt wurde abfiltriert und getrocknet. Man erhielt 2,94 g D ,L-a-Difluormethylornithin-monohydrochlorid- monohydrat (Gehalt96,5%, HPLC) in einer Ausbeute von 49,8%, bezogen auf das Edukt.

Beispiel 6
Herstellung von 3-Difluormethyl-3-azidocarbonyl-2-piperidon
2,09 g (10 mmol) 3-Difluormethyl-3-hydrazinocarbonyl-2- piperidon wurden in 30 ml Eisessig und 4 ml Wasser vorgelegt und bei 10 C mit 2,03 g(20 mmol) konz. Salzsäure versetzt. Bei einer Temperatur von 5 C gab man innert 10 min eine Lösung von 0,90 g (13 mmol) Natriumnitritin 2 ml Wasser zu und liess das
Gemisch 3 h bei 0 C rühren. Anschliessend wurde mit 1n Natronlauge auf pH 5 gestellt und mit 3 x50 ml Ethylacetat extrahiert.
Die vereinigten organischen Phasen wurden mit 70 ml Wasser gewaschen, mit Natriumsulfat getrocknet und zur Trok kene eingeengt, wodurch man2,10 g 3-Difluormethyl-3-azido carbonyl-2-piperidon erhielt.
Ausbeute: 96%

Beispiel 7
Herstellung von 3-Difluormethyl-3-isocyanato-2-piperidon
2,09 g (10 mmol) 3-Difluormethyl-3-hydrazinocarbonyl-2- pipendon wurden in 30 ml Eisessig und 4 ml Wasser vorgelegt und bei 10 C mit 2,03 g (20 mmol) konz. Salzsäure versetzt. Bei einer Temperatur von 5 C gab man innert 10 min eine Lösung von 0,90 g (13 mmol) Natriumnitrit in 2 ml Wasser zu und liess das Gemisch 2 h bei 0 C rühren. Nach Zugabe von 3,12 g (29 mmol) Natriumcarbonat wurde das Gemisch mit 3 x 50 ml Methylenchlorid extrahiert.
Die vereinigten organischen Phasen wurden mit 70 ml Wasser gewaschen, mit Natriumsulfat getrocknet, auf 50 ml eingeengt und mit 50 ml Toluol versetzt. Unter Abdestillieren von Methylenchlorid erhitzte man die Mischung während 1h auf 80 C und engte anschliessend zur Trockene ein, wodurch 1,81 g 3-Difluormethyl-3-isocyanato-2-piperidon erhalten wurden.
Ausbeute: 95%

DESCRIPTION
The invention relates to the new 2-difluoromethyl-2-cyanoethylmalonic acid lower alkyl esters, a process for the preparation of these compounds and the use of these compounds for the preparation of racemic difluoromethylornithine.
It is known to produce difluoromethylornithine from ornithine (US Pat. Nos. 4,309,442 and 4,413,141). Ornithine is an expensive starting material, which is largely lost due to the low yields achieved by this process.
During the synthesis, both amino groups of ornithine must be protected, preferably by converting ornithine with benzaldehyde into the dibenzylidene ornithine ester, and later released again with loss of the protective group.
The aim of the present invention was to provide a new way starting from easily accessible starting products.
According to the invention, this is achieved by the new intermediates 2-difluoromethyl-2-cyanoethylmalonic acid lower alkyl ester of the formula

where R represents a lower alkyl radical.
Preferred representatives of these intermediates are the 2-difluoromethyl-2-cyanoethylmalonic acid methyl ester and the 2-difluoromethyl-2-cyanoethylmalonic acid ethyl ester.

in which pure is lower alkyl radical, with acrylonitrile in the presence of an alkali metal, an alkali metal hydride, an alkali metal alcoholate or an alkali metal hydroxide to give 2-cyanoethylmalonic acid lower alkyl ester, which is reacted in the presence of an alkali metal hydride or an alkali metal alcoholate and in a polar organic solvent and with a difluoromethylchloromethane 2-difluoromethyl-2-cyanoethylmalonic acid lower alkyl ester isolated.
A lower alkyl radical is preferably understood to mean the methyl and the ethyl radical. Of course, longer-chain ester residues can also be produced by the process according to the invention. However, such compounds are of little importance for the further use of the intermediates for the preparation of difluoromethylornithine.
Potassium, lithium or sodium can be used as the alkali metal; sodium is preferably used.
With corresponding success, alkali metal hydrides such as sodium hydride, alkali metal alcoholates such as sodium methylate or alkali metal hydroxides such as sodium hydroxide can also be used.
In the reaction with acrylonitrile, the procedure is appropriately such that the alkali metal or. the alkali metal alcoholate, the alkali metal hydride or the alkali metal hydroxide is dissolved at elevated temperature in the lower alkyl malonate and the acrylic flitrile is then added dropwise at temperatures between 10 to 90 ° C. The resulting 2-cyanoethyl malonyl lower alkyl ester is advantageously isolated by distillation.
The alkylation with difluorochloromethane is carried out in the presence of an alkali metal hydride or alkali alcoholate.
Potassium or sodium hydride can be used as the alkali metal hydride; sodium hydride is preferably used. The alkali alcoholate used is advantageously alcoholates of lower aliphatic alcohols, such as sodium methylate, sodium ethylate or potassium tert-butoxide.
Suitable representatives of a polar organic solvent are N, N-dimethylformamide, tetrahydrofuran or 1,2 dimethoxyethane, but N, N-dimethylformamide is preferred.
The gaseous difluorochloromethane is expediently used in an excess of 1 to 5 mol, based on 1 mol of cyanethyl malonic ester used.
The alkylation is expediently carried out at temperatures between 10 to 80 ° C., preferably between 20 and 40 ° C., and at pressures between normal pressure and 3 bar, preferably between 2 and 3 bar.
The desired reaction product, the 2-difluoromethyl-2-cyanoethylmalonic acid lower alkyl ester, is advantageously isolated by distillation.
According to the invention, the lower alkyl 2-difluoromethyl-2-cyanoethylmalonic acid can be further converted to difluoromethylornithine.
For this purpose, hydrogenation with hydrogen is first carried out in the presence of a hydrogenation catalyst. Raney nickel, Raney iron, Raney copper or Raney cobalt catalysts or customary noble metal catalysts, such as palladium or platinum catalysts, generally applied to a support material such as carbon or aluminum oxide, can be used as catalysts. A Raney nickel catalyst is preferably used. The amount of catalyst suitably ranges between 5 and 15 mol%, based on the 2-difluoromethyl-2-cyanoethylmalonic acid ester used.
Suitable solvents are alcohols, such as methanol or ethanol, but also toluene or mixtures of alcohols with low-boiling petroleum fractions, such as gasolines.
Alcohols which correspond to the ester radical of the malonic ester are preferably used.
The hydrogenation temperature is preferably 60 to 110 ° C. and the pressure 5 to 10 bar.
The 3-difluoromethyl-3-lower alkoxycarbonyl2-piperidone obtained in this way can be isolated in a conventional manner and then further converted into the difluoromethylornithine in two different ways.
One route is via 3-difluoromethyl-3-aminocarbonyl-2-piperidone, which is obtained by treating 3-difluoromethyl-3-lower alkoxycarbonyl-2-piperidone with ammonia. It is expedient to work under an ammonia pressure between normal pressure and 10 bar, preferably between 5 and 8 bar, and temperatures between room temperature and 70 ° C.
In order to avoid an additional solvent, it is advantageous to work with concentrated aqueous ammonia solution. The 3-difluoromethyl-3-aminocarbonyl-2-piperidone is then reacted by treatment with an alkali metal hypochlorite, preferably with sodium hypochlorite, and an alkali metal hydroxide, preferably with sodium hydroxide, at temperatures of 25 to 60 ° C. and further via the intermediate 3 difluoromethyl 3-amino-2-piperidone hydrolyzed at temperatures from 60 to 90 ° C. with aqueous hydrochloric acid or sodium hydroxide solution to the difluoromethylornithine and isolated as monohydrochloride monohydrate.
The alkali metal hypochlorite is advantageously used in an excess of 5 to 20% and the alkali metal hydroxide in an excess of 1 to 20%.
The second route leads via the 3-difluoromet'nyl-3-hydrazinocarbonyl-2-piperidone, which is obtained by treating the 3-difluoromethyl-3-lower alkoxycarbonyl-2-piperidone with hydrazine hydrate.
The reaction is expediently carried out in an organic solvent, preferably in alcohols such as methanol or ethanol, and at temperatures from 30 to 60 ° C.
The hydrazine hydrate is advantageously used in a considerable excess of 5 to 15 mol, based on the piperidone used.
The resulting 3-difluoromethyl-3-hydrazinocarbonyl-2-piperidone can be converted into the 3-difluoromethyl3-azidocarbonyl-2-piperidone, if necessary after purification by recrystallization, with an alkali metal nitrite.
The reaction is expediently carried out with sodium nitrite in a dilute mineral acid, preferably with hydrochloric acid, in the presence of a chlorinated hydrocarbon, preferably methylene chloride. However, the reaction can also be carried out in a water-diluted organic acid, such as acetic acid. The product can finally be extracted with a water-immiscible organic solvent, preferably methylene chloride or ethyl acetate.
The reaction temperatures are advantageously between 0 and 10 "C.
After the reaction, the organic phase containing the azido compound can be separated off, dried in a customary manner and optionally with an additional higher-boiling solvent, such as. As toluene, mixed, then subjected to a temperature treatment from 60 to 110 "C, the low-boiling solvent being removed.
The 3-difluoromethyl-3-isocyanato-2-piperidone thus obtained can finally be converted into the desired difluoromethylornithine monohydrochloride monohydrate by treatment with an acid, preferably with hydrochloric acid, at from 60 to 120 ° C.
