BESCHREIBUNG
Die vorliegende Erfindung betrifft ein chromatographisches Trennelement für eine Planar-Chromatographieeinrichtung, welches eine untere Tragplatte, eine obere Deckplatte und ein zwischen den beiden angebrachtes adsorbierendes Medium enthält.
Bei der herkömmlichen Vorrichtung ist das adsorbierende Medium auf die den Rotor bildende oder auf den Rotor befestigte Glasplatte mit geeignetem Bindemittel aufgeklebt.
(S. US-Patentschrift Nr. 4 139 458.) Der Nachteil dieser Lösung ist, dass nur solche adsorbierende Medien angewendet werden können, die klebbar sind.
Es wurden Versuche gemacht diesen Nachteil zu beseiti gen, und zwar mit einer Scheibe, bei welcher das adsorbie rende Medium zwischen zwei parallelen kreisförmigen Glas platten angeordnet wird. (S. die Vorrichtung von Hitachi
Corp.) Der Nachteil dieser Lösung ist, dass der auf den
Halbmesser senkrechte Querschnitt des adsorbierenden Me diums nicht konstant ist, d.h. die Massenkraft verändert sich mit der Entfernung von dem Mittelpunkt, deshalb kommt eine Streifenverbreiterung zustande.
Die Aufgabe der Erfindung ist ein Trennelement zustan dezubringen, bei welchem das adsorbierende Medium die
Erfordernisse der Säulenchromatographie erfüllt.
Erfindungsgemäss wird diese Aufgabe dadurch gelöst, dass zwischen der unteren Tragplatte und der oberen Deckplatte ein Spalt vorhanden ist, und in dem Spalt das adsorbierende Medium kreisförmig angeordnet ist, und sich die Höhe des Spaltes von der Achse des Trennelementes ab in Richtung des Halbmessers gemäss der Gleichung

ändert, wo K, a, b und c konstant sind, und am Rand des Trennelementes in dem Spalt ein Abdeckfilter angeordnet ist.
Zweckmässig verringert sich die Höhe des Spaltes in umgekehrtem Verhältnis zu einer von der Achse des Elementes ab in Richtung des Halbmessers nach aussen gemessenen Entfernung, d.h. der auf die Richtung des Halbmessers senkrechte Querschnitt des Spaltes ist konstant: h = Kr.
Ist der Querschnitt des Spaltes in Richtung des Halbmessers konstant, so wird das Erfordernis der Säulenchromatographie, namentlich, dass der Querschnitt des adsorbierenden Mediums auf die Strömungsrichtung des Lösungsmittels senkrecht streng konstant sein muss, erfüllt. Davon ergibt sich die in der Säulenchromatographie vorkommenden Füllungen ohne Bindemittel angewendet werden können. Ein weiterer Vorteil dieser Lösung ist, dass die Auffüllung der Säule einfach und gut reproduzierbar ist.
Die Ausführung, gemäss welcher sich die Höhe des Spaltes gemäss der Gleichung h = K/r2 ändert, d.h. die Massenkraft entlang des Halbmessers von der Achse des Trennelementes ab gemessen konstant ist, ist deshalb vorteilhaft, weil dadurch die Strömungsgeschwindigkeit des Lösungsmittels konstant ist, so ergibt sich die Möglichkeit, die vom Standpunkt der Trennung aus optimale Strömungsgeschwindigkeit einzustellen.
Bei einer anderen Ausführungsform ist das Trennelement um die Achse drehbar angeordnet, und in einer der Platten sind auf die Achse konzentrische ringförmige Kanäle zum Einführen des Lösungsmittels ausgebildet und die dem adsorbierenden Medium zugewendeten Enden der Kanäle sind mit einer Filterplatte abgedeckt. Diese Ausführungsform hat den Vorteil, dass das Lösungsmittel zwischen der Achse und dem Rand des Trennelementes an irgendeinem Punkt oder an irgendwelchen Punkten eingeführt werden kann, wodurch die Wirksamkeit der Trennung beeinflusst werden kann.
Im folgenden wird die Erfindung anhand der Zeichnung näher erläutert. Es zeigen rein schematisch: Fig. 1 das erfindungsgemässe Trennelement in Seitenansicht, teilweise im Schnitt, in ungefülltem Zustand; Fig. 2 das erfindungsgemässe Trennelement in Seitenansicht, teilweise im Schnitt, in mit adsorbierendem Medium gefülltem Zustand; Fig. 3 eine andere Ausführungsform des Trennelementes in Seitenansicht, teilweise im Schnitt, in welchem konzentrische ringförmige Kanäle ausgebildet sind.
In Fig. 1 und 2 ist ein Trennelement dargestellt, bei welchem auf eine Achse 4 eine untere Tragplatte 1 und eine obere Deckplatte 2 montiert sind. Zwischen den beiden Platten ist ein Spalt 3, in dem ein adsorbierendes Medium 5 kreisförmig angeordnet ist. Die Höhe h des Spaltes 3 ändert sich von der Achse 4 des Trennelementes ab in Richtung des Halbmessers r gemäss der Gleichung

wo K, a, b und c konstant sind. Am Rand 7 des Trennelementes ist ein ringförmiger Abdeckfilter 6 angeordnet, welcher den Spalt 3 nach aussen absperrt und das in dem Spalt angebrachte adsorbierende Medium 4 in dem Spalt hält, wobei der Abdeckfilter 6 das Lösungsmittel durchlässt.
Die Höhe h des zwischen der unteren Tragplatte 1 und der oberen Deckplatte 2 vorhandenen Spaltes 3 kann sich in Richtung des Halbmessers r von der Achse 4 ab gemessen nach aussen gemäss der Gleichung h = Kir ändern, falls a = c = 0 und b = 1. In diesem Fall ist der auf die Richtung des Halbmessers senkrechte Querschnitt des Spaltes 3 konstant.
Die Höhe des zwischen der unteren Tragplatte 1 und der oberen Deckplatte 2 vorhandenen Spaltes 3 kann sich in Richtung des Halbmessers r von der Achse 4 ab gemessen nach aussen auch gemäss der Gleichung h = K/r2 ändern, falls a = b = 0 und c = 1. In diesem Fall ist die Massenkraft entlang des Halbmessers r konstant.
Es kann auch ein Trennelement angewendet werden, das um die Achse 4 drehbar ist, und bei welchem entweder in der Tragplatte 1 oder in der Deckplatte 2 auf die Achse 4 konzentrische ringförmige Kanäle 8 ausgebildet sind. Diese Kanäle 8 dienen zum Einführen des Lösungsmittels an verschiedenen Punkten. Die dem adsorbierenden Medium zugewandten Enden der Kanäle 8 sind mit einer Filterplatte 9 abgedeckt, damit das adsorbierende Medium 5 nicht in die Kanäle 8 hineindringt.
Das erfindungsgemässe Trennelement wird auf folgende Weise angewendet:
Das Lösungsmittel wird in der Nähe der Achse 4 in Richtung des Pfeiles F(Fig. 1) eingeführt und strömt durch das adsorbierende Medium 5 in Richtung des Halbmessers r nach aussen. Inzwischen geht die chromatographische Trennung vor sich. Das Lösungsmittel verlässt das Trennelement in Richtung des Pfeiles G.
Das Lösungsmittel strömt durch das adsorbierende Medium 5 entweder unter Wirkung der Zentrifugalkraft. falls das Trennelement dadurch in Drehung gesetzt wird, dass die Achse 4 in bekannter Weise mit einem elektrischen Motor angetrieben wird. Das Trennelement kann aber auch stehend verwirklicht werden, dann wird das Lösungsmittel pneumatisch durch das adsorbierende Medium 5 getrieben. In diesem Fall kann das Durchströmen des adsorbierenden Mediums auch in verkehrter Richtung stattfinden, d.h. das Lösungsmittel kann am Rand 7 des Trennelementes eingeführt und in der Nähe der Achse 4 entfernt werden.
Falls ein Trennelement angewendet wird, in welchem konzentrische ringförmige Kanäle 8 ausgebildet sind, dann besteht die Möglichkeit, das Lösungsmittel zwischen der Achse 4 und dem Rand 7 des Trennelementes an irgendeinem Punkt einzuführen, wodurch die Wirksamkeit der Trennung beeinflusst werden kann.

The height of the gap expediently decreases in inverse proportion to a distance measured outwards from the axis of the element in the direction of the radius, i.e. the cross section of the gap perpendicular to the direction of the radius is constant: h = Kr.
If the cross section of the gap is constant in the direction of the radius, then the requirement of column chromatography, namely that the cross section of the adsorbing medium must be strictly constant perpendicular to the direction of flow of the solvent, is met. This means that the fillings found in column chromatography can be used without binders. Another advantage of this solution is that the filling of the column is easy and reproducible.
The design according to which the height of the gap changes according to the equation h = K / r2, i.e. the mass force along the radius from the axis of the separating element is constant, is advantageous because it makes the flow rate of the solvent constant, so there is the possibility of setting the optimal flow rate from the point of view of the separation.
In another embodiment, the separating element is arranged so as to be rotatable about the axis, and in one of the plates there are formed annular channels concentric to the axis for introducing the solvent, and the ends of the channels facing the adsorbent medium are covered with a filter plate. This embodiment has the advantage that the solvent can be introduced at any point or at any point between the axis and the edge of the separating element, whereby the effectiveness of the separation can be influenced.

The separating element according to the invention is used in the following way:
The solvent is introduced in the vicinity of the axis 4 in the direction of the arrow F (FIG. 1) and flows out through the adsorbing medium 5 in the direction of the radius r. The chromatographic separation is now taking place. The solvent leaves the separating element in the direction of arrow G.
The solvent flows through the adsorbing medium 5 either under the action of the centrifugal force. if the separating element is rotated in that the axis 4 is driven in a known manner with an electric motor. However, the separating element can also be realized in an upright position, then the solvent is pneumatically driven through the adsorbing medium 5. In this case, the flow through the adsorbing medium can also take place in the wrong direction, i.e. the solvent can be introduced at the edge 7 of the separating element and removed in the vicinity of the axis 4.
If a separating element is used, in which concentric annular channels 8 are formed, then there is the possibility of introducing the solvent at any point between the axis 4 and the edge 7 of the separating element, whereby the effectiveness of the separation can be influenced.
