Verfahren gemäss Patentanspruch 3, dadurch gekennzeich net. dass man in einem ersten Schritt a) 4-Chloracetessigsäuremethylester mit Orthoameisensäuretrimethylester in Gegenwart eines sauren Ionenaustauschers umsetzt. und den entstehenden 4-Chlor-3-methoxy-2-E-butensäuremethylester isoliert, dann in einem zweiten Schritt b) das Reaktionsprodukt mit wässriger NH3-Lösung zum 4 Methoxy-3-pyrrolin-2-on umsetzt, isoliert und dann in einem letzten Schritt c) mit Bromessigsäureethylester in Gegenwart von Natriumhydrid zum Endprodukt alkyliert.
5. Verfahren gemäss einem der Patentansprüche 3 oder 4.
dadurch gekennzeichnet. dass man für die Umsetzung zum 4 ( C l-C )-alkoxy-3-pyrrolin-2-on wässrige NH-Lösung einer Kon zentration zwischen 10% und 25C/c anwendet.
6. Verfahren gemäss einem der Patentansprüche 3. 4 oder 5 dadurch gekennzeichnet dass man für die Umsetzung zum 4 (C -C )-alkoxy-3-pvrrolin-2-on das Molverhältnis 4-Haiogen-3- ( C -C'* )-allioxv-'-E-hutensäure-(C,-C4)-alkvlester zurwässrigen Nfl-Losune zwischen 1 zu 2 und 1 zu 10 wählt.

BESCHREIBUNG
Die Erfindung betrifft neue 4-Alkoxy-3-pyrrolin-2-on-l-yl- essigsäurealkylester sowie ein Verfahren zu deren Herstellung.
4-Alkoxy-3-pyrrolin-2-on-1-yl-essigsäurealkylester sind wertvolle, stabile Zwischenprodukte für die Synthese des cerebral aktiven 4-Hydroxy-2-oxo-pyrrolidin-1-yl-acetamids.
Sämtlichen bisher bekannten Verfahren zur Herstellung des 4-Hydroxy-2-oxo-pyrrolidin-1-yl-acetamids haftet (wegen aufwendigen chemischen Prozessen mit teuren Ausgangsmaterialien und schlechten Ausbeuten) der Makel der Unrentabilität an.
Es stellte sich daher die Aufgabe, einen Weg zu finden, der die genannten Nachteile ausschaltet.
Diese Aufgabe konnte auf überraschende Weise durch das Auffinden der neuen Zwischenprodukte der 4-Alkoxy-3-pyrrolin-2-on-1-yl-essigsäurealkylester der allgemeinen Formel

worin R1 = Alkyl mit 1 bis 2 C-Atomen und R2 = Alkyl mit 1 bis 4 C-Atomen bedeutet, gelöst werden.
Diese neuen Verbindungen, bevorzugt der 4-Methoxy-3pyrrolin-2-on-1-yl-essigsäureethylester, sind erfindungsgemäss auf einfache Weise, gemäss einem Verfahren nach Patentanspruch 3, herstellbar.
So wird auf literaturbekannte Weise in einem ersten Schritt ein 4-Halogenacetessigsäure-C1-C4-alkylester. vorzugsweise ein 4-Chloracetessigsäure-C1-C4-alkylester, besonders bevorzugt der 4-Chloracetessigsäuremethylester mit Orthoameisensäure C,-C2-trialkylester. bevorzugt mit dem Orthoameisensäuretrimethylester, in Gegenwart eines sauren Ionenaustauschers umgesetzt.
Der resultierende 4-Halogen-3-(C1-C2)-alkoxy-2-E-buten- säure-(Cl-C4)-alkylester wird in der zweiten Stufe mit wässriger NH-Lösung einer Konzentration von zweckmässig 10% bis 25Z, vorzugsweise 20% bis 25%, bei Temperaturen von zweckmässig 20 bis 100 "C. vorzugsweise 60 bis 80 "C. während 1 bis 10 h umgesetzt.
Das Molverhältnis 4-Halogen-3-(C1-C2)-alkoxy-2-E-buten- säure-(C 1-C4)-alkylester zu NH; liegt zweckmässig zwischen 1 zu 2 und 1 zu 10. vorteilhaft zwischen 1 zu 5 und 1 zu 7.
Nach üblicher Aufarbeitung. z. B. durch Extraktion mit geeigneten Lösungsmitteln aus der Gruppe der halogenierten Kohlenwasserstoffe, wie u. a. Methylenchlorid und Chloroform.
kann das entsprechende 4-(C,-C2)-alkoxy-3-pyrrolin-2-on in guter Ausbeute erhalten werden.
Dieses kann gegebenenfalls nach zusätzlicher Reinigung durch z. B. Umkristallisieren in aromatischen Kohlenwasserstoffen bevorzugt in Toluol, für die letzte Stufe eingesetzt werden.
Darin wird das 4-(C1-C2)-alkoxy-3-pyrrolin-2-on. bevorzugt das 4-Methoxy-3-pyrrolin-2-on. mit einem 2-Bromessigsäure-C1-C4- alkylester. bevorzugt mit dem 2-Bromessigsäureethylester. in Gegenwart eines Alkalihydrids. vorteilhaft mit Natriumhydrid.

Das Molverhältnis der eingesetzten Verbindungen 4-(C,-C2)- alkoxy-3-pyrrolin-2-on zu 2-Bromessigsäure-C1-C4-alkylester zu Alkalihydrid liegt zweckmässig zwischen 1 zu 1 zu 1 und 1 zu 1,5 zu 1.4, vorzugsweise 1 zu 1,2 zu 1,2 und 1 zu 1,4 zu 1,4.
Die Reaktionstemperatur wählt man zweckmässig zwischen 0 und 50 "C.
Die Reaktion führt man vorteilhaft in gegenüber Alkalihydri den inerten Lösungsmitteln. zweckmässig in Acetonitril, Tetrahydrofuran oder Dimethoxyethan, vorzugsweise in Acetonitril, durch.
Nach üblicher Aufarbeitung, z. B. durch Extraktion, gegebenenfalls anschliessender Reinigung durch Kristallisation, können in sehr guter Ausbeute die erfindungsgemässen Zwischenprodukte erhalten werden.
Beispiel a) Herstellung von 4-Chlor-3-methoxy-2-E-butensäuremethyl ester
31,0 g (0,2 mol) 4-Chloracetessigsäuremethylester wurden mit 106,0 g (1,0 mol) Orthoameisensäuretrimethylesterver- mischt. Unter Argon gab man 30,0 g Amberlyst-15-Ionenaustauscherharz unter Rühren hinzu. Unter heftiger Gasentwicklung stieg die Reaktionstemperatur auf 40 "C. Nach 5 h Rühren liess sich im Dünnschichtchromatogramm kein Edukt mehr nachweisen. Man filtrierte vom Ionenaustauscherharz ab und destillierte den Rückstand im Wasserstrahlvakuum. Das Destillat wurde mit 1,0 g p-Toluolsulfonsäure-monohydratversetztundlangsam auf 150 "C erhitzt, wobei Methanol abdestillierte.
Die Reaktionsmasse wurde anschliessend im Wasserstrahlvakuum destilliert.
Man erhielt 24,7 g einer farblosen Flüssigkeit mit einem Siedepunkt Kp12 = 93 "C
NMR (CDCl3) Ï = 5,16 (s, 1H); 4,67 (s, 2H); 3,73 (s, 6H)
Ausbeute: 75% b) Herstellung von 4-Methoxy-3-pvrrolin-2-on
32,9 g (0,2 mol) 4-Chlor-3-methoxy-2-E-butensäuremethylester wurden mit 82,0 g (1,2 mol) 25ScigwässrigerNH3-Lösung versetzt und unter kräftigem Rühren auf 80 "C erhitzt. Nach 1 h Nachrühren liess man auf Raumtemperatur abkühlen und extrahierte die wässrige Lösung viermal mit je 200,0 ml Methylenchlorid. Nach Trocknen der organischen Phase über Na2SO4 wurde abfiltriert, eingedampft und der Rückstand aus 120,0 ml Toluol heiss umkristallisiert.
Man erhielt 16,9 gweisses. kristallines Produkt vom Schmelzpunkt 132 bis 133 "C.
Ausbeute: 74,8%
NMR (CDCl3) Ï = 6,92 (br. s, IH); 5,07 (s, 1H); 3,93 (s, 2H); 3,82 (s, 3H).
c) Herstellung von 4-Methoxy-3-pyrrolin-2-on-1-yl-essigsäure methylester
11,3 g (0,1 mol) 4-Methoxy-3-pyrrolin-2-on und 24,2 g (0,137 mol) Bromessigsäuremethylester (95%ig) wurden in 150,0 ml Acetonitril suspendiert. Bei O0Cversetzte man die Suspension mit 4,1 g (0,137 mol) 80%igemNatriumhydrid.
Anschliessend liess man die Reaktionslösung auf Raumtemperatur erwärmen und rührte 90 min nach. Nach Abdampfen des Lösungsmittels wurde der Rückstand mit 150,0 ml Eiswasser aufgeschlämmtund viermal mit je 400,0 ml Methylenchlorid extrahiert. Nach üblicher Aufarbeitung wurde der obige Rückstand mit 100,0 ml Petrolether (50 bis 70 "C) versetzt und über Nacht im Kühlschrank aufbewahrt. Das auskristallisierte Produkt wurde abgenutscht und im Hochvakuum getrocknet.
Man erhielt 19,6 g gelblich gefärbtes Produkt vom Schmelzpunkt 55 bis 58 "C.
Ausbeute: 98,5% NMR (CDCl3) Ï = 5,11 (s, 1H);4,69 (q, J=7,0Hz, 2H);4,66 (s, 2H); 3,99 (3, 2H); 3,83 (s, 3H); 1,28 (t, J=7,0 Hz, 3H);
MS (70 eV) m/z = 199 (M+, 25); 126 (100).

4-Alkoxy-3-pyrrolin-2-one-1-yl-acetic acid alkyl esters are valuable, stable intermediates for the synthesis of cerebrally active 4-hydroxy-2-oxo-pyrrolidin-1-yl-acetamide.
All previously known processes for the production of 4-hydroxy-2-oxo-pyrrolidin-1-yl-acetamide are subject to the flaw of unprofitability (due to complex chemical processes with expensive starting materials and poor yields).
It was therefore the task to find a way that eliminates the disadvantages mentioned.
