Description
The present invention relates to novel compositions containing cephradine and clavulanic acid, in which the clavulanic acid has a strong synergistic effect on cephradine, the antibiotic strength of which is almost doubled.
It is known that cephradine is a broad spectrum antibiotic which is widely used in human therapy, while the clavulanic acid has only a weak antibiotic activity. It was therefore not foreseeable that the combination of clavulanic acid with cephradine would lead to such an increase in the activity of the latter as has been found in practice.
Zur Bestimmung der antibakteriellen Aktivität der neuen Kombination wurden zwei Untersuchungen durchgeführt: Two tests were carried out to determine the antibacterial activity of the new combination:
A - Bestimmung der geringsten wachstumshemmenden Konzentration (MIC) an Gram-positiven und Gramnegativen Bakterien; A - Determination of the lowest growth-inhibiting concentration (MIC) of Gram-positive and Gram-negative bacteria;
B - Determination of the sensitivity of Gram-positive and Gram-negative bacteria in agar.
A - Die Aktivität von Cephradin und Cephradin plus Clavulansäure (2:1) wurde an einigen Bakterienstämmen unersucht, die vom Institut Universitaire de Microbiologie - Centre de Collection de type microbien CH -1011 - Lausanne, erhalten wurden: A - The activity of cephradin and cephradin plus clavulanic acid (2: 1) was investigated on some bacterial strains obtained from the Institut Universitaire de Microbiologie - Center de Collection de type microbien CH -1011 - Lausanne:

The above-mentioned bacteria were examined both from the morphological point of view and in terms of their biochemical characteristics before they were used.
The nutrient medium used was broth (Difco), while agar was preferred to preserve the strains.
The various concentrations of both cephradin and the combination of cephradin + clavulanic acid (2: 1) were obtained by preparing a mother liquor, each of which was made double dilutions in 5 ml of the nutrient medium; each of these series was used for a different bacterial strain.
Die Animpfungen der Bakterienstämme wurden aus den vorerwähnten Kulturen der einzelnen Stämme 12 bis 14 The inoculations of the bacterial strains were made from the cultures of the individual strains 12 to 14 mentioned above

35 It is clear from the data above that the MIC of cephradine is heavily dependent on the combination with clavulanic acid and decreases by 10 to 50% of the initial value depending on the strain.
B - The sensitivity of the bacterial 40 strains in agar was determined for the purpose of evaluating the activity of cephradin in comparison with the combination cephradin -1-clavulanic acid (2: 1) on different bacterial strains in vitro in a solid medium and also for the purpose the determination of the Dif-45 fusion ability of the antibiotic in agar.
Concentrations were used which had proven to be proportionally more suitable for the samples in the liquid medium.
The nutrient medium used was that of Müller-Hinton so (20 ml per Petri dish with a diameter of 9 cm), to which 1 ml of a suspension of each of the germs to be examined, prepared as described above, was added while still molten. After appropriate mixing in the Petri dish, the mass is allowed to solidify on a horizontal plane; then dimples with a diameter of 8 mm are made in the layer of the solidified nutrient medium; 0.05 ml of a 0.9% sodium chloride solution of the antibiotic in different concentrations are introduced into each of them. 6o The samples were read 12 hours after incubation at 37 ° C and made it possible to measure the growth inhibition zones depending on the sensitivity of the germ used and the concentration of the antibiotic.
65 The inhibition zones given in mm make it possible to quantitatively estimate the growth-inhibiting ability of the bacteria examined. The results obtained are shown in Table 2.

It is clear from the data in Table 2 that 5 pg of composition containing only 2/3 cephradine have an inhibitory effect equal to or greater than that of 10% pure cephradine.
It is therefore clear from the microbiological data obtained that the synergistic effect of clavulanic acid on cephradine - an effect which occurs in mixtures in which the two active compounds are present in a weight ratio of 3: 1 to 1: 1, but the better results are obtained with compositions in which cephradine and clavulanic acid are contained in a weight ratio of 2: 1 - the use of cephradine is possible in much lower doses than before and can therefore be used for the patient without any risk of side effects, even with long-term treatment.
