Technisches Gebiet
Die vorliegende Erfindung betrifft ein neue Thiazolopyri midin-Derivat mit gefässerweiternder Wirkung, hypotensiver Wirkung, inhibierender Wirkung auf die Plättchen-Aggregation und reduzierende Wirkung auf den Cholesteringehalt im
Blut.
Stand der Technik
Cardiovaskuläre Erkrankungen, z.B. ischämische Herzkrankheiten, cerebrovaskuläre Erkrankungen, Atherosklerose und Bluthochdruck, zusammen mit bösartigen Tumoren, stellen unter den Haupttodesursachen sehr wesentliche Erkrankungen dar. Unter diesen nimmt man an, dass cardiovaskuläre Erkrankungen, ischämische Herzkrankheiten und Insuffizienz des cerebralen oder peripheren Kreislaufes im allgemeinen verursacht werden durch Mangel an Nährstoff- und Sauerstoffzuführung, die ihre Ursache in einem ungenügenden Blutstrom haben, der von arterieller Thrombusbildung oder von Atherosklerose herrührt. Deshalb sind die Arzneimittel bei der Behandlung dieser Erkrankungen wirksam, die den Blutfluss des betroffenen Organs auf der Basis von gefässerweiternder Wirkung im ischämischen Bereich, wie z.B.
den Herzkranzarterien, verbessern. Es wird auch angenommen, dass die Drogen wirksam sind bei der Verhinderung der Pathogenese dieser Erkrankungen, die die Plättchenbildung inhibieren, welche eine Ursache für die arterielle Thrombusbildung ist, oder Arzneimittel, die das erhöhte Serumcholesterin reduzieren, durch welches Atherosklerose verursacht wird.
Vasodilatoren, z.B. Hydralazin, besitzen eine hypotensive bzw. blutdrucksenkende Wirkung und wurden häufig in der Behandlung von Bluthochdruckstörungen verwendet. In letzter Zeit wurden Vasodilatoren auch bei der Behandlung von Herzinsuffizienz eingesetzt, was auf deren Wirkung, die periphere Vaskularresistenz zu reduzieren, beruht; davon wird von Kei Itoh et al. (Nippon Rinshon, 36[11]51 [1978]) und von Shinobu Matsui et al. (Abstract No. 165, The 43rd annual congress of the Japanese Circulation Society) berichtet.
Darstellung der Erfindung
Die Erfinder haben erkannt, dass cardiovaskuläre Erkrankungen, wie vorstehend genannt, miteinander in Beziehung stehen und dass die optimale Behandlung dieser Erkrankungen eine Behandlung darstellen würde, die zu einer umfassenden Verbesserung dieser Erkrankungen führt. Als Ergebnis der Suche nach Verbindungen, die diesem Zweck entsprechen, haben die Erfinder gefunden, dass 7-Diethylamin5-methylthiazolo[5,4-d]pyrimidin gefässerweiternde Wirkung hypotensive wirkung, inhibierende Wirkung auf die Plättchenaggregation und eine senkende Wirkung auf den Cholesteringehalt im Blut besitzt.
Die vorliegende Erfindung betrifft eine Verbindung des 7-Diethylamino-5-methylthiazolo[5,4-d]pyrimidin-Derivates, das im allgemeinen wie folgt hergestellt werden kann:
4-Diethylamino-6-mercapto-2-methyl-5-nitropyrimidin wird nach bekannten Methoden reduziert, um die Nitrogruppe in 5-Stellung zur Aminogruppe umzuwandeln. Das auf diese Weise erhaltene Aminopyrimidin-Derivat wird direkt unter Verwendung von Ameisensäure zyklisiert, wobei ein Thiazolopyrimidin-Derivat erhalten wird.
Die Verbindung gemäss der Erfindung, wie sie vorstehend erhalten wird, kann zu einem pharmakologisch annehmbaren Salz umgewandelt werden, wie z.B. einem Säureaddukt mit anorganischen Säuren, z.B. Salzsäure, Bromwasserstoffsäure, Jodwasserstoffsäure, Schwefelsäure, Salpetersäure oder Phosphorsäure, oder mit organischen Säuren, z.B. Essigsäure, Maleinsäure, Zitronensäure, Weinsäure, Oxalsäure oder Bernsteinsäure.
Die erfindungsgemässe Verbindung besitzt als therapeutisches Mittel vorteilhafte Eigenschaften, insbesondere als Vasodilator, da es über eine starke gefässerweiternde Wirkung, hypotensive Wirkung, inhibierende Wirkung auf die Plättchenaggregation und reduzierende Wirkung auf den Blutcholesteringehalt besitzt und ausserdem in Wasser löslich ist. Die Verbindung entsprechend der vorliegenden Erfindung oder deren Säureaddukte können zu Produkten formuliert werden, die therapeutisch Verwendung finden, und zwar allein oder zusammen mit anderen pharmakologisch aktiven Verbindungen, nach einer beliebigen konventionellen Methode, wobei man gegebenenfalls pharmazeutisch annehmbare Bindemittel, Füllstoffe oder Aromastoffe verwendet.
Im folgenden wird ein Beispiel des Verfahrens zur Herstellung der erfindungsgemässen Verbindung angegeben.
Beispiel: 7-Diethylamino-5-methylthiazolo[5,4-d]pyrimidin
7,3 g 4-Chloro-6-diethylamino-2-methyl-5-nitropyrimidin wurden in 100 ml eines Gemisches aus Methanol-Wasser (9:1) aufgelöst und 4,7 g 70%iges Natriumhydrosulfid langsam zugegeben. Das Reaktionsgemisch wurde bei Raumtem- peratur 30 Minuten lang gerührt und das Lösungsmittel unter vermindertem Druck abdestilliert und der pH des Rückstandes durch Zugabe von 5N Salzsäure auf 5 bis 6 eingestellt.
Die ausgefallenen gelben Kristalle wurden durch Filtration abfiltriert. Die Kristalle wurden aus einem Gemisch von Chloroform und n-Hexan umkristallisiert, wobei gereinigtes 4-Diethylamino-6-mercapto-2-methyl-5-nitropyrimidin erhalten wurde (5,2 g, Schmelzpunkt 181,3 - 183,3 "C).
5 g der 4-Diethylamino-6-mercapto-2-methyl-5-nitropyrimidin-Kristalle und 7,0 g metallisches Zinn wurden gut miteinander vermischt und 50 ml konzentrierte Salzsäure langsam unter Kühlung zu dem Gemisch zugegeben. Nach der Zugabe wurde das Reaktionsgemisch bei Raumtemperatur 3 h lang gerührt. Dann wurde eine 3N Natriumhydroxidlösung zu dem Gemisch zugegeben, während dieses mit Eis gekühlt wurde, und der pH des Gemisches auf 8 bis 9 eingestellt, wobei eine ölige Substanz erhalten wurde. Die ölige Substanz wurde mit 200 ml Ethylacetat extrahiert und die organische Schicht über Magnesiumsulfatanhydrid getrocknet. Dann wurde das Lösungsmittel unter vermindertem Druck abdestilliert und der Rückstand aus einem Gemisch von Chloroform und n-Hexan umkristallisiert, wobei 5-Amino-4-diethyl amino-6-mercapto-2-methylpyrimidin- Kristalle (3,9 g) erhalten wurden.
3 g 5-Amino-4-diethylamino-6-mercapto-2-methylpyrimidin wurden in 50 ml 90%iger Ameisensäure aufgelöst und 2 h lang unter Rückfluss gehalten. Nach- Beendigung der Reaktion wurde die Ameisensäure unter reduziertem Druck abdestilliert. Dem Rückstand wurden 30 ml Wasser zugegeben und dieser mit Ammoniumhydroxid neutralisiert. Diese wässrige Lösung wurde mit 200 ml Ethylacetat extrahiert und die organische Schicht über Natriumsulfatanhydrid getrocknet.
Dann wurde das Lösungsmittel unter reduziertem Druck abdestilliert und der Rückstand durch Säulenchromatografie auf einer Silicagelsäule gereinigt, wobei 7-Diethylamino5-methylthiazolo[5,4-d]pyrimidin (1,6 g) erhalten wurde.
Schmelzpunkt: 41,6-43 C. Massenspektrum: M+ 222.
Elementaranalyse CIOH15N4S Berechnet C 54,05%; H 6,37%; N 25,23% Gefunden: C 54,11%; H 6,32%; N 25,29% NMR(oppm, CDCL3): 8,48 IM s, 3,91 4H q (7Hz), 2,49 3H s, 1,36 6H t (7Hz).

The present invention relates to a compound of the 7-diethylamino-5-methylthiazolo [5,4-d] pyrimidine derivative, which can generally be prepared as follows:
4-Diethylamino-6-mercapto-2-methyl-5-nitropyrimidine is reduced according to known methods in order to convert the nitro group in the 5-position to the amino group. The aminopyrimidine derivative thus obtained is cyclized directly using formic acid, whereby a thiazolopyrimidine derivative is obtained.
The compound according to the invention as obtained above can be converted to a pharmacologically acceptable salt, e.g. an acid adduct with inorganic acids, e.g. Hydrochloric acid, hydrobromic acid, hydroiodic acid, sulfuric acid, nitric acid or phosphoric acid, or with organic acids, e.g. Acetic acid, maleic acid, citric acid, tartaric acid, oxalic acid or succinic acid.
The compound according to the invention has advantageous properties as a therapeutic agent, in particular as a vasodilator, since it has a strong vasodilating effect, hypotensive effect, inhibitory effect on platelet aggregation and a reducing effect on blood cholesterol content and is also soluble in water. The compound according to the present invention or its acid adducts can be formulated into products which are used therapeutically, alone or together with other pharmacologically active compounds, by any conventional method, optionally using pharmaceutically acceptable binders, fillers or flavorings.
An example of the process for the preparation of the compound according to the invention is given below.
Example: 7-diethylamino-5-methylthiazolo [5,4-d] pyrimidine
7.3 g of 4-chloro-6-diethylamino-2-methyl-5-nitropyrimidine were dissolved in 100 ml of a mixture of methanol-water (9: 1) and 4.7 g of 70% sodium hydrosulfide were slowly added. The reaction mixture was stirred at room temperature for 30 minutes and the solvent was distilled off under reduced pressure and the pH of the residue was adjusted to 5 to 6 by adding 5N hydrochloric acid.
The precipitated yellow crystals were filtered off by filtration. The crystals were recrystallized from a mixture of chloroform and n-hexane to give purified 4-diethylamino-6-mercapto-2-methyl-5-nitropyrimidine (5.2 g, melting point 181.3-183.3 "C). .
5 g of the 4-diethylamino-6-mercapto-2-methyl-5-nitropyrimidine crystals and 7.0 g of metallic tin were mixed well and 50 ml of concentrated hydrochloric acid were slowly added to the mixture with cooling. After the addition, the reaction mixture was stirred at room temperature for 3 hours. Then, a 3N sodium hydroxide solution was added to the mixture while cooling with ice, and the pH of the mixture was adjusted to 8 to 9 to obtain an oily substance. The oily substance was extracted with 200 ml of ethyl acetate and the organic layer was dried over magnesium sulfate anhydride. Then, the solvent was distilled off under reduced pressure and the residue was recrystallized from a mixture of chloroform and n-hexane, whereby 5-amino-4-diethylamino-6-mercapto-2-methylpyrimidine crystals (3.9 g) were obtained.
3 g of 5-amino-4-diethylamino-6-mercapto-2-methylpyrimidine were dissolved in 50 ml of 90% formic acid and kept under reflux for 2 hours. After the reaction was completed, the formic acid was distilled off under reduced pressure. 30 ml of water were added to the residue and this was neutralized with ammonium hydroxide. This aqueous solution was extracted with 200 ml of ethyl acetate and the organic layer was dried over sodium sulfate anhydride.
Then the solvent was distilled off under reduced pressure and the residue was purified by column chromatography on a silica gel column to obtain 7-diethylamino5-methylthiazolo [5,4-d] pyrimidine (1.6 g).
Melting point: 41.6-43 C. Mass spectrum: M + 222.
