Die Erfindung betrifft ein Verfahren zur Herstellung von Vincamin-Derivaten der Formel I

in Form der freien Base oder als Säureadditionssalz. Die Verbindung der Formell weist die Konfiguration 3S,14S,16S auf. Die Verbindung der Formel list beschrieben und wird beansprucht in der schweizerischen Patentschrift 593283 und im USP 4146643.
Der Inhalt derselben wird durch die angeführten Zitate mit einbezogen. Die Verbindung der Formel I findet als Heilmittel bei der Behandlung der Cerebralinsuffizienz Anwendung. Während in einem der genannten Patentschriften beschriebenen Verfahren Vincamin direkt bromiert wird, erfolgt beim vorliegenden Verfahren die Bromierung erst nach Einführung einer Schutzgruppe. Hierdurch werden überraschende Vorteile erhalten, z. B. wird die Epimerisierung in Stellung 14 verhindert, wodurch wenig Nebenprodukte gebildet werden, und die Verbindung der Formel I in hoher Ausbeute und Reinheit erhalten.
Erfindungsgemäss gelangt man zu dem Vincamin-Derivat der Formeln in Form der freien Base oder als Säureadditionssalz, indem man aus einer Verbindung der Formel II,

worin R eine Schutzgruppe bedeutet, die Schutzgruppe abspaltet und die so erhaltene Verbindung der Formel 1 in Form der freien Base oder als ein Säureadditionssalz gewinnt.
Vorzugsweise soll die Schutzgruppe R so gewählt werden, dass unter Bromierungsbedingungen eine möglichst kleine Spaltung der ROH-Gruppe stattfindet. R bedeutet vorzugsweise Trimethylsilyl. Die Abspaltung der Trimethylsilylschutzgruppe erfolgt nach an sich bekannten Methoden. Die Reaktion kann beispielsweise so erfolgen, dass man eine Verbindung der Formel II in einem unter den Reaktionsbedingungen inerten Lösungsmittel wie Methanol aufschlämmt und/oder hierauf Säure, wie Schwefelsäure, zufliessen lässt. Geeignete Temperaturen liegen zwischen 0 bis 50"C.
Das entstandene Vincamin-Derivat der Formeln kann hierauf auf an sich bekannte Weise, z. B. mittels Chromatographie und/ oder Kristallisation isoliert und gereinigt werden.
Die als Ausgangsmaterial verwendete Verbindung der Formel II wird zweckmässig hergestellt, indem man eine Verbindung der Formel III,

worin Reine Schutzgruppe bedeutet, bromiert. Der Bromie rungsschrittkann auf an sich bekannte Weise erfolgen, z. B. mit BrombeiTemperaturenzwischen 700 und -100C,so wie er z. B. in der schweizerischen Patentschrift 593283, imUSP 4146643 und im Beispiel b) beschrieben wird.
Die als Ausgangsmaterial verwendete Verbindung der Formel III wird hergestellt, indem man Vincamin schützt, z. B. durch eine Trimethylsilylschutzgruppe, z.B. wie in Beispiel a) beschrieben wird.
In dem nachfolgenden Beispiel erfolgen alle Temperaturangaben in Celiusgraden.
Beispiel (3S,14S,16S)-11-Brom-vincamin
Der unter b) erhaltene Eindampfrückstand, enthaltend (3S,14S,16S)-11-Brom-vincamin-trimethylsilyläther wird in
530 ml Methanol aufgeschlämmt, worauf man unter Kühlung eine Lösung von 120 g Schwefelsäure in 550 ml Wasser zufliessen lässt. Die entstandene feine Suspension lässt man während einigen Stunden bei Raumtemperatur nachreagieren. Hierauf gibt man obige Reaktionslösung zu einer auf 10 abgekühlten Mischung von 295 ml ca. 25 % Ammoniak-Lösung und 420 ml Wasser. Bei der Zugabe fällt das Reaktionsprodukt sofort aus.
Die Suspension wird nach ca. 30 Minuten bei 10 nachgerührt und dann durch eine Glassinternutsche filtriert. Der Nutschenrückstand wird mit 400 ml Wasser gewaschen. Man erhält feuchtes Kristallisat, das am Vakuum getrocknet wird.
Reines (3S ,14S,16S)-11-Brom-vincamin erhält man beim Chromatographieren roher Kristalle auf Kieselgel mit Methylenchlorid/Methanol (98:2) als Lösungsmittel und nachfolgendem Kristallisieren aus 2-Propanol.
Smp.: 214 (Zers.); [a]r20 = +8,70(1%, CHCl3).
Auf an sich bekannte Weise erhält man aus (3S,14S,16S)-11- Brom-vincamin und Fumarsäure (3S,14S,16S)-11-Brom- vincamin-hydrogenfumarat; Smp.: 1800.
Der als Ausgangsmaterial verwendete (3S,14S,16S)-11-Bromvincamin-trimethylsilyläther wird wie folgt hergestellt: a) (35,145 16S)-Vincamin-trimethylsilyläther
Man lässt unter Stickstoffspülung zu 31,18 g Natriumhydrid 900 ml Tetrahydrofuran zufliessen. Zu der so erhaltenen grauen Suspension gibt man unter Kühlung 100 g (3S,14S,16S) Vincamin. Man spült 100 ml Tetrahydrofuran und lässt 2 bis 3 Stunden nachreagieren. Dann kühlt man auf -2" ab und tropft bei 20 bis 0" 38,3 g Trimethylchlorsilan zu. Der Tropftrichter
wird mit 5 ml Tetrahydrofuran gespült. Bei gleicher Temperatur werden 42,5 g Essigsäure zugegeben. Nach beendeter Zugabe erwärmt man das Reaktionsgemisch auf Zimmertemperatur, lässt nachrühren, worauf man 300 ml Wasser zugibt. Das 2 Phasengemisch wird durch Zugabe von ca. 18 ml einer 1:2 G/G Mischung Ammoniak/Wasser auf pH 9 bis 10 gestellt. Dann werden die Phasen getrennt. Die untere Wasserphase wird mit 100 ml Tetrahydrofuran extrahiert.
Die vereinigten Tetrahydrofuranphasen werden möglichst vollständig eingeengt. Dann gibt man 270 ml Methanol zum Rückstand, rührt bis man eine feine Suspension erhält und engt nochmals möglichst vollständig ein. Zu diesem Rückstand gibt man zuerst 100 ml Methanol und dann 200 ml Wasser. Man rührt eine Stunde bei Zimmertemperatur und filtriert über eine Sinternutsche. Der Rückstand wird mit 60 ml Wasser gewaschen und das so erhaltene feuchte Kristallisat getrocknet. Das Rohkristallisat kann direkt zur Bromierung eingesetzt werden.
Reines Material erhält man durch Umkristallisation aus der 4,5fachen Menge Äthylmethylketon.
Smp.: 182,5 bis 184,5".
b) (3S,14S,16S)-11-Brom-vincaminsilyläther
Unter Stickstoff werden 35,1 g Zinkchlorid und 820 ml Essigsäureäthylester vorgelegt. Man rührt bis eine klare Lösung entsteht und kühlt auf 43Q ab. Dazu tropft man bei -43 bis -40" eine Lösung von 43,1 gBromin480 ml Essigsäureäthylester. Parallel dazu trägt man in 12 Portionen 100 g (3S,14S,16S)- Vincamin-trimethylsilyläther ein. Nachher lässt man 3 Stunden nachreagieren und gibt das Reaktionsgemisch auf eine Mischung von 200 ml Ammoniak und 500 ml Wasser. Es entsteht ein 2-Phasengemisch, das getrennt wird. Die untere wässrige Phase wird mit total 840 ml Essigsäureäthylester extrahiert. Die vereinten organischen Phasen werden möglichst vollständig eingeengt.
Zum Rückstand gibt man 150 ml Methanol und engt vollständig ein. Der Eindampfrückstand kann direkt in die Silylätherspaltung eingesetzt werden. Reines Material erhält man durch Umkristallisation aus der 9fachen Menge 2-Propanol.

The evaporation residue obtained under b), containing (3S, 14S, 16S) -11-bromo-vincamin-trimethylsilyl ether is in
530 ml of methanol slurried, whereupon a solution of 120 g of sulfuric acid in 550 ml of water is allowed to flow in with cooling. The resulting fine suspension is left to react for a few hours at room temperature. Then the above reaction solution is added to a mixture of 295 ml of approximately 25% ammonia solution and 420 ml of water cooled to 10. The reaction product precipitates immediately upon addition.
The suspension is stirred at 10 after about 30 minutes and then filtered through a glass suction filter. The filter residue is washed with 400 ml of water. Moist crystals are obtained which are dried in vacuo.
Pure (3S, 14S, 16S) -11-bromo-vincamine is obtained when crude crystals are chromatographed on silica gel with methylene chloride / methanol (98: 2) as solvent and subsequent crystallization from 2-propanol.

The (3S, 14S, 16S) -11-bromvincamine trimethylsilyl ether used as starting material is prepared as follows: a) (35.145 16S) -vincamine trimethylsilyl ether
9.18 g of sodium hydride and 900 ml of tetrahydrofuran are allowed to flow in under a nitrogen purge. 100 g (3S, 14S, 16S) vincamine are added to the gray suspension thus obtained with cooling. 100 ml of tetrahydrofuran are rinsed and the mixture is left to react for 2 to 3 hours. is rinsed with 5 ml of tetrahydrofuran. 42.5 g of acetic acid are added at the same temperature.
