Die erfindungsgemässen Verbindungen der Formel I sind wertvolle Vorläufer bei der Herstellung von 1-Methyl -5-aroylpyrrol-2-essigsäuren, die eine ausgezeichnete ent zündungshemmende Wirkung zeigen [J. Pharmacology and Experimental Therapeutics, 185, 127 (1973)1.
Die erfindungsgemässe Verbindung der Formel I ist neu und kann durch Umsetzen von Bromoform zusammen mit Ethylmercaptan unter basischen Bedingungen in 1-Methyl -5-aroylpyrrol-2-essigsäure umgewandelt werden (siehe das nachfolgend beschriebene Referenzbeispiel 2).
Üblicherweise sind die folgenden Verfahren bekannt zur Herstellung dieser 1-Methyl-5-aroylpyrrol-2-essigsäuren:
1. Ein Verfahren, das eine Reaktion mit einem Aroylchlorid in Gegenwart einer Lewis-Säure wie Aluminiumchlorid usw. [Japan. Patent Nr. 34153/763 umfasst.
2. Ein Verfahren, das ein gemischtes Säureanhydrid, erhalten aus einer Arylcarbonsäure und Fluoracetanhydride, verwendet [Japan. Patent Nr. 418/711.
3. Ein Verfahren, das ein Reaktionsprodukt eines Dimethylaroylamides und Phosphoroxidchlorid verwendet [Japan. Patent Nr. 418/713.
4. Ein Verfahren, das ein gemischtes Säureanhydrid, erhalten aus einem Arylcarbonsäureanhydrid und einer Säure wie Methansulfonsäure usw. verwendet [Japan. Patent Nr. 126660/75].
5. Ein Verfahren, das Umsetzen der Säure mit Phosgen zum 5-Chlorcarbonylderivat und anschliessendem Umsetzen des Derivates mit einer Arylmetallverbindung umfasst [Japan. Patent Nr. 55659/743.
Diese Verfahren haben jedoch die Nachteile, dass sie eine niedere Produktausbeute und als Nebenprodukte Isomere geben, die eine Aroylgruppe in der 4-Stellung enthalten und die abgetrennt werden müssen.
Es wurde nun ein neues Formylierungs-Verfahren gefunden, durch das ein 2-Aroylformyl substituiertes Pyrrol leicht mit hoher Ausbeute hergestellt werden kann, indem man ein 2-Aroylpyrrol verwendet, das leicht aus einem industriell verfügbaren Pyrrol als Ausgangsmaterial erhalten werden kann, und durch das, anders als bei allgemeinen Verfahren, eine Formylierung heterozyklischer Verbindungen, substituiert durch eine Aroylgruppe erfolgen kann.

X eine Alkyl- oder Alkyloxygruppe oder ein Halogenatom bedeuten kann leicht hergestellt werden, indem man ein Addukt, gebildet durch die Reaktion von Phosphoroxidchlorid mit einem Arylamid, mit einem Pyrrol umsetzt (siehe das nachfolgende Referenzbeispiel 1).
Beispiele von 2-Aroylpyrrolen der Formel II schliessen die folgenden ein: 2-Benzoylpyrrol, 2-p-Chlorbenzoylpyrrol, 2-p-Toluoylpyrrol, 2-p-Methoxybenzoylpyrrol, 2-Benz oyl-1 -methylpyrrol, 2-p-Chlorbenzoyl- 1 -methylpyrrol, 2-p -Toluoyl- 1 -methylpyrrol, 2-Methoxybenzoyl- 1-methylpyrrol und ähnliche.
Das als ein anderes Ausgangsmaterial verwendete Addukt von Dimethylformamid (DMF) und Phosphoroxidchlorid ist ein wohlbekanntes Formylierungsmittel, das durch einfaches Mischen der beiden obigen Verbindungen erhalten werden kann.
Beim Herstellen der erfindungsgemässen Verbindungen wird die Verwendung eines Lösungsmittels bevorzugt. Beispiele von Lösungsmittel schliessen 1,2-Dichlorethan, Dichlormethan, Diethylether und ähnliche ein. Auch das Ausgangs-DMF kann als Lösungsmittel agieren, wenn es in einem Überschuss verwendet wird.
Üblicherweise erfolgt die Reaktion in einem Temperaturbereich von -200C bis 80"C, vorzugsweise, unter Berücksichtigung der Produktausbeute-Verbesserung, von - IOOC bis 40"C. Darüber hinaus ist es für das in dieser Erfindung eingesetzte Verfahren charakteristisch, dass das Bildungs- verhältnis zunimmt, wenn die Reaxtion von - 100C bis Raumtemperatur durchgeführt wird.
Die Erfindung wird weiter durch die folgenden Referenzbeispiele und Beispiele detailliert veranschaulicht.
Referenzbeispiel 1
Zu 10,7 g (0,07 Mol) Phosphoroxidchlorid fügte man tropfenweise 25 ml einer Lösung von N,N-Dimethyl-p- tolylamid (11,43 g, 0,07 Mol), das bei Raumtemperatur in 1,2-Dichlorethan gelöst worden war, und erhitzte die Mischung während 2 Stunden auf Rückfluss-Temperatur. Nach Abkühlen der Mischung auf Raumtemperatur wurde dazu langsam eine Lösung von N-Methylpyrrol (5,7 g, 0,07 Mol) in 15 ml Dichlorethan getropft. Die Reaktionsmischung wurde auf 50 bis 60"C erhitzt und nachfolgend über Nacht gerührt.
Zu dieser neuerlich auf Raumtemperatur gekühlten Mischung tropfte man langsam unter Eiswasser-Kühlung eine Lösung von 48 g Natriumacetat, gelöst in 150 ml Wasser und erhitzte die Mischung während 30 Minuten auf Rückfluss-Temperatur. Die Reaktionsmischung wurde in Eiswasser geleert, mit Chloroform extrahiert und dann getrocknet.
Nach Entfernen des Lösungsmittels wurde die entstehende Lösung destilliert (B 170 - 1800C/ 1 mmHg) und durch Säulen-Chromatographie (Silicagel, 20% Ethylacetat/n-Hexan) gereinigt. Man erhielt 2-p-Toluoyl- 1 -methylpyrrol (8,98 g). Ausbeute 64%.
Referenzbeispiel 2
Zu einem Lösungsgemisch aus 25 ml Acetonitril und 10 ml Wasser fügte man 1-Methyl-2-formyl-5-p-methylbenzoylpyrrol (1,135 g, 5 mMol), Bromoform (1,54 g, 6,1 mMol) und Ethylmercaptan (1,61 g, 26 mMol, 1,94 ml). Zu dieser Mischung tropfte man unter Rühren bei Raumtemperatur Kaliumhydroxid (1,57 g, 28 mMol) gelöst in 7,5 ml Wasser und 25 ml Acetonitril. Nach beendeter Zugabe wurde die Reaktionsmischung über Nacht bei Raumtemperatur gerührt, gefolgt von Rühren und Heizen auf einem Wasserbad während 2 Stunden auf 800C. Nach abkühlen lassen verdünnte man die Mischung mit 50 ml Wasser und wusch mit Äther. Beim Ansäuren der abgetrennten wässrigen Schicht mit Salzsäure wurde die erstere trübe weiss und es bildete sich der Feststoff. Diesen Feststoff extrahierte man mit Äther, trocknete den Extrakt und dampfte ein.
Man erhielt 5-p-Toluoyl- 1-methylpyrrol-2-essigsäure (0,90 g).
Ausbeute 70%.
Mp. 155-156"C.
NMR (CDCl3): 8 2,37 (3H, s), 3,69 (2H, s), 3,89 (3H, s), 6,06 (1H, d, J=4Hz), 6,62 (1H, d, J=4Hz), 7,18 (2H, d, J=8Hz), 7,66 (2H, d, J=8Hz).
IR (KBr): 3425, 2940, 2900, 1700, 1600 cm-1.
Beispiel I
Unter Eiswasser-Kühlung fügte man langsam tropfenweise zu Dimethylformamid (0,365 g, 5 mMol) 1 ml Phos poroxidchloridi. Nach beendeter Zugabe wurde die Mischung während 15 Minuten bei Raumtemperatur gerührt, dann wieder mit Eiswasser gekühlt und dazu 25 ml einer Lösung von 2-p-Toluoyl-1-methylpyrrol (0,995 g, 5 mMol) in 1,2-Dichlorehtan getropft. Nach beendeter Zugabe wurde die Mischung über Nacht bei Raumtemperatur stehen gelassen.
Die Reaktionsmischung wurde in 10%ige wässrige Natriumcarbonat-Lösung geleert und für 15 Minuten auf Raumtemperatur und für weitere 10 Minuten auf 60"C erwärmt.
Die entstehende Mischung wurde wieder auf Raumtemperatur gekühlt, getrocknet und eingeengt. Der Rückstand (1,22 g) wurde durch Säulenchromatographie gereinigt. Man erhielt 5-p-Toluoyl-2-formyl-1-methylpyrrol (0,78 g). Ausbeute 68%.
Mp. 81-82"C.
NMR (CDCld: a 2,50 (3H, s), 4,33 (3H, s), 6,77 (1H, d, J=3,6Hz), 7,03 (1H, d, J=3,6Hz), 7,42 (2H, d, J=7,3 Hz), 7,90 (2H, d, J=7,3Hz), 9,98 (1H, s).
IR (KBr): 3025, 2950, 2830, 1640, 1610, 1360, 1270, 1095, 895, 745 cm-1.
Beispiel 2
Man löste 1,0 g Phosphoroxidchlorid in 9 ml Dimethylformamid. Dazu tropfte man unter Eiswasser-Kühlung nach und nach 3 ml Ätherlösung mit 2-p-Toluoyl-1-methylpyrrol (0,995 g, 5 mMol). Nach beendeter Zugabe rührte man die Mischung unter Eiswasser-Kühlung während weiteren 5 Minuten und entfernte dann das Eiswasserbad aus dem System. Während 19 Stunden liess man die Reaktionsmischung bei Raumtemperatur stehen und kühlte dann wieder mit Eiswasser. Dann gab man 15 ml Wasser zu und rührte die Mischung während 15 Minuten.
Dazu fügte man gesättigte wässrige Natriumcarbonat Lösung und extrahierte die Mischung mit Chloroform. Der Rückstand wurde getrocknet, eingeengt und durch Säulenchromatographie gereingt (Silicagel; Ethylacetat: n-Hexan = 1:9 v/v). Man erhielt 5-p-Toluoyl-3-formyl-1-methylpyr- rol (0,80 g). Ausbeute 70%.
NMR (CDCl3): ô 2,50 (3H, s), 4,15 (3H, s), 7,32 (1H, d, J=1,7Hz), 7,45 (2H, d, J=6,7Hz), 7,67 (1H, d, J=1,7 Hz), 7,92-(1H, d, J=6,7Hz), 10,0 (1H, s).
Beispiel 3
Unter Eiswasser-Kühlung tropfte man zu Dimethylformamid (3,34 g, 46 mMol) Phosphoroxidchlorid (6,99 g, 0,046 mMol). Die festgewordene Reaktionsmischung wurde bei Raumtemperatur in 1, 2-Dichlorethan gelöst. Die Lösung wurde wieder mit Eiswasser gekühlt und darauf langsam tropfenweise mit einer Lösung von 2-p-Toluoyl-1 -methylpyrrol (7,22 g, 36 mMol) in 50 ml Dichlorethan versetzt. Das Eiswasserbad wurde aus dem System entfernt und die Mischung während 20 Stunden bei Raumtemperatur stehen gelassen. Die Reaktionsmischung wusde dann unter Rückfluss während 1 Stunde auf 100 C erhitzt, auf Raumtemperatur gekühlt und durch Zufügen einer Lösung von 75 g Natriumacetat, 3H2O (75 g) gelöst in 100 ml Wasser hydrolisiert.
Die organische Schicht wurde mit Methylenchlorid extrahiert, getrocknet, eingeengt und ergab 12,84 g Restlösung.
Diese wurde durch Säulenchromatographie gereinigt (Silicagel; 10% Ethylenacetat/n-Hexan). Man erhielt 4,26 g 5-p-Toluoyl-2-formyl-1-methylpyrrol (Ausbeute 52%) und 3,34 g 5-p-Toluoyl-3-formyl-methylpyrrol (Ausbeute 41%).

The compounds of the formula I according to the invention are valuable precursors in the preparation of 1-methyl -5-aroylpyrrole-2-acetic acids which have an excellent anti-inflammatory effect [J. Pharmacology and Experimental Therapeutics, 185, 127 (1973) 1.
The compound of the formula I according to the invention is new and can be converted into 1-methyl -5-aroylpyrrole-2-acetic acid by reacting bromoform together with ethyl mercaptan under basic conditions (see reference example 2 described below).
The following processes are usually known for the preparation of these 1-methyl-5-aroylpyrrole-2-acetic acids:

Examples of 2-aroylpyrroles of formula II include the following: 2-benzoylpyrrole, 2-p-chlorobenzoylpyrrole, 2-p-toloylpyrrole, 2-p-methoxybenzoylpyrrole, 2-benzoyl-1-methylpyrrole, 2-p-chlorobenzoyl- 1-methylpyrrole, 2-p-toluoyl-1-methylpyrrole, 2-methoxybenzoyl-1-methylpyrrole and the like.
The adduct of dimethylformamide (DMF) and phosphorus oxychloride used as another starting material is a well known formylating agent which can be obtained by simply mixing the above two compounds.
When preparing the compounds according to the invention, the use of a solvent is preferred. Examples of solvents include 1,2-dichloroethane, dichloromethane, diethyl ether and the like. The starting DMF can also act as a solvent when used in excess.
The reaction usually takes place in a temperature range from -200 ° C. to 80 ° C., preferably, taking into account the improvement in product yield, from −IOOC to 40 ° C. In addition, it is characteristic of the method used in this invention that the formation ratio increases when the reaction is carried out from −100 ° C. to room temperature.
The invention is further illustrated in detail by the following reference examples and examples.
Reference example 1

Potassium hydroxide (1.57 g, 28 mmol) dissolved in 7.5 ml of water and 25 ml of acetonitrile was added dropwise to this mixture with stirring at room temperature. After the addition was complete, the reaction mixture was stirred at room temperature overnight, followed by stirring and heating on a water bath at 800C for 2 hours. After allowing to cool, the mixture was diluted with 50 ml of water and washed with ether. Example I
With ice-water cooling, 1 ml of phosporoxidchloridi was slowly added dropwise to dimethylformamide (0.365 g, 5 mmol). Example 2
1.0 g of phosphorus oxychloride was dissolved in 9 ml of dimethylformamide. 3 ml of ether solution with 2-p-toluoyl-1-methylpyrrole (0.995 g, 5 mmol) were gradually added dropwise while cooling with ice water. After the addition was complete, the mixture was stirred with ice-water cooling for a further 5 minutes and then the ice-water bath was removed from the system. The reaction mixture was left at room temperature for 19 hours and then cooled again with ice water. Then 15 ml of water was added and the mixture was stirred for 15 minutes.

The ice water bath was removed from the system and the mixture was left at room temperature for 20 hours. The reaction mixture was then heated under reflux at 100 C for 1 hour, cooled to room temperature and hydrolyzed by adding a solution of 75 g sodium acetate, 3H2O (75 g) dissolved in 100 ml water.
The organic layer was extracted with methylene chloride, dried, concentrated and gave 12.84 g of residual solution.
This was purified by column chromatography (silica gel; 10% ethylene acetate / n-hexane). 4.26 g of 5-p-toluoyl-2-formyl-1-methylpyrrole (yield 52%) and 3.34 g of 5-p-toluoyl-3-formyl-methylpyrrole (yield 41%) were obtained.
