L'invention se rapporte à de nouveaux dérivés carboxyméthylés du B-1,3-glucane, utilisables pour le traitement préventif et curatif de tumeurs.
Depuis des années, des recherches ont été effectuées en vue de trouver une substance capable de provoquer, par administration parentérale ou orale, I'inhibition de croissance de tumeurs et la prolongation du temps de survie d'animaux à sang chaud atteints de tumeurs, sans effets secondaires sérieux.
L'invention a pour but de fournir de nouveaux dérivés carboxyméthylés du B-1,3-glucane ayant la formule suivante:

On sait déjà par les brevets japonais Nos 32673/1973 et 32674/1973, et par le brevet anglais No 1352938, que certaines souches de micro-organismes du genre Alcaligenes et du genre Agrobacterium produisent un ss-1,3-glucane thermogélifiable insoluble dans l'eau (désigné ci-aprés, de façon abrégée, par TAK-N). Aucun usage pharmaceutique du glucane n'a toutefois été indiqué.
Les études poussées effectuées par les inventeurs sur le TAK-N, ses polyméres inférieurs obtenus par hydrolyse partielle (ci-après désignés par TAK-D) et les dérivés carboxyméthylés de TAK-N ou desdits polymères inférieurs (ci-après désignés par CMTAK) ont abouti à la découverte selon laquelle ces substances possèdent une forte activité antitumorale, se traduisant par un taux d'inhibition des tumeurs remarquablement élevé et par une régression complète de ces tumeurs.
Le CMTAK peut être obtenu par carboxyméthylation de TAK
N ou TAK-D de façon connue, par exemple en faisant réagir le
TAK-N ou le TAK-D avec l'acide monochloracétique en présence d'un alcali. Outre cette méthode, on peut employer tout autre procédé conventionnel utilisé pour la carboxyméthylation des hydrates de carbone.
Pour récupérer le CMTAK â partir du mélange réactionnel, il est possible d'avoir recours à des procédés conventionnels qui sont couramment utilisés pour la purification des hydrates de carbone, tels que la précipitation par addition d'un solvant organique. En outre, d'autres techniques, telles que chromatographie échangeuse d'ions, filtration par gel, etc., peuvent être utilisées suivant la teneur en groupes carboxyméthylés dans le CMTAK, ou suivant la solubilité dans l'eau du CMTAK.
Alors que le CMTAK ainsi obtenu renferme des groupes car boxyméthylés dans sa molécule, la teneur des groupes carboxymé thylés varie largement en fonction des conditions de la réaction de carboxymèthylation. Comme déterminé par titration, la teneur en groupes carboxyméthylés de CMTAK obtenu par réaction normale de carboxyméthylation est généralement moins de trois groupes carboxyméthylés par résidu de glucbse dans la molécule de CMTAK.
Le CMTAK, en tant que produit convenant pour les buts de l'invention, renferme toutes les substances qui peuvent être obtenues par carboxyméthylation de TAK-N ou de TAK-D, dont les molécules renferment des groupes carboxyméthylés décelables, indépendamment du degré de carboxyméthylation.
Comme cela ressort de la formule générale indiquée préèèdem- ment, le CMTAK, sous la forme de son acide libre, est apte à réagir avec des bases variées, pour former les sels correspondants, tels que sels de sodium, potassium, calcium, aluminium, magnésium et d'amines. Dans le contexte de l'invention, le CMTAK comprend non seulement son acide libre, mais également ses sels, en particulier ceux de faible toxicité.
Le tableau 1 illustre les propriétés physiques de quelques espèces
types de CMTAK préparées à partir de TAK-N et de TAK-D, suivant les techniques précédemment indiquées. Les déterminations de ces propriétés physiques ont été effectuées après que chaque échantillon a été préalablement séché sous pression réduite et sur pentoxyde de phosphore à 60 C, pendant 10 h.

Les fig. 1 et 2 illustrent les spectres I.R. dans le bromure de po
tassium, de CMTAK, respectivement Nos 2 et 11 au tableau 1. Les chiffres en haut à gauche et en bas de chaque figure indiquent respectivement les longueurs d'onde (,u), les coefficients de transmission ou transmittances (%) et les nombres d'ondes (cm - 1).
Les absorptions caractéristiques ont été trouvées à 1440-1400 et 891 cm5.
Le CMTAK de DP allant de 2 à environ 1000, de préférence de
15 à 800, et plus avantageusement de 40 à 600, présente une activité inhibitrice remarquable contre différentes sortes de tumeurs chez les animaux à sang chaud comprenant les animaux domestiques, volailles, chiens, chats, lapins, rats, souris, etc., et notamment contre de sérieuses tumeurs connues comme étant moins sensibles aux traitements par des médicaments antitumoraux cytotoxiques généralement connus.
Par exemple, la croissance du sarcome 180, du carcinome d'Ehrlich, de la leucémie SN-36, de l'adénocarcinome CCM, ou du sarcome à cellules réticulaires NTF, chaque tumeur étant transplantée par voie sous-cutanée dans les souris, fut notablement inhibée en administrant le TAK-N, le TAK-D ou le CMTAK par voire intrapéritonéale, intraveineuse ou sous-cutanée, avant, après ou au moment de la transplantation, en une fois ou à plusieurs reprises, à des doses d'environ 1 à 1000 mg/kg par administration, par jour.
Les toxicités de CMTAK sont extrêmement faibles. Par exemple, aucun effet toxique n'est observé, à la fois chez les souris et les rats, à une dose de 3000 mg/kg par voie intrapèritonéale, ou de 10000 mg/kg par voie orale. Ainsi, ces polysaccharides peuvent être administrés, de façon sûre et à plusieurs reprises, à des animaux à sang chaud.
L'administration peut être réalisée suivant les techniques générales habituellement appliquées pour le traitement des cancers. Sont ainsi applicables les injections intratumorales, sous-cutanées, intramusculaires ou intraveineuses, si nécessaire, des administrations orales et rectales, ou encore des applications externes, badigeonnages, instillations et autres méthodes d'administration.
Le CMTAK est administré en une quantité suffisante à un animal à sang chaud pour inhiber la croissance de sa tumeur.
Le dosage et la méthode d'administration de CMTAK à des animaux à sang chaud affectés de néoplasies peuvent varier suivant le type d'agents, d'animaux et de patients, de symptomes de tumeurs, de formes d'administration et autres facteurs. En général, la quantité suffisante de dosage par administration est d'environ 0,02 à 2000 mg/kg de poids vif, de préférence de 0,2 à 2000 mg/kg de poids vif, la limite supérieure préférée étant d'environ 500 mg/kg de poids vif. Dans bien des cas, la limite supérieure la plus avantageuse est d'environ 2000 mg/kg.
L'administration pet être effectuée, par exemple, une à six fois parjour, pendant plusieurs jours consécutifs ou par intermittence.
Les TAK-N, TAK-D et CMTAK peuvent être administrés en combinaison avec d'autres agents antitumoraux. La combinaison avec de tels agents entraînant une activité immunologique accrue chez les animaux atteints de tumeurs est également avantageuse.
L'invention sera maintenant décrite en se référant aux exemples ci-après. Certains de ces exemples illustrent la technique de production de CMTAK, les propriétés physiques du CMTAK et de certaines formes injectables de ce produit. D'autres exemples mettent en évidence les activités antitumorales du CMTAK contre les tumeurs chez les souris, par l'intermédiaire de modèles types.
Comme indiqué précédemment, le traitement des animaux atteints de tumeurs au moyen des composés produit une régression énergique des différents types de tumeurs, tels que sarcomes, carcinomes, sarcomes à cellules réticulaires et leucémies. Tous ces types de tumeurs, chez les rongeurs, se sont avérés être des types classiques exacts de tumeurs chez d'autres animaux à sang chaud.
Ces exemples ne présentent bien entendu aucun caractère limitatif de l'invention.
La plupart des essais suivants sur l'activité antitumorale de
CMTAK sont évalués en employant approximativement les mêmes méthodes que celles décrites pour l'activité antitumorale du lentinane, glucane ramifié provenant d'un champignon comestible, selon
Chihara, G. et al. [voir Nature , 222, 687-688 (1969), et Cancer
Research , 30, 2776-2781, 1970)].
Les tumeurs des souris utilisées dans ces essais, telles que sarcome 180 (S 180), carcinome d'Ehrlich (Ehrlich) et sarcome à cellules réticulaires NTF (NTF), adénocarcinome CCM (CCM) et leucémie SN-36 (SN-36), sont maintenues chez des souris du type ICR-
JCL, de 5 à 7 semaines, chacune sous des formes ascites. Un élément approprié de ces cellules tumorales est transplanté par voie souscutanée sur des souris ICR-JCL, et les nodosités tumorales qui se sont développées sont excisées pour la pesée, le 35e jour après transplantation de la tumeur.
Un TAK-N ou TAK-D a été mis en suspension ou dissous dans l'eau distillée ou dans une solution physiologique, et le CMTAK a été dissous dans l'eau distillée ou dans une solution saline physiologique à des concentrations appropriés, et à des pH voisins de la neutralité. Chaque préparation a été administrée par voie intrapéritonéale, sous-cutanée ou intraveineuse à des souris (0,2 ml/20 g de poids vif), quotidiennement pendant 10 d, en commençant 24 h après la transplantation de la tumeur, quotidiennement pendant 5 d avant la transplantation de la tumeur, ou seulement en une fois durant le 7e jour avant et le 2lejour après l'implantation de la tumeur.
Exemple 1:
Effet de suppression d'une tumeur par le TA K-N (DP 540): injection unique.
4 millions de cellules S 180 sont transplantées par voie souscutanée dans la région inguinale gauche de souris femelles du type
ICR-JCL, de 4 à 5 semaines. Les nodosités de la tumeur sont excisées le 35e jour après la transplantation et pesées. Le poids moyen d'une tumeur d'un groupe traité (T) comprenant 5 souris a été comparé avec celui d'un groupe témoin (C) de 10 souris, et l'on a calculé le taux d'inhibition de la tumeur (inhibition %: (C-T) x100).

On a également compté le nornbre d'animaux exempts de tumeur (régression complète) au 35e jour.
Du TAK-N (DP 540) mis en suspension dans une solution saline physiologique (10 mg/ml) a été administré par voie intrapéritonéale (0,2 ml/20 g de poids vif) une fois à chaque animal, à la dose de 100 mg/kg, le 7e, 3e ou lez jour avant, ou le 1er, 2e, 3e, 5e, 10e, 14e ou 21' jour après la transplantation de la tumeur, ou lejour de la transplantation de la tumeur.
Comme on le voit au tableau 2, le TAK-N à la dose de 100 mg/kg inhibe de façon marquée la croissance de S 180, lorsque l'échantillon test est administré une fois durant le 7e jour avant et le 21e jour après la transplantation de la tumeur.
En outre, par administration unique de TAK-N (DP 540) le 7e jour après la transplantation de la tumeur à des doses de 20 mg/kg, 60 mg/kg et 100 mg/kg, on a observé une suppression marquée de la croissance de la tumeur aux deux doses plus élevées, 60 mg/kg et 100 mg/kg.

Ces résultats indiquent que le TAK-N (DP 540) est efficace dans la suppression de la croissance de la tumeur, à tous les stades de progression de la tumeur sur des animaux à sang chaud, même par administration unique.
Exemple 2:
Effet de suppression d'une tumeur par le TAK-N (DP 540) et le TAK N flo culé en autoclave (DP 540).
6 millions de cellules S 180 sont transplantées par voie souscutanée dans la région inguinale droite de souris du type ICR-JCL d'un poids vif d'environ 23 g. Les nodosités tumorales sont excisées le 35e jour après transplantation et pesées. Le taux d'inhibition de la tumeur dans un groupe traité a été calculé comme dans l'exemple 1.
On a utilisé du TAK-N (DP 540) et du TAK-N floculé (DP 540).
Ce dernier a été obtenu en chauffant une suspension à 2% de TAK
N (DP 540) à 1200 C pendant 25 min, puis en séchant et en pulvérisant le gel formé. Les échantillons furent mis en suspension dans l'eau distillée aux concentrations voulues, et chaque supension a été administrée par voie intrapéritonéale à un groupe de 5 souris, quotidiennement pendant 10 d, en commençant 24 h après la transplantation de la tumeur.
Comme on le voit au tableau 3, le TAK-N (DP 540) exerce un effet prononcé sur la croissance des tumeurs à des doses de I à 50 mg/kg, cet effet étant particulièrement important à des doses de 5 à 25 mg/kg.
Le TAK-N (DP 540) s'est avéré également efficace dans la suppression de la croissance des tumeurs à des doses de 10 mg/kg. On n'a observé aucune toxicité.

Ces résultats montrent que le TAK-N est utilisé dans le ralentissement de la croissance de tumeurs, à des doses relativement faibles, et que l'échantillon peut être stérilisé par traitement à l'autoclave, sans perte de son activité.
Exemple 3:
Effets de suppression de tumeurs par le CMTAK et le CMTAK traité il l'autoclave.
L'activité antitumorale de trois types de CMTAK préparés à partir du TAK-N (CMTAK Nos 1,4, 5) et de CMTAK traité à l'autoclave a été essayée, comme décrit dans l'exemple 2. Les échantillons furent dissous dans l'eau distillée, en vue de l'injection, et le pH a été ajusté à environ 7,0. Dans certains essais, I'un de ces échantillons (CMTAK No 1) a été stérilisé par traitement à l'autoclave (1200 C, 2,5 min) avant tout autre administration.
Comme on le voit au tableau 4, tous les échantillons de

Ces résultats montrent que le CMTAK peut être stérilisé sans
perte de son activité antitumorale et utilisé sous une forme injectable
pour le traitement du cancer.
Exemple 4:
Effets de suppression de tumeurs par les TAK-N, TAK-D et CMTAK suivant des méthodes variées d'administration.
Les TAK-N (DP 540), TAK-D (F-III: DP 50), TAK-D (F-VII:
DP 16) et CMTAK No 1 ont été administrés à des doses de 5 mg/kg, par voie intraveineuse, sous cutanée ou intrapéritonéale pendant
10 d consécutifs rades souris ICR-JCL qui avaient préalablement subi une transplantation avec 4 > < x 106 10 cellules, 24 h avant la pre-
mière injection. Les taux d'inhibition de la croissance de la tumeur
ont été calculés comme dans l'exemple 1.
Comme on le voit au tableau 5, on a observé, dans chaque cas, une inhibition importante de la croissance de la tumeur. Par administration intraveineuse, le TAK-N, le CMTAK et le même le F-VII (DP 16) inhibent presque complètement la croissance de la tumeur, le taux d'inhibition étant supérieur à 85%, avec régression complète de la tumeur sur 4 ou 5 des 5 souris traitées.

Exemple 5:
Effets de suppression de tumeurs par TAK-N, TAK-D et CMTAK ad-
ministrés avant la transplantation de la tumeur: prétraitement.
Les TAK-N, TAK-D de différents degrés de polymérisation et le
CMTAK No 1 furent administrés par voie intrapéritonéale à des souris ICR-JCL de poids vif d'environ 23 g, à des doses de 3 mg/kg à 80 mg/kg, quotidiennement, pendant 5 d consécutifs. Le TAK-N et le TAK-D furent mis en suspension, et le CMTAK dissous, dans l'eau distillée. Dans le cas du CMTAK, le pH final fut ajusté à
environ 7,0. Puis, le ler ou le 3e jour après la dernière administration, 6 x 106 cellules de S 180 furent transplantées par voie souscutanée dans la région inguinale droite de chaque animal. Le 35e jour après la transplantation, les nodosités tumorales furent excisées et pesées. Les taux d'inhibition de la tumeur furent calculés comme dans l'exemple 1.
Comme on le voit au tableau, on observe pour chaque échantillon, une inhibition marquée de la croissance de la tumeur. En particulier, TAK-N (DP 540); CMTAK et TAK-D de DP supérieur à 50 présentent une forte activité inhibitrice. Le CMTAK fut efficace pour le retardement de tumeurs, à des doses faibles, de-l'ordre de 3 mg/kg.

Ces résultats montrent que les polysaccharides mentionnés précé demment sont utiles en tant qu'agents prophylactiques dans le traitement du cancer.
Exemple 6:
Activité répressive des tumeurs par les TAK-N et CMTAK vis-à-vis de divers types de tumeurs.
Le carcinome d'Ehrlich, I'adénocarcinome CCM, le sarcome à cellules réticulaires NTF et la leucémie SN-36 sont utilisés pour examiner l'activité de TAK-N et CMTAK pour le retardement de divers types de tumeurs.
On a transplanté sur des souris ICR-JCL, d'un poids moyen de 23 g, par voie sous-cutanée, 3,1 x 106 cellules d'Ehrlich, 1,1 x 107 cellules CCM, 4,5 x 106 cellules NTF ou 7,2 x 104 cellules SN-36, et l'on a administré à ces souris, par voie intrapéritonéale, du TAK
N (DP 540) en suspension dans l'eau distillée, ou du CMTAK No 1 dissous dans l'eau distillée (pH 7,0) et passé à autoclave, à des doses de 10 à 50 mg/kg, quotidiennement, pendant 10 d, en commençant 24 h après la transplantation. Les tumeurs furent excisées et pesées 35 d après la transplantation, et les taux d'inhibition des tumeurs dans les groupes traités furent calculés comme dans l'exemple 1.
Comme on le voit au tableau 7, on a observé, dans chaque cas, une inhibition importante de la croissance de la tumeur.

Le TAK-N (TAK-D ou CMTAK) et le lactose sont mélangés
dans les proportions indiquées, pastillés et pulvérisés. On ajoute
ensuite le stéarate de magnésium. Le mélange est distribué en capsu
les.
Exemple 9:
TAK-N (remplaçable par TAK-D ou CMTAK) 400 mg
Lactose 95 mg
HPC-L (hydroxypropylcellulose) 5 mg
Total 500 mg
La quantité précitée forme une dose unique.
Les trois ingrédients indiqués sont mélangés dans les proportions
mentionnées et, avec addition d'une petite quantité d'eau, le mélange
est passé au malaxeur, granulé, séché, de nouveau granulé, tamisé et
aggloméré suivant les doses indiquées ci-dessus.
Exemple 10:
a) On dissout 1 g de TAK-D (DP 16) dans 1000 ml d'eau distillée pour l'injection (ou d'une solution saline physiologique), et l'on filtre la solution, le filtrat étant réparti dans des ampoules, en portions de 500 ml. Après scellage, les ampoules sont stérilisées à chaud de la façon habituelle.
b) On dissout 2 g de CMTAK (produit de l'exemple 14) dans 100 ml d'eau distillée pour l'injection (ou d'une solution saline physiologique), et l'on filtre la solution. Le filtrat est réparti dans des ampoules, en portion de 20 ml et, après scellage, les ampoules sont stérilisées à chaud de la façon habituelle.
Exemple 11:
TAK-N (remplaçable par TAK-D ou CMTAK) 160 mg
Sorbitol 200 mg
Carboxyméthylcellulose, sel de sodium 10 mg
Polysorbate 80 3,2 mg
Méthyl-p-hydroxybenzoate 4 mg
Propyl-p-hydroxybenzoate 0,4 mg
Les ingrédients ci-dessus sont mélangés dans l'eau distillée pour l'injection, pour former un volume total de 4 ml. (Lorsque le
CMTAK est employé, la solution est neutralisée, si nécessaire, avec de l'hydroxyde de sodium N/10.)
Exemple 12:
Dans 80 ml d'alcool isopropylique, on met en suspension 3 g de
TAK-N (DP 540), et l'on agite cette suspension à la température
ambiante pendant 30 min. On ajoute ensuite lentement, tout en
agitant, pendant environ 60 min, 8 ml d'une solution à 30% d'hy
droxyde de sodium.
On continue d'agiter vigoureusement le mélange
à température ambiante, pendant environ 90 min, en vue d'empê
cher la formation d'un gel. On ajoute ensuite 3,6 g d'acide monochloracétique.et le mélange est agité à 60-70 C pendant 5 h, afin que la carboxyméthylation s'effectue. Le produit est recueilli par filtration et lavé à fond avec un mélange de méthanol et d'acide acétique (7/3, v/v). On recueille le précipité par filtration, on le lave convenablement avec du méthanol aqueux à 80%, du méthanol et de l'acétone, dans l'ordre mentionné, et on le sèche sous pression réduite.
On obtient, selon cette technique, 2,9 g de CMTAK. Teneur en carboxyméthyle (nombre de groupes de carboxyméthyle par résidu de glucose; la même définition s'appliquant également ci-après): 0,54.
Exemple 13:
Dans 40 mi d'alcool isopropylique, on met en suspension 1,5 g de TAK-N (DP 540), et on agite à température ambiante pendant 30 min. On ajoute ensuite, sous agitation, 2 ml d'une solution à 30% d'hydroxyde de sodium, en 4 fractions, chacune de 0,5 ml, à intervalles de 15 min. Le mélange est ensuite agité à température ambiante pendant 90 min. On ajoute 0,9 g d'acide monochloracétique, en 3 fractions, chacune de 0,3 g, à intervalles de 10 min. La carboxy méthylation est ainsi conduite à 50 C, sous agitation pendant 150 min. Le produit est rassemblé par centrifugation, dissous dans 50 ml d'eau et neutralisé à l'acide acétique. A cette solution neutre, on ajoute 120 ml de méthanol, et le précipité formé est rassemblé par centrifugation.
Le précipité est lavé par un mélange de 300 ml de méthanol aqueux à 80% et 100 ml d'éthanol, puis par un mélange de 300 ml de méthanol aqueux à 80% et 200 ml d'éther. Le produit est finalement lyophilisé, de manière à obtenir 1,7 g de CMTAK.
Teneur en carboxyméthyle: 0,75.
Exemple 14:
Dans 40 d'alcool isopropylique, on met en suspension 1,5 g de TAK-N (DP 540), et on agite la suspension à température ambiante pendant 30 min. On ajoute ensuite 4 ml d'une solution à 30% d'hydroxyde de sodium, en 4 fractions de 1 ml chacune, à intervalles de 15 min, puis on continue l'agitation à température ambiante pendant 90 min.
On ajoute ensuite 1,8 g d'acide monochloracétique, en 3 fractions, chacune de 0,6 g, à intervalles de 10 min. On agite le mélange à 50c C pendant 150 min, ce qui permet d'effectuer la carboxyméthylation. Le produit est rassemblé par centrifugation, dissous dans 40 ml d'eau et neutralisé par l'acide acétique. A cette solution neutre, on ajoute 90 ml de méthanol et l'on rassemble le précipité formé par centrifugation. Le précipité est bien lavé avec un mélange de 200 ml de méthanol aqueux à 80% et 100 ml d'éthanol, puis avec un mélange de 200 ml d'éthanol aqueux à 80% et 200 ml d'éther. On lyophilise ensuite pour obtenir 2,0 g de CMTAK.
Teneur en carboxyméthyle: 1,07.
Exemple 15:
Dans 33 ml d'eau, on met en suspension 3,2 g de TAK-N (DP 255), et on ajoute, sous agitation à température ambiante, 1 g d'hydroxyde de sodium, puis 2,4 g de monochloracétate de sodium.
La réaction de carboxyméthylation est ainsi conduite à température ambiante, sous agitation constante pendant 2 h. On ajoute ensuite
I g d'hydroxyde de sodium et, de nouveau, 2,4 g de monochloroacétate de sodium et on continue la réaction à température ambiante sous agitation pendant 3 h. On ajoute encore g d'hydroxyde de sodium et 2,4 g de monochloracétate de sodium. On poursuit la réaction à température ambiante sous agitation pendant 2 h supplémentaires. On ajoute au mélange réactionnel il d'éthanol, et le précipité formé est lavé convenablement à 1'éthanol sur filtre en verre fritté, jusqu'à ce que le filtrat cesse de donner une coloration rouge à la phénolphtaléine. On sèche ensuite à 50 C sous pression réduite.
La poudre formée (3,7 g) est dissoute dans 90 ml d'eau et neutralisée à l'acide acétique, puis on ajoute 210 ml d'éthanol. Le précipité formé est rassemblé par centrifugation, lavé à l'méthanol aqueux à 80% et lyophilisé. On obtient par cette méthode 2,6 g de CMTAK.
Teneur en carboxyméthyle: 0,30.
Exemple 16:
Dans 66 ml d'eau, on met en suspension 6,4 g de TAK-N (DP 255), et on ajoute, sous refroidissement à la glace et agitation, 4 g d'hydroxyde de sodium, puis 9,6 g de monochloracétate de sodium. La carboxyméthylation est conduite sous refroidissement à la glace et agitation pendant 2 h. On ajoute de nouveau 4 g d'hydroxyde de sodium et 9,6 g de monochloracétate de sodium et on agite le mélange pendant 3 h en refroidissant avec de la glace. On ajoute ensuite 4 g d'hydroxyde de sodium et, une nouvelle fois, 9,6 g de monochloracétate de sodium, et on poursuit la réaction avec refroidissement à la glace et agitation pendant 3 h.
On ajoute au mélange réactionnel Il d'éthanol, et le précipité formé est bien lavé à l'méthanol sur filtre en verre fritté, jusqu'à ce que le filtrat cesse de donner une couleur rouge à la phénolphtaléine, puis on sèche à 50 C sous pression réduite. La poudre formée (8,6 g) est dissoute dans 172 ml d'eau et neutralisée à l'acide acétique. Puis, après addition de 480 ml d'éthanol, le précipité est rassemblé par centrifugation, lavé à l'méthanol aqueux à 80% et lyophilisé. On obtient, suivant cette méthode, 5,6 g de CMTAK. Teneur en carboxyméthyle: 0,36.
Exemple 17:
Dans 40 ml d'alcool isopropylique, on met en suspension 1,5 g de TAK-D (F-I, DP 299), et l'on effectue la carboxyméthylation comme dans l'exemple 13. Le produit est lavé, comme dans l'exemple 14, pour obtenir 1,9 g de CMTAK. Teneur en carboxyméthyle: 0,59.
On met en suspension dans 40 ml d'alcool isopropylique le même
TAK-D que ci-dessus, et on effectue la carboxyméthylation suivant la même méthode que celle décrite dans l'exemple 14. Le produit réactionnel est lavé comme dans l'exemple 14, ce qui permet d'obtenir 2,2 g de CMTAK. Teneur en carboxyméthyle: 1,15.
Exemple 18:
Dans 40 ml d'alcool isopropylique, on met en suspension 1,5 g, respectivement, de TAK-D (F-II, DP 113) et de TAK-D (S-III,

The invention relates to new carboxymethylated derivatives of B-1,3-glucan, which can be used for the preventive and curative treatment of tumors.
For years, research has been carried out with a view to finding a substance capable of causing, by parenteral or oral administration, the inhibition of tumor growth and the prolongation of the survival time of warm-blooded animals suffering from tumors, without serious side effects.
The object of the invention is to provide new carboxymethylated derivatives of B-1,3-glucan having the following formula:

wherein at least one of R is -CH2COOH; the rest, if any, being H; n is an integer between 2 and 1000, and its salt.
It is already known from Japanese patents Nos. 32673/1973 and 32674/1973, and from English patent No. 1352938, that certain strains of microorganisms of the genus Alcaligenes and of the genus Agrobacterium produce a thermogellable ss-1,3-glucan insoluble in water (hereinafter abbreviated as TAK-N). However, no pharmaceutical use of glucan has been reported.
The advanced studies carried out by the inventors on TAK-N, its lower polymers obtained by partial hydrolysis (hereinafter designated by TAK-D) and the carboxymethylated derivatives of TAK-N or of said lower polymers (hereinafter designated by CMTAK) led to the discovery that these substances have strong anti-tumor activity, resulting in a remarkably high rate of tumor inhibition and complete regression of these tumors.
CMTAK can be obtained by carboxymethylation of TAK
N or TAK-D in a known manner, for example by reacting the
TAK-N or TAK-D with monochloroacetic acid in the presence of an alkali. In addition to this method, any other conventional process used for the carboxymethylation of carbohydrates can be used.
To recover CMTAK from the reaction mixture, it is possible to use conventional methods which are commonly used for the purification of carbohydrates, such as precipitation by addition of an organic solvent. In addition, other techniques, such as ion exchange chromatography, gel filtration, etc., can be used according to the content of carboxymethyl groups in CMTAK, or according to the water solubility of CMTAK.
While the CMTAK thus obtained contains carboxymethyl groups in its molecule, the content of the thylated carboxymethyl groups varies widely depending on the conditions of the carboxymethylation reaction. As determined by titration, the content of carboxymethyl groups in CMTAK obtained by normal carboxymethylation reaction is generally less than three carboxymethyl groups per glucase residue in the CMTAK molecule.
CMTAK, as a product suitable for the purposes of the invention, contains all the substances which can be obtained by carboxymethylation of TAK-N or TAK-D, the molecules of which contain detectable carboxymethyl groups, independently of the degree of carboxymethylation .
As is apparent from the general formula indicated above, CMTAK, in the form of its free acid, is capable of reacting with various bases, to form the corresponding salts, such as sodium, potassium, calcium, aluminum salts, magnesium and amines. In the context of the invention, CMTAK includes not only its free acid, but also its salts, in particular those of low toxicity.
Table 1 illustrates the physical properties of some species
types of CMTAK prepared from TAK-N and TAK-D, according to the techniques previously indicated. The determinations of these physical properties were carried out after each sample was previously dried under reduced pressure and on phosphorus pentoxide at 60 ° C. for 10 h.

Figs. 1 and 2 illustrate the I.R. spectra in po bromide
tassium, from CMTAK, respectively Nos 2 and 11 in table 1. The figures at the top left and bottom of each figure indicate respectively the wavelengths (, u), the transmission coefficients or transmittances (%) and the numbers of waves (cm - 1).
Characteristic absorptions were found at 1440-1400 and 891 cm5.
The CMTAK of DP ranging from 2 to approximately 1000, preferably from
15 to 800, and more advantageously from 40 to 600, has a remarkable inhibitory activity against different kinds of tumors in warm-blooded animals including domestic animals, poultry, dogs, cats, rabbits, rats, mice, etc., and in particular against serious tumors known to be less susceptible to treatment with generally known cytotoxic anti-tumor drugs.
For example, the growth of sarcoma 180, Ehrlich's carcinoma, SN-36 leukemia, CCM adenocarcinoma, or NTF reticular cell sarcoma, each tumor being transplanted subcutaneously in mice, was significantly inhibited by administering TAK-N, TAK-D or CMTAK by intraperitoneal, intravenous or subcutaneous injection, before, after or at the time of transplantation, in one or more doses, in doses of approximately 1 to 1000 mg / kg per administration, per day.
The toxicities of CMTAK are extremely low. For example, no toxic effect is observed, in both mice and rats, at a dose of 3000 mg / kg intraperitoneally, or 10000 mg / kg orally. Thus, these polysaccharides can be safely and repeatedly administered to warm-blooded animals.
The administration can be carried out according to the general techniques usually applied for the treatment of cancers. Are thus applicable intratumoral, subcutaneous, intramuscular or intravenous injections, if necessary, oral and rectal administrations, or even external applications, brushing, instillations and other methods of administration.
CMTAK is administered in an amount sufficient to a warm-blooded animal to inhibit the growth of its tumor.
The dosage and method of administration of CMTAK to warm-blooded animals with neoplasia may vary depending on the type of agents, animals and patients, tumor symptoms, forms of administration and other factors. In general, the sufficient dosage amount per administration is from about 0.02 to 2000 mg / kg bodyweight, preferably from 0.2 to 2000 mg / kg bodyweight, the preferred upper limit being about 500 mg / kg bodyweight. In many cases, the most advantageous upper limit is around 2000 mg / kg.
The administration can be carried out, for example, one to six times a day, for several consecutive days or intermittently.
TAK-N, TAK-D and CMTAK can be administered in combination with other anti-tumor agents. Combination with such agents causing increased immunological activity in animals with tumors is also advantageous.
The invention will now be described with reference to the examples below. Some of these examples illustrate the technique for producing CMTAK, the physical properties of CMTAK and certain injectable forms of this product. Other examples demonstrate the anti-tumor activities of CMTAK against tumors in mice, using standard models.
As indicated above, the treatment of animals suffering from tumors with the compounds produces an energetic regression of the various types of tumors, such as sarcomas, carcinomas, reticular cell sarcomas and leukemias. All of these types of tumors in rodents have been shown to be exact classic types of tumors in other warm-blooded animals.
These examples do not of course have any limiting character of the invention.
Most of the following trials on the anti-tumor activity of
CMTAK are evaluated using approximately the same methods as those described for the antitumor activity of lentinan, a branched glucan from an edible fungus, according to
Chihara, G. et al. [see Nature, 222, 687-688 (1969), and Cancer
Research, 30, 2776-2781, 1970)].
The tumors of the mice used in these trials, such as sarcoma 180 (S 180), Ehrlich carcinoma (Ehrlich) and reticular cell sarcoma NTF (NTF), adenocarcinoma CCM (CCM) and leukemia SN-36 (SN-36) , are maintained in ICR- type mice
JCL, from 5 to 7 weeks, each in ascites forms. An appropriate element of these tumor cells is transplanted subcutaneously into ICR-JCL mice, and the tumor nodules which have developed are excised for weighing, on the 35th day after transplantation of the tumor.
A TAK-N or TAK-D was suspended or dissolved in distilled water or in physiological solution, and the CMTAK was dissolved in distilled water or in physiological saline at appropriate concentrations, and at pH close to neutrality. Each preparation was administered intraperitoneally, subcutaneously or intravenously to mice (0.2 ml / 20 g bodyweight) daily for 10 d, starting 24 h after tumor transplant, daily for 5 d before the transplant of the tumor, or only once during the 7th day before and the 2nd day after the implantation of the tumor.
Example 1:
Effect of tumor suppression by TA K-N (DP 540): single injection.
4 million S 180 cells are transplanted subcutaneously into the left inguinal region of female mice of the type
ICR-JCL, 4 to 5 weeks. The nodules of the tumor are excised on the 35th day after transplantation and weighed. The average weight of a tumor in a treated group (T) comprising 5 mice was compared with that of a control group (C) in 10 mice, and the inhibition rate of the tumor was calculated ( inhibition%: (CT) x100).

TAK-N (DP 540) suspended in physiological saline solution (10 mg / ml) was administered intraperitoneally (0.2 ml / 20 g bodyweight) once to each animal, at the dose of 100 mg / kg, on the 7th, 3rd or 1st day before, or on the 1st, 2nd, 3rd, 5th, 10th, 14th or 21st day after the tumor transplant, or on the day of the tumor transplant.
As seen in Table 2, TAK-N at the dose of 100 mg / kg markedly inhibits the growth of S 180, when the test sample is administered once during the 7th day before and the 21st day after the tumor transplant.
In addition, by single administration of TAK-N (DP 540) on the 7th day after transplantation of the tumor at doses of 20 mg / kg, 60 mg / kg and 100 mg / kg, marked suppression of the tumor growth at the two higher doses, 60 mg / kg and 100 mg / kg.

These results indicate that TAK-N (DP 540) is effective in suppressing tumor growth, at all stages of tumor progression in warm-blooded animals, even by single administration.
Example 2:
Effect of tumor suppression by TAK-N (DP 540) and TAK N flo culated in autoclave (DP 540).
6 million S 180 cells are transplanted subcutaneously into the right inguinal region of ICR-JCL mice of a live weight of approximately 23 g. The tumor nodes are excised on the 35th day after transplantation and weighed. The tumor inhibition rate in a treated group was calculated as in Example 1.
TAK-N (DP 540) and flocculated TAK-N (DP 540) were used.
The latter was obtained by heating a suspension of 2% TAK
N (DP 540) at 1200 C for 25 min, then drying and spraying the gel formed. The samples were suspended in distilled water at the desired concentrations, and each suspension was administered intraperitoneally to a group of 5 mice, daily for 10 d, starting 24 h after transplantation of the tumor.
As seen in Table 3, TAK-N (DP 540) exerts a pronounced effect on tumor growth at doses of I to 50 mg / kg, this effect being particularly important at doses of 5 to 25 mg / kg.
TAK-N (DP 540) has also been shown to be effective in suppressing tumor growth at doses of 10 mg / kg. No toxicity was observed.

These results show that TAK-N is used in slowing the growth of tumors, at relatively low doses, and that the sample can be sterilized by autoclave treatment, without losing its activity.
Example 3:
Effects of tumor suppression by CMTAK and CMTAK treated it autoclave.
The antitumor activity of three types of CMTAK prepared from TAK-N (CMTAK Nos 1,4, 5) and autoclaved CMTAK was tested, as described in Example 2. The samples were dissolved in distilled water for injection, and the pH was adjusted to about 7.0. In some tests, one of these samples (CMTAK No 1) was sterilized by autoclave treatment (1200 ° C., 2.5 min) before any other administration.
As seen in Table 4, all of the samples of
CMTAK, at doses of 10 mg / kg, significantly inhibited tumor growth. Growth inhibition by CMTAK has been observed at doses of I to 40 mg / kg.

These results show that CMTAK can be sterilized without
loss of its antitumor activity and used in an injectable form
for cancer treatment.
Example 4:
Effects of tumor suppression by TAK-N, TAK-D and CMTAK following various methods of administration.
TAK-N (DP 540), TAK-D (F-III: DP 50), TAK-D (F-VII:
DP 16) and CMTAK No 1 were administered at doses of 5 mg / kg, intravenously, subcutaneously or intraperitoneally for
10 consecutive dice ICR-JCL mice which had previously undergone a transplant with 4> <x 106 10 cells, 24 h before the first
first injection. Inhibition rates of tumor growth
were calculated as in Example 1.
As seen in Table 5, in each case, significant inhibition of tumor growth was observed. By intravenous administration, TAK-N, CMTAK and even F-VII (DP 16) almost completely inhibit tumor growth, the inhibition rate being greater than 85%, with complete tumor regression out of 4 or 5 of the 5 mice treated.

Example 5:
Effects of tumor suppression by TAK-N, TAK-D and CMTAK ad-

As can be seen in the table, a marked inhibition of tumor growth is observed for each sample. In particular, TAK-N (DP 540); CMTAK and TAK-D with DP greater than 50 have strong inhibitory activity. CMTAK was effective in delaying tumors at low doses, on the order of 3 mg / kg.

Example 6:
Repressive activity of tumors by TAK-N and CMTAK against various types of tumors.
Ehrlich's carcinoma, CCM adenocarcinoma, NTF reticular cell sarcoma and SN-36 leukemia are used to examine the activity of TAK-N and CMTAK for the retardation of various types of tumors.
ICR-JCL mice, 23 g mean weight, were transplanted subcutaneously, 3.1 x 106 Ehrlich cells, 1.1 x 107 TLC cells, 4.5 x 106 NTF cells or 7.2 x 104 SN-36 cells, and these mice were administered intraperitoneally with TAK
N (DP 540) suspended in distilled water, or CMTAK No 1 dissolved in distilled water (pH 7.0) and autoclaved, at doses of 10 to 50 mg / kg, daily, for 10 d, starting 24 hours after transplantation. The tumors were excised and weighed after transplantation, and the tumor inhibition rates in the treated groups were calculated as in Example 1.

Example 12:
In 80 ml of isopropyl alcohol, 3 g of
TAK-N (DP 540), and this suspension is stirred at temperature
room for 30 min. Then add slowly, while
shaking, for approximately 60 min, 8 ml of a 30% hy solution
sodium hydroxide.
Continue to vigorously stir the mixture
at room temperature, for about 90 min, to prevent

1.5 g of TAK-N (DP 540) are suspended in 40 ml of isopropyl alcohol and the mixture is stirred at room temperature for 30 min. Then added, with stirring, 2 ml of a 30% sodium hydroxide solution, in 4 fractions, each of 0.5 ml, at 15 min intervals. The mixture is then stirred at room temperature for 90 min. 0.9 g of monochloroacetic acid are added, in 3 fractions, each of 0.3 g, at 10 min intervals. The carboxy methylation is thus carried out at 50 ° C., with stirring for 150 min. The product is collected by centrifugation, dissolved in 50 ml of water and neutralized with acetic acid. To this neutral solution, 120 ml of methanol are added, and the precipitate formed is combined by centrifugation.
The precipitate is washed with a mixture of 300 ml of 80% aqueous methanol and 100 ml of ethanol, then with a mixture of 300 ml of 80% aqueous methanol and 200 ml of ether. The product is finally lyophilized, so as to obtain 1.7 g of CMTAK.
Carboxymethyl content: 0.75.
Example 14:
1.5 g of TAK-N (DP 540) are suspended in isopropyl alcohol, and the suspension is stirred at room temperature for 30 min. 4 ml of a 30% sodium hydroxide solution are then added, in 4 fractions of 1 ml each, at 15 min intervals, then stirring is continued at room temperature for 90 min.
Then 1.8 g of monochloroacetic acid are added, in 3 fractions, each 0.6 g, at 10 min intervals. The mixture is stirred at 50 ° C. for 150 min, which makes it possible to carry out the carboxymethylation. The product is collected by centrifugation, dissolved in 40 ml of water and neutralized with acetic acid. To this neutral solution, 90 ml of methanol are added and the precipitate formed by centrifugation is combined. The precipitate is washed well with a mixture of 200 ml of 80% aqueous methanol and 100 ml of ethanol, then with a mixture of 200 ml of 80% aqueous ethanol and 200 ml of ether. Example 15:
3.2 g of TAK-N (DP 255) are suspended in 33 ml of water and 1 g of sodium hydroxide is added, with stirring at room temperature, followed by 2.4 g of monochloroacetate. sodium.

The powder formed (3.7 g) is dissolved in 90 ml of water and neutralized with acetic acid, then 210 ml of ethanol are added. The precipitate formed is combined by centrifugation, washed with 80% aqueous methanol and lyophilized. This method gives 2.6 g of CMTAK.

Example 16:
6.4 g of TAK-N (DP 255) are suspended in 66 ml of water, and 4 g of sodium hydroxide are added with ice cooling and stirring, followed by 9.6 g of sodium monochloroacetate. The carboxymethylation is carried out under ice cooling and stirring for 2 h. Again 4 g of sodium hydroxide and 9.6 g of sodium monochloroacetate are added and the mixture is stirred for 3 h while cooling with ice. Then 4 g of sodium hydroxide and, again, 9.6 g of sodium monochloroacetate are added, and the reaction is continued with cooling in ice and stirring for 3 h.
Ethanol is added to the reaction mixture II, and the precipitate formed is washed well with methanol on a sintered glass filter, until the filtrate stops giving a red color to the phenolphthalein, then it is dried at 50 ° C. under reduced pressure. 1.5 g of TAK-D (FI, DP 299) are suspended in 40 ml of isopropyl alcohol, and the carboxymethylation is carried out as in Example 13. The product is washed, as in Example 14, to obtain 1.9 g of CMTAK. Carboxymethyl content: 0.59.
The same is suspended in 40 ml of isopropyl alcohol
TAK-D as above, and the carboxymethylation is carried out according to the same method as that described in Example 14. The reaction product is washed as in Example 14, which allows 2.2 g of CMTAK to be obtained . Carboxymethyl content: 1.15.
Example 18:
1.5 g, respectively, of TAK-D (F-II, DP 113) and TAK-D (S-III,
DP 68), then the carboxymethylation is carried out as in Example 13. The respective reaction products are combined by centrifugation, dissolved in 40 ml of water and neutralized with acetic acid.
90 ml of methanol are added to each of these two neutral solutions, and the resulting precipitate is combined by centrifugation and washed well, first with 200 ml of 80% aqueous methanol and then with 200 ml of aqueous ethanol, then freeze-drying. Two types of CMTAK are obtained by this method.
