PATENTANSPRÜCHE
1. Verfahren zur Herstellung eines nach dem Osmoseprinzip arbeitenden Wirkstoff-Spenders durch Verformen einer wirkstoffhaltigen, osmotisch wirksamen Zubereitung zu einer festen Masse, dem Kern (13, 22), Aufsprühen eines semipermeablen Materials auf diesen Kern unter Bildung einer den Kern umschliessenden Wand und Bildung einer Aus trittsöffliung in der Wand, dadurch gekennzeichnet, dass man vor dem Aufsprühen des semipermeablen Materials (16) in dem Kern (13, 22) eine Vertiefung (15, 23) anbringt, die so breit und tief ist, dass sie beim Besprühen mit dem semipermeablen Material (16) mindestens teilweise nicht überzogen wird.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die Vertiefung gleichzeitig mit der Bildung des Kerns (22) in die Masse (10) einpresst.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass die Breite der Vertiefung mindestens zweimal so gross ist wie die Dicke der Wand und dass das Verhältnis von Tiefe zu Breite mindestens 1,5:1 beträgt.
4. Verfahren nach Anspruch 3, dadurch gekennzeichnet, dass die Breite der Vertiefung mindestens fünfmal so gross ist wie die Dicke der Wand und dass das Verhältnis von Tiefe zu Breite mindestens 2:1 beträgt.
Die Erfindung betrifft ein Verfahren zur Herstellung eines nach dem Osmoseprinzip arbeitenden Wirkstoff-Spenders, wie er in der DE-OS 2 328 409 beschrieben ist. Dieser Spender besteht im wesentlichen aus einem inneren Kern aus einem osmotisch wirksamen, den Wirkstoff enthaltenden Mittel und einer äusseren formbeständigen semipermeablen Wand, die den inneren Kern umschliesst. Diese Wand besitzt eine kleine Öffnung, die als Austrittsöffnung dient, durch die das wirksame Mittel in Lösung abgegeben wird. Dieser Spender wird hergestellt, indem man zunächst den Kern bildet, z. B. durch Verpressen, diesen mit einem semipermeablen Polymer zur Bildung der Wand überzieht und dann in der Wand bzw. dem Überzug ein Loch bildet. Zur Erzeugung dieses Loches wurden verschiedene Verfahren angewandt, wie Durchbohren der Wand mit einem Bohrer oder einem Laserstrahl.
Das erfindungsgemässe Verfahren zur Herstellung eines nach dem Osmoseprinzip arbeitenden Wirkstoff-Spenders durch Verformen einer wirkstoffhaltigen, osmotisch wirksamen Zubereitung zu einer festen Masse, dem Kern, Aufsprühen eines semipermeablen Materials auf diesen Kern unter Bildung einer den Kern umschliessenden Wand und Bildung einer Austrittsöffnung in der Wand ist dadurch gekennzeichnet, dass man vor dem Aufsprühen des semipermeablen Materials in dem Kern eine Vertiefung anbringt, die so breit und tief ist, dass sie beim Besprühen mit dem semipermeablen Material mindestens teilweise nicht überzogen wird.
Die Erfindung wird anhand der beiliegenden Zeichnung und der folgenden Beschreibung beispielsweise näher erläutert. Dabei wird ein erfindungsgemäss hergestellter Spender mit einem nach dem bekannten Verfahren hergestellten Spender verglichen. Bei den Zeichnungen ist:
Fig. 1 ein Teilschnitt durch einen tablettenförmigen Kern eines Spenders, der mit Hilfe einer Pressform und eines Stempels hergestellt wird,

Die Fig. 1 bis 3 zeigen ein 3stufiges erfindungsgemässes
Verfahren zur Herstellung eines tablettenförmigen Wirk stoffspenders, wie er zur oralen Verabreichung von Arz neimitteln angewandt werden kann. In der ersten in Fig. 1 gezeigten Stufe wird eine osmotisch wirksame, den Wirkstoff enthaltende Zubereitung 10 in den Hohlraum der Pressform
11 gegeben und mit Hilfe des Stempels 12 zu dem festen
Kern 13 (Fig. 2) verpresst. Nachdem der Kern 13 geformt ist, wird der Stempel 12 aus der Pressform entfernt. In der in Fig. 2 gezeigten zweiten Stufe wird ein zweiter Stempel 14 in den Hohlraum der Pressform 11 eingeführt und gegen die Oberseite des Kerns 13 gepresst, wobei die konische Vertiefung 15 entsteht. Nach Bildung der Vertiefung 15 wird der Stempel 14 entfernt und der Kern 13 aus dem Hohlraum der Pressform 11 entnommen.
Die dritte Stufe des Verfahrens ist in Fig. 3 angegeben und umfasst das Überziehen des mit der Vertiefung versehenen Kerns 13 mit einem semipermeablen Polymer 16. Das angewandte Verfahren zum Uberziehen ist ein übliches Versprühen (air suspension coating), bei dem das Polymer 16 mit Hilfe eines Luftstroms versprüht und der Kern 13 in diesen Luftstrom gebracht wird. Das in dem Luftstrom enthaltene Polymer ist durch die gekrümmten punktierten Linien in der Fig. 3 angegeben. Dieses Polymer verteilt sich gleichmässig auf der Oberfläche des Kerns 13 mit Ausnahme der Vertiefung 15 unter Bildung der Wand 19 um den Kern 13. Die Dimensionen der Vertiefung 15 sind so gewählt, dass das Polymer 16 nur teilweise in die Vertiefung 15 eindringt. So erreicht das aufgebrachte Polymer 16 nicht den Boden der Vertiefung 15, und die Öffnung der Vertiefung 15 wird nicht vollständig bedeckt.
Dadurch entsteht in der Wand 19 ein Loch bzw. eine Offnung 18 an der Stelle der Vertiefung 15 beim Uberziehen. Der fertige Spender, der in Fig. 4 als 17 bezeichnet ist, besteht (a) aus dem Kern 13 aus der den Wirkstoff enthaltenden, osmotisch wirksamen Zubereitung 10, die von (b) der Wand 19 aus einem semipermeablen Polymer 16 umgeschlossen ist, und (c) dem Loch bzw. der Austrittsöffnung 18, die sich durch die Wand 19 zu dem Kern 13 an der Stelle der Vertiefung 15 erstreckt.
Die Fig. 5 und 6 zeigen eine alternative Arbeitsweise zur Durchführung des erfindungsgemässen Verfahrens, die nur zwei Schritte umfasst. In der ersten in Fig. 5 angegebenen Stufe wird die den Wirkstoff enthaltende osmotisch wirksame Zubereitung 10 in den Hohlraum einer Form 20 gegeben und mit einem Stempel 21 verpresst, der in die Pressform 20 eingeführt wird. Die Form des Stempels 21 ist so gewählt, dass die Zubereitung 10 zu einem scheibenförmigen festen Kern 22 verpresst und gleichzeitig in dem Kern 22 eine zylin derförmige Vertiefung 23 gebildet wird. Nachdem der Kern mit der Vertiefung gebildet worden ist, wird der Stempel 21
entfernt und der Kern 22 aus der Pressform entnommen. In der zweiten Stufe, die in Fig. 6 angegeben ist, wird der Kern 22 nach dem bei Fig. 3 beschriebenen Verfahren mit dem semipermeablen Polymer 16 überzogen. Der nach der in den Fig. 5 und 6 angegebenen Arbeitsweise erhaltene Wirkstoffspender ist in Fig. 7 gezeigt (24). Dieser Spender 24 besteht aus einem inneren Kern 22 aus der den Wirkstoff enthaltenden osmotisch wirksamen Zubereitung 10, die von der äusseren Wand 25 aus dem Polymer 16 umgeben ist. Die Wand 25 besitzt eine Öffnung 26 an der Stelle der Vertiefung 23.
Die Öffnung 26 ist auf die gleiche Weise entstanden, wie die Öffnung 18 bei der Vorrichtung 17.
Die Dimensionen der Vertiefung, die vor dem Überziehen in dem Kern gebildet wird, sind ein wichtiger Faktor.
Die Breite der Vertiefung ist üblicherweise mehr als zweimal so gross wie die Dicke des auf den Kern aufzubringenden Überzugs. Im allgemeinen beträgt sie ungefähr das Fünffache der Dicke des aufzubringenden Überzugs. Die Dicke des Überzugs liegt üblicherweise in der Grössenordnung von 100 bis 300 llm. Bei Vertiefungen, deren Breite sich ändert, z. B.
bei konischen Vertiefungen, kann die äussere Weite zur Bestimmung der oben angegebenen Beziehung zur Dicke des Überzugs dienen. Das Verhältnis Tiefe: Breite der Vertiefung beträgt üblicherweise mindestens ungefähr 1,5:1 und insbesondere mindestens ungefähr 2:1. Wieder kann bei Vertiefungen deren Weite variiert die Weite an der äusseren Öffnung der Vertiefung zur Bestimmung dieses Verhältnisses angewandt werden. Die Form der Vertiefung wird nicht als kritisch angesehen. Der Einfachheit halber besitzt sie üblicherweise eine gleichmässige Form und ist z. B. zylindrisch, konisch, kegelstumpfförmig, halbkreisförmig oder kubisch.
Die Vertiefung oder Einbuchtung kann nach bekannten Verfahren, wie Pressen, Bohren, Laserbohren und ähnlichem hergestellt werden.
Die Zusammensetzung der osmotisch wirksamen wirkstoffhaltigen Zubereitung, wie sie zur Herstellung des Kerns der Vorrichtung angewandt wird, und die Zusammensetzung des semipermeablen Materials, das zur Bildung der Wand um den Kern angewandt wird, sind nicht Gegenstand der Erfindung. Geeignete wirkstoffhaltige Mittel und semipermeable Substanzen sind z. B. in der erwähnten DE-OS 2 328 409 angegeben. Ebenso sollte die Grösse der Austritts öffnung in der Wand des Wirkstoffspenders nach der Erfindung mit den für die Austrittsöffnung in der DE-OS 2328 409 angegebenen Dimensionen übereinstimmen.
Die Erfindung wird durch das folgende Beispiel näher erläutert.
Beispiel
Es wurden nach dem Osmoseprinzip arbeitende Spender zur oralen Verabreichung von Theophyllin hergestellt. Kugelförmige (sphärische) Pressformen mit einem Durchmesser von ungefähr 0,8 cm wurden mit einem Gemisch aus 125 mg Theophyllinmonoäthanolamin einer Spur des kohäsiven Bindemittels Natriumalginat und einer Spur Magnesiumstearat als Gleitmittel gefüllt und das Gemisch zu Kugel verpresst. Eine zylinderförmige Vertiefung, mit einem Durchmesser von ungefähr 625 m und einer Tiefe von ungefähr 1250 ,um wurde in die Oberfläche jedes Kernes gebohrt. Anschliessend wurden die Kerne in einem Wirbelbett mit 6% Cellulose in Methylenchlorid: Methanol 80:20 (Gew.-%) überzogen. Das Celluloseacetat besass einen Acetylgehalt von 32%. Der entstehende Überzug war ungefähr 170 cm dick.
Die Grösse (Weite) der Öffnung betrug ungefähr 375 ilm an der Stelle der Vertiefung.
Es wurden Kerne wie oben hergestellt, gebohrt und überzogen mit der Ausnahme, dass die Vertiefung einen Durchmesser von ungefähr 940 pm und eine Tiefe von ungefähr 1775 um besass. Nach dem Überziehen besass der Uberzug bzw. die Wand an der Stelle der Vertiefung eine Öffnung von ungefähr 650 um. Ferner wurden Kerne wie oben hergestellt, gebohrt und überzogen, wobei die Vertiefung einen Durchmesser von ungefährt 1320 um und eine Tiefe von ungefähr 25401um besass. Nach dem Überziehen war in dem Überzug eine Öffnung von ungefähr 1060 um an der Stelle der Vertiefung entstanden.
Die in vitro-Freisetzungsgeschwindigkeit von Theophyllin, angegeben als freie Base, aus dem beschriebenen Spender mit einer 650 um grossen Öffnung in der Wand wurde bestimmt. Sie ist grafisch in Fig. 9 angegeben. Zum Vergleich ist die in vitro-Freisetzungsgeschwindigkeit für einen Spender, der in Struktur und Zusammensetzung im wesentlichen identisch ist, aber nach dem Verfahren der DE-OS 2 328 409 hergestellt worden ist, angegeben. Der Kern des Vergleichsspenders wurde nicht angebohrt, und die Auslassöffnung in dem Uberzug wurde mechanisch, nachdem der Überzug auf den Kern aufgebracht worden war, gebohrt. Die Freisetzungsgeschwindigkeit aus dem Vergleichsspender ist grafisch in Fig. 8 angegeben.

PATENT CLAIMS
1. A process for producing an active substance dispenser working according to the osmosis principle by shaping an active substance-containing, osmotically active preparation into a solid mass, the core (13, 22), spraying a semipermeable material onto this core to form a wall surrounding the core and forming it From an outlet opening in the wall, characterized in that, before spraying on the semipermeable material (16) in the core (13, 22), a recess (15, 23) is made which is so wide and deep that it is sprayed with the semipermeable material (16) is at least partially not coated.

The invention relates to a method for producing an active substance dispenser working according to the osmosis principle, as described in DE-OS 2 328 409. This dispenser consists essentially of an inner core of an osmotically active agent containing the active substance and an outer shape-retaining semi-permeable wall which encloses the inner core. This wall has a small opening which serves as an outlet opening through which the active agent is released in solution. This dispenser is made by first forming the core, e.g. B. by pressing, this with a semipermeable polymer to form the wall and then forms a hole in the wall or the coating. Various methods were used to create this hole, such as drilling the wall with a drill or a laser beam.
The method according to the invention for producing an active substance dispenser working according to the osmosis principle by shaping an active substance-containing, osmotically active preparation into a solid mass, the core, spraying a semipermeable material onto this core with formation of a wall enclosing the core and formation of an outlet opening in the wall is characterized in that, before spraying on the semipermeable material, a recess is made in the core which is so wide and deep that it is at least partially not covered when sprayed with the semipermeable material.
The invention is explained in more detail, for example, with reference to the accompanying drawing and the following description. A dispenser manufactured according to the invention is compared with a dispenser manufactured according to the known method. In the drawings:

Process for the preparation of a tablet-shaped active substance dispenser, as can be used for the oral administration of medicinal products. In the first stage shown in FIG. 1, an osmotically active preparation 10 containing the active ingredient is placed in the cavity of the mold

Core 13 (Fig. 2) pressed. After the core 13 is formed, the punch 12 is removed from the die. In the second stage shown in FIG. 2, a second punch 14 is inserted into the cavity of the mold 11 and pressed against the top of the core 13, the conical recess 15 being formed. After the depression 15 has been formed, the stamp 14 is removed and the core 13 is removed from the cavity of the mold 11.
The third stage of the process is indicated in FIG. 3 and involves coating the core 13 provided with the recess with a semipermeable polymer 16. The process used for coating is a conventional spraying (air suspension coating) in which the polymer 16 is coated with the aid of of an air stream is sprayed and the core 13 is brought into this air stream. The polymer contained in the air stream is indicated by the curved dotted lines in FIG. 3. This polymer is evenly distributed on the surface of the core 13 with the exception of the recess 15, forming the wall 19 around the core 13. The dimensions of the recess 15 are chosen such that the polymer 16 only partially penetrates into the recess 15. Thus, the applied polymer 16 does not reach the bottom of the recess 15 and the opening of the recess 15 is not completely covered.
This creates a hole or opening 18 in the wall 19 at the location of the recess 15 when pulled over. The finished dispenser, which is designated as 17 in FIG. 4, consists of (a) the core 13 from the active substance-containing, osmotically active preparation 10, which is enclosed by (b) the wall 19 from a semipermeable polymer 16, and (c) The hole or outlet opening 18 which extends through the wall 19 to the core 13 at the location of the recess 15.
5 and 6 show an alternative mode of operation for carrying out the method according to the invention, which comprises only two steps. In the first stage shown in FIG. 5, the osmotically active preparation 10 containing the active ingredient is placed in the cavity of a mold 20 and pressed with a stamp 21 which is inserted into the mold 20. The shape of the stamp 21 is chosen so that the preparation 10 is pressed into a disk-shaped solid core 22 and at the same time a cylindrical recess 23 is formed in the core 22. After the core with the recess has been formed, the stamp 21
removed and the core 22 removed from the mold. In the second stage, which is indicated in FIG. 6, the core 22 is coated with the semi-permeable polymer 16 according to the method described in FIG. 3. The active substance dispenser obtained according to the procedure given in FIGS. 5 and 6 is shown in FIG. 7 (24). This dispenser 24 consists of an inner core 22 made of the osmotically active preparation 10 containing the active ingredient, which is surrounded by the outer wall 25 made of the polymer 16. The wall 25 has an opening 26 at the location of the recess 23.

The dimensions of the indentation that is formed in the core before coating is an important factor.
The width of the depression is usually more than twice as large as the thickness of the coating to be applied to the core. In general, it is approximately five times the thickness of the coating to be applied. The thickness of the coating is usually in the order of 100 to 300 llm. For depressions whose width changes, for. B.
in the case of conical depressions, the outer width can be used to determine the above-mentioned relationship to the thickness of the coating. The depth: width ratio of the depression is usually at least approximately 1.5: 1 and in particular at least approximately 2: 1. Again, in the case of depressions whose width varies, the width at the outer opening of the depression can be used to determine this ratio. The shape of the recess is not considered critical. For the sake of simplicity, it usually has a uniform shape and is e.g. B. cylindrical, conical, frustoconical, semi-circular or cubic.
The depression or indentation can be produced by known methods, such as pressing, drilling, laser drilling and the like.
The composition of the osmotically active substance-containing preparation, as it is used for producing the core of the device, and the composition of the semi-permeable material, which is used for forming the wall around the core, are not the subject of the invention. Suitable active substance-containing agents and semipermeable substances are e.g. B. specified in the aforementioned DE-OS 2 328 409. Likewise, the size of the outlet opening in the wall of the active substance dispenser according to the invention should correspond to the dimensions specified for the outlet opening in DE-OS 2328 409.
The invention is illustrated by the following example.

Oral theophylline donors were made to work according to the osmosis principle. Spherical (spherical) molds with a diameter of approximately 0.8 cm were filled with a mixture of 125 mg of theophylline monoethanolamine, a trace of the cohesive binder sodium alginate and a trace of magnesium stearate as a lubricant, and the mixture was pressed into a sphere. A cylindrical recess approximately 625 m in diameter and approximately 1250 µm deep was drilled in the surface of each core. The cores were then coated in a fluidized bed with 6% cellulose in methylene chloride: methanol 80:20 (% by weight). The cellulose acetate had an acetyl content of 32%. The resulting coating was approximately 170 cm thick.
The size (width) of the opening was approximately 375 µm at the location of the recess.
Cores were made, drilled and coated as above except that the recess was approximately 940 µm in diameter and approximately 1775 µm deep. After coating, the cover or wall had an opening of approximately 650 µm at the location of the recess. Cores were also made, drilled and coated as above, the recess having a diameter of approximately 1320 µm and a depth of approximately 25401 µm. After coating, an opening of approximately 1060 µm was created in the coating at the location of the recess.
The in vitro release rate of theophylline, expressed as free base, from the described donor with a 650 µm opening in the wall was determined. It is indicated graphically in FIG. 9. For comparison, the in vitro release rate for a donor, which is essentially identical in structure and composition, but was produced by the process of DE-OS 2 328 409, is given. The reference dispenser core was not drilled and the outlet opening in the coating was mechanically drilled after the coating was applied to the core. The rate of release from the reference donor is shown graphically in FIG. 8.
