PATENTANSPRÜCHE
1. Verfahren zur Herstellung von p-Hydroxybenzylcyanid, dadurch gekennzeichnet, dass man a-(p-Hydroxyphenyl) -glycin mit Cyanidionen umsetzt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die Cyanidionen in Form eines Alkalimetallcyanids, vorzugsweise Natrium- oder Kaliumcyanid, verwendet.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass man es in einem hochsiedenden, dipolaren, aprotischen Lösungsmittel, vorzugsweise N,N-Dimethylformamid, 2-Pyrrolidon, N-Methyl-2-pyrrolidon oder Dimethylsulfoxid, ausführt.
4. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass man es in n-Butanol, 3-Methylbutanol, Acetamid oder 2-Äthoxyäthanol als Lösungsmittel ausführt.
5. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass man es in Wasser, Äthylenglykol, Formamid oder Harnstoff als Lösungsmittel ausführt.
6. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass man es bei einer Temperatur zwischen 100 und 250"C, vorzugsweise zwischen 120 und 1900C, ausführt.
7. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man es in Gegenwart einer Base, vorzugsweise eines Alkalimetallhydroxids, insbesondere Natrium- oder Kaliumhydroxid, ausführt.
Die Erfindung bezieht sich auf ein neues Verfahren zur Herstellung der Verbindung p-Hydroxybenzylcyanid, welche ein wertvolles chemisches Zwischenprodukt ist.
Erfindungsgemäss wird p-Hydroxybenzylcyanid hergestellt, indem man z-(p-Hydroxyphenyl)-glycin [a-Amino-x- (p-hydroxyphenyl)-essigsäurel mit Cyanidionen umsetzt.
Die Cyanidionen werden zweckmässig in Form eines Alkalimetallcyanids, beispielsweise Natrium- oder Kaliumcyanid, verwendet.
Die Umsetzung wird zweckmässig in einem verhältnismässig hochsiedenden, dipolaren, aprotischen Lösungsmittel, wie z.B. N,N-Dimethylformamid, 2-Pyrrolidon, N-Methyl-2 -pyrrolidon oder Dimethylsulfoxid, ausgeführt. Andere verhältnismässig hochsiedende polare Lösungsmittel, wie z.B.
n-Butanol, 3-Methylbutanol, Acetamid oder 2-Athoxyätha- nol, sind ebenfalls zufriedenstellende Lösungsmittel, und einige andere solcher Lösungsmittel, wie z.B. Wasser, Äthylenglykol, Formamid und Harnstoff, sind ebenfalls geeignet, obwohl weniger zufriedenstellend.
Die Reaktion wird zweckmässig bei einer Temperatur zwischen 100 und 250"C, vorzugsweise bei einer Temperatur zwischen 120 und 1900C, ausgeführt.
Die Reaktion kann gegebenenfalls in Gegenwart einer Base, wie z.B. eines Alkalimetallhydroxids, beispielsweise Natrium- oder Kaliumhydroxid, ausgeführt werden.
Das als Ausgangsmaterial verwendete a-(p-Hydroxyphe- nyl)-glycin ist eine bekannte Verbindung.
Wie oben bereits festgestellt, ist p-Hydroxybenzylcyanid ein wertvolles Zwischenprodukt. Insbesondere eignet es sich zur Herstellung von p-Hydroxyphenylacetamid durch Hydrolyse, das seinerseits ein wertvolles Zwischenprodukt für die Herstellung des adrenergischen ,e;-Rezeptorenblockers p-(2 -Hydroxy-3-isopropylaminopropoxy)-phenylacetamid ist.
Die Erfindung wird durch die folgenden Beispiele näher erläutert.
Beispiel 1
3,0 g Natriumcyanid werden unter Rühren bei 80"C zu einer Lösung von 10,0 g p-Hydroxyphenylglycin und 2,4 g Natriumhydroxid in 20 ml N,N-Dimethylformamid zugegeben, und das Gemisch wird dann 1 Stunde lang unter Rühren auf 1300C erhitzt und dann auf 20"C abgekühlt. 25 ml Wasser werden zugegeben, und das Gemisch wird mit konzentrierter wässriger Salzsäure auf pH 5 angesäuert und 4mal mit jeweils 40 ml Diäthyläther extrahiert. Die vereinigten Extrakte werden getrocknet und zur Trockene eingedampft, und der Rückstand wird bei einem Druck von 0,05 mm Hg destilliert. Auf diese Weise wird als Destillat, das sich beim Abkühlen verfestigt, p-Hydroxybenzylcyanid, Fp 68 - 69"C (6,37 g; 80% Ausbeute), erhalten.
Beispiel 2
Das in Beispiel 1 beschriebene Verfahren wird wiederholt, wobei aber Isobutylmethylketon anstelle von Diäthyl äther zur Extraktion des Reaktionsgemischs verwendet wird.
Die vereinigten Extrakte werden getrocknet, aber nicht zur Trockene eingedampft. Auf diese Weise wird eine Lösung erhalten, die p-Hydroxybenzylcyanid enthält, das sich für die Hydrolyse zum p-Hydroxyphenylacetamid ohne weitere Isolierung oder Reinigung eignet.

PATENT CLAIMS
1. A process for the preparation of p-hydroxybenzyl cyanide, characterized in that reacting a- (p-hydroxyphenyl) glycine with cyanide ions.
2. The method according to claim 1, characterized in that the cyanide ions are used in the form of an alkali metal cyanide, preferably sodium or potassium cyanide.
3. The method according to claim 1 or 2, characterized in that it is carried out in a high-boiling, dipolar, aprotic solvent, preferably N, N-dimethylformamide, 2-pyrrolidone, N-methyl-2-pyrrolidone or dimethyl sulfoxide.

The invention relates to a new process for the preparation of the compound p-hydroxybenzyl cyanide, which is a valuable chemical intermediate.
According to the invention, p-hydroxybenzyl cyanide is prepared by reacting z- (p-hydroxyphenyl) glycine [a-amino-x- (p-hydroxyphenyl) acetic acid with cyanide ions.
The cyanide ions are expediently used in the form of an alkali metal cyanide, for example sodium or potassium cyanide.
The reaction is conveniently carried out in a relatively high-boiling, dipolar, aprotic solvent, such as e.g. N, N-dimethylformamide, 2-pyrrolidone, N-methyl-2-pyrrolidone or dimethyl sulfoxide. Other relatively high boiling polar solvents such as e.g.
n-Butanol, 3-methylbutanol, acetamide or 2-ethoxyethanol are also satisfactory solvents, and some other such solvents such as e.g. Water, ethylene glycol, formamide and urea are also suitable, although less satisfactory.
The reaction is expediently carried out at a temperature between 100 and 250 ° C., preferably at a temperature between 120 and 1900 ° C.
The reaction may optionally be carried out in the presence of a base, e.g. an alkali metal hydroxide, for example sodium or potassium hydroxide.
The a- (p-hydroxyphenyl) glycine used as the starting material is a known compound.
As stated above, p-hydroxybenzyl cyanide is a valuable intermediate. It is particularly suitable for the production of p-hydroxyphenylacetamide by hydrolysis, which in turn is a valuable intermediate for the production of the adrenergic, e; -receptor blocker p- (2-hydroxy-3-isopropylaminopropoxy) phenylacetamide.
The invention is illustrated by the following examples.
example 1
3.0 g of sodium cyanide are added with stirring at 80 ° C. to a solution of 10.0 g of p-hydroxyphenylglycine and 2.4 g of sodium hydroxide in 20 ml of N, N-dimethylformamide, and the mixture is then stirred for 1 hour 1300C heated and then cooled to 20 "C. 25 ml of water are added and the mixture is acidified to pH 5 with concentrated aqueous hydrochloric acid and extracted 4 times with 40 ml of diethyl ether each time. Example 2
The procedure described in Example 1 is repeated, but using isobutyl methyl ketone instead of diethyl ether to extract the reaction mixture.
The combined extracts are dried, but not evaporated to dryness. In this way a solution is obtained which contains p-hydroxybenzyl cyanide which is suitable for hydrolysis to p-hydroxyphenylacetamide without further isolation or purification.
