Das erfindungsgemässe Verfahren kann beispielsweise ausgeführt werden, indem man zu einer Lösung oder Suspension einer Verbindung der Formel V in einem unter Mannichreaktionsbedingungen inerten Lösungsmittel z.B. in einem cyclischen Äther wie Dioxan eine Mischung aus einer Formalinlösung und einer Verbindung der Formel VI oder Piperazin in einem unter den Reaktionsbedingungen inerten Lösungsmittel z.B. in einem cyclischen Äther wie Dioxan zusetzt und vorzugsweise bei Raumtemperatur reagieren lässt. Zur Ver
vollständigung der Reaktion kann gegebenenfalls anschliessend noch bei höherer Temperatur z.B. bei 40 bis 60"C weiter reagieren gelassen werden. Aus dem Reaktionsgemisch kann das Reaktionsprodukt nach bekannten Methoden isoliert und gegebenenfalls gereinigt werden.
Die Verbindungen der Formel I können in ihre Säureadditionssalze überführt werden und umgekehrt.

Die Verbindungen der Formel I besitzen bei geringer Toxizität interessante pharmakodynamische Eigenschaften und können daher als Heilmittel verwendet werden. Sie entfalten eine Hemmwirkung gegen Bakterien, wie sie durch Untersuchungen in vitro mit dem Agarplattentest unter Verwendung verschiedener Bakterienstämme und durch Versuche in vivo an der Maus, insbesondere im Inokulationspyelonephritis-Infektionsmodell zeigen lässt. Diese Hemmwirkung wurde ab einer Konzentration von ca. 0,8 bis 50 Fg/ml festgestellt. Daher können die Verbindungen als Hemmstoffe des Bakterienwachstums verwendet werden.
Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustands. Im allgemeinen werden jedoch bei Testtieren befriedigende Resultate mit einer Dosis von ca. 10 bis 300 mg/kg Körpergewicht erhalten.
Diese Dosis kann nötigenfalls in 2 bis 4 Anteile oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 250 bis 3000 mg.
Weiters zeigen die Verbindungen der Formel I eine Wirkung gegen Protozoen, insbesondere gegen Amöben, Trichomonaden und Kokzidien, wie sich durch Untersuchungen in vitro zeigen lässt. Zur Feststellung der Wirksamkeit von Substanzen gegen Amöben und Trichomonaden in vitro wird die minimale letale Konzentration (MLK) in einem Reihenverdünnungstest nach 48stündiger Inkubation bei 37"C ermittelt.
Zur Austestung der trichomonaziden Wirkung werden die Substanzen T.vaginalis-Kulturen, die im CACH-Medium (nach Müller und Mitarb., Angew.Parasit. 11/170, 1970) gehal.
ten werden, zugesetzt. Die amöbizide Wirksamkeit wird an monoxenisch gezüchteten E. histolytica-Amöben (Diamond L.S., J. Parasit. 54/715, 1968) ermittelt. Die Wirksamkeit gegen Trichomonaden konnte in vivo durch Versuche an der Maus, die gegen Amöben durch Versuche an der Ratte und am Hamster gesichert werden. Die Wirksamkeit gegen Kokzidien wurde durch in vivo Versuche an Eimeria tenella infizierten Küken nachgewiesen.
Die Verbindungen der Formel I erwiesen sich in vitro aber einer Konzentration von ca. 0,1 bis 25 Fg/ml amöbizid und in vivo nach peroraler Gabe von 60 bis 300 mg/kg wirksam. Sie können daher zur Behandlung aller Formen der Amöbiasis verwendet werden.
Aufgrund ihrer trichomonaziden Wirkung, die sich sowohl in vitro (MLK 0,1 bis 0,35 Fg/ml) wie auch in vivo durch Ver suche an der Maus in Dosierungen von 3 > < X 20 20 bis 100 mg/kg bestätigen lässt, eignen sich die Substanzen sowohl für die perorale wie auch lokale Anwendung zur chemotherapeutischen Behandlung der Trichomoniasis. In Futterkonzentrationen an 125 ppm führen die Verbindungen der Formel I zur Hemmung des Infektionsverlaufs der Blinddarmkokzidiose.
Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustands. Im allgemeinen werden jedoch bei Testtieren befriedigende Resultate mit einer Dosis von ca. 100 bis 300 mg/kg Körpergewicht erhalten. Diese Dosis kann in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 1 bis 3 g.
Als Heilmittel können die Verbindungen der Formel I allein oder in geeigneten Arzneiformen gemeinsam mit anorganischen oder organischen, pharmakologisch indifferenten Hilfsstoffen verabreicht werden. Beispielsweise werden sie als Bestandteil von Kapseln oder Tabletten eingesetzt, wobei die Konzentration in der zubereiteten Form ca. 50 bis 500 mg Wirkstoff pro Einheit der Dosisform betragen kann.
Chemotherapeutische Wirkungen strukturverwandter Stoffe sind bekannt. So weisen gemäss der irischen Patentanmeldung 31/65 3-(5-nitro-2-furyl)-4H-1,2,4-triazole mit in Stellung 5 eine Alkylgruppe, eine Hemmwirkung gegen bestimmte Bakterien auf, und besitzt gemäss J. Med. Chem. 11, 1287-1291 (1973) das 3-[2-(5-nitro-2-furyl)vinyl]-1,2,4-triazol eine Aktivität gegen Schistosomen.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1 4-(4-Methylpiperazinyi) -methyl-3 -(5-nitro -2-furyl)-1,2,4-triazol
Zu einer Suspension von 2,5 g 5-(Nitro-2-furyl)-4H-1,2,4- -triazol in 50 ml Dioxan wird eine Mischung von 1,39 g N-Methylpiperazin, 0,4 g Formalinlösung (37%) und 20 ml Dioxan unter Rührung zugetropft. Nach ca. 3 Stunden kristallisiert das Endprodukt teilweise aus. Nach 12 Stunden Stehen bei +50 wird abgesaugt und gut mit Dioxan, dann mit Äther gewaschen und anschliessend über P2O5 im Vakuum getrocknet.

The N-tert.B utoxycarbonyl-5-nitro-2-furyl-carboxamide hydrazone required as the starting product can be obtained as follows:
12.3 g of free 5-nitro-2-furylcarbimino ether and 8.86 g of tert. Butyl carbazate are dissolved in a total of 100 ml of methanol, combined and left to stand at room temperature for 15 hours.
Then it is filtered, washed with methanol and dried over PIO.
