Geeignete Ester von Verbindungen der Formel I und IV sind unter anderem Silylester, aliphatische oder cycloaliphatische Ester und Ester, die einen aromatischen oder heterocyclischen Ring enthalten. Geeignete Silylester sind Triniederalkylsilylester, wie Trimethylsilylester, Triäthylsilylester usw. Geeignete aliphatische oder cycloaliphatische Ester sind gesättigte oder ungesättigte niedere oder höhere aliphatische Ester, die verzweigt oder durch einen cycloaliphatischen Ring substituiert sein können, und niedere oder höhere cycloaliphatische Ester, z.B.
Niederalkylester, wie Methylester, Äthylester, Propylester, Isopropylester, 1-Cyclopropyläthylester, Butylester, tert.-Butylester usw., Höheralkylester, wie Octylester, Nonylester, Undecylester usw., Niederalkenylester, wie Vinylester, 1-Propenyl- ester, Allylester, 3-Butenylester usw., Niederalkinylester, wie 2 Propinylester, 3-Butinylester, 4-Pentinylester usw., niedere oder höhere Cycloalkylester, wie Cyclopentylester, Cyclohexylester, Cycloheptylester usw., und niedere oder höhere aliphatische Ester, die ein Stickstoff-, Schwefel- oder Sauerstoffatom enthalten, z.B.

Geeignete Ester von heterocyclischen Hydroxylverbindungen sind z.B. solche mit einem gesättigten oder ungesättigten 3- bis 8-gliedrigen heterocyclischen Ring mit 1-4 Heteroatomen, wie Sauerstoff-, Schwefel- oder Stickstoffatomen und gegebenenfalls anellierten aromatischen Ringen, wie Pyridylester, Piperidinylester, 2-Pyri don- 1 -ylester, Tetrahydropyranylester, Chinolylester, Pyrazolylester usw.
Geeignete Niederalkylester mit einem heterocyclischen Substituenten im Niederalkylrest sind z.B. solche mit einem gesättigten oder ungesättigten 3- bis 8-gliedrigen heterocyclischen Ring mit 1-4 Heteroatomen, wie Sauerstoff, Schwefel und Stickstoff; gegebenenfalls mit anellierten aromatischen Ringen, wie durch Pyridyl, Piperidinyl, 2-Pyridon-1-yl, Tetra hydropyranyl, Chinolyl, Pyrazolyl usw. substituierte Niederalkylester, z.B. Methylester, Äthylester, Propylester usw. Die Silylester, die aliphatischen Ester und die einen aromatischen oder heterocyclischen Ring enthaltenden Ester können 1 bis 10 geeignete Substituenten aufweisen, z.B.

Geeignete Salze der Verbindungen der Formel I und IV sind Salze von Säuren mit Metallen, wie Natrium, Kalium, Magnesium usw., oder organischen Aminen, wie Methylamin, Diäthylamin, Trimethylamin, Triäthylamin, Anilin, Pyridin, Dicyclohexylamin, N,N'Dibenzyläthylendiamin usw.
Unter niederen aliphatischen Gruppen werden solche mit einer Kohlenstoffkette mit 1-6 Kohlenstoffatomen verstanden, während höhere aliphatische Gruppen solche mit einer Kohlenstoffkette mit 7-16 Kohlenstoffatomen sind.
Für das Verfahren geeignete quaternäre Ammoniumsalze sind Tetraalkylammoniumhalogenide, wie Tetramethylammoniumchlorid, Tetraäthylammoniumbromid, Dimethyldiäthylammoniumjodid, Tetrabutylammoniumchlorid, Methyltriisobutylammoniumbromid usw., Dimethyldiphenylammoniumchlorid, Triäthyltolylammoniumbromid, Tetraaralkylammoniumhalogenide, wie Tetrabenzylammoniumchlorid, Tetraphenyläthylammoniumbromid usw., Dialkyldiaralkylammoniumhalogenide, wie Dimethyldibenzylammoniumjodid, Diäthyldibenzylammoniumchlorid usw., und Trialkylaralkylammoniumhalogenide, wie Triäthylbenzylammoniumbromid, Tripropylphenyläthylammoniumjodid usw. Für das Verfahren geeignete Phosphoniumsalze sind z.B.
Tetraalkylphosphoniumhalogenide, wie Tetra methylphosphoniumchlorid, Tetraäthylphosphoniumbromid, Trimethyläthylphosphoniumj odid, Tetrapropylphosphoniumchlorid, Tetralsobutylphosphoniumbromid usw., Tetraphenylphosphoniumjodid und Tetraaralkylphosphoniumhalogenide, wie Tetrabenzylphosphoniumchlorid, Tetraphenyläthylphosphoniumbromid usw.
Wenn das Verfahren in einem wässrigen organischen Lösungsmittel ausgeführt wird, kann man als organisches Lösungsmittel beispielsweise Methylenchlorid, Chloroform, Tetrachlormethan, Tetrachloräthan usw., die mit Wasser nicht mischbar sind, verwenden.
Das Verfahren wird vorzugsweise unter schwach sauren oder schwach basischen Bedingungen ausgeführt; zu diesem Zweck wird vorzugsweise eine nucIeophile Verbindung in Form eines Metallsalzes, z.B. des Natriumsalzes, Kaliumsalzes usw., verwendet, oder wenn die Verbindung der Formel IV in freier Form verwendet wird, wird das Verfahren B vorteilhaft in Gegenwart einer anorganischen Base, z.B. eines Alkalimetallhydroxydes, wie Natriumhydroxyd, Kaliumhydroxyd usw., eines Erdalkalimetallhydroxydes, wie Calciumhydroxyd, Magnesiumhydroxyd usw. eines Alkalimetallbicarbonates, eines Alkalimetallcarbonates oder eines Erdalkalimetallcarbonates oder dergl., oder einer organischen Base, z.B. eines Trialkylamins, wie Trimethylamin, Triäthylamin usw., oder einer tertiären organischen Base, wie Dimethylanilin usw., ausgeführt.
Hinsichtlich der Reaktionstemperatur bestehen keine speziellen Beschränkungen; sie kann je nach der verwendeten Verbindung der Formel IV und der verwendeten nucleophilen Verbindung in der Praxis in geeigneter Weise gewählt werden.
Bei dem Verfahren kann der 3-Cephemkern, falls die Verbindung der Formel IV einen solchen enthält, bei der Umsetzung oder Nachbehandlung in den 2-Cephemkern umgelagert werden.
Bisher wurden Verbindungen der Formel I mit dem 3 Cephemkern hergestellt, indem man eine entsprechende Ausgangsverbindung der Formel IV, worin X Acyloxy bedeutet, mit einer stark nucleophilen Verbindung umsetzte (japanische Patentveröffentlichung No. 17936/1964).
Das Verfahren hat gegenüber diesem bekannten Verfahren z.B. den Vorteil, dass die Verbindungen der Formel I in höherer Ausbeute und Reinheit und unter Bildung von weniger Nebenprodukten erhalten werden, dass die Reaktionsdauer kürzer ist und dass kleinere Mengen der Reaktionsteilnehmer verwendet werden können, wobei die Reaktion ausserdem in heterogener Phase ausgeführt werden kann.
Das Verfahren hat beispielsweise den Vorteil, dass keine Nebenreaktionen, wie Lactonbildung oder Lactamspaltung, eintreten.
Falls A eine Gruppierung der Formel

darstellt, kann diese Gruppierung bei dem Verfahren erforderli chenfalls zu einer Gruppierung -S-reduziert werden. Falls die Carboxylgruppe der erhaltenen Verbindungen der Formel I in Form eines Salzes oder eines Esters geschützt ist, kann das Salz oder die Estergruppierung durch eine Eliminierungsreaktion gewünschtenfalls in die Carboxylgruppe übergeführt werden.
Die Reduktion der Gruppierung der Formel:

zu einer Gruppierung -5- kann unter Verwendung von Stan nochlorid oder einem Metallthiosulfat, wie Natriumthiosulfat, Kaliumthiosulfat usw., unter Verwendung einer solchen Verbindung zusammen mit einem Säurechlorid, wie Acetylchlorid usw., unter Verwendung von Phosphortrichlorid, Phosphorpentachlorid, Siliciumtrichlorid usw. ausgeführt werden; es kann aber auch jedes beliebige andere für diesen Zweck geeignete Reduktionsverfahren angewandt werden.
Die Reduktion erfolgt gewöhnlich in Gegenwart eines Lösungsmittels, wie Dimethylformamid, Acetonitril, Acetessigester, Tetrahydrofuran, Chloroform, Dichlormethan, Dioxan oder eines beliebigen anderen Lösungsmittels, das die Reaktion nicht in nachteiliger Weise beeinflusst.
Hinsichtlich der Reaktionstemperatur bestehen keine Beschränkungen; diese kann je nach der Ausgangsverbindung und dem Reduktionsverfahren, das in der Praxis verwendet werden soll, in geeigneter Weise gewählt werden.
Die Überführung von in den Verbindungen der Formel I als Salz oder Ester vorliegenden Carboxylgruppen in freie Carboxylgruppen kann nach für diesen Zweck üblichen Verfahren, beispielsweise durch Reduktion, Hydrolyse usw., ausgeführt werden.
Diese Reaktion kann durch das folgende Schema wiedergegeben werden:

Wenn die geschützte Carboxylgruppe ein aktiver Ester ist, kann sie durch Hydrolyse, gewöhnlich unter milden Hydrolysenbedingungen, wie durch Inberührungbringen mit Wasser, eliminiert werden. 2-Jodäthylester, 2,2,2-Trichloräthylester, Benzylester usw. können z.B. durch Reduktion eliminiert werden. Die Eliminierung unter Verwendung einer Säure eignen sich für geschützte Gruppen, wie p-Methoxybenzylester, tert. - Butylester, tert.-Pentylester, Tritylester, Diphenylmethylester, Bis-(methoxyphenyl)-methylester, 3 ,4-Dimethoxybenzylester, 1-Cyclopropyläthylester und dergleichen. Die Eliminierung mit einem wasserfreien basischen Katalysator eignet sich z.B. für geschützte Gruppen, wie Äthinylester, 4-Hydroxy-3,5-di-(tert.
butyl)-benzylester und dergleichen.
Für die Freisetzung der Carboxylgruppen geeignete Reduktionsverfahren sind z.B. die Reduktion unter Verwendung eines Metalles, wie Zinn, Zink, Zinkamalgam usw., oder eines Chromsalzes, wie Chromdichlorid, Chromacetat usw., und einei organischen oder anorganischen Säure, wie Essigsäure, Propionsäure, Salzsäure usw., sowie die Reduktion in Gegenwart eines Metallkatalysators. Metallkatalysatoren für die katalytische Reduktion sind z.B. Platinkatalysatoren, wie Platindraht, Platinschwamm, Platinschwarz, kolloidales Platin usw., Palladiumkatalysatoren, wie Palladiumschwamm, Palladiumschwarz, Palladiumoxyd, Palladium auf Bariumsulfat, Palladium auf Bariumcarbonat, Palladium auf Kohle, Palladium auf Kieselgel, kolloidales Palladium usw., Nickelkatalysatoren, wie reduziertes Nickel, Nickeloxyd, Raneynickel, Urushibaranickel usw.
Für die Freisetzung der Carboxylgruppe geeignete Säuren sind z.B. Ameisensäure, Trihalogenessigsäuren, wie Trichloressigsäure, Trifluoressigsäure usw., Salzsäure, Flusssäure, p Toluolsulfonsäure, Trifluormethansulfonsäure, Gemische aus Salzsäure und Essigsäure usw.
Für die Freisetzung der Carboxylgruppe geeignete wasserfreie basische Katalysatoren sind z.B. Natriumthiophenophenolat, (CH3)2LiCu usw.
Wenn die Schutzgruppe durch Behandlung mit Wasser oder einer flüssigen Säure eliminiert wird, kann diese Reaktion mit oder ohne Lösungsmittel ausgeführt werden. Für diesen Zweck können beliebige Lösungsmittel, die die Reaktion nicht in nachteiliger Weise beeinflussen, verwendet werden, z.B. Dimethylformamid, Methylenchlorid, Chloroform, Tetrahydrofuran, Aceton und dergleichen.
Es gibt keine speziellen Beschränkungen hinsichtlich der Reaktionstemperatur; diese kann je nach der verwendeten Ausgangsverbindung und dem angewandten Eliminierungsverfah- ren in geeigneter Weise gewählt werden.
Die folgenden Beispiele erläutern die Erfindung. In den NMR-Spektren (magnetischen Kernresonanzspektren) bedeutet s Singulett, d Dublett, q Quartett und m Multiplett.
Beispiele (A) Ein Gemisch aus 29,15 g 7-[5-(Benzamido)-adipin amido]-cephalosporansäure, 7,45 g 5-Methyl-1,3,4-thiadiazol2-thiol und 14,2 g Natriumbicarbonat wurde vorsichtig mit 291 ml Wasser und dann mit 1,45 g Triäthylbenzylammoniumbromid versetzt, worauf das Gemisch 6 Stunden lang bei 60 C gerührt wurde. Nach dem Abkühlen wurde das Reaktionsgemisch mit Äthylacetat versetzt und mit 6-normaler Salzsäure auf pH = 5,0 gebracht. Die wässrige Schicht wurde abgetrennt und unter Eiskühlung mit 6-normaler Salzsäure auf pH = 1,0 - 1,5 gebracht, wobei sich ein öliges Material ausschied.
Nach Entfernen des grössten Teils des Wassers durch Dekantieren wurde das resultierende ölige Material in Aceton gelöst und das Lösungsmittel abdestilliert, wobei man 25,4 g 7-[5-(Benz amido)-adipinamido]-3-(5-methyl-1,3,4-thiadiazol-2-yl)-thio- methyl-3-cephem-4-carbonsäure erhielt.
(B) Ein Gemisch aus 242 g 7-[5-(Benzamido)-adipin- amido]cephalosporansäure, 2,42 Liter Wasser, 117 g Natrium bicarbonat, 61,5 g 5-Methyl-1,3,4-thiadiazol-2-thiol und 12 g Triäthylbenzylammoniumbromid wurde 6 Stunden lang bei 60 + 1" C gerührt. Das Reaktionsgemisch wurde auf pH = 5,0 gebracht, mit Äthylacetat gewaschen und die wässrige Schicht zu einem Gemisch aus 500 ml 6-normaler Salzsäure und 500 g Eis gegeben. Der Niederschlag wurde abfiltriert und unter vermindertem Druck über Phosphorpentoxyd getrocknet, wobei man 158 g 7-[5-(Benzamido)-adipinamido]-3-(5-methyl 1,3 ,4-thiadiazol-2-yl)-thiomethyl-3 -cephem-4-carbonsäure erhielt.
Die folgenden Verbindungen wurden nach dem Verfahren von Beispiel A und B hergestellt.

(G) Eine Lösung von 0,1 g 7-(2-Phenylacetamido)-3-chlor methyl-2-cephem-4-carbonsäure-tert.-butylester in 20 ml Chloroform wurde mit 20 ml eines Phosphatpuffers vom pH = 7,0, 0,1 g Natriumbicarbonat und 0,1 g Triäthylbenzylammoniumbromid versetzt, worauf das Gemisch 3 Stunden lang bei 70 C gerührt wurde. Nach dem Abkühlen wurde das Reaktionsgemisch mit Chloroform versetzt und das Gemisch mit Chloroform extrahiert. Der Extrakt wurde getrocknet und das Lösungsmittel unter vermindertem Druck abdestilliert, wobei man 0,07 g 7-(2-Phenylacetamido)-3-hydroxymethyl-2cephem-4-carbonsäure-tert.-butylester erhielt.
(H) 1,0 g 7-[2-(1H-Tetrazol-1-yl)-acetamido]-3-(5-methyl- 1,3,4-thiadiazol-2-yl)-thiomethyl-3-cephem-4-carbonsäure-1- oxyd, das ähnlich wie in Beispiel (A) bis (G) beschrieben erhalten worden war, wurde in 20 ml Dimethylformamid gelöst.
Diese Lösung wurde mit 400 mg Stannochiorid und dann unter Rühren und Kühlen auf 0 - 10 C tropfenweise mit 1 ml Acetylchlorid versetzt, worauf das Gemisch 1 Stunde lang gerührt wurde. Nachdem die Reaktion beendet war, wurde das Reaktionsgemisch in Wasser gegossen und mit Äthylacetat extrahiert. Der Extrakt wurde mit Wasser gewaschen, über Magnesiumsulfat getrocknet und das Lösungsmittel abdestilliert. Die zurückbleibenden Kristalle wurden aus einem Gemisch aus Aceton und Wasser umkristallisiert,wobei man 0,45 g farblose Nadeln von 7-[2-(1H-Tetrazol-1-yl)-acet- amido]-3-(5-methyl- 1,3 ,4-thiadiazol-2-yl)-thiomethyl-3cephem-4-carbonsäure vom Smp. 198 - 200 C unter Zersetzung erhielt.
UV-Spektrum (Phosphatpuffer, pH 6,4) Xma 271 mR, E = 286 (I) Ein Gemisch aus 1,3 g 7-[2-(1H-Tetrazol-1-yl)-acet amido]-3-(5-methyl-1,3,4-thiadiazol-2-yl)-thiomethyl-3- cephem-4-carbonsäurediphenylmethylester, der ähnlich wie in Beispiel (A) bis (G) beschrieben erhalten worden war, 1,3 ml Trifluoressigsäure und 1,3 ml Anisol wurde 20 Minuten lang bei Umgebungstemperatur gerührt. Das Reaktionsgemisch wurde unter vermindertem Druck auf etwa das halbe Volumen eingeengt und der Rückstand mit Wasser versetzt, worauf das Gemisch mit Äthylacetat extrahiert wurde. Der Extrakt wurde mit einer gesättigten wässrigen Natriumbicarbonatlösung rückextrahiert. Der Extrakt wurde mit 10 %-iger Salzsäure neutralisiert und wieder mit Äthylacetat rückextrahiert.
Der Extrakt wurde mit Wasser gewaschen, getrocknet und das Lösungsmittel unter vermindertem Druck abdestilliert, wobei man 0,8 g Kristalle von 7-[2-(1H-Tetrazol-1-yl)-acetamido]-3-(5-methyl 1,3,4-thiadiazol-2-yl)-thiomethyl-3-cephem-4-carbonsäure erhielt.

The remaining crystals were recrystallized from a mixture of acetone and water, whereby 0.45 g of colorless needles of 7- [2- (1H-tetrazol-1-yl) -acetamido] -3- (5-methyl-1 , 3, 4-thiadiazol-2-yl) thiomethyl-3cephem-4-carboxylic acid of melting point 198-200 ° C. with decomposition.
UV spectrum (phosphate buffer, pH 6.4) Xma 271 mR, E = 286 (I) A mixture of 1.3 g of 7- [2- (1H-tetrazol-1-yl) -acetamido] -3- ( 5-methyl-1,3,4-thiadiazol-2-yl) -thiomethyl-3-cephem-4-carboxylic acid diphenylmethyl ester, which had been obtained in a manner similar to that described in Examples (A) to (G), 1.3 ml of trifluoroacetic acid and 1.3 ml of anisole was stirred for 20 minutes at ambient temperature. The reaction mixture was concentrated to about half volume under reduced pressure, water was added to the residue, and the mixture was extracted with ethyl acetate. The extract was back extracted with a saturated aqueous sodium bicarbonate solution. The extract was neutralized with 10% hydrochloric acid and back-extracted again with ethyl acetate.
The extract was washed with water, dried and the solvent was distilled off under reduced pressure, whereby 0.8 g of crystals of 7- [2- (1H-tetrazol-1-yl) acetamido] -3- (5-methyl 1, 3,4-thiadiazol-2-yl) thiomethyl-3-cephem-4-carboxylic acid.
