PATENTANSPRÜCHE
1. Verfahren zur Gewinnung von Einzellerprotein auf Basis von Methanol, dadurch gekennzeichnet, dass in einem begasten
Reaktor eine wachsende Submerskultur mit dem obligat metha nol-verwertenden Bakterium Methylomonas sp. DSM 580 unter aeroben Bedingungen erzeugt wird, wobei sich in der
Submerskultur Methanol als einzige Kohlenstoff- und Energie quelle sowie anorganische Nährstoffe befinden und ausserdem in dieses System Luft oder sauerstoffangereicherte Luft bei einer Reaktionstemperatur von 20-45 C zugeführt wird, und danach aus dem entstandenen Dreiphasensystem die Zellmasse abgetrennt und getrocknet wird, wobei eine Zellmasse erzeugt wird mit einem Rohproteingehalt von mindestens 60-70 Gew.- %, einem Nucleinsäuregehalt von 2-17 Gew.- %, einem
Aschegehalt von 3-6 Gew.- %,
einem Fettgehalt von 3-8 Gew.-%.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die Submerskultur mechanisch rührt und ihr gegebe nenfalls Wuchsstoffe zusetzt.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekenn zeichnet, dass Methylomonas sp. DSM 580 in chargenweiser
Prozessführung bei einer Anfangskonzentration von
0.5-5.0 Vol.-% Methanol, insbesondere bei 2-3 Vol.-% gewonnen wird.
4. Verfahren nach Anspruch 1 oder 2, dadurch gekenn zeichnet, dass Methylomonas sp. DSM 580 in chargenweiser
Prozessführung bei gesteuerter Aufrechterhaltung einer kon stanten Methanolkonzentration von 0.01-2.0 Vol.-%, bis zu einem Gesamtumsatz von 25 Vol.-% Methanol gezüchtet wird.
5. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass Methylomonas sp. DSM 580 in kontinuierlicher
Prozessführung mit einer Durchflussrate von 0.1-0.5 Vol./ Vol./Std. unter chemostatischen oder turbidostatischen Bedingungen gewonnen wird.
6. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass während des Wachstums durch Zusatz von Alkalien bzw. Säuren ein praktisch konstanter pH-Wert von 4.5-9.0 eingestellt wird.
7. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass während der Züchtung in den Reaktor Luft oder sauerstoffangereicherte Luft mit einer Belüftungsrate von 0.5-1.5 Vol./Vol./Min. insbesondere von 0.1-0.2 Vol./ Vol./Min. zugeführt wird und das eingeleitete Gasgemisch einen Sauerstoffgehalt von 20-60 Vol.-% aufweist.
8. Verfahren nach einem der Ansprüche 1 bis 7, dadurch gekennzeichnet, dass die wässrige Nährlösung als Stickstoffquelle Ammonium- und/oder Nitrat-Salze und/oder Harnstoff, sowie die für das Wachstum notwendigen Kationen Natrium, Kalium, Magnesium, Calcium, Eisen, Zink, Mangan und die Anionen Phosphat, Sulfat, Nitrat, Chlorid und Wuchsstoffe enthält.
9. Verfahren nach einem der Ansprüche 1-8, dadurch gekennzeichnet, dass man die von der Zellmasse befreite flüssige Phase mindestens teilweise in den Prozess zurückführt.
Der zunehmende Bedarf an Protein für die menschliche und tierische Ernährung machen es erforderlich, neue Verfahren zur Gewinnung hochwertigen Proteins zu entwickeln. So wurde in den letzten Jahren die Produktion von Protein aus Mikroorganismen untersucht, die als Kohlenstoffquelle gasförmige Kohlenwasserstoffe wie Methan oder flüssige Kohlenwasserstoffe aus Erdölfraktionen verwerten können. Die gasförmigen Kohlenwasserstoffe haben jedoch die Nachteile, dass sie eine geringe Wasserlöslichkeit aufweisen und bei den aeroben Züchtungsbedingungen explosive Gasgemische entstehen. Die flüssigen Kohlenwasserstoffe haben die Nachteile, dass sie ebenfalls nur eine minimale Wasserlöslichkeit besitzen, was zur Verteilung der zwei flüssigen Phasen einen erhöhten Energiebedarf bedingt.
Ausserdem muss bei der auf flüssigen Kohlenwasserstoffen gewachsenen Zellmasse mindestens eine Extrak tion mit organischen Lösungsmitteln durchgeführt werden.
Als besonders günstige Kohlenstoff- und Energiequelle für die Züchtung von Mikroorganismen in grossem Massstabe wird seit einigen Jahren Methanol diskutiert. Dieses Substrat hat die
Vorteile, dass es in chemisch definierter reiner Form und preis würdig aus Synthesegas zugänglich ist, welches aus verschiede nen Rohstoffen wie Erdgas, Fraktionen des Erdöls, Steinkohle oder Braunkohle erzeugt werden kann.
Aus der DT-OS 2 040 358 ist ein Verfahren zur Herstellung von Einzellerprotein bekannt, bei dem aus der Oxidation von flüssigen Alkanen entstandenen Alkoholen, Aldehyden, Ketonen, Carbonsäuren und deren Derivate als Kohlenstoffquellen verwendet werden.
Aus der DT-AS 2 152 039 ist die Gewinnung von Bakterien-Zellmasse in einem methanolhaltigen Nährmedium durch die Verwendung der Bakterien-Stämme: Protaminobacter rubervar. machidanus ATCC 21 611, ATCC 21 612, ATCC 21 613 oder ATCC 21 614 bekannt.
Aus der DT-OS 2 311 006 ist ein Verfahren zur Proteinherstellung bekannt, bei welchem der aerobe Mikroorganismus Methylomonas methanolica NRRL-B-5458 auf Methanol als einziger Kohlenstoffquelle gezüchtet wird.
Aus der DT-OS 2 059 277 ist ein mikrobiologisches Verfahren zur Herstellung von Protein bekannt, welches in geeigneter Nährlösung mit Methanol als einziger Kohlenstoffquelle unter aeroben Bedingungen die folgenden Bakterien-Stämme verwendet: Pseudomonas methanica, Pseudomonas sp. ATCC 21 438, Pseudomonas sp. ATCC 21 439, Pseudomonas sp.
PRL-W 4, Corynebacterium sp. ATCC 21 232, Corynebacterium sp. ATCC 21 235 und Corynebacterium sp. ATCC 21 236.
Aus der DT-OS 2 407 740 ist ein Verfahren zur Züchtung von Mikroorganismen bekannt, bei welchem eine Mischkultur, bestehend aus einem fakultativ Methanol-verwertenden Bakterium, und mehreren nicht Methanol-verwertenden Bakterien, angewandt wird. Das Methanol-verwertende Bakterium ist nicht beweglich und kann neben Methanol auch andere Kohlenstoffsubstrate wie z. B. Glucose oder Glycerin als Kohlenstoffquelle verwerten.
Aus der DT-OS 2 418 385 ist ein Verfahren zur Herstellung eines proteinreichen Produktes bekannt, bei welchen ein nichtrosapigmentierter Stamm, abgeleitet von dem Mikroorganismus Pseudomonas extorquens (NCIB Nr. 9399), eingesetzt wird.
Für die Züchtung von Protein aus Methanol wird nach diesem Stand der Technik kein obligat Methanol-verwertendes Bakterium, sondern es werden fakultativ methylotrophe Mikroorganismen verwendet. Dieser Stand der Technik wird durch das Verfahren der Erfindung überwunden, welches ein obligat methylotrophes Bakterium zur Züchtung von Einzeller-Protein verwertet.
Aus einer Bodenprobe vom Rheinufer bei Ludwigshafen wurde ein obligat Methanol verwertendes Bakterium isoliert.
Es wurden 1 g der Bodenprobe in 100 ml wässrigem anorganischem Nährmedium = KH2PO4 3,75 g; Na2HPO4 2,50 g; (NH4)2 SO4 4,00 g; MgSO4 7H2O 0,5 g; Ca(NO3)2 4H2O 0,025 g; FeSO4 7H2O 0,005 g; ZnSO4 H2O 0,005 G in 1000 ml dest. H2O pH=7,0 mit 1% (V/V) Methanol als einzige Kohlenstoff- und Energiequelle suspendiert und bei 30 C in einem 500 ml Erlenmeyer-Kolben auf der Schüttelmaschine (100 U/min) inkubiert. Nach 5 Tagen Inkubation wurden davon je 0,1 ml auf Petrischalen ausgestrichen, die dasselbe Medium mit 2 % Agar zur Verfestigung enthielten.
Von diesen Platten wurden nach weiteren 3 Tagen Inkubation bei 30" C Einzelkolonien abgeimpft und durch Verdünnungsausstriche
auf Platten mit demselben Medium wurde nach 5 Flüssig-Passagen die Reinkultur von dem Bakterienstamm erhalten. Der Bakterienstamm wurde als Methylomonas sp. bezeichnet und erhielt die Nr. DSM 580.
Es wurde nun ein Verfahren zur Gewinnung von Einzellerprotein auf Basis von Methanol gefunden, welches dadurch gekennzeichnet ist, dass in einem begasten Reaktor mit oder ohne einer mechanischen Rührung eine wachsende Submerskultur mit dem obligat Methanol-verwertenden Bakterium Methylomonas sp. DSM 580 unter aeroben Bedingungen erzeugt wird, in der sich Methanol als einzigste Kohlenstoffund Energiequelle, anorganische Nährstoffe und gegebenenfalls Wuchsstoffe befinden und ausserdem in dieses System Luft oder sauerstoffangereicherte Luft bei einer Reaktionstempera tur von 20-45 C zugeführt wird, danach aus dem entstandenen Dreiphasensystem die Zellmasse abgetrennt und getrocknet wird,
wobei eine Zellmasse erzeugt wird mit einem Rohproteingehalt von mindestens 60-70 Gew.-%, einem Nucleinsäuregehalt von 2-17 Gew.-% und die von der Zellmasse befreite flüssige Phase gegebenenfalls ganz oder teilweise in dem Prozess recyclisiert wird.
Das Verfahren der Erfindung zur Gewinnung von Einzellerprotein erzeugt das obligat Methanol-verwertende Bakterium Methylomonas sp. DSM 580 mit folgenden charakteristischen Eigenschaften: 1. Zellmorphologie:

Das Bakterium Methylomonas sp. wurde bei der Deutschen Sammlung von Mikroorganismen, Griesebachstrasse 8, Göttingen, am 29.8.1974 mit der Nummer DSM 580 hinterlegt.
Methylomonas sp. DSM 580 kann nur auf Methanol als Kohlenstoff- und Energiequelle wachsen, nicht dagegen auf Methan, Methylamin, Äthanol oder Glucose, wie aus Tabelle ersichtlich ist.
Wachstum von Methylomonas sp. Ferner wurde gefunden, dass Methylomonas sp. DSM 580 bei chargenweiser Prozessführung bei einer Anfangskonzentration von 0.5-5.0 Vol.-% Methanol, insbesondere bei 2-3 Vol-% gewonnen wird und dass Methylomonas sp.
DSM 580 bei chargenweiser Prozessführung bei gesteuerter Aufrechterhaltung einer konstanten Methanolkonzentration von 0.01-2.0 Vol.-% bis zu einem Gesamtumsatz von 25 Vol.- % Methanol gezüchtet wird.
Weiter wurde gefunden, dass Methylomonas sp. DSM 580 bei kontinuierlicher Prozessführung mit einer Durchflussrate von 0.1-0.5 Vol./Vol./Std. unter chemostatischen oder turbidostatischen Bedingungen gewonnen wird.
Ausserdem wurde gefunden, dass während des Wachstums durch Zusatz von Alkalien bzw. Säuren ein praktisch konstanter pH-Wert von 4.5-9.0, vorzugsweise von 6.5-7.5, eingestellt wird.
Ferner wurde gefunden, dass die Züchtung von Methylomonas sp. DSM 580 bei einer Temperatur zwischen 20 und 45" C, vorzugsweise bei 33 bis 36 C, durchgeführt wird.
Weiter wurde gefunden, dass während der Züchtung in den Reaktor Luft oder sauerstoffangereicherte Luft mit einer Belüftungsrate von 0.5-1.5 Vol./Vol./Min. zugeführt wird und das eingeleitete Gasgemisch einen Sauerstoffgehalt von 20-60 Vol.-% aufweist.
Ausserdem wurde gefunden, dass während der Züchtung in den Raktor Luft oder sauerstoffangereicherte Luft mit einer Belüftungsrate von 0.1-0.2 Vol./Vol./Min. zugeführt wird und das eingeleitete Gasgemisch einen Sauerstoffgehalt von 20-60 Vol.-% aufweist.
Ferner wurde gefunden, dass die wässrige Nährlösung als Stickstoffquelle Ammonium- und/oder Nitrat-Salze und/oder Harnstoff, sowie die für das Wachstum notwendigen Kationen Natrium, Kalium, Magnesium, Calcium, Eisen, Zink, Mangan und die Anionen Phosphat, Sulfat, Nitrat, Chlorid und Wuchsstoffe enthält.
Das Verfahren der Erfindung wird durch folgende Beispiele näher erläutert:
Beispiel 1
Ein 80 1 Bioreaktor, gefüllt mit 50 1 Nährlösung (Zusammensetzung: 200 g (NH4)2 SO4, 150 g KH2PO4, 125 g Na2HPO4, 25 gMgS04 7H,O, 1.25 gCa(NO3)2 4H2O, 0.25 g FeSO4 7H2O, 0.25 g ZnSO4 H2O, 0.25 g KC1 in 50 1 Wasser) wird 10 Min. bei 121" C sterilisiert, auf 35" C abgekühlt, aseptisch mit 1000 ml Methanol versetzt, mit 500 ml Inoculum von Methylomonas sp. DSM 580 beimpft, 28 Stdn. bei 35" C mit einem Turborührer (300 Upm) suspendiert und bei einer Belüftungsrate von 0.7 Vol./Vol./Min. gezüchtet.
Während der Züchtung wird in der Submerskultur durch automatische Zugabe von 6 Vol.-% Ammoniaklösung ein konstanter pH-Wert von 7.0 aufrechterhalten. Nach 22 Stdn. wird der Reaktor auf 15 " C gekühlt, der pH-Wert durch Zusatz von Schwefelsäure auf pH 3.0 eingestellt und die ausgefällte Zellmasse abfiltriert, mit Wasser gewaschen und getrocknet (324 g Zelltrockenmasse). Die Zellzusammensetzung beträgt:
71% Rohprotein und 9% Nukleinsäuren, 4% Asche und 7% Fett, bezogen auf die Zelltrockenmasse.
Beispiel 2
Ein 340 1 Bioreaktor, ausgerüstet mit einem Intensor , Hersteller: Biologische Verfahrenstechnik AG, Basel, wird mit 200 1 Nährsalzlösung (Zusammensetzung: 500 g (NH4)2 S04, 500 g NH4NO3, 600g KH2PO4, 500g Na2HPO4, 140g MgSO4 7H20, 15 g Ca(N03)2 4H20, 6 g FeSO4- 7H2O,2 g KCI in 2001 Leitungswasser) beschickt, der pH-Wert auf 6.8 eingestellt, 15 Min. bei 121" C sterilisiert, auf 33 " C abgekühlt, aseptisch mit 2000 ml Methanolversetzt, mit 4000ml einer 18 Stdn.-Vorkultur vonMethylomonas sp.
DSM580 beimpft und bei 33 C, einer Belüftungsrate von 0.5 Vol./Vol./Min. und einerUmdrehungszahl von 1800 Upm gezüchtet. Während des Wachstums wird in der Submerskultur automatisch die Methanolkonzentration auf 0.5 Vol. % konstant gehalten, indem man die der Flüssigkeitskonzentration entsprechende Dampfkonzentration an Methanol im Abgas mittels eines Flammenionisationsdetektors kontinuierlich erfasst und über einen Grenzwert- geber die notwendige Methanolzugabe steuert, ausserdem wird automatisch mit einer pH-Regulierstation durch Zusatz einer 12 Vol.-%igen Ammoniaklösung ein pH-Wert von 6.8 aufrechterhalten.
Nachdem nach 25 Stdn. bereits eine Zellkonzentration von 14 g Zelltrockenmasse pro Liter erreicht ist, wird unter Konstanthaltung der Belüftungsrate mit sauerstoffangereicherter Luft mit einem Sauerstoffgehalt von 40 Vol.-% begast. Der Prozess wird nach 60 Stdn. auf 15" C abgekühlt, in einer Durchlaufzentrifuge bei 10 000 g die entstandene Zellmasse abgetrennt und getrocknet. Unter diesen Prozessbedingungen beträgt die Zellausbeute 0.44 g Zelltrockenmasse/g Methanol, mit einem Gehalt an 76% Rohprotein, 6,5% Nukleinsäuren, 5% Asche und 4,5 % Fett, bezogen auf die Zelltrockenmasse.
Beispiel 3
Ein 80 1 Bioreaktor, ausgerüstet mit einem Intensor wie in Beispiel 2, wird mit 50 l Nährsalzlösung mit einer Zusammensetzung wie in Beispiel 1 beschrieben, mit 1000 ml einer Kultur von Methylomonas sp. DSM 580 beimpft, die zuvor 15 Stdn. bei 35" C kultiviert wurde, bei 35" C, einer Belüftungsrate von 0.5 Vol./Vol./Min. und einer Umdrehungszahl von 1200 Upm bei einem konstanten pH-Wert von 7.0 unter statischen Bedingungen gezüchtet. Die kontinuierliche Kultur wird nach 18 Stdn. gestartet bei einer Durchflussrate von 0.05, nach weiteren 48 Stdn. wird die Durchflussrate auf 0.1 erhöht und dann innerhalb von 120 Stdn. stufenweise auf 0.35 gesteigert.
Unter diesen Bedingungen wird der Gleichgewichtszustand aufrechterhalten, bei Zellausbeutekoeffizienten von 0.46 (g Zelltrockenmasse/g Methanol) und einer Produktivität von 10.8 g/l/h. Das anfallende Kulturfiltrat wird nach Erreichung des Gleichgewichtszustandes in einer Menge von 50 Vol.-% in den Reaktor zurückgeführt unter entsprechender Verringerung der eingeführten Nährlösung.
Die Zellzusammensetzung beträgt: 72% Rohprotein, 4,5% Nukleinsäuren,3,5 Asche und 5,5% Fett, bezogen auf die Zelltrockenmasse.
Das Verfahren der Erfindung bietet den Vorteil, dass erstmalig ein obligat methylotrophes Bakterium verwendet wird, das eine wesentlich höhere Produktivität aufweist gegen über bekannten fakultativ methylotrophen Mikroorganismen.
Bei dem Verfahren der Erfindung können sich keine Muttern entwickeln, die die Fähigkeit auf Methanol zu wachsen verloren haben. Das Bakterium nach dem Verfahren der Erfindung ist somit sehr stabil, sein Stoffwechsel ist auf ein Minimum reduziert, weitere genetischen Veränderungen würden zum Absterben der Zellen führen. Ein weiterer Vorteil des Verfahrens der Erfindung liegt darin, dass die Methanol-dissimilatorischen und -assimilatorischen Enzyme konstitutiv sind. Wenn das Verfahren der Erfindung unter Methanol-limitierenden Bedingungen kontinuierlich durchgeführt wird, ist der Nucleinsäuregehalt der Zellmasse sehr niedrig und der Verlust an Methanol durch Verdampfung praktisch gleich null.

From DT-OS 2 407 740 a method for cultivating microorganisms is known in which a mixed culture consisting of an optional methanol-utilizing bacterium and several non-methanol-utilizing bacteria is used. For the cultivation of protein from methanol, according to this prior art, no bacterium that uses methanol is used; instead, methylotrophic microorganisms are optionally used. The method of the invention cannot develop nuts which have lost the ability to grow on methanol. The bacterium according to the method of the invention is thus very stable, its metabolism is reduced to a minimum, and further genetic changes would lead to the death of the cells.
