Gegenstand der Erfindung ist ein neues Verfahren zur Herstellung von Desacetoxycephalosporin C (3-Methyl-7-(6- -aminodipoyl)-ceph-em-carbonsäure) der Formel I

und seiner Salze. Diese Verbindung ist bekannt, vgl. Stedman et al. J. Med. Chem., 7 (1964), 117-119 und US-Patent 3 124576. Sie wurde durch katalytische Hydrierung von Cephalosporin C erhalten und, ohne charakterisiert zu werden, direkt in Derivate übergeführt. Auch Derivate von Verbindung I, z.B. 3-Methyl-7- (phenoxyacetyl)-ceph-3 -em-4-carbon- säure, wurden durch katalytische Hochdruckhydrierung der entsprechenden 3-Acetoxymethylverbindung hergestellt, vgl.
Mprin et al., J. Am. Chem. Soc. 85 (1963), 1896-97. Ein anderer Weg zur Herstellung solcher Derivate besteht in der Sulfoxidumlagerung entsprechender Penicilline, vgl. Morin et al., 1. c.
Die Verbindung der Formel I ist von erheblichem prak tischem Interesse, und zwar weniger wegen ihrer relativ schwachen antibiotischen Wirkung, als wegen ihrer Verwendbarkeit als Ausgangsmaterial zur Herstellung wertvoller Derivate der Formel II

worin R ein anderer Acylrest als der Aminoadipoylrest ist, insbesondere ein Acylrest, wie er in therapeutisch verwendbaren Acylderivaten der 6-Aminopenicillansäure und 7-Aminocephalosporansäure vorkommt, vgl. z.B. Sassiver et al., Adv. Appl. Microbiol. 13 (1970), 163-235.
Um solche Acylderivate herzustellen, wird aus Verbindung I zunächst der b-Aminoadipoylrest abgespalten, z.B. nach dem Verfahren des französischen Patentes Nr. 1 394820 oder des belgischen Patentes Nr. 720 185, wobei die Verbindung der obigen Formel II, worin R für Wasserstoff steht, erhalten wird. Diese Verbindung kann dann in erwünschter Weise acyliert werden.
Die vorliegende Erfindung macht Verbindung I durch Fermentation zugänglich. Es wurde gefunden, dass man einen Cephalosporin C produzierenden Mikroorganismus so mutieren kann, dass er Desacetoxycephalosporin C produziert.
Die Biosynthese von Cephalosporin C im Mikroorganismus ist bisher nicht aufgeklärt. Als Nebenprodukte in der Cephalosporin C-Produktion war Desacetylcephalosporin C bekannt, nicht aber Desacetoxycephalosporin C. Es war daher sehr überraschend; dass es gelang, Mutanten herzustellen, die Desacetoxycephalosponn C produzieren. Die Auffindung der Mutanten lässt den Schluss zu, dass Desacetoxycephalosporin C ein Zwischenprodukt in der Biosynthese von Cephalosporin C durch Cephalosporin C-bildende Mikroorganismen ist und dass bei der Mutante die Reaktionsschritte, welche Desacetoxycephalosporin C in Cephalosporin C bzw. Desacetylcephalosporin C überführen, blockiert sind.
Da biosynthetische Reaktionen unter Mitwirkung von Enzyme men zustande kommen, kann man vermuten, dass bei der neuen Mutante ein oder mehrere Enzyme, die an der der-Bioo synthese von Cephalosporin C aus Desacetoxycephalosporin
C beteiligt sind, nicht mehr vorhanden oder ganz oder teil weise blockiert sind. Das bedeutet, dass ein oder mehrere
Enzyme ihre Funktion der Katalyse der Umwandlung von
Desacetoxycephalosporin C in Cephalosporin C (als Endpro dukt) nicht oder nicht vollständig erfüllen können. Im ersten
Fall wird gar kein Cephalosporin C produziert, im zweiten
Fall könnte neben Desacetoxycephalosporin C eine kleinere oder grössere Menge von Cephalosporin C oder Desacetyl cephalosporin C gebildet werden.
Für die Auffassung, dass die neuen Mutanten einen Defekt aufweisen hinsichtlich eines oder mehrere Enzyme, die an der Überführung von
Desacetoxycephalosporin C in Cephalosporin C im Mikro organismus beteiligt sind, spricht die Tatsache, dass aus Ce phalosporin C - produzierenden Mikroorganismen bzw. in deren zellfreien Extrakten ein Enzym nachgewiesen werden konnte, das in Gegenwart von Sauerstoff und einem Co faktor (H-Donator) Desacetoxycephalosporin C in Desace tvlcephalosporin C überführt. Das Enzym (Desacetoxyce phalosporin C-Hydroxylase) ist ein lösliches Protein, das ein pH-Optimum von 6,5 aufweist und das durch Hitze (5 Min.
bei 100"C) denaturiert wird. In den neuen Mutanten bzw.
deren zellfreien Extrakten konnte diese Hydroxylase nicht nachgewiesen werden. Man kann daraus schliessen, dass durch den Mangel an Hydroxylase in den neuen Mutanten die Bio synthese des Cephalosporin C auf der'Stufe des Desacetoxy cephalosporin C abgebrochen wird. Die neuen Mutanten, die man als Desacetoxycephalosporin C-Hydroxylase-Ausfall mutanten (hydroxylase-defective or leaky mutants) bezeich nen kann, wurden in der Natur bisher nicht gefunden.
Gegenstand der Erfindung sind daher Mutanten von Ce phalosporin C-produzierenden Mikroorganismen, bei denen die Überführung von Desacetoxycephalosporin C in Cephalo sporin C bzw. Desacetylcephalosporin C ganz oder teilweise unterbunden ist. Die Erfindung betrifft auch die Verwen dung der genannten Desacetoxycephalosporin C-Hydroxylase Ausfailmutanten zur Gewinnung von Desacetoxycephalo sporin C durch Fermentation der genannten Mutanten. Fer ner ist Gegenstand der Erfindung ein Verfahren zur Herstel lung der neuen Mutanten nach an sich bekannten Methoden, nämlich durch Behandlung mit mutagenen Mitteln.
Weiter ist ein Erfindungsgegenstand das Verfahren zur Herstellung von Desacetoxycephalosporin C, das dadurch gekennzeich net ist, dass man eine Mutante eines Cephalosporin C-pro duzierenden Mikroorganismus, bei der die Überführung von
Desacetoxycephalosporin C in Cephalosporin C ganz oder teilweise unterbunden ist,-in an sich bekannter Weise züchtet und Desacetoxycephalosporin C isoliert.
Als der Mutagenese zu unterwerfende Cephalosporin C produzierende Microorganismen verwendet man die bekann ten Pilze der Gattung Emericellopis oder Cephalosporium, wie Cephalosporium acremonium, z.B. den Brotzu-Stamm
I.M.I. 49 137 (ATCC 11550) oder Mutanten hiervon, z.B. die
Clevedon Mutante 8650 (ATCC 14533), vgl. die U.S. Patente
2 831 797 und 3 396 083, ferner deren Mutanten, die mehr
Cephalosporin C produzieren, z.B. die in der deutschen Of fenlegungsschrift 2 101 345 beschriebenen Methionin-auxo trophen Mutanten oder die Mutanten gemäss der deutschen
Offenlegungsschrift 2239 321.
Die Herstellung der neuen Mutanten erfolgt nach den für die Gewinnung von Mutanten bekannten Methoden durch
Einwirkung von mutationsauslösenden Strahlen oder chemi schen Agenzien auf Konidiien des Pilzstammes.
So können zur Mutationsauslösung radioaktive Strahlen, wie < 4-, a- - oder y-Strahlen, oder Neutronen, ferner ultravio- lette Strahlen verwendet werden. Bekannte chemische Mutagene sind z.B. alkylierende, besonders niederalkylierende Agenzien, wie Diäthylsulfat, Äthylmethansuifonat, Äthyl äthansulfonat, oder Analoge von Nucleotidbasen wie 5-Bromuracil oder 2-Aminopurin, oder Verbindungen, welche die Nucleotidbasen chemisch verändern, wie Hydroxylamin, salpetrige Säure oder l-Methyl-3-nitro-nitrosoguanidin oder Verbindungen, die einen Ausfall oder einen zusätzlichen Einschub einer oder mehrerer Nucleotidbasen bewirken, z.B.
Acridine wie Proflavin. Die mutationsauslösenden Strahlen oder chemischen Agenzien werden zur Einwirkung auf Konidien des Pilzstammes gebracht. Die Einwirkungszeit wird so bemessen, dass die Zahl der Mutanten möglichst gross ist; bei einer solchen Einwirkungsdauer werden die meisten Konidien (ca. 90 - 99%) abgetötet, vgl. z.B. die deutsche Offenlegungsschrift 2 101 345, Fig. 1. Vorzugsweise wird die mutagene Behandlung so durchgeführt, dass die Überlebensrate ca. 1 % beträgt.
Es wurde experimentell gefunden, dass das Verhältnis der Anzahl der die mutagene Behandlung überlebenden Konidien zur Anzahl der erhaltenen Desacetoxycephalosporin Cproduzierenden Mutanten unabhängig von der Behandlungsmethode innerhalb statistischer Grenzen konstant und rerro- duzierbar ist. Im Durchschnitt findet man eine Desacetoxy cephalosporin C-produzierende Mutante unter 5000 bis 10 000 die mutagene Behandlung überlebenden Konidien.
Die mutagene Behandlung kann beispielsweise wie folgt durchgeführt werden:
Die in M/15 Phosphatpuffer vom pH 7 suspendierten Konidien (2,5 104/mol) werden gleichmässig auf der Oberfläche eines festen Agarnährmediums, das alle benötigten Nährstoffe enthält, z.B. auf dem komplexen Vollmedium 1 H (10 g Glucose, 4 g Difco-Bacto-Hefeextrakt, 4 g D,L Methionin, 25 g Difco-Agar, Leitungswasser ad 1000 ml, Sterilisation 20 Minuten bei 1200C und 1,2 atü, pH nach der Sterilisation 7,0) verteilt; pro Agarplatte werden 0,1 ml ausgestrichen. Auf diese Weise gelangen 2,5 103 lebende Konidien auf die Agaroberfläche.
Diese wird während 14 Sekunden dem UV-Licht einer Philips TUV-Sterilisationslampe mit einer maximalen Strahlenintensität bei der Wellenlänge von 2537 Ä ausgesetzt. Dabei gelangen 10 000 erg/sec/cm2 auf die Oberfläche. Die Strahlenmenge kann mittels eines Präzisions-UV-Messgerätes gemessen und so dosiert werden, dass die gewünschte Zahl von überlebenden Kondien, z.B.
1% (30 + 5) erhalten wird.
Die Isolierung der gewünschten Mutanten aus den mutagen behandelten Konidien erfolgt in der Weise, dass man die Agarplatten inkubiert, z.B. 7 Tage bei 25 , und die separaten Kolonien, im obigen Fall 25-35 Kolonien (Durchmesser ca. 8 mm), testet, ob sie Desacetoxycephalosporin C bilden, bzw. ob sie das Hydroxylase-Enzym enthalten, welches für die Überführung von Desacetoxycephalosporin C in Desacetylcephalosporin C verantwortlich ist.
Diese Prüfungen werden wie üblich nicht an den einzelnen Kolonien selbst, sondern an der Tochtergeneration, die man durch Züchtung der einzelnen Kolonien in einem flüssigen Nährmedium, z.B. in Schüttelkolben während 3 bis 7 Tagen bei 23 CC erhält, vorgenommen. Man kann entweder das Kulturfiltrat untersuchen, um festzustellen, ob darin Desacetoxycephalosporin C bzw. kein oder nur wenig Cephalosporin C vorhanden ist oder man kann das Mycel untersuchen, um festzustellen, ob die erwähnte Hydroxylase darin vorhanden ist.
Da man, wie bereits erwähnt, im Durchschnitt unter 5000 bis 10 000 Kolonien, die aus den die mutagene Behandlung überlebenden Konidien hervorgegangen sind, nur eine Desacetoxycephalosporin C-produzierende Kolonie findet, ist es arbeitstechnisch wichtig, ein Vorausleseverfahren zur Verfügung zu haben, mit dem man die Cephalosporin C-bildenden Kolonien ausscheiden kann. Hierfür geeignet ist vor allem der mikrobiologische Plattendiffusionstest mit auf Cephalosporin C, nicht aber gleich empfindlich auf Desacetoxycephalosporin C ansprechenden Mikroorganismen, z.B. mit Alcaligenes faecalis, Sarcina lutea oder Neisseria catarrhalis.
Diesen Stämmen gegenüber zeigt Desacetoxycephalosporin C nur eine sehr schwache Aktivität, also einen kleinen Hemmhof, während Cephalosporin C stark aktiv ist und dementsprechend einen grossen Hemmhof bildet. Von der weiteren Prüfung auf Desacetoxycephalosporin C-enthaltende Kulturfiltrate können daher diejenigen, die einen grosen Hemmhof gegenüber den genannten Mikroorganismen erzeugens ausgeschieden werden. Im Durchschnitt können auf diese Weise 950/0 der Kulturen von der weiteren Prüfung ausgeschlossen werden. In den gegenüber den genannten Microorganismen weniger aktiven Fermentationsbrühen kann z.B.
biologisch, biochemisch und dünnschichtchromatographisch auf Anwesenheit von Desacetoxycephalosporin C geprüft werden.
Die biologische Prüfung kann z.B. vorgenommen werden, indem man von mehreren, beispielsweise 5 - 10, Kulturfiltraten und einer Ständardmenge von authentischem Desacetoxycephalosporin C ein Bioautogramm mit den genannten Mikroorganismen, z.B. Alcaligenes faecalis, herstellt. Zu diesem Zweck chromatographiert man die Kulturfiltrate und das Standard-Desacetoxycephalosporin C auf Papier (z.B. Whatman Nr. 1) in einem der in Tabelle 1 genannten Lösungsmittelsysteme (laufzeit ca. 8 - 10 Stunden) und legt das Papier dann, auf eine Agarplatte mit dem betreffenden Mikroorganismus.
Nach 30 Minuten bei 4"C wird das Papier wieder entfernt und die Agarplatte inkubiert (ca. 12 - 20 Stunden bei 37"C). Desacetoxycephalosporin C weist an der Stelle des Rf-Wertes (s. Tabelle 1) einen Hemmhof auf. Man kann daher ablesen, welche Kulturfiltrate Desacetoxycephalosporin C enthalten und aus der Grösse des Hemmhofs auf die Menge an Desacetoxycephalosporin C schliessen. Auf diese Weise kann man auch erkennen, ob die geprüfte Mutante ausschliesslich Desacetoxycephalosporin C produziert oder ob die daneben auch noch Cephalosporin C, Desacetylcephalosporin C oder eine andere mikrobiol. aktive Substanz produziert.
Die biochemische Prüfung auf Anwesenheit von Desactoxycephalosporin C-Hydroxylase kann beispielsweise so durchgeführt werden, dass man im Mycel des mutierten Stammes bzw. in daraus erhaltenen zellfreien Extrakten auf Vor handensein bzw. Nichtvorhandensein der erwähnten Hy. droxylase prüft.
Das Mycel kann beispielsweise erhalten werden, indem man den zu untersuchenden Stamm (aus einer Kolonie der oben erwähnten Agarplatten) in einer synthetischen Nährlösung ( synthetisches Grundmedium C3 , s. unten), der 4 g D.L-Methionin zugesetzt sind, während 96 Stunden bei 23 0C inkubiert und abtrennt, z.B. durch Zentrifugieren. (Man verwendet in diesem Fall vorzugsweise ein synthetisches N hrmedium, um zu vermeiden, dass andere unbekannte oder ähnliche Enzyme aus den Nährlösungsbestandteilen, z.B. aus Erdnussmehl, zugegen sind.) Zwecks Herstellung des zellfreie Extraktes wird das Mycel mit M/15 Phosphatpuffer vom pH 7,0 zweimal gewaschen, dann werden die Zeilen mit einer X-Presse (der Fa.
AB Biox, NACKA 2, Schweden) in analoger Weise wie z.B. für D-Aminosäureoxidase von Benz et al., Eur. J. Biochem. 20 (1971) 82, beschrieben aufgeschlossen. Der so gewonnene zellfreie Rohextrakt in M/15 Phosphatpuffer pH 7 wird auf seine Fähigkeit geprüft, Desacetoxycephalosporin C in Gegenwart von Sauerstoff in Desacetylcephalosporin C umzuwandeln, z.B.
indem man ihn während 4 Stunden bei 23"C unter Schüt tein mit Desacetoxycephalosporin C reagieren lässt. Falls die Hydroxylase vorhanden ist, wird Desacetylcephalosporin C gebildet Dieses wird mittels eines weiteren Enzyms, (Desace tylcephaiosporin GO-Acetyl-Transferase) und Acetyl-l-C14- -Co enzym A in Cepharlosporin C übergeführt. Wenn keine Hydroxylase vorhanden ist, bildet sich kein Desacetylcephalosporin C und somit auch kein (radioaktives) Cephalosporin C. Das Desacetoxycephalosporin C häuft sich während der Züchtung des Pilzstammes an und ist dann im Kulturfiltrat nachweisbar.

Die Dünnschichtchromatographische Prüfung auf Anwesenheit von Desacetoxycephalosporin C kann in der vorgereinigten Fermentationslösung des zu untersuchenden Stammes (aus einer Kolonie der oben erwähnten Agarplatten) vorgenommen werden. Die Fermentation des Stammes und die Vorreinigung der Fermentationsbrühe erfolgen vorzugsweise in der für die Gewinnung von Cephalosporin C bekannten Weise.
So wird der Stamm beispielsweise in einem komplexen Vollmedium 144 Stunden fermentiert und die Fermentationsbrühe wie im belgischen Patent 750 292 (Case 4-6770) beschrieben vorgereinigt, z.B. durch Ansäuern, Filtration, Extraktion, Adsorption an nicht-ionischen Harzen mit grosser Oberfläche, wie Amberlites XAD-2, gegebenenfalls Absorption an schwach basischen Ionenaustauschern wie Amberlitee IR-68, Elution und LyophilisatiQn. Das so erhaltene Lyophilisat wird dünnschichtchromatographisch untersucht. In der Tabelle 1 sind die Rf-Werte tan Cellulose in 9 verschiedenen Lösungsmittelsysternen angcgeben. Ausserdem enthält die Tabelle die entsprechenden Rf-Werte für Cephalosporin C, Desacetylcephalosporin C und Desacetylcephalosporin C-Lacton.
Die Lösungsmittel sind: A: Isopropanol-Ameisensäure-Wasser (77:4:19)' B: n-Butanol-Aceton-Diäthylamin-Wasser (37:37:8:18) C: n-Butanol-Essigsäure-Pyridin-Wasser (37,5:7,5:15:30) D: n-Butanol-Äthanol-Wasser-Eisessig (50:15:20:15) E: 66%iges wässeriges Acetonitril F: 80%iges wässeriges Phenol G: n-Butanol-Ameisensäure-Wasser (4:1:4) H: n-Butanol-Eisessig-Wasser (obere Phase) [11:3:7] I: 70 %ges wässeriges n-Propanol.

Das durch Fermentation gewonnene Desacetoxycephalosporin C weist die gleichen Rf-Werte wie die synthetisch erhaltene Verbindung auf. Wie Tabelle 1 zeigt, kann Desacetoxycephalosporin C, wenn erwünscht, dünnschichtchromatogra- phisch von den übrigen Verbindungen abgetrennt werden.
Zur Abtrennung eignet sich auch der Aminosäureanla- lysator [AA] (Technicon TSM 1 der Fa. Technicon Corp., Torryton, N.Y., USA). In der Tabelle 2 sind die Retentionszeiten der oben genannten Verbindungen (20 , l-Proben einer 5 millimolaren Lösung der Verbindung in 0,2 M Natriumcitratpuffer vom pH 2) angegeben. Weiter kann Desacetoxy cephalosporin C nachgewiesen und von den genannten übrigen Verbindungen getrennt werden durch Hochdruckflüssigkeits chromatographle [HPLC] (s. J. Konecny et al., J. Antibiotics 26, 135 (19731 > . In Tabelle 2, äusserste rechte Spalte sind die.
entsprechenden Retentionszeiten in Minuten angegeben.
nicht mischbaren Lösungsmittel oder Lösunigsmittelgemisch und/oder Adsorption an Ionenaustauschern.
Als mit Wasser nicht mischbare Lösungsmittel kommen beispielsweise in Betracht aliphatische, cycloaliphatische, araliphatische und aromatische Kohlenwasserstoffe mit höchstens 12 Kohlenstoffatomen, die gegebenenfalls durch Halogenatome, wie Brom, Fluor, besonders Chlor, substituiert sind, z.B. Hexan, Heptan, Cyclohexan, Benzin, Petrol- äther (Kp. 110-1400C), Kerosen (Kp. 210.2400C), Tetrachlorkohlenstoff, Chloroform, Methylenchlorid, Methylchloroform, Äthylenchlorid, Perchloräthylen, Perfluoräthylen, Isopropylbromid, Benzol, Toluol, Xylole, ferner Ester, besonders Niederalkylester von niederen Fettsäuren wie Essigester, Butylacetat, Amylacetat, Ketone wie Methylisobutylketon, Methylisoamylketon, Äther wie Diisopropyläther,
mit WzaF ser nicht oder wenig mischbare Alkohole wie Butanol, 2 Äthylbutanol, Äthylhexanol, Cyclopentanol, Cyclohexanoi.
Die Extraktion mit einem mit Wasser nicht mischbaren Lösungsmittel wird vorzugsweise in Gegenwart eines flüssigen Ionenaustauschers, z.B. Amberlite LA-2 (der Fa.
Rohm und Haas) durchgeführt. Zweckmässig arbeitet man im sauren pH-Bereich, z.B. pH 1-6.
Statt das Kulturfiltrat durch Säurebehandlung und/oder Lösungsmittelextraktion vorzureinigen, kann man es auch einer aufeinanderfolgenden Behandlung mit einem starken Kationenaustauscherharz, z.B. Amberlite IR-120 oder Dowex 50-8 (der Fa. Dow Chemical Co.,) und einem starken Anionenaustauscherharz, z.B. Amberlite IRA-400 oder Dowex 1 unterwerfen, wie im englischen Patent 968 324 beschrieben.
Um Desacetoxycephalosporin C dem gegebenenfalls wie erwähnt vorgereinigten Kulturfiltrat zu entziehen, sind verschiedene Methoden geeignet, die ein- oder mehrmals, einzeln oder in beliebiger Kombination angewandt werden können. Zu nennen sind insbesondere Absorption oder Adsorption, Lösungsmittelverteilung und Präzipitation.
Als Absorptions- oder Adsorptionsmittel kann man beispielsweise Aktivkohle, Aluminiumoxid, Cellulose, Ionen austauschharze und nichtionische Adsorptionsharze, zur Elution vor allem wässerige Lösungen verwenden. Die Verwendung von Aktivkohle und Aluminiumoxid sind beispielsweise im englischen Patent 810 196 beschrieben. Als Ionenaustauscher sind z.B. zu nennen schwach basische Anionenaustauscher wie Amberlite IR- 4B, oder Deacidite E, aus denen das gewünschte Produkt z.B. mit wässerigem Pyridinacetat bei einem pH von ca. 6 eluiert werden kann. Es können auch stark basische Anionenausiauscherharze wie Am berlite IRA-400, Dowex 1, Dowex 2 oder Deacidite FF verwendet werden, aus denen die Produkte mit ca. l-n.
Essigsäure eluiert werden können, vgl. englische Patente 810 196 und 968 324. Als nichtionische Adsorptionsharze hervorzuheben sind mit Divinylbenzol vernetzte Styrolpolymerisate mit grosser Oberfläche wie Amberlite XAD-2, aus denen das Desacetoxycephalosporin C beispielsweise mittels wässerigen Alkoholen, z.B. Niederalkanolen wie Methanol, Äthanol, besonders Isopropanol, eluiert werden kann, vgl. belgisches Patent 750 292.
Die Methode der Lösungsmittelverteilung kann beispielsweise in Form der Gegenstromverteilung angewandt werden.
Als Lösungsmittelsystem eignen sich Gemische von Wasser und Phenol oder Niederalkvl-substituierten Phenolen, vgl. das englische Patent 810 196.
Zur Präzipitation von Desacetoxycephalosporin C aus wässerigen Lösungen eignen sich z.B. mit Wasser mischbare organische Lösungsmittel wie Ketone, z.B. Aceton, oder Niederalkanole, z.B. Isopropanol. Aus konzentrierten wässse- rigen Lösungen können die Verbindungen auch in Form ihrer Metall- wie Alkalimetall, z.B. Natrium-, oder Erdalkalime-

Die Fermentation der erfindungsgemässen Mutanten und die Isolierung von Desacetoxycephalosporin C aus der Fer mentationsbrühe erfolgt nach den für die Gewinnung von Antibiotika bekannten Methoden.
Da Desacetoxycephalosporin C ein dem Cephalosporin C sehr ähnliches chemischphysikalisches Verhalten zeigt, sind alle bekannten Verfahren zur Gewinnung von Cephalosporin C aus Fermentationsflüssigkeiten anwendbar.
Die Gewinnung von Desacetoxycephalosporin C aus der Fermentationsbrühe kann daher z.B. nach den Verfahren der englischen Patente 810 196; 938 758; 968 324; 1 036 125; 1109 362; 1 205 226, der französischen Patente 1 429 873 oder 2011 520, des belgischen Patentes 750 292 oder der deutschen Offenlegungsschriften 2 101 345, 1 933 187, 2031 754 oder 1 939 341 erfolgen.
Der Pilz wird aerob gezüchtet, vorzugsweise submers, unter Schütteln oder Rühren mit Luft oder Sauerstoff in Schüttelflaschen oder den bekannten Fermentern. Die Fer mentationsiösung enthält eine Kohlenstoff- und Stickstoffquelle und gegebenenfalls achstumsfördernde Stoffe sowie anorganische Salze. Als Kohlenstoffquelle kommen z.B. assimilierbare Kohlenhydrate wie Glucose, Saccharose, Laktose, Maltose, Stärke, ferner Mannit, Inosit, Glyzerin, in Betracht. Als stickstoffhaltige Nährstoffe und gegebenenfalls wachstumsfördernde Stoffe seien genannt: Aminosäuren, insbesondere Methionin, Peptide und Proteine sowie deren Abbauprodukte, z.B. Erdnussmehl, Sojabohnenmehl. Pepton oder Trypton, ferner Fleischextrakte, wasserlösliche Anteile von Getreidekörnern oder Destiilationsrückstände der Alkoholherstellung, z.B.
Cornsteep dry, Ammoniumsalze und Nitrate. Von anderen anorganischen Salzen kann die Nährlösung beispielsweise Chloride, Carbonate, Sulfate von Alkalioder Erdalkalimetalien, Eisen, Kupfer, Zink und Mangan enthalten.
Die Züchtung erfolgt bei einer Temperatur zwischen 18 und 37"C, vorzugsweise bei ca. 23"C während 4-7 Tagen.
Zur Isolierung des Desacetoxycephalosporin C kann man sich verschiedener Methoden bedienen. Vorzugsweise wird Desacetoxycephalosporin C aus der Fermentationsbrühe nach den für die isolierung von Cephalosporin C bekannten Methoden isoliert.
Die Isolierung kann entweder nach dem Whole-Broth Verfahren direkt aus der Fermentationsbrühe oder vorzugsweise nach Abtrennung des Mycels, z.B. durch Filtrieren, gegebenenfalls in Anwesenheit von Filterhiifsmitteln wie Diatomeenerde, aus dem Kulturfiltrat erfolgen. Vor oder nach dem Filtrieren kann eine Behandlung mit Säuren, z.B. Mineralsäuren wie Schwefelsäure oder organischen Säuren wie Oxalsäure, zur Ausfällung schwerlöslicher Salze, z.B. Cal ciumsaize, aus derNährlösung und zur Zerstörung gegebenenfalls vorhandenen Penicillins N vorgenommen werden Auch können zur weiteren Vorreinigung des Kulturfiltrates lipophile Verunreinigungen daraus entfernt werden. Hierzu eignen sich beispielsweise Extraktion mit einem mit Wasser tall-, z.B.
Calcium- oder Bariumsalze gefällt werden. Besonders hervorzuheben ist die Fällung als schwerlösliche Schwermetallkomplexe, z.B. als Komplexe mit Kupfer, Quecksilber, Cadmium, Blei, Mangan, Eisen, Kobalt, Nickel, oder insbesondere mit Zink, vgl. franz. Patent 2011 520.
Zur Trennung von Desacetoxycephalosporin C und gegebenenfalls durch Fermentation ebenfalls gebildeten Cephalosporin C eignet sich besonders Chromatographie an Cellulose. Bei hohem Gehalt an Cephalosporin C kann es erforderlich sein, die Chromatographie in mehr als einer Stufe durchzuführen. Es kann auch vorteilhaft sein, das Cephalosporin C vor einer der Chromatographie-Stufen in Desacetylcephalosporin C überzuführen, da dieses besser von Desacetoxycephalosporin C abgetrennt werden kann.
Die Überfüh- rung von Cephalosporin C in Desacetylcephalosporin C erfolgt in bekannter Weise durch Desacetylierung, z.B. mittels Desacetylasen wie Citrus-Desacetvlase oder Desacetylase aus Mikroorganismen wie sie im englischen Patent 1 080 904 für die Desacetylierung von 7-Amino-cephalosporansäure beschrieben sind, insbesonder mit der Desacetylnse aus Bac.
subtilis ATCC 6633. Man kann mit dem Zell-Lyophllisat des Mikroorganismus oder mit aus dem Mikroorganismus ge wonnenem Enzym desacetylieren.
Die Elution wird z.B. mit wässerig-alkoholischen oder wÅasserig-phenolischen Lösungen wie wässerigen Niederalka- nolen, z.B. Propanol oder Butanol oder wässerigem Phenol durchgeführt.
Desacetoxycephalosporin C der Formel I hat die folgenden Eigenschaften:
Im UV-Spektrum In Wasser ist man = 260 nm. Im NMR Spektrum (in D2O) ist ein scharfes Signal bei 1,92 8 vorhanden, das den 3 Protonen der CH3-Gruppe in 3-Stellung zugeordnet werden kann. Im IR-Spektrum (in Kaliumbromid) fehlen die für Cephalosporin C typischen Banden bei 8,15 x und 9,7,S" stattdessen ist bei 3,36 {w, eine Bande vorhanden.
Die optische Drehung von Desacetoxycephalosporin C beträgt [JD20 = + 1250 + 10 (c = 1 in Wasser). Bei der Hydrolyse von Desacetoxycephalosporin C mit 6N HCI im Bombenrohr (24 h, llO"C) neben anderen Zersetzungsprodukten oc-Aminoadipinsäure erhalten. Weitere chemisch-physikalische Charakteristika sind oben in den Tabellen 1 und 2 angegeben.
In allen genannten Charakteristica stimmt das fermentativ hergestellte Desacetoxycephalosporin C mit chemisch hergestelltem Desacetoxycephalosporin C völlig überein.
Die Erfindung wird im folgenden Beispiel beschrieben.
Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Der Inhalt einer Ampulle mit lyophilisierter Mycel-Spo ren > Suspension der Desacetoxycephalosporin C-Hydrolyase Ausfalimutante 127a/2, die man durch Bestrahlung von Cephalosporium acremonium ATCC 14533 mit W-Licht, wie im allgemeinen Teil der Beschreibung angegeben, erhal ten hat, wird in 5 mi M/15 Phosphatpuffer pH 7 aufgelöst.
Ein 500 ml-Erlenmeyerkolben mit 4 Strömungsbrechern und
100 ml der Nährlösung a) wird mit den 5 mi der Cephalospo riumsuspension beimpft und 72 Stunden auf einer rotierenden Schüttelmaschine mit 250 Umdrehungen pro Minute (upm), Amplitude 50 mm, bei 250C inkubiert. Mit je 5 ml der so gewonnenen Vorkultur beimpft man die Hauptkulturlösung b), die sich ebenfalls in 500 ml-Erlenmeyerkolben mit 1 Strömungsbrecher und 100 ml Nährlösung befindet. Die Kolben werden 168 Stunden auf einer rotierenden Schüttelmaschine mit 250 upm bei 25"C inkubiert. Vom 4. Tag an werden täglich Proben zur biologischen (Plattendiffusionstest) und chemischen (Dünaschichtchromatogramm) Aktivitätsbtestimmung entnommen.
Nach 168 Stunden hat sich die maximale Menge Desacetoxycephalosporin C gebildet. Die Kulturlösung hat dann ein pH von 7,5 und eine Konzentration von ca. 750 mg/l Desacetoxycephalosporin C.

3 Liter Kulturiösung, die 0,75 g Desacetoxycephalosporin C pro Liter enthält, wird mit 50%iger wässeriger Schwefelsäure (V: V) auf pH 3,0 angesäuert und zentrifugiert. Das Kulturfiltrat mit einem Gehalt von 680 mg Desacetoxycephalosporin C pro Liter wird mit 1,5 Liter einer Lösung von 10% Amberlite LA-2 (Acetatform) in 2-Äthylhexanol in 3 Portionen à 0,5 Liter extrahiert, wobei das Desacetoxycephalosporin C in der wässerigen Phase bleibt. Das extrahierte Kulturfiltrat wird mit 50%iger Schwefelsäure auf pH 2,6 angesäuert und durch 15 Liter Amberlite XAD-2-perkoliert.
Das Harz wird mit 750 ml deionisiertem Wasser gewaschen und anschliessend mit 3 Liter 12%igem wässerigem Isopropanol eluiert. Das Eluat wird in 4 Fraktionen à 600 ml aufgefangen. Die Fraktion mit der höchsten- Konzentration an Desacetoxycephalosporin C wird lyophilisiert. Das Rohlyophilisat enthält 24% Desacetoxycephalosporin C. Es kann wie im Beispiel 2 beschrieben in den Zinkkomplex und sodann in das freie Desacetoxycephalosporin C übergeführt wer den.
Beispiel 2
27 Liter Kulturbrühe werden mit 50%iger Schwefelsäure auf pH 2,8 gestellt und nach Zusatz von 810 g Filterhilfsmittel auf einer Filterpresse filtriert Das saure Filtrat wird an 18 Liter Amberlite XAD-2 (Polystyrol-divinylbenzolharz) adsorbiert. Die Durchflussgeschwinadigkeit beträgt 18 1 /Stunden. Die beladene XAD-2 Säule wird mit 20% V/V wässerigem Isopropanol eluiert. Die Eluiergeschwindigkeit beträgt 36 1/Std. Das Eluat wird in Fraktionen zu 2 Liter aufgefangen. Über 70% der absorbierten Aktivität sind in den Fraktionen 7-18, d.h. in 24 Liter Eluat. Diese werden an 500 ml Amberlite IRA-68 (Acetat-Form) adsorbiert. Die Durchflussgeschwindigkeit beträgt 5 1/sud. Die Säule wird alsdann mit Pyridinacetatpuffer vom pH 5,5, der 0,44 molar an Pyridin und 0,2 molar an Essigsäure ist, eluiert.
Die Eluiergeschwindigkeit beträgt 1 I jSbd. Es werden Eluate zu 200 ml aufgefangen. Die Fraktionen 2-13, d.h. 2,4 Liter, enthalten die gesamte Aktivität. Sie werden im Vakuum bei 30-35"C auf 100 ml eingeengt. Das Konzentrat wird unter Rühren in 1200 ml Isopropanol eingetragen. Die dabei entstanfdene Fällung wird abgenutscht, mit Isopropanol gewaschen und saugtrocken in 200 ml Wasser gelöst. Zur klaren Lösung gibt man 20 g Zinkacetat, versetzt dann die homogene Lösung innert 15 Minuten mit 250 ml Isopropanol. Dabei fällt Desacetoxy Cephalosporin C als Zinkkomplex aus.
Nach 4-stündigem Kühlen auf 0 C wirrdabgenutscht. Man wäscht den Filterrück- stand mit Wasser und Isopropanol und trocknet bei 50"C im Vakuum. Man erhält das Produkt in Form hellbeiger Kristalle, welche gemäss HPLC und Dünnschichtchromatogramm mit Desacetoxy-Cephalosporin C identisch sind (da der Zinkkomplex sich bei diesen beiden analytischen Nachweismethoden gleich wie das freie Desacetoxy-Cephalosporin C ver hält).
Aus dem Zinkkomplex kann das freie Desacetoxy-Cephalo sporin C in gleicher Weise freigesetzt werden wie Cephalosporin C aus dem Zinkkomplex, vgl. U.S. Patent 3 661 901.
Beispielsweise wird eine ca. 10%ige wässerige Suspension des Zinkkomplexes langsam unter Rühren mit einem stark sauren Kationenaustauscher in der H-Form wie Dowex 50-WX l2 (sulfoniertes, mit Divinylbenzol vernetztes Polystyrol) versetzt, bis ein pH von 2,5 erreicht ist. Dann wird die Lösung filtriert und eingedampft. Man kann auch das Natriumsalz herstellen, indem man bis zu einem pH-Wert von 6,5 Natronlauge zugibt, dann einengt und das Natriumsalz auskristallisieren lässt.
