Die Erfindung betrifft ein Verfahren zur Herstellung von 2',3'-Didesoxy-3'-fluor-pyrimidin-nucleosiden der allgemeinen Formel:

worin X das Sauerstoff- oder Schwefelatom oder die NH Gruppe, R das Wasserstoffatom, einen substituierten oder unsubstituierten Alkylrest, ein Halogenatom, die Hydroxy- oder SH-Gruppe bedeuten.
Es ist aus DWP 75.084 bekannt, Verbindungen der Formel I aus den entsprechenden Pyrimidin-2,3'-anhydro-2'desoxynucleosiden herzustellen. So wird z. B. das als selektiver Hemmstoff der DNS-Synthese bekannte 3'-Desoxy-3' fluor-thymidin (Formel I, X = O, R = CH3) durch Umsetzung von 2,3' -Anbydro-2' -desoxy-ss -D-xylofuranosylthymin mit HF in Dioxan in Gegenwart von Aluminiumfluorid als Katalysator hergestellt und anschliessend chromatographisch gereinigt.
Die als Ausgangsprodukt des Verfahrens verwendete Anhydro-Verbindung ist jedoch nur entweder in einer mit mässiger Ausbeute verlaufenden Mehrstufensynthese (Thymidine 5'-O-Trityl-thymidino 3'-O-Mesyl-5'-O-trityi-thymidino 2,3'-Anhydro-1-[2-desoxy-5-O-trityl-ss-D-xylofuranosyl]- thymin 2,3'-Anhydro-2'-desoxy-ss-D-xylofuranosylthymin) oder in einer Direktsynthese, die jedoch eine aufwendige Trennung erfordert (DWP 77.233), aus Thymidin zugänglich.
Deshalb sind der Anwendung des Verfahrens nach DWP 75.084 Grenzen gesetzt.
Der Erfindung liegt die Aufgabe zugrunde, Pyrimidin Nucleoside der Formel I ausgehend von gut zugänglichen Ausgangsstoffen herzustellen.
Erfindungsgemäss werden 2',3' -Didesoxy-3'-fluor-pyri- midin-nucleoside hergestellt, indem man 2',3'-Didesoxy-3' fiuor-5' -O-mesyl-pyrimidin-nucleoside der allgemeinen Formel:

worin X und R die oben genannten Bedeutungen haben und R' Methansulfonyl bedeutet, in alkalischem Milieu oder ein Alkalisalz der Verbindung der Formel II mit einem von einer organischen Carbonsäure abgeleiteten Acylierungsmittel behandelt, die entstandene Acylverbindung der allgemeinen Formel:

worin R2 eine von einer organischen Carbonsäure abgeleitete Acylgruppe bedeutet, isoliert und anschliessend die Acylgruppe abspaltet.
Gemäss einer ersten Verfahrensvariante wird so verfahren, dass man die Verbindung der Formel II mit wässrig-alkoholischer Natronlauge unter Rückfiuss behandelt, anschliessend neutralisiert und das nach Einengen im Vakuum erhaltene getrocknete Substanz-Salz-Gemisch mit Acetylierungsmitteln, vorzugsweise mit Acetanhydrid in Pyridin, behandelt, die gebildete 5'-O-Acetylverbindung (Formel III, R2 = COCH3) mit vorzugsweise Chloroform extrahiert, das Lösungsmittel entfernt und mit vorzugsweise methanolischem Ammoniak entacetyliert.
Eine weitere vorteilhafte Ausführungsvariante des Verfahrens besteht darin, die als Zwischenstufe zur Isolierung dienenden Fluornucleoside der allgemeinen Formel III, in der X und R die oben genannte Bedeutung haben und R2 einen Acetyl- oder Benzoylrest bedeuten, direkt aus den 5'-O-Mesyl-Nucleosiden der allgemeinen Formel II (R1 = Mesyl) durch Behandlung mit Alkaliacetaten oder -benzoaten, vorzugsweise mit Kaliumacetat, in einem geeigneten Lösungsmittel, vorzugsweise Dimethylformamid oder Acetanhydrid, bei erhöhter Temperatur herzustellen. Durch Entacylierung mit vorzugsweise methanolischem Ammoniak werden die gewünschten 2',3'-Didesoxy-3'-fluor-pyrimidin-nucleoside der Formel 1 erhalten.
Die als Ausgangsmaterial des Verfahrens eingesetzten Mesylverbindungen der Formel II sind nach DWP 83.142 aus den mesylierten Anhydroverbindungen in guter Ausbeute zugänglich. So wird 3'-Desoxy-3' -fiuor-5' -O-mesyl-thymidin (Formel II, X = O, R = CH3, R1 = Mesyl) aus 2,3'-Anhydro-l (2-desoxy-5-O-mesyl-ss-D-xylo-furanosyl)-thymin in 6 1pro- zentiger Ausbeute gewonnen.
Gegenüber der Umsetzung nach DWP 83.142 kann die erforderliche Lösungsmittelmenge um ca. 60% verringert und die Reaktionszeit um die Hälfte verkürzt werden, wenn die Reaktion in einem Schüttel(Rühr-)autoklav durchgeführt wird.
Die erfindungsgemässe Umsetzung beschreitet neue Wege, da Entmesylierungen an Verbindungen dieser Art bisher nicht bekannt sind und durch die Anwesenheit eines alkalilabilen Substituenten in 3'-Position (Fluor) bei der alkalischen Behandlung von Fluor-Nucleosiden der allgemeinen Formel II oder III eine Vielzahl von Nebenreaktionen zu befürchten waren. Überraschenderweise entstehen dabei jedoch praktisch nur Verbindungen der Formel I in guter Ausbeute, so dass keine ungewöhnlichen Reinigungsoperationen erforderlich sind. Es ist lediglich die Abtrennung vom gebildeten bzw. eingesetzten überschüssigen Salz notwendig, die entweder (analog DWP 75.084) durch Chromatographie an Kieselgel oder erfindungsgemäss durch Extraktion des $'-O-Acetyl-Derivates (Formel III, R2 = COCH3) mittels organischer Lösungsmittel, vorzugsweise Chloroform, erfolgt.
Die Acetylierung von Verbindungen der Formel I, Extraktion der Acetylverbindung und nachfolgende Entacetylierung verlaufen ohne Ausbeuteverluste.
Die Erfindung soll anschliessend an Ausführungsbeispielen näher erläutert werden.
Beispiel 1 3'-Desoxy-3'-fluor-thymidin (Formel I, X = O, R = CH3) a) Herstellung der Ausgangsverbindung
1 g 2' ,3'-Anhydro- 1-(2-desoxy-5-O-mesyl-ss-D-xylo- furanosyl)-thymin werden mit der gleichen Gewichtsmenge AIF3 in 200 ml einer 0,1prozentigen HF/Dioxan-Lösung suspendiert und in einem verschlossenen Stahlgefäss 1 Stunde lang in einem auf 1700 vorgeheizten Ölbad erhitzt. Man schüttelt während der Reaktionszeit mehrmals kräftig oder benutzt einen heizbaren Schüttelautoklav. Danach wird rasch abgekühlt, die Lösung mit 50 ml Wasser verdünnt und durch Zugabe von überschüssigem festem Calciumcarbonat neutralisiert. Dann wird vom festen Niederschlag abgetrennt und dieser mehrmals mit Aceton gewaschen.
Aus insgesamt 131 derartigen Ansätzen werden 163 g rohes 3'-Desoxy-3'-fluor-5'- O-mesyl-thymidin (Formel II, X = O, R = CH3, R1 = Mesyl) erhalten.
b) Umsetzung zu Verbindungen der Formel I (X= O,R=CH3)
Jeweils 50 g des nach a) hergestellten Rohproduktes werden in 3 Liter Äthanol gelöst, mit 600 ml 1n NaOH versetzt und 1 Stunde lang unter Rückfiuss erhitzt. Nach dem Erkalten wird mit 1n HCI neutralisiert. Die Lösung wird im Vakuum zur Trockne eingeengt. Der so aus den oben erwähnten 163 g Rohprodukt erhaltene Rückstand wird in 500 ml Pyridin gelöst und unter Rühren und Eiskühlung mit 50 ml Acetanhydrid versetzt. Man belässt 12 Stunden lang bei Raumtemperatur und engt danach im Vakuum ein. Der ölige Rückstand wird in 500 ml Wasser gelöst und in mehreren Portionen mit insgesamt 2,5 Liter Chloroform extrahiert. Die vereinten Extrakte werden im Vakuum zur Trockne eingeengt.
Der sirupöse Rückstand wird in 200 ml methanolischem Ammoniak (bei 0 gesättigt) gelöst und über Nacht bei Raumtemperatur aufbewahrt. Der nach Entfernen des methanolischen Ammoniaks verbleibende Rückstand (34 g) wird an 1 kg Kieselgel (0,05-0,2 mm Korngrösse) mit Chloroform/Methanol (98 :2, v/v) als Fliessmittel gereinigt. Nach Umkristallisieren aus Methanol erhält man 20,3 g 3'-Desoxy-3'-fluor-thymidin vom Smp. 173-174". Das Produkt ist mit authentischer Substanz identisch.
Beispiel 2 5'-O-Acetyl-3' -desoxy-3' -fluor-thymidin (Formel III, X = O, R = CH3, R2 = COCH3)
235 mg 5'-O-Mesyl-3'-desoxy-3'-fluor-thymidin (Formel II, X = O, R = CH3) werden mit der gleichen Gewichtsmenge trockenem Kaliumacetat in 30 ml Acetanhydrid 3 Stunden lang auf 135 erhitzt. Darauf wird im Vakuum zur Trockne eingeengt, der Rückstand in 50 ml Wasser gelöst und das Produkt durch dreimaliges Ausschütteln mit je 50 ml Chloroform extrahiert. Die Chloroform-Extrakte werden über Natriumsulfat getrocknet und danach im Vakuum eingeengt. Das Rohprodukt wird an 50 g Kieselgel H mit Chloroform-Methanol (95 :5) als Laufmittel gereinigt. Ausbeute 166 mg (80%).
Smp. 98-99" (Äthanol).
Entacetylierung dieses Produkts analog nach Beispiel 1 führt quantitativ zu 3'-Desoxy-3'-fluor-thymidin vom Smp.

Beispiel 3
2',3'-Didesoxy-3'-fluor-uridin (Formel I, X = O, R = H)
Analog Beispiel 1 werden aus 600 mg 2,3'-Anhydro-1 (2-desoxy-5-O-mesyl-ss-D-xylqfuranosyl)-uracil 90 mg 2',3' Didesoxy-3'-fluor-uridin erhalten, das bei Verreiben mit Essigester kristallisiert und daraus umkristallisiert wird. Smp.

It is known from DWP 75.084 to produce compounds of the formula I from the corresponding pyrimidine-2,3'-anhydro-2'-deoxynucleosides. So z. The anhydro compound used as the starting product of the process is, however, only either in a multi-stage synthesis proceeding with moderate yield (thymidine 5'-O-trityl-thymidino 3'-O-mesyl-5'-O-trityi-thymidino 2,3'- Anhydro-1- [2-deoxy-5-O-trityl-ss-D-xylofuranosyl] -thymine 2,3'-anhydro-2'-deoxy-ss-D-xylofuranosylthymine) or in a direct synthesis, which, however, is an expensive one Separation requires (DWP 77.233), accessible from thymidine.

According to a first variant of the process, the procedure is such that the compound of the formula II is treated with refluxing aqueous-alcoholic sodium hydroxide solution, then neutralized and the dried substance-salt mixture obtained after concentration in vacuo is treated with acetylating agents, preferably with acetic anhydride in pyridine, the 5'-O-acetyl compound formed (formula III, R2 = COCH3) is extracted with preferably chloroform, the solvent is removed and deacetylated with preferably methanolic ammonia.
A further advantageous variant of the process consists in that the fluoronucleosides of the general formula III, in which X and R are as defined above and R2 are acetyl or benzoyl radicals, used as intermediate for isolation, are obtained directly from the 5'-O-mesyl Nucleosides of the general formula II (R1 = mesyl) by treatment with alkali acetates or benzoates, preferably with potassium acetate, in a suitable solvent, preferably dimethylformamide or acetic anhydride, at elevated temperature. The desired 2 ', 3'-dideoxy-3'-fluoropyrimidine nucleosides of the formula 1 are obtained by deacylation with, preferably, methanolic ammonia.
The mesyl compounds of the formula II used as starting material for the process are obtainable in good yield from the mesylated anhydro compounds according to DWP 83.142. So 3'-deoxy-3'-fluor-5 '-O-mesyl-thymidine (formula II, X = O, R = CH3, R1 = mesyl) from 2,3'-anhydro-l (2-deoxy- 5-O-mesyl-ss-D-xylo-furanosyl) -thymine obtained in 6 1 percent yield.
Compared to the implementation according to DWP 83.142, the required amount of solvent can be reduced by approx. 60% and the reaction time shortened by half if the reaction is carried out in a shaking (stirring) autoclave.
The inventive reaction breaks new ground, since demesylations of compounds of this type are not previously known and, due to the presence of an alkali-labile substituent in the 3'-position (fluorine), a large number of side reactions in the alkaline treatment of fluorine nucleosides of the general formula II or III were to be feared. Surprisingly, however, practically only compounds of the formula I are formed in good yield, so that no unusual cleaning operations are necessary. It is only necessary to separate off the excess salt formed or used, which is either (analogous to DWP 75.084) by chromatography on silica gel or, according to the invention, by extraction of the $ '- O-acetyl derivative (formula III, R2 = COCH3) using organic solvents, preferably chloroform.
The acetylation of compounds of the formula I, extraction of the acetyl compound and subsequent deacetylation proceed without loss of yield.
The invention will then be explained in more detail using exemplary embodiments.
Example 1 3'-Deoxy-3'-fluoro-thymidine (Formula I, X = O, R = CH3) a) Preparation of the starting compound
1 g of 2 ', 3'-anhydro- 1- (2-deoxy-5-O-mesyl-ss-D-xylo-furanosyl) -thymine with the same amount by weight of AIF3 in 200 ml of 0.1 percent HF / dioxane Solution suspended and heated in a closed steel vessel for 1 hour in an oil bath preheated to 1700. Shake vigorously several times during the reaction time or use a heatable shaking autoclave. b) conversion to compounds of formula I (X = O, R = CH3)
In each case 50 g of the crude product prepared according to a) are dissolved in 3 liters of ethanol, 600 ml of 1N NaOH are added and the mixture is refluxed for 1 hour. After cooling, it is neutralized with 1N HCl. The solution is concentrated to dryness in vacuo. The residue obtained in this way from the above-mentioned 163 g of crude product is dissolved in 500 ml of pyridine, and 50 ml of acetic anhydride are added while stirring and while cooling with ice. It is left at room temperature for 12 hours and then concentrated in vacuo. The oily residue is dissolved in 500 ml of water and extracted in several portions with a total of 2.5 liters of chloroform. The combined extracts are concentrated to dryness in vacuo.
The syrupy residue is dissolved in 200 ml of methanolic ammonia (saturated at 0) and stored at room temperature overnight. The residue (34 g) remaining after removal of the methanolic ammonia is purified on 1 kg of silica gel (0.05-0.2 mm particle size) using chloroform / methanol (98: 2, v / v) as the flow agent. After recrystallization from methanol, 20.3 g of 3'-deoxy-3'-fluoro-thymidine with a melting point of 173-174 "are obtained. The product is identical to an authentic substance.
Example 2 5'-O-Acetyl-3 '-deoxy-3' -fluorothymidine (Formula III, X = O, R = CH3, R2 = COCH3)
235 mg of 5'-O-mesyl-3'-deoxy-3'-fluoro-thymidine (formula II, X = O, R = CH3) are heated to 135 for 3 hours with the same amount by weight of dry potassium acetate in 30 ml of acetic anhydride. It is then concentrated to dryness in vacuo, the residue is dissolved in 50 ml of water and the product is extracted by shaking out three times with 50 ml of chloroform each time. The chloroform extracts are dried over sodium sulfate and then concentrated in vacuo. The crude product is purified on 50 g of silica gel H using chloroform-methanol (95: 5) as the mobile phase. Yield 166 mg (80%).

Analogously to Example 1, 90 mg of 2 ', 3' dideoxy-3'-fluorouridine are obtained from 600 mg of 2,3'-anhydro-1 (2-deoxy-5-O-mesyl-ss-D-xylqfuranosyl) uracil which crystallizes when triturated with ethyl acetate and is recrystallized from it. M.p.
