Die vorliegende Erfindung bezieht sich auf neue Verfahren zur Herstellung von 1,2,3 ,4-Tetrahydroisochinolinen, welche auf die glatte Muskulatur eine erholende Wirksamkeit ausüben. Insbesondere bezieht sich die vorliegende Erfindung auf ein Verfahren zur Herstellung von 1,2,3,4-Tetrahydroisochinolinen der folgenden allgemeinen Formel:

worin R1 ein Wasserstoffatom oder einen niederen Alkylrest bedeutet, R'b die Hydroxylgruppe, die Aminogruppe, eine- Mono- oder Di-niederalkylaminogruppe, einen niederen Alkoxyrest, einen Arylrest, die Nitrogruppe, ein Halogenatom oder einen Halogen-niederalkylrest bedeutet und R'6 das Wasserstoffatom, die Hydroxylgruppe, die Aminogruppe, einen niederen Alkoxyrest oder ein Halogenatom darstellt, R, das Wasserstoffatom oder einen niederen Alkoxyrest darstellt und X das Sauerstoff- oder Schwefelatom darstellt, sowie ihre pharmazeutisch zulässigen Salze.
Die Bezeichnung Niederalkyl und Niederalkoxy bedeutet Reste mit 1 bis 6 Kohlenstoffatomen, während unter der Bezeichnung Halogen Chlor-, Brom-, Fluor- oder Jodatome in Frage kommen.
Geeignete Beispiele von niederen Alkoxyresten sind niedere Alkoxyreste mit 1 bis 4 Kohlenstoffatomen wie z.B.
die Methyl-, Äthyl-, Propyl-, Isopropoyl-, Butyl-, Isobutyl-, tert.-Butyl-, Pentyl- oder Hexylreste, und vorzugsweise niedere Alkylreste mit 1 bis 4 Kohlenstoffatomen. Am liebsten verwendet man niedere Alkylreste mit 1 bis 2 Kohlenstoffatomen.
Geeignete Beispiele von niederen Alkoxyresten sind niedere Alkoxyreste mit 1 bis 6 Kohlenstoffatomen, z.B. die Methoxy-, Äthoxy-, Propoxy-, Isopropoxy-, Butoxy-, Isobutoxy-, tert.-Butoxy-, Pentoxy- Isopentoxy- oder Hexyloxyreste, und vorzugsweise niedere Alkoxyreste mt 1 bis 4 Kohlenstoffatomen. Insbesondere wird man niedere Alkoxyreste mit 1 bis 2 Kohlenstoffatomen bevorzugen.
Geeignete Beispiele für Halogen-niederalkylreste sind Halogen-niederalkylreste mit 1 bis 6 Kohlenstoffatomen, wie z.B. die Chlormethyl-, Fluoräthyl-, Chlorpropyl-, Brompropyl-, Jodbutyl-, Chlorpentyl-, Bromchloräthyl-, Dichlormethyl-, Dichloräthyl-, Dibrommethyl-, Dibromäthyl-, Difluormethyl-, Dichlorpropyl-, Dichlorbutyl-, Dibrompropyl-, Difluorpropyl-, Trichlormethyl-, Tribrommethyl-, Trifluormethyl-, Trichloräthyl- oder Tribromäthylreste, und insbesondere wird man eine Halogen-niederalkylgruppe mit 1 bis 4 Kohlenstoffatomen verwenden. Insbesondere bevorzugt man Halogen-niederalkylreste mit 1 bis 2 Kohlenstoffatomen.
Als geeignete Arylreste kann man beispielsweise Phenyl-, Tolyl- oder Xylylreste nennen.
Die erfindungsgemäss erhältlichen 1,2,3,4-Tetrahydroisochinoline der Formel I und deren pharmazeutisch zulässigen Salze besitzen eine besänftigende Wirkung auf die glatte Muskulatur, insbesondere auf die glatte Gefässmuskulatur und die glatte Eingeweidemuskulatur. Demzufolge besitzen sie vasodilatierende Wirkungen, hemmende Wirkung auf die Darmkontraktion und hemmende Wirkung auf die Gallenblasekontraktion, besitzen keine bronchodilatierende Wirkung und sind daher wertvoll als vasodilatierende Mittel, Hemmittel für die intenstinale Kontraktion und für die Gallenkontraktion.
Das erfindungsgemässe Verfahren besteht darin, dass man ein in l-Stellung substituiertes 6,7-Dihydroxy-1,2,3,4 -tetrahydroisochinolinderivat der Formel:

worin Rt, R, und X die obigen Bedeutungen haben, jedes der Symbole R' und R" ein Wasserstoffatom oder eine Schutzgruppe für die Hydroxylgruppe bedeutet, R4 eine Schutzgruppe für die Iminogruppe darstellt, R5 die Hydroxylgruppe, eine geschützte Hydroxylgruppe, die Aminogruppe, eine geschützte Aminogruppe, eine Mono- oder Di-niederalkylaminogruppe, einen niederen Alkoxyrest, einen Arylrest, die Nitrogruppe, ein Halogenatom oder eine Halogen-niederalkylgruppe bedeutet und R6 das Wasserstoffatom, die Hydroxylgruppe, eine geschützte Hydroxylgruppe, die Aminogruppe, eine geschützte Aminogruppe, einen niederen Alkoxyrest oder ein Halogenatom darstellt,

Die als Ausgangsverbindungen verwendeten, neuen in l-Stellung substituierten 6,7-Dihydroxy-1,2,3,4-tetrahydro- isochinolinderivate der Formel IV können dadurch erhalten werden, dass man eine Verbindung der Formel:

In den obigen Verbindungen der Formeln IV, V und VI kann man als einzelne Schutzgruppen für die Hydroxylgruppe beispielsweise leicht entfernbare Acylgruppen verwenden, wie z.B. der Acetyl-, Benzyloxycarbonyl-, 4-Nitrobenzyloxy- carbonyl-, 4-Brombenzyloxycarbonyl-, 4-Methoxybenzyloxycarbonyl-, 3 ,4-Dimethoxybenzyloxycarbonyl-, 4-(Phenylazo)-benzyloxycarbonyl-, 4-(4-Methoxyphenylazo)-benzyloxycarbonyl-, tert.-Butoxycarbonyl-, l,l-Dimethylpropoxy- carbonyl-, Isopropoxycarbonyl-, Diphenylmethoxycarbonyl-, 2-Pyridylmethoxycarbonyl-, 2,2,2-Trichloräthoxycarbonyl-, 2,2,2-Tribromäthoxycarbonyl-, 3-Jodpropoxycarbonyl-, 2-Furfuryloxycarbonyl-, l-Adamantyloxycarbonyl-, l-Cyclopro- pyläthoxycarbonyl-, 8-Chinolyloxycarbonyl- und Trifluoracetylrest, femer ein Benzyl-, Trityl-, Methoxymethyl-,
2-Ni trophenylthio-, Tetrahydropyranyl-, 2,4-Dinitrophenylthiorest usw. Als geeignete Schutzgruppe für die Hydroxylgruppen R' und R" kommen beispielsweise Alkylengruppen, z.B.
Methylen-, Äthylen- und Dimethylmethylenreste in Frage, welche gebildet werden, wenn R' und R" miteinander verbunden sind.
Als geeignete Schutzgruppe für Amino- und Iminogruppen kommen beliebige Schutzgruppen für Amino- und/oder Iminogruppen in Frage. Beispiele sind leicht entfernbare Acylgruppen, wie z.B. eine Trichloräthoxycarbonyl-, Tribromäthoxycarbonyl-, Benzyloxycarbonyl-, p-Toluolsulfonyl-, p-Nitrobenzyloxycarbonyl-, o-Brombenzyloxycarbonyl-, o-Nitrophenylsulfenyl-, Acetyl-, Chloracetyl-, Trifluoracetyl-, Formyl-, tert.-Butoxycarbonyl-, p-Methoxybenzyloxycarbonyl-, 3 ,4-Dimethoxybenzyloxycarbonyl-, 4-(Phenylazo)-benzyloxycarbonyl-, 4- (4-Methoxyphenylazo) -benzyloxycarb- onyl-, Pyrdin-l-oxyd-2-methoxycarbonyl-, 2-Pyridylmethoxy carbonyl-, 2-Furyloxycarbonyl-, Diphenylmethoxycarbonyl-, l,l-Dimethylpropoxycarbonyl-. Isopropoxycarbonyl-, l-Cy clopropyläthoxycarbonyl-,

1 -[N-(4-Methoxvphenyl > -carb- amoyl]-2-propyliden-, 2-Äthoxycarbonylcyclohexyliden-, 2-Äthoxycarbonylcyclopentyliden-, 2-Acetylcyclohexyliden-, 3,3-Dimethyl-5-oxocyclohexyliden- und Di- oder Trialkylsilylgruppe.
Die Abspaltung der Schutzgruppen kann in an sich bekannter Weise durchgeführt werden, beispielsweise durch Hydrolyse mit einer Säure oder einem Alkali oder durch katalytische Hydrierung, je nach der Art der Schutzgruppen.
Unter den üblichen Methoden wird man vorzugsweise zur Hydrolyse mit einer Säure schreiten. Dieser Fall wird vor allem im Falle von Schutzgruppen, wie z.B. Benzyloxycarbonyl-, substituierten Benzyloxycarbonyl-, Alkoxycarbonyl-, substituierten Alkoxycarbonyl-, Adamantyloxycarbonyl-, Toluolsulfonyl-, Trityl-, Methoxymethyl-, substituierten Phenylthiogruppen und niederen Alkylengruppen, welche dadurch gebildet werden, dass die Symbole R' und R" miteinander verbunden werden, eintreten. Die geeignetsten Beispiele für die Hydrolyse verwendbaren Säuren sind Bromwasserstoffsäure, Salzsäure, Ameisensäure, Essigsäure, Trifluoressigsäure und dergleichen, d.h. Säuren, welche sich leicht unter vermindertem Druck aus dem Reaktionssystem abdestillieren lassen.
Die Hydrolyse kann ohne Lösungsmittel oder in einem geeigneten Lösungsmittel, z.B. in einem hydrophilen organischen Lösungsmittel, Wasser oder einem Mischlösungsmittel davon, durchgeführt werden.
Die katalytische Hydrierung erfolgt im Falle von Schutzgruppen, wie z.B. Benzyloxycarbonylgruppen, substituierten Benzyloxycarbonyl-, 2-Pyridylmethoxycarbonyl-, Diphenylmethoxycarbonyl-, Benzyl- oder Tritylgruppen. Besonders geeignet als Katalysator ist Palladiumkatalysator, doch kann man auch andere Katalysatoren verwenden. Sofern R5 die Nitrogruppe darstellt, kann die Hydrierung auch die- sen Rest in eine Aminogruppe überführen, wobei auch diese Reduktion zusammen mit der soeben genannten Nebenreaktion unter die vorliegende Erfindung fällt.
Man kann auch andere übliche Methoden für die Beseitigung der Schutzgruppen anwenden. So kann man beispielsweise Trifluoracetylgruppen durch Behandlung der so geschützten Verbindungen mit Wasser, halogensubstituierte Alkoxycarbonylreste und 8 -Chinolyloxycarbonylreste durch Behandeln der geschützten Gruppe mit einem Schwermetall, z.B. Kupfer oder Zink, und andere Methansulfonyl-, Toluolsulfonylreste und dergleichen durch Behandeln der geschützten Verbindung mit metallischem Natrium in flüssigem Ammoniak entfernen.
Die Ausgangsverbindungen der Formel V, welche neue Verbindungen darstellen, können beispielsweise dadurch erhalten werden, dass man 4-(2-Aminoäthyl)-catechol oder ein Derivat davon mit einem entsprechenden substituierten Essigsäurechlorid umsetzt und das erhaltene Produkt in Gegenwart von Phosphoroxychlorid kondensiert und das dann erhaltene Produkt reduziert.
In den obigen Verbindungen der Formeln V und VI können die einzelnen Schutzgruppen für die Hydroxylgruppe die bereits erwähnten sein. Geeignete Schutzgruppen für Amino- und Iminogruppen sind solche der oben erwähnten Art. Als geeignete Reste einer Säure Y kann beispielsweise ein Rest einer Säure, z.B. Salzsäure, Schwefelsäure, Bromwasserstoffsäure, Jodwasserstoffsäure, einer Alkylschwefelsäure, Toluolsulfonsäure, Benzolsulfonsäure und einer Dialkylcarbaminsäure, in Frage kommen.
Als geeignetes Salz einer Verbindung der Formel V kann man ein Salz einer anorganischen Säure, z.B. ein Chlorhydrat, Bromhydrat oder Sulfat, oder einer organischen Säure.
z.B. einem Maleat oder Lactat, nennen.
Die Umsetzung der Verbindungen der Formel V oder eines Salzes davon mit einer Verbindung der Formel VI erfolgt vorzugsweise in Gegenwart einer Base, z.B. eines Alkali- oder Erdalkalimetalles, wie z.B. Natrium, Kalium, Magnesium oder Calcium, oder eines Hydroxyds, Alkoxyds, z.B. Äthoxyds, Propoxyds oder tert.-Butoxyds, eines Carbonats oder Bicarbonats davon, oder in Gegenwart einer starken organischen Base, z.B. Pyridin. Die Umsetzung kann in Gegenwart oder in Abwesenheit eines geeigneten Lösungsmittels, z.B. eines niederen Alkohols, wie z.B. Methanol oder Äthanol, eines Äthers, Benzol, Aceton, Dioxan, Acetonitril, Chloroform, Athylenchlorid, Tetrahydrofuran, Äthylacetat oder Pyridin, oder in Gegenwart eines anderen üblichen Lösungsmittels, welches für die Reaktion inert ist, durchgeführt werden. Bezüglich der Reaktionstemperatur sind keine Schranken gesetzt.
Man kann bei beliebigen Temperaturen zwischen Zimmertemperatur und erhöhten Temperaturen arbeiten.
Die erhaltenen Verbindungen I' können in Salze übergeführt werden, indem man sie mit einer Säure, z.B. einer anorganischen Säure, wie z.B. Salzesäure oder Schwefelsäure, oder einer organischen Säure, z.B. Essigsäure, Weinsäure oder Picrinsäure, umsetzt.
Die Erfindung sei durch die folgenden Beispiele erläutert Bemerkung: Die Eigenschaften der Produkte in der folgenden
Tabelle 1 entsprechen dem Produkt in Form des Hydrobro mids für das Beispiel 20 und den Produkten in Form des Chlor hydrates für die übrigen Beispiele.
Beispiel I
Ein Gemisch von 1- (4-Chlorphenyl)-oxymethyl-2-acetyl- -6,7-diacetoxy- 1,2,3 ,4-tetrahydroisochinolin (450 mg), konz.
Salzsäure (10 ml) und 95%igem Äthanol (15 ml) wird während 16 Stunden unter Rückfluss zum Sieden erhitzt. Diesem Reaktionsgemisch gibt man 20 ml Wasser und 100 ml Äthylacetat hinzu, worauf man die wässerige Schicht abtrennt und unter vermindertem Druck zur Trockne eindampft. Der entstandene Rückstand wird in Isopropylalkohol gelöst und die Lösung dann mit Äther versetzt. Das Gemisch wird stehen gelassen. Der entstandene Niederschlag wird durch Dekantieren abgetrennt und getrocknet, wobei man 1-(4-Chlorphenyl)-oxymethyl-6,7-dihydroxy-1,2,3,4-te- trahydroisochinolinhydrochlorid (150 mg), Smp. 2260C, erhält.
Beispiel 2
Ein Gemisch von 1-(4-Chlorphenyl)-oxymethyl-2-acetyl- -6,7-dihydroxy-l ,2,3,4-tetrahydroisochinolin (720 mg), konz.
Salzsäure (20 ml) und Methanol (100 ml) wird während 16 Stunden unter Rückfluss zum Sieden erhitzt. Dann wird das Reaktionsgemisch unter vermindertem Druck zur Trockne eingedampft. Der entstandene Rückstand wird mit Äthylacetat und Chloroform gewaschen und hierauf in Isopropanol gelöst. Dieses Gemisch wird mit Äther versetzt und das Gemisch hierauf stehen gelassen. Die ausgefällten Kristalle werden durch Filtrieren abgetrennt und getrocknet, wobei man 1 -(4-Chlorphenyl)-oxymethyl-6,7-dihydroxy-1,2,3 ,2,3,4-tetrahydroisochinolin-hydrochlorid (420 mg), Smp. 226 C, erhält.
Beispiel 3
In Methanol (40 ml) löst man 1-(4-Benzyloxyphenyl)-oxy- methyl-2-benzyl-6,7-dibenzyloxy-1 2,3 ,4-tetrahydroisochinolin (500 mg), worauf man 10%iges Palladium-auf-Kohle (200 mg) und konz. Salzsäure (2 ml) zugibt. Dann wird das Gemisch bei Zimmertemperatur und bei Atmosphärendruck einer katalytischen Reduktion unterworfen. Nach dem Abbrechen der Wasserstoffabsorption wird das Reaktipnsge misch fiItriert. Das Filtrat wird hunter vermindertem Druck zur Trockne eingedampft. Der erhaltene Rückstand wird aus einer Mischung von Methanol und Äther umkristallisiert, wobei man 1-(4Hydroxyphenyl)-oxymethyl-6,7-dihydroxy- -I,Z,3,4-tetrahydroisochinolin-hydrochlorid (160 mg), Smp.262 C, erhält.

Arbeitet man in der gleichen Weise wie in den Beispielen 1 bis 3, so kann man verschiedene Verbindungen herstellen.
Diese Verbindungen finden sich in der nachstehenden Tabelle 1.

The present invention relates to new processes for the production of 1,2,3,4-tetrahydroisoquinolines which have a restorative effect on the smooth muscles. The designations lower alkyl and lower alkoxy denote radicals with 1 to 6 carbon atoms, while chlorine, bromine, fluorine or iodine atoms are possible under the designation halogen.
Suitable examples of lower alkoxy groups are lower alkoxy groups having 1 to 4 carbon atoms, e.g.
the methyl, ethyl, propyl, isopropyl, butyl, isobutyl, tert-butyl, pentyl or hexyl radicals, and preferably lower alkyl radicals having 1 to 4 carbon atoms. It is best to use lower alkyl radicals with 1 to 2 carbon atoms.
