In den erfindungsgemäss erhältlichen Verbindungen können die Hydroxygruppen zu den entsprechenden Estern verestert werden und als Acylreste kommen hierzu solche Reste in Frage, die in der Steroidchemie gebräuchlicherweise für Veresterungen angewandt werden. Bevorzugte Säuren sind solche mit bis zu 15 Kohlenstoffatomen, insbesondere niedere und mittlere aliphatische Carbonsäuren. Die Säuren können aber auch ungesättigt, verzweigt, mehrbasisch oder in üblicher Weise z. B. durch Hydroxy-, Oxo- oder Aminogruppen oder Halogenatome substituiert sein. Geeignet sind auch cycloaliphatische, aromatische, gemischt aromatischaliphatische oder heterocyclische Säuren, die ebenfalls in geeigneter Weise substituiert sein können. Solche Säuren sind z. B.
Ameisensäure, Essigsäure, Propionsäure, Buttersäure, Valeriansäure, Capronsäure, Önanthsäure, Undecylsäure, Trimethylessigsäure, Diäthylessigsäure, tert.-Butylessig- säure, Phenylessigsäure, Cyclopentylpropionsäure, Ölsäure, Milchsäure, Mono-, Di- und Trichloressigsäure, Aminoessigsäure, Diäthylamino-, Piperidino- und Morpholinoessigsäure, Bernsteinsäure, Adipinsäure, Benzoesäure, Nicotinsäure.
Die erfindungsgemäss herstellbaren Verbindungen sind entweder selbst wertvolle Arzneimittel oder dienen als Zwischenprodukte zur Herstellung derselben. Sie haben im Prinzip dieselben pharmakologischen Eigenschaften wie die entsprechenden 6-Desoxysteroide, jedoch meistens mit wesentlich verstärkter Wirksamkeit.
Insbesondere zeigt das 3,17ss-Dihydroxy-17a-äthinyl- 1,3,5(10pöstratrien-6-on im s.c. Allen-Doisy-Test zwar die gleiche östrogene Wirksamkeit wie das bekannte Äthinylöstradiol, jedoch mit einer um das Zehnfache verringerten ovulationshemmenden Wirkung.
Die erfindungsgemäss erhältlichen Verbindungen können in der Human- und Veterinärmedizin immer dann appliziert werden, wenn allein die östrogene Wirksamkeit erwünscht ist. Die erfindungsgemäss herstellbaren Verbindungen, insbesondere das 3,17p-Dihydroxy-17a-äthinyl-1,3,5(10)-östratrien- 6-on, sind somit vorteilhaft zur Behandlung von Krankheiten bzw. krankhaften Erscheinungen geeignet, bei denen Östrogene gemeinhin, gegebenenfalls in Kombination mit Gestagenen, angewendet werden, wie z. B. beim Klimakterium und dessen Folgeerkrankungen.
Die erfindungsgemäss erhältlichen Verbindungen können oral oder parenteral appliziert werden. Zum therapeutischen Gebrauch werden sie mit den in der galenischen Pharmazie entsprechenden üblichen Zusätzen, Trägerstoffen und Geschmackskorrigentien zu den üblichen Arzneimittelformen verarbeitet. Für die orale Applikation kommen insbesondere Tabletten, Dragees, Kapseln, Pillen, Suspensionen oder Lösungen in Frage. Für die parenterale Applikation kommen insbesondere ölige Lösungen in Frage, wie z. B. Sesamöloder Rhizinusöllösungen, die gegebenenfalls noch zusätzlich ein Verdünnungsmittel, wie Benzylbenzoat oder Benzylalkohol, enthalten können.
Die Konzentration der genannten Verbindungen in der zu applizierenden Arzneiform ist abhängig von der Applikationsform. So enthalten Tabletten 10-200 y, vorzugsweise etwa 20 , und ölige Lösungen zur i.m. Injektion 25-400 y in einem ml, vorzugsweise etwa 50 ylml.
Methoden zur Aromatisierung des A-Ringes von Steroiden sind aus der Literatur bekannt. So kann beispielsweise 3-Keto-A5(10 > -19-nor-androsten mit einer Persäure zur entsprechenden 5,10-Epoxyverbindung epoxydiert werden, die dann mit Alkalien zum 10ss-Hydroxy-A4-3-keto-19-nor-androstan und mit Säuren in 3-Hydroxy-1,3,5(10)-östratrien umgewandelt wird [J. Org. Chem. 23(1958)1744]. Die Aromatisierung lässt sich aber auch in einem Schritt mit starken Säuren ausführen, wenn man von ,-disubstituierten 3-Keto-19-nor-steroiden ausgeht (DT AS 1 223 379).
Des weiteren sind auch Methoden zur Einführung einer 6-Ketogruppierung in der Östranreihe seit langem bekannt.
Hierzu wird 3,17ss-Diacetoxy-1,3,5(10)-östratrien unter milden Bedingungen mit Chromsäure oxydiert [J. Biol. Chem. 133 (1940) 219].
Es wurde nun gefunden, dass sich die Aromatisierung des Ringes A unter gleichzeitiger Oxydation in 6-Stellung erfindungsgemäss überraschend einfach ausführen lässt, wenn man ein 3-Keto-A4-Steroid der Formel II

oder ein im Steroidgerüst substituiertes Derivat davon, in welcher Formel Rt und R2 die obenangegebene Bedeutung haben, in einem hochsiedenden polaren Lösungsmittel mit basischen Reagenzien in Gegenwart von Sauerstoff bei Temperaturen von 70-150 "C behandelt. Freie Hydroxygruppen können anschliessend in an sich bekannter Weise verestert werden.
Als hochsiedende polare Lösungsmittel seien beispielsweise genannt Dimethylformamid, Sulfolan, Dimethylsulfoxid, N-Methylpyrrolidin u. a.
Als basische Reagenzien seien die Alkalisalze schwacher Säuren, wie Essigsäure und Kohlensäure, wie z. B. Kaliumacetat oder Natriumcarbonat, und von Alkoholen, wie z. B. Kalium-tritert.-butylat, genannt.
Der Sauerstoff kann in Form von Luftsauerstoff oder als reiner Sauerstoff in das Reaktionsgemisch eingebracht werden.
Der Temperaturbereich der erfindungsgemässen Reaktion liegt, wie gesagt, zwischen 70 und 150 C. Höhere Temperaturen sind unzweckmässig, da sie zu Nebenreaktionen führen. Tiefere Temperaturen sind im Prinzip zwar möglich, jedoch muss man dann mit unangemessen langen Reaktionszeiten rechnen.
Die sich gegebenenfalls anschliessende Veresterung freier Hydroxygruppen geschieht gewöhnlich nach Methoden, wie sie dem Fachmann allgemein bekannt sind. Beispielsweise genannt sei die Veresterung mit Säurehalogeniden oder Säureanhydriden in Gegenwart von starken Säuren, wie Salzsäure, Schwefelsäure und p-Toluolsulfonsäure oder in Gegenwart von basischen Katalysatoren, wie Pyridin und Kaliumhydrogencarbonat.
Die als Ausgangsmaterial verwendeten Steroide der Formel II können darüber hinaus auch noch in üblicher Weise substituiert sein, ohne dass die Substituenten einen Einfluss auf die erfindungsgemässe Reaktion hätten. So kann das Steroidgerüst beispielsweise Alkylgruppen, vorzugsweise Methyl, in 15- oder 16-Stellung besitzen. Des weiteren ist nicht ausgeschlossen, dass Sauerstoffunktionen, wie eine Ketogruppe in 11-Stellung oder Hydroxygruppen in 11- und/oder 16-Stellung, vorhanden sind.
Beispiel 1
2 g 17ss-Hydroxy-17α-äthinyl-4-östren-3-on werden unter Einleiten von Luftsauerstoff mit 2 g wasserfreiem Kaliumacetat 6 Stunden bei 100 C gerührt. Das Reaktionsprodukt wird dann in Eiswasser eingerührt. Den Niederschlag saugt man ab, wäscht mit Wasser nach und nimmt in Methylenchlorid auf. Die Lösung wird über Natriumsulfat getrocknet.
Nach Chromatographieren an Kieselgel und Umkristallisation aus Aceton/Hexan werden 450 mg 3,17ss-Dihydroxy-17a- äthinyl-1,3,5(10)-östratrien-6-on vom Schmelzpunkt 224-226 C erhalten.

2 g 17ss-Acetoxy-17α-äthinyl-4-östren-3-on werden unter Einleiten von Luftsauerstoff mit 2 g wasserfreiem Natriumacetat 6 Stunden bei 100 C in 20 ml Dimethylformamid gerührt. Nach Fällung in Eiswasser und Aufarbeitung analog Beispiel 1 wird an Kieselgel mit 13,5-16,5% Aceton/Hexan chromatographiert. Es werden 1,1 g 3-Hydroxy-17ss-aceto- xy-17a-äthinyl-1,3,5(1 0)-östratrien-6-on vom Schmelzpunkt 192-193 C erhalten.

2 g 17ss-Acetoxy-4-östren-3-on werden in 15 ml Dimethylsulfoxid mit 1 g wasserfreiem Kaliumacetat unter langsamen Einleiten von reinem Sauerstoff 4 Stunden bei 120 C gerührt. Nach Fällung in Eiswasser und Aufarbeitung analog Beispiel 1 wird an Kieselgel mit 11,7-17% Aceton/Hexan chromatographiert. Es werden 466 mg 3-Hydroxy-17ss-aceto- xy-1,3,5(10)-östratrien-6-on vom Schmelzpunkt 276-278 C (Zersetzung) erhalten.

2 g 17-Acetoxy-19-nor-4-pregnen-3,20-dion werden in 40 ml Dimethylformamid mit 1 g wasserfreiem Kalimacetat 24 Stunden unter Einleiten von Luftsauerstoff bei 120 C gerührt. Nach Fällung in Eiswasser und Aufarbeitung analog Beispiel 1 wird das Rohprodukt in 8 ml Pyridin mit 4 ml Acetanhydrid bei Raumtemperatur acetyliert. Die Reaktionslösung wird in Eiswasser eingerührt. Der Niederschlag wird abgesaugt, mehrmals mit Wasser gewaschen und in Methylenchlorid aufgenommen.
Nach Chromatographieren an Kieselgel mit 15,5-17,5% Aceton/Hexan werden 446 mg 3,17-Diacetoxy 19nor-l ,3,5(10)-pregnatrien-6,20-dion vom Schmelzpunkt 263-265 C isoliert.

3 g 17ss-Hydroxy-18-methyl-17a-äthinyl-4-östren-3-on werden in 60 ml Dimethylformamid mit 1,5 g wasserfreiem Kaliurn- acetat unter Einleiten von Sauerstoff 8 Stunden bei 120 C gerührt.
Nach Fällung in Eiswasser und Aufarbeitung analog Beispiel 1 erhält man nach Chromatographieren an Kieselgel mit 20-25% Aceton/Hexan 1,5 g 3,17ss-Dihydroxy-18-met- hyl-17α-äthinyl-1,3,5(10)-östratrien-6-on als Schaum.

1,4 g 3,17ss-Dihydroxy-18-methyl-17a-äthinyl-1,3,5(10)-östra- trien-6-on werden in 6 ml Pyridin mit 3 ml Acetanhydrid innerhalb einer Stunde bei Raumtemperatur acetyliert. Die Reaktionslösung wird dann in Eiswasser eingerührt. Der Niederschlag wird abgesaugt, mit Wasser gewaschen, in Methylenchlorid aufgenommen und über Natriumsulfat getrocknet. Nach Umkristallisation des Rohproduktes aus Aceton Hexan werden 1,3 g 17ss-Hydroxy-3-acetoxy-18-methyl-17α-ät- hinyl- 1,3,5,(10)-östratrien-6-on vom Schmelzpunkt 216-218 C erhalten.
