Die Erfindung betrifft ein Verfahren zur Herstellung von 2,4-Diamino-thiophen-3,5-dicarbonsäurenitril aus Malonsäuredinitril und ist dadurch gekennzeichnet, dass man in einer ersten Stufe Malonsäuredinitril dimerisiert und das dimere Malonsäuredinitril in einer zweiten Stufe durch Behandlung mit elementarem Schwefel in 2,4-Diamino-thiophen-3,5dicarbonsäurenitril überführt.
Zweckmässig wird sowohl die Dimerisierung als auch die Umsetzung des dimeren Malonsäuredinitrils in einem geeigneten Lösungsmittel durchgeführt. Solche Lösungsmittel sind beispielsweise Wasser, Alkohole, wie Methanol, Äthanol, Butanol, Ispropanol, Methylglykol, Äthylglykol, Isopropylglykol, ferner aromatische Kohlenwasserstoffe, wie Benzol, Toluol, Xylol, Chlorbenzol, Dioxan.
Vorzugsweise wird die Umsetzung des dimeren Malonsäuredinitril mit elementarem Schwefel in Gegenwart geringer Mengen einer Base, nämlich in Mengen von 0,7 molar bis 3 molar, bezogen auf das dimere Malonitril, durchgeführt. Solche Basen sind beispielsweise Diäthylamin, Morpholin, Äthylendiamin, Triäthylendiamin, Piperidin usw.
Als besonders günstig hat es sich erwiesen, sowohl die Dimerisierung als auch die Umsetzung der zweiten Stufe im gleichen Lösungsmittel durchzuführen. Dabei kann zweckmässig so vorgegangen werden, dass das Dimer, ohne es zu isolieren, direkt weiter mit Schwefel umgesetzt wird.
Die Reaktionszeiten für die zweite Stufe liegen je nach Reaktionstemperatur, die zwischen 20 und 800C liegen kann, bei 1 bis 90 Minuten.
Die erzielten Ausbeuten, bezogen auf Malondinitril, hängen von der Art des Lösungsmittels ab. Die höchsten Ausbeuten werden mit Alkoholen als Lösungsmittel erzielt. Wird Methanol verwendet, liegen die Ausbeuten bei etwa 80% der Theorie.

Das 2,4-Diamino-thiophen-3 ,5-dicarbonsäurenitril kristallisiert in nahezu farblosen Nadeln vom Schmelzpunkt 2500C.
Durch saures Verseifen erhält man das 2,4-Diamino-thiophen3,5-dicarbonsäureamid.
Durch Kondensation mit aromatischen Aldehyden werden Monoazomethine des Thiophens erhalten. Solche Kondensationen gelingen beispielsweise mit Benzaldehyd, p-Dimethylaminobenzaldehyd, p-Chlorbenzaldehyd, Phthaldialdehyd und anderen.
Das Kondensationsprodukt der Verbindung der Erfindung mit p-Dimethylaminobenzaldehyd führt zu einem tiefgefärbten Körper mit einem X max von 460 nm.
Beispiel 1
Dimeres Malonitril wurde nach der Vorschrift von E. C. Taylor und K. S. Hartke, J. Amer. hem. Soc. 81, 2454 (1959) hergestellt. 5,8 gmetallisches Natrium wurden unter Kühlung in 105 ml mindestens 98%igem Äthanol gelöst und unter weiterer Kühlung und unter Rühren eine Lösung von 33,0 g Malonitril gelöst in 50 ml Äthanol eingetragen. Danach wurde eine Stunde zum Sieden erhitzt und nach dem Erkalten das ausgeschiedene Na-Salz des dimeren Malonitrils abfiltriert.
Dieses wurde bei 800C getrocknet und in möglichst wenig Wasser gelöst. Diese Lösung wurde mit HCI konz. angesäuert (mindestens pH 4), das ausgeschiedene Produkt abgesaugt und mit Wasser gewaschen. Umkristallisation aus Wasser. Schwach gefärbte Nadeln vom Smp. 173-1750C. Ausbeute 26,0 g = 79% d. Th.
6,6 g (0,05 Mol) dimeres Malonitril und 1,6 g Schwefel wurden in 40 ml Äthanol unter Rühren aufgeschlämmt. Dazu tropfte man 3 ml Diäthylamin und rührte noch 90 Minuten bei Raumtemperatur weiter. Das ausgefallene Produkt wurde abgesaugt, mit Wasser gewaschen und aus Wasser umkristallisiert.
Farblose Nadeln vom Smp. 250OC, Ausbeute 6,1 g = 75% d. Th.

Beispiel 2
1,1 g metallisches Natrium (0,05 Mol) wurden in 20 ml absolutem Äthanol gelöst. Zu dieser Lösung gab man unter Rühren eine Lösung von 6,6 g (0,1 Mol) Malonitril in 10 ml Äthanol. Danach wurde eine Stunde unter Rückfluss zum Sieden erhitzt. Nach dem Erkalten wurde mittels eines Kühlgemisches auf 5 C weiter gekühlt und die Lösung mit 4,2 ml HCI konz. versetzt. In diese nun neutrale Lösung wurden 3,2 g Schwefel und 3 ml Diäthylamin eingetragen und 90 Minuten bei Raumtemperatur gerührt. Der ausgefallene Niederschlag wurde abfiltriert, mit Wasser gewaschen und getrocknet.
Ausbeute 4,2 g = 43 % d. Th., Smp. 2480C.
PATENTANSPRUCH
Verfahren zur Herstellung von 2,4-Diamino-thiophen3,5-dicarbonsäurenitril aus Malonsäuredinitril, dadurch gekennzeichnet, dass man in einer ersten Stufe Malonsäuredinitril dimerisiert und das dimere Malonsäuredinitril in einer zweiten Stufe durch Behandeln mit elementarem Schwefel in 2,4-Diamino-thiophen-3,5-dicarbonsäurenitril überführt.
UNTERANSPRÜCHE
1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man die Dimerisierung und die Umsetzung des Dimers mit elementarem Schwefel im gleichen Lösungsmittel durchführt.
2. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man die Umsetzung des Dimers mit elementarem Schwefel ohne das Dimer nach der Dimerisierung zu isolieren durchführt.

The 2,4-diamino-thiophene-3,5-dicarboxamide is obtained by acidic saponification.
Monoazomethines of thiophene are obtained by condensation with aromatic aldehydes. Such condensations succeed, for example, with benzaldehyde, p-dimethylaminobenzaldehyde, p-chlorobenzaldehyde, phthaldialdehyde and others.

1.1 g of metallic sodium (0.05 mol) was dissolved in 20 ml of absolute ethanol. A solution of 6.6 g (0.1 mol) of malonitrile in 10 ml of ethanol was added to this solution with stirring.
