Die Erfindung betrifft ein Verfahren zur Herstellung von neuen l-Phenoxy-2-hydroxy-3-amino-propanen der Formel I

worin Ri ein Zuckerrest ist und R2 Isopropyl, tert.-Butyl oder im Phenylteil gegebenenfalls substituiertes a-Methylphen äthyl ist, sowie deren Salzen.
Ein Zuckerrest R1 ist z. B. ein Pentosidylrest oder ein Hexosidylrest, insbesondere ein Aldopentosidylrest oder ein Aldohexosidylrest, vorzugsweise ein l-Aldopentosidylrest oder ein l-Aldohexosidylrest. Beispielsweise sei besonders hervorgehoben der l-Glucopyranosidylrest. Ein Zuckerrest kann auch Schutzgruppen enthalten.
Ein im Phenylteil gegebenenfalls substituiertes a-Methylphenäthyl R2 ist z. B. im Phenylteil durch Hydroxy oder Alkoxy substituiertes a-Methylphenäthyl. Alkoxy ist darin insbesondere solches mit bis zu 7 C-Atomen, vor allem mit bis zu 4 C-Atomen, wie Äthoxy, n- oder iso-Propoxy, n-, sek.- oder tert.-Butoxy und vor allem Methoxy. So ist im Phenylteil gegebenenfalls substituiertes a-Methylphenäthyl z. B. o-, moder p-Hydroxy-a-methyl-phenäthyl, o-, m- oder p-Methoxya-methyl-phenäthyl und vor allem das im Phenylteil unsubstituierte a-Methylphenäthyl.
Die neuen Verbindungen haben vorteilhafte pharmakologische Eigenschaften. So zeigen sie eine positiv chronotrope und insbesondere eine positiv inotrope Wirkung, wie sich durch eine Steigerung der myocardialen Kontraktibilität (wahrscheinlich durch direkte Stimulation der B-Receptoren) und der Herzfrequenz zeigen lässt, z. B. am nicht-narkotisierten Hund mittels Registrierung von diversen Ilontraktibili- tätsparametern, wie maximale Beschleunigung der Blutströmung in der Aorta, sowie der Herzfrequenz bei oraler Gabe in Dosen von etwa 0,1 bis etwa 10 mg/kg. Die neuen Verbindungen können daher als positiv inotrope Mittel insbesondere bei der Behandlung von Herzmuskelinsuffizienz verwendet werden.
Sie können ferner auch als wertvolle Zwischenprodukte für die Herstellung anderer nützlicher Stoffe, insbesondere pharmazeutisch wirksamer Verbindungen, verwendet werden.
Hervorzuheben sind l-Phenoxy-2-hydroxy-3-amino-pro- pane Ia der Formel I, worin R1 l-Glucopyranosidyl ist und R2 Isopropyl, tert.-Butyl oder im Phenylteil gegebenenfalls substituiertes a-Methylphenäthyl ist.
Besonders hervorzuheben sind l-Phenoxy-2-hydroxy-3amino-propane Ib der Formel I, worin R1 l-Glucopyranosidyl ist und R2 Isopropyl, tert.-Butyl oder im Phenylteil durch Hydroxy oder Alkoxy mit bis zu 7 C-Atomen substituiertes a-Methylphenäthyl ist.
Vor allem zu nennen sind l-Phenoxy-2-hydroxy-3-aminopropane Ic der Formel I, worin R1 l-Glucopyranosidyl ist und R2 Isopropyl oder tert.-Butyl ist.
Namentlich hervorzuheben ist insbesondere das l-[p-(ss-D- Glucopyranosidyloxy) -phenoxy]-2-hydroxy-3 -isopropyl-amino-propan mit der obengenannten vorteilhaften Wirkung.
Nach dem erfindungsgemässen Verfahren setzt man eine Verbindung der Formel II

worin R1 obige Bedeutung hat, mit einer Verbindung der Formel III Z2-R2 (III), worin R2 obige Bedeutung hat, einer der Reste Z1 und Z2 Amino ist und der andere reaktionsfähiges verestertes Hydroxy ist und X1 Hydroxy ist, oder Z1 zusammen mit X1 Epoxy ist und Z2 Amino ist, oder einem Salz davon, um.
Reaktionsfähiges verestertes Hydroxy ist insbesondere eine durch eine starke anorganische oder organische Säure, vor allem eine Halogenwasserstoffsäure, wie Chlorwasserstoffsäure, Bromwasserstoffsäure oder Jodwasserstoffsäure, ferner Schwefelsäure oder eine organische Sulfonsäure, wie eine aromatische Sulfonsäure, beispielsweise Benzolsulfonsäure, 4-Brombenzolsulfonsäure oder 4-Toluolsulfonsäure, veresterte Hydroxygruppe. So steht reaktionsfähiges verestertes Hydroxy insbesondere für Chlor, Brom oder Jod.
Diese Umsetzung kann in üblicher Weise durchgeführt werden. Bei Verwendung eines reaktionsfähigen Esters wird vorzugsweise in Gegenwart eines basischen Kondensationsmittels und/oder mit einem Überschuss an Amin der Formel II gearbeitet. Geeignete basische Kondensationsmittel sind z. B. Alkalihydroxyde, wie Natriumhydroxyd oder Kaliumhydroxyd, Alkalicarbonate, wie Kaliumcarbonat, und Alkalialkoholate, wie Alkaliniederalkanolate, zf B. Natriummethylat, Kaliumäthylat oder Kaliumtertiärbutylat.
Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe in freier oder in Form ihrer Säureadditionssalze. So können beispielsweise basische, neutrale oder gemischte Salze, gegebenenfalls auch Hemi-, Mono-, Sesqui- oder Polyhydrate davon, erhalten werden. Die Säureadditionssalze der neuen Verbindungen können in an sich bekannter Weise in die freie Verbindung übergeführt werden, z. B. mit basischen Mitteln, wie Alkalien oder Ionenaustauschern. Andererseits können die erhaltenen freien Basen mit organischen oder anorganischen Säuren Salze bilden. Zur Herstellung von Säureadditionssalzen werden insbesondere solche Säuren verwendet, die zur Bildung von therapeutisch verwendbaren Salzen geeignet sind.
Als solche Säuren seien beispielsweise genannt: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure, aliphatische, alicyclische, aromatische oder heterocyclische Carbonoder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Apfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxymalein- oder Brenztraubensäure-, Fumar-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxybenzoe-, Salicyl- oder p-Aminosalicylsäure, Embonsäure, Methansulfon-, iithansulfon-, Hydroxyäthansulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäure oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin oder Cyclohexylsulfaminsäure.
Diese oder andere Salze der neuen Verbindungen, wie z. B. die Pikrate, können auch zur Reinigung der erhaltenen freien Basen dienen, indem man die freien Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen frei macht. Infolge der engen Beziehungen zwischen den neuen Verbindungen in freier Form und in Form ihrer Salze sind im Vorausgegangenen und nachfolgend unter den freien Verbindungen sinn- und zweckmässig gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die neuen Verbindungen können je nach der Wahl der Ausgangs stoffe und Arbeitsweisen als optische Antipoden oder Racemate oder, sofern sie mindestens zwei asymmetrische Kohlenstoffatome enthalten, auch als Isomerengemische (Racematgemische) vorliegen.
Erhaltene Isomerengemische (Racematgemische) können aufgrund der physikalisch-chemischen Unterschiede der Bestandteile in bekannter Weise in die beiden stereoisomeren (diastereomeren) reinen Racemate aufgetrennt werden, beispielsweise durch Chromatographie und/oder fraktionierte Kristallisation.
Erhaltene Racemate lassen sich nach bekannten Methoden, beispielsweise durch Umkristallisation aus einem optisch aktiven Lösungsmittel, mit Hilfe von Mikroorganismen oder durch Umsetzen mit einer, mit der racemischen Verbindung Salze bildenden optisch aktiven Säure und Trennung der auf diese Weise erhaltenen Salze, z. B. auf Grund ihrer verschiedenen Löslichkeiten, in die Diastereomeren, aus denen die Antipoden durch Einwirkung geeigneter Mittel freigesetzt werden können, zerlegen. Besonders gebräuchliche optisch aktive Säuren sind z. B. die D- und L-Formen von Weinsäure, Di-o-Toluylweinsäure, Äpfelsäure, Mandelsäure, Camphersulfonsäure oder Chinasäure. Vorteilhaft isoliert man der wirksameren der beiden Antipoden.
Zweckmässig verwendet man für die Durchführung der erfindungsgemässen Reaktionen solche Ausgangsstoffe, die zu den eingangs besonders erwähnten Gruppen von Endstoffen und besonders zu den speziell beschriebenen oder hervorgehobenen Endstoffen führen.
Die Ausgangsstoffe sind bekannt oder können, falls sie neu sind, nach an sich bekannten Methoden erhalten werden
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem z. B. für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen, festen oder flüssiger Trägermaterial enthalten. Für die Bildung desselben kommer solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z. B. Wasser, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Benzylalkohole, Gummi, Polyalkylenglykole, Vaseline, Cholesterin oder an dere bekannte Arzneimittelträger. Die pharmazeutischen Prä parate können z. B. als Tabletten, Dragees, Kapseln oder Sup positorien oder in flüssiger Form als Lösungen (z.
B. als Elixier oder Sirup), Suspensionen oder Emulsionen vorliegen.
Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Die Präparate, welche auch in der Veterinärmedizin Verwendung finden können, werden nach üb lichen Methoden gewonnen. Die Tagesdosis für einen Warmblüter von etwa 75 kg Körpergewicht beträgt etwa 10-100 mg, vorzugsweise etwa 20-40 mg.
Die folgenden Beispiele erläutern die Erfindung, ohne sie jedoch einzuschränken. Die Temperaturen sind in Celsiusgra den angegeben.
Beispiel 1
16,4 g (0,05 Mol) 4-(2,3-Epoxy-propoxy)-phenyl-ss-D-glu- copyranosid werden mit 11,8 g (0,20 Mol) Isopropylamin in 250 ml Methanol gelöst und 5 Stunden unter Rückfluss erhitzt. Anschliessend wird im Wasserstrahlvakuum total eingedampft. Der ölige Rückstand wird in Äthanol gelöst und mit 8,95 g (0,05 Mol) N-Cyclohexyl-sulfaminsäure, gelöst in Isopropanol, versetzt. Das kristallin ausgefallene Reaktionsprodukt wird unter Ausschluss von Feuchtigkeit abgesaugt. Man erhält nach Umkristallisieren aus Äthanol-Isopropanol das 1-[p-(fl-D-Glucopyranosidyloxy) -phenoxy] -2-hydroxy-3-isopropylamino-propan-N-cyclohexylsulfaminat, F. 70-1100, [oc] r, -28,80 in Methanol.
Das Ausgangsmaterial kann wie folgt erhalten werden:
27,2 g (0,1 Mol) Hydrochinon-ss-D-glucopyranosid und 250 ml Epichlorhydrin werden in 1 Liter Äthanol gelöst, mit 6,9 g (0,05 Mol) Kaliumcarbonat versetzt und 6 Stunden unter Rückfluss erhitzt. Der ausgefallene Niederschlag wird abgesaugt. Das Filtrat wird im Wasserstrahlvakuum eingedampft. Der Rückstand wird heiss in Äthanol gelöst und filtriert. Das Filtrat wird abgekühlt und bis zur beginnenden Trübung mit Äther versetzt. Das dabei kristallin ausgefallene Reaktionsprodukt wird abgesaugt und mit Äther gewaschen.
Man erhält so das 4-(2,3-Epoxy-propoxy)-phenyl-4-D-gluco- pyranosid, F. 154-1560, [aJ20 52,40 in Methanol.
Beispiel 2 a) Eine Lösung von 9,4 g 4-(2S,3-Epoxypropoxy)-phenyl- B-D-glucopyranosid in 94 ml Äthanol und 46,7 ml Isopropylamin wird 45 Minuten auf 700 erwärmt und anschliessend im Wasserstrahlvakuum vom Lösungsmittel und überschüssigem Amin befreit. Der kristalline Rückstand wird aus 230 ml Methanol umkristallisiert, wonach man das 4-(3-Isopropylamino 2S-hydroxy-propoxy)-phenyl-,8-D-glucopyranosid erhält, F.
186-1880; [a]r, = -460 + 10; [a]E g = -1610 + 10 (Methanol, c = 1,286).
b) Eine Lösung von 500 mg 4-(3-Isopropylamino-2S-hy droxy-propoxy)-phenyl-ss-D-glucopyranosid in 10 ml Methanol wird mit 230 mg N-Cyclohexylsulfaminsäure versetzt, nach 30 Minuten Stehen bei etwa 250 im Wasserstrahlvakuum vom Lösungsmittel befreit und im Hochvakuum getrocknet, wonach man das amorphe 4-(3-Isopropylamino-2S hydroxy-propoxy)-phenyl-4-D -glucopyranosid-N-cyclohexylsulfaminsäuresalz erhält. [a] r) - =400 j 1", [alHp 1380 + 10 (Methanol, c = 1,008).
Das als Ausgangsmaterial verwendete 4-(2S,3-Epoxy propoxy) -phenyl-fl-D-glucopyranosid wird wie folgt hergestellt: c) Eine Lösung von 100 g 4-Hydroxyphenyl-B-D-gluco- pyranosid und 60,5 g 1-O-Benzyl-2S-anhydro-D-glycerin in 366 ml 0,1-n. Natronlauge und 740 ml Acetonitril wird 50 Stunden in einer Stickstoffatmosphäre unter Rückfluss gekocht. Anschliessend wird das Lösungsmittel im Wasserstrahlvakuum abdestilliert und der Rückstand in 1 Liter Wasser aufgenommen.
Das auskristallisierte Produkt wird abgesaugt, aus 1 Liter Wasser umkristallisiert und getrocknet, wonach man das 4-(3-B enzyloxy-2S-hydroxy-propoxy) -phenyl-B- D-glucopyranosid erhält, F. 146-1470; Rf-Wert 0,45 auf Kieselgeldünnschichtplatten im System Methylenchlorid-Methanol 3:1; [a] DO = ¯ 340 + 10 (Methanol, c = 1,027).
d) Eine Lösung von 123 g 4-(3-Benzyloxy-2S-hydroxy propoxy)-phenyl-ss-D-glucopyranosid in 1400 ml Pyridin wird unter Eiskühlung mit 270 ml Acetanhydrid versetzt, 16 Stunden bei etwa 250 stehengelassen und auf 3000 ml Eiswasser gegossen. Nach 2 Stunden Stehen wird das kristalline Produkt abgesaugt, 3mal mit 500 ml Wasser gewaschen, getrocknet und aus 700 ml Äthanol umkristallisiert, wonach man das 4-(2S-Acetoxy-3-benzyloxy-propoxy)-phenyl-2,3,4,6-tetra-O- acetyl-,B-D-glucopyranosid erhält, F. 93-950; Rf-Wert 0,37 auf Kieselgeldünnschichtplatten im System Methylenchlorid Essigsäureäthylester 85:15; bald = 20 - 10 form, c = 0,967).
e) Eine Lösung von 144 g 4-(2S-Acetoxy-3-benzyloxy propoxy) -phenyl-2,3,4,6-tetra-O-acetyl-fl-D-glucopyranosid in 1500 ml Methanol wird mit 14 g 5 0/obigem Palladium-Kohle Katalysator bei Normaldruck hydriert. Der Katalysator wird abfiltriert und das Lösungsmittel im Wasserstrahlvakuum abdestilliert. Nach Umkristallisieren aus Äther-Petroläther erhält man das 4-(2S-Acetoxy-3-hydroxy-propoxy) -phenyl-2,3, 4,6-tetra-O-acetyl--D-glucopyranosid, F. 103-1050; Rf Wert 0,22 auf Kieselgeldünnschichtplatten im System Me thylenchlorid-Essigsäureäthylester 85:15; zahl - = 290 1 10 (Chloroform, c = 1,141).
f) Eine Lösung von 116 g 4-(2S-Acetoxy-3-hydroxypropoxy)-phenyl-2,3,4,6-tetra-O-acetyl-ss-D-glucopyranosid in 700 ml Pyridin wird unter Eiskühlung innerhalb einer Stunde mit 17,4 ml Methansulfonsäurechlorid tropfenweise versetzt und 5 Stunden bei etwa 250 stehengelassen. Das Reaktionsgemisch wird auf 5 Liter Eiswasser gegossen, wobei das Produkt auskristallisiert.
Die Kristalle werden abgesaugt, mit Wasser gründlich gewaschen, getrocknet und aus 800 ml heis sem Äthanol umkristallisiert, wonach man das 4-(2R-Acetoxy-3 -methansulfonyloxy-propoxy)-phenyl-2,3,4,6-tetra-O- acetyl-ss-D-glucopyranosid erhält, F. 110-1150; Rf-Wert 0,20 auf Kieselgeldünnschichtplatten im System Methylenchlorid-Essigsäureäthylester 85:15; [a] r, 280 + 10 (Chloroform, c = 1,223).
g) Eine Suspension von 103 g 4-(2R-Acetoxy-3-methan sulfonyloxy-propoxy)-phenyl-2,3,4,6-tetra-O-acetyl-ss-D-glu- copyranosid in 3 Liter abs. Methanol wird mit 250 g frisch regeneriertem und mit abs. Methanol gewaschenem, stark ba sischem Anionenaustauscherharz Dowex 1 (OH-), 50-100 mesh, versetzt und 30 Minuten bei etwa 250 stark gerührt.
Anschliessend wird der Anionenaustauscher abfiltriert und mit 500 ml Methanol gewaschen. Das Filtrat wird im Wasser strahlvakuum auf etwa 300 ml eingeengt und mit 1 Liter Äther unter Rühren versetzt. Das auskristallisierte Produkt wird abgesaugt und aus Athanol-Ather umkristallisiert, wobei man das 4-(2S,3-Epoxypropoxy)-phenyl-"-D-glucopyranosid erhält, F. 134=1360; Rf-Wert 0,37 auf Kieselgeldünnschicht.

Beispiel 3 a) Eine Lösung von 7,3 g 4-(2S,3-Epoxypropoxy)-phenyl- 2,3,4,6-tetra-O-methyl-fl-D-glucopyranosid in 50 ml Äthanol und 27 ml Isopropylamin wird während 45 Minuten auf 700 erwärmt und das Lösungsmittel sowie überschüssiges Amin anschliessend im Wasserstrahlvakuum abdestilliert. Der Kolbenrückstand wird in 30 ml Methanol aufgenommen, mit 27,6 ml einer 4 0/oigen Lösung von Fumarsäure in Methanol versetzt und im Wasserstrahlvakuum zur Trockne eingedampft.
Der Rückstand wird aus Isopropanol-Äther umkristallisiert, wobei man das 4-(3-Isopropylamino-2S-hydroxy-1- propoxy)-phenyl-2,3,4,6-tetra-O-methyl-ft-D-glucopyranosid fumarat (2:1) erhält, F. 132-1340; [ DO = ¯ 290 + 1" (Methanol, c = 1,193).
Das als Ausgangsmaterial verwendete 4-(2S,3-Epoxypropoxy)-phenyl-2,3,4,6-tetra-O-methyl-fl-D-glucopyranosid wird wie folgt hergestellt: b) Eine Lösung von 30,6 g 4-Benzyloxyphenyl-ss-D-gluco- pyranosid in 320 ml Dimethylsulfoxid wird unter Rühren und Feuchtigkeitsausschluss in einer Stickstoffatmosphäre mit 33,2 g Kaliumhydroxidpulver versetzt und auf 100 abgekühlt.
Nun tropft man bei Aussenkühlung während 4 Stunden 31,8 g Methyljodid zu und rührt weitere 16 Stunden bei etwa 250.
Das Reaktionsgemisch wird auf Eiswasser gegossen, mit Äther extrahiert, die Ätherphase mit Wasser neutral gewaschen, über Magnesiumsulfat getrocknet und im Wasserstrahlvakuum vom Lösungsmittel befreit. Man erhält das 4 Benzyloxyphenyl-2,3,4,6-tetra-O-methyl-fl-D-glucopyranosid als hellgelbes öl; Rf-Wert 0,40 auf Kieselgeldünnschichtplatten im System Methylenchlorid-Essigsäureäthylester 85:15; [zJ r - -31" + 10 (Chloroform, c = 1,081).
c) Eine Lösung von 15,9 g 4-Benzyloxyphenyl-2,3,4,6-te tra-O-methyl-ss-D-glucopyranosid in 160 ml Äthanol wird in Gegenwart von 1,6 g 5 0/obigem Palladium-Kohle-Katalysator bei Normaldruck hydriert. Man filtriert den Katalysator ab und dampft das Filtrat im Wasserstrahlvakuum ein. Der Rückstand wird aus Äther-Petroläther kristallisiert, wobei man das 4-Hydroxyphenyl-2,3,4, 6-tetra-O-methyl-/l'-D-gluco- pyranosid erhält, F. 111-1120; Rf-Wert 0,36 auf Kieselgeldünnschichtplatten im System Methylen chlorid-Essigsäure- äthylester S5:15.
d) Eine Lösung von 12,2 g 4-Hydroxyphenyl-2,3,4,6-tetra O-methyl-ss-D-glucopyranosid in 90 ml çaithylenglykolmono- äthyläther und 4 ml n-Natronlauge wird mit 6,1 g l-O-Benz- yl-2,3-anhydro-D-glycerin während 3 Stunden unter Rückfluss gekocht. Anschliessend wird das Lösungsmittel im Wasserstrahlvakuum abdestilliert und der Rückstand in Äther aufgenommen. Man wäscht diese Lösung mit Wasser neutral, trocknet über Magnesiumsulfat und dampft das Lösungsmittel ab, wobei man das 4-(3-Benzyloxy-2S-hydroxy-propoxy) phenyl-2,3,4,6-tetra-O-methyl-ss-D-glucopyranosid als gelbes öl erhält; Rf-Wert 0,35 auf Kieselgeldünnschichtplatten mit Äther.
e) Eine Lösung von 18 g 4-(3-Benzyloxy-2S-hydroxy prop oxy) -phenyl-2,3 4, 6-tetra-O-methyl-fl-D-glucopyranosid in 20 ml Pyridin lässt man mit 5 ml Acetanhydrid während 18 Stunden bei etwa 250 stehen. Das Reaktionsgemisch wird auf Eiswasser gegossen und nach 30 Minuten mit Äther extrahiert. Man wäscht die Ätherphase mit eiskalter 2-n. Salzsäure, mit Wasser, mit einer gesättigten Natriumhydrogencarbonatlösung und nochmals mit Wasser, trocknet über Magesiumsulfat und dampft das Lösungsmittel ab.
Man erhält das 4-(2S-Acetoxy-3 -benzyloxy-propoxy) -phenyl-2,3 ,4,6-tetra-O- methyl-ss-D-glucopyranosid als hellgelbes Öl; Rf-Wert 0,51 auf Kieselgeldünnschichtplatten im System Äther; [zj r, = -350 + 10 (Chloroform, c = 1,031).
f) Eine Lösung von 19 g 4-(2S-Acetoxy-3-benzyloxy-prop oxy) -phenyl-2,3,4,6-tetra-O-methyl-D-glucopyranosid in 190 ml Methanol wird mit 2 g 5 obigem Palladium-Kohle-Katalysator bei Normaldruck hydriert. Man filtriert den Katalysator ab und dampft die Lösung im Wasserstrahlvakuum ein.
Der Rückstand wird säulenchromatographisch auf 500 g Kieselgel mit Äther gereinigt, wonach man das 4-(2-S-Acetoxy -hydroxy-propoxy)-phenyl-2,3 ,4,6-tetra-O -methyl-fl-D-glu- copyranosid erhält; Rf-Wert 0,17 auf Kieselgeldünnschicht platten mit Äther: [zj 20 = = 310 :2: 10 (Chloroform, c = 2,321).
g) Eine Lösung von 14,3 g 4-(2S-Acetoxy-3-hydroxy propoxy) -phenyl-2,3 ,4,6-tetra-O-methyl-P-D-glucopyranosid in 70 ml Pyridin wird unter Rühren und Eiskühlung während 20 Minuten mit 2,9 ml Methansulfonsäurechlorid tropfenweise versetzt. Nach weiteren 5 Stunden Rühren bei etwa 250 wird das Reaktionsgemisch auf Eiswasser gegossen, lässt 30 Minuten stehen und extrahiert mit Äther. Man wäscht die Ätherphase mit eiskalter 2-n. Salzsäure, mit Wasser, einer gesättigten Natriumhydrogencarbonatlösung und schliesslich wieder mit Wasser, trocknet über Magnesiumsulfat und dampft das Lösungsmittel ab.
Man erhält das 4-(2R-Acetoxy3-methansulfonyloxy-propoxy) -phenyl-2,3,4,6-tetra-O-methyl ss-D-glucopyranosid als farbloses Öl; Rf-Wert 0,12 auf Kie- selgeldünnschichtplatten im System Methylenchlorid-Essigsäureäthylester 85:15; [z] 20 = 180 + 10 (Chloroform, c =1,718).
h) Eine auf -50 abgekühlte Lösung von 15,5 g 4-(2R Acetoxy-3 -methansulfonyloxy-propoxy)-phenyl-2,3,4,64etra O-methyl-ss-D-glucopyranosid in 100 ml Chloroform wird mit einer Lösung von 0,76 g Natrium in 30 ml Methanol versetzt und 2 Stunden im Eisbad stehengelassen. Anschliessend wird das Reaktionsgemisch mit Kohlendioxid neutralisiert und da Lösungsmittel im Wasserstrahlvakuum abgedampft. Der Rückstand wird in Chloroform aufgenommen, mit Wasser gewaschen, über Magnesiumsulfat getrocknet und im Wasserstrahlvakuum vom Lösungsmittel befreit.
Nach Umkristallisation aus Äther-Petroläther erhält man das 4-(2S-3-Epoxy propoxy)-phenyl-2,3,4,6-tetra-O-methyl-ss-D-glucopyranosid; F. 68-690; Rf-Wert 0,45 auf Kieselgeldünnschichtplatten mit Äther; [a] D0 = ¯ 350 + 10 (Chloroform, c = 1,265).
Beispiel 4 a) Eine Suspension von 3,7 g 4-(2,3-Epoxypropoxy) phenyl-2,3,4,6-tetra-O-benzyl-ss-D-glucopyranosid in 50 ml Äthanol wird mit 6 ml Isopropylamin versetzt und auf 700 erwärmt. Nach 2 Stunden Rühren wird die klare Lösung abgekühlt und im Wasserstrahlvakuum zur Trockne eingedampft. Der Kolbenrückstand wird aus Äther-Petroläther umkristallisiert, wonach man das 4-(3-Isopropylamino-2-hy droxy-1-propoxy)-phenyl-2,3,4,6-tetra-O-benzyl-ss-D-gluco- pyranosid erhält, F. 95-980; [α]D20 = -30 + 1 (Chloroform, c = 1,045).
Eine Lösung von 4,1 g 4-(3-Isopropylamino-2-hydroxy-1- propoxy)-phenyl-2,3,4,6-tetra-O-benzyl-ss-D-glucopyranosid in 50 ml Methanol wird mit einer Lösung von 400 mg D Weinsäure in 10 ml Methanol versetzt, wonach 4-(3-Isopropylamino-2-hydroxy-1-propoxy)-phenyl-2,3,4,6-tetra-O-benzyl- ss-D-glucopyranosid-D-tartrat auskristallisiert; es wird aus Methanol umkristallisiert, F. 189-1900; [a] 20 = = -110 + 10 (Chloroform, c = 1,112).
Das als Ausgangsmaterial verwendete 4-(2,3-Epoxyprop oxy)-phenyl-2,3,4,6-tetra-O-benzyl-ss-D-glucopyranosid wird wie folgt hergestellt: b) Eine Lösung von 13,6 g 4-Hydroxyphenyl-ss-D-gluco- pyranosid in 50 ml 1-n. Natronlauge und 100 ml Methanol wird mit 4,2 g Natriumhydrogencarbonat versetzt, dann tropft man unter Rühren bei etwa 250 während einer Stunde 6,2 g Allylbromid zu. Nach weiteren 16 Stunden Rühren wird das Reaktionsgemisch mit 300 ml Methanol verdünnt, filtriert und das Lösungsmittel im Wasserstrahlvakuum abgedampft.
Das erhaltene 4-Allyloxyphenyl-B-D-glucopyranosid wird aus 100 ml Wasser umkristallisiert, F. 144-1450; [zJ D0 = ¯ 540 + 10 (Methanol, c = 1,101); Rf-Wert 0,46 auf Kieselgeldünnschichtplatten im System Methylenchlorid-Methanol 3:1.
c) Eine Lösung von 14 g 4-Allyloxyphenyl-B-D-gluco- pyranosid in 100 ml Dimethylsulfoxid wird unter Rühren und Feuchtigkeitsausschluss in einer Stickstoff-Atmosphäre mit 25 g Kaliumhydroxidpulver versetzt und während 8 Stunden 24,7 g Benzylchlorid zugetropft. Nach weiteren 5 Stunden Rühren wird das Reaktionsgemisch auf Eiswasser gegossen, mit Äther extrahiert, die Ätherphase mit Wasser neutral gewaschen, über Magnesiumsulfat getrocknet und eingedampft.
Aus Äthanol kristallisiert das 4-Allyloxyphenyl-2,3,4,6-tetra O-benzyl-B-D-glucopyranosid, welches nochmals aus Äthanol umkristallisiert wird, F. 70-710; val 20 = -40 + 10
D (Chloroform, c = 1,000); Rf-Wert 0,42 auf Kieselgeldünnschichtplatten im System Äther-Petroläther 1:1.
d) Eine Lösung von 16,5 g 4-Allyloxyphenyl-2,3,4,6-tetra O-benzyl-4-D-glucopyranosid in 250 ml Methylenchlorid wird mit 8,2 g m-Chlorperbenzoesäure während 18 Stunden unter Rückfluss gekocht. Das Gemisch wird abgekühlt, mit einem Liter Äther verdünnt und mit verdünnter Natriumcarbonatlösung, dann mit einer 10 obigen Eisen(II)-sulfatlösung, Wasser, einer verdünnten Natriumcarbonatlösung und wieder mit Wasser neutral gewaschen, über Magnesiumsulfat getrocknet und das Lösungsmittel abgedampft. Der Rückstand wird säulenchromatographisch auf 500 g Kieselgel (Merck) durch Eluieren mit Methylenchlorid gereinigt.
Das so erhaltene 4 (2,3-Epoxypropoxy)-phenyl-2,3,4,6-tetra-O-benzyl-ss-D-glu- copyranosid wird aus Äther-Petroläther umkristallisiert, F.
85870; [a] D0 -40 + 10 (Chloroform, c = 1,207); Rf-Wert 0,1 auf Kieselgeldünnschichtplaften mit Methylenchlorid.
Beispiel 5 a) 27,2 g (0,1 Mol) Hydrochinon-ss-D-glucopyranosid und 250 ml Epichlorhydrin werden in 1 Liter Äthanol gelöst, mit 6,9 g (0,05 Mol) Kaliumcarbonat versetzt und 6 Stunden unter Rückfluss erhitzt. Der ausgefallene Niederschlag wird abgesaugt. Das Filtrat wird im Wasserstrahlvakuum eingedampft. Der Rückstand wird heiss in Äthanol gelöst und filtriert. Das Filtrat wird abgekühlt und bis zur beginnenden Trübung mit Äther versetzt. Das dabei kristallin ausgefallene Reaktionsprodukt wird abgesaugt und mit Äther gewaschen.
Man erhält so das 4-(2,3-Epoxy-propoxy)-phenyl-ss-D-gluco- pyranosid als Diastereomerengemisch, F. 154-1560, [al r, = -52,4 in Methanol.
b) 25 g des Diastereomerengemisches werden in 300 ml Methanol gelöst und mit 18,5 g Isopropylamin 6 Stunden zum Rückfluss erwärmt. Anschliessend wird das Reaktionsgemisch vollständig eingedampft. Der Rückstand wird in 35 ml Wasser gelöst und mit 11,6 g Cyclohexylsulfaminsäure gelöst in Aceton versetzt. Zu dieser Lösung gibt man Aceton bis zur beginnenden Trübung. Dabei beginnt sich das Reaktionsprodukt kristallin abzuscheiden. Zur Vervollständigung der Kristallisation lässt man 15 Stunden bei 0 stehen. Die ausgefallenen Kristalle werden abgesaugt und dreimal aus Aceton-Wasser umkristallisiert.
Man erhält 10 g des reinen Diastereomeren von 1-[p-(ss-D-Glucopyranosidyloxy)-phen oxy] -3 -isopropylamino-2R-propanol-N-cyclohexylsulfaminat- dihydrat, [ 20 16,5 + 0,50 (C = 1,04% g/v in Methanol).
Die beim Kristallisationsprozess anfallenden Mutterlaugen werden vereinigt und im Wasserstrahlvakuum zum Sirup eingeengt, der in 50 ml Wasser gelöst wird. g) A suspension of 103 g of 4- (2R-acetoxy-3-methane sulfonyloxy-propoxy) -phenyl-2,3,4,6-tetra-O-acetyl-ss-D-glucopyranoside in 3 liters of abs. Methanol is freshly regenerated with 250 g and with abs. Methanol-washed, strongly basic anion exchange resin Dowex 1 (OH-), 50-100 mesh, added and stirred vigorously for 30 minutes at about 250.
The anion exchanger is then filtered off and washed with 500 ml of methanol. The filtrate is concentrated to about 300 ml in a water jet vacuum, and 1 liter of ether is added while stirring. The product which has crystallized out is filtered off with suction and recrystallized from ethanol-ether, giving 4- (2S, 3-epoxypropoxy) phenyl - "- D-glucopyranoside, mp 134 = 1360; Rf value 0.37 on a thin layer of silica gel.

Example 3 a) A solution of 7.3 g of 4- (2S, 3-epoxypropoxy) -phenyl-2,3,4,6-tetra-O-methyl-fl-D-glucopyranoside in 50 ml of ethanol and 27 ml of isopropylamine is heated to 700 over 45 minutes and the solvent and excess amine are then distilled off in a water jet vacuum. The residue on the flask is taken up in 30 ml of methanol, 27.6 ml of a 40% solution of fumaric acid in methanol are added and the mixture is evaporated to dryness in a water jet vacuum.
The residue is recrystallized from isopropanol ether, the 4- (3-isopropylamino-2S-hydroxy-1-propoxy) -phenyl-2,3,4,6-tetra-O-methyl-ft-D-glucopyranoside fumarate (2: 1) obtained, m.p. 132-1340; [DO = ¯ 290 + 1 "(methanol, c = 1.193).
The 4- (2S, 3-epoxypropoxy) -phenyl-2,3,4,6-tetra-O-methyl-fl-D-glucopyranoside used as starting material is prepared as follows: b) A solution of 30.6 g of 4 -Benzyloxyphenyl-ss-D-gluco- pyranosid in 320 ml of dimethyl sulfoxide is admixed with 33.2 g of potassium hydroxide powder in a nitrogen atmosphere while stirring and with exclusion of moisture, and the mixture is cooled to 100%.
With external cooling, 31.8 g of methyl iodide are then added dropwise over the course of 4 hours, and the mixture is stirred at about 250 for a further 16 hours.
The reaction mixture is poured onto ice water, extracted with ether, the ether phase washed neutral with water, dried over magnesium sulfate and freed from the solvent in a water jet vacuum. The 4-benzyloxyphenyl-2,3,4,6-tetra-O-methyl-fl-D-glucopyranoside is obtained as a pale yellow oil; Rf value 0.40 on silica gel thin-layer plates in the methylene chloride-ethyl acetate system 85:15; [zJ r - -31 "+ 10 (chloroform, c = 1.081).
c) A solution of 15.9 g of 4-benzyloxyphenyl-2,3,4,6-te tra-O-methyl-ss-D-glucopyranoside in 160 ml of ethanol is in the presence of 1.6 g of 5 0 / above palladium -Carbon catalyst hydrogenated at normal pressure. The catalyst is filtered off and the filtrate is evaporated in a water jet vacuum. The residue is crystallized from ether-petroleum ether, 4-hydroxyphenyl-2,3,4,6-tetra-O-methyl- / l'-D-glucopyranoside being obtained, F. 111-1120; Rf value 0.36 on thin-layer silica gel plates in the methylene chloride-ethyl acetate system S5: 15.
d) A solution of 12.2 g of 4-hydroxyphenyl-2,3,4,6-tetra O-methyl-ss-D-glucopyranoside in 90 ml of ethylene glycol monoethyl ether and 4 ml of n-sodium hydroxide solution is mixed with 6.1 g of lO -Benzyl-2,3-anhydro-D-glycerol boiled under reflux for 3 hours. The solvent is then distilled off in a water jet vacuum and the residue is taken up in ether. This solution is washed neutral with water, dried over magnesium sulphate and the solvent is evaporated off, the 4- (3-benzyloxy-2S-hydroxypropoxy) phenyl-2,3,4,6-tetra-O-methyl-ss -D-glucopyranoside obtained as a yellow oil; Rf value 0.35 on thin-layer silica gel plates with ether.
e) A solution of 18 g of 4- (3-benzyloxy-2S-hydroxy prop oxy) -phenyl-2,3,4,6-tetra-O-methyl-fl-D-glucopyranoside in 20 ml of pyridine is left with 5 ml Acetic anhydride stand at about 250 for 18 hours. The reaction mixture is poured onto ice water and extracted after 30 minutes with ether. The ether phase is washed with ice-cold 2-n. Hydrochloric acid, with water, with a saturated sodium hydrogen carbonate solution and again with water, dried over magnesium sulfate and the solvent evaporated.
4- (2S-acetoxy-3-benzyloxy-propoxy) -phenyl-2,3,4,6-tetra-O-methyl-ss-D-glucopyranoside is obtained as a pale yellow oil; Rf value 0.51 on silica gel thin-layer plates in the ether system; [zj r, = -350 + 10 (chloroform, c = 1.031).
f) A solution of 19 g of 4- (2S-acetoxy-3-benzyloxy-prop oxy) -phenyl-2,3,4,6-tetra-O-methyl-D-glucopyranoside in 190 ml of methanol is mixed with 2 g of 5 the above palladium-carbon catalyst hydrogenated at normal pressure. The catalyst is filtered off and the solution is evaporated in a water jet vacuum.
The residue is purified by column chromatography on 500 g of silica gel with ether, after which 4- (2-S-acetoxy-hydroxy-propoxy) -phenyl-2,3, 4,6-tetra-O -methyl-fl-D-glu - receives copyranosid; Rf value 0.17 on thin-layer silica gel plates with ether: [zj 20 = = 310: 2: 10 (chloroform, c = 2.321).
g) A solution of 14.3 g of 4- (2S-acetoxy-3-hydroxy propoxy) -phenyl-2,3,4,6-tetra-O-methyl-PD-glucopyranoside in 70 ml of pyridine is stirred and ice-cooled 2.9 ml of methanesulfonyl chloride were added dropwise over the course of 20 minutes. After stirring for a further 5 hours at about 250, the reaction mixture is poured onto ice water, left to stand for 30 minutes and extracted with ether. The ether phase is washed with ice-cold 2-n. Hydrochloric acid, with water, a saturated sodium hydrogen carbonate solution and finally again with water, dried over magnesium sulfate and the solvent evaporated.
4- (2R-acetoxy3-methanesulfonyloxy-propoxy) -phenyl-2,3,4,6-tetra-O-methyl-ss-D-glucopyranoside is obtained as a colorless oil; Rf value 0.12 on thin-layer silica plates in the methylene chloride-ethyl acetate system 85:15; [z] 20 = 180 + 10 (chloroform, c = 1.718).
h) A solution, cooled to -50, of 15.5 g of 4- (2R acetoxy-3-methanesulfonyloxy-propoxy) -phenyl-2,3,4,64etra O-methyl-ss-D-glucopyranoside in 100 ml of chloroform is mixed with a solution of 0.76 g of sodium in 30 ml of methanol and left to stand for 2 hours in an ice bath. The reaction mixture is then neutralized with carbon dioxide and the solvent is evaporated off in a water jet vacuum. The residue is taken up in chloroform, washed with water, dried over magnesium sulfate and freed from the solvent in a water-jet vacuum.
After recrystallization from ether-petroleum ether, 4- (2S-3-epoxy propoxy) -phenyl-2,3,4,6-tetra-O-methyl-ss-D-glucopyranoside is obtained; F. 68-690; Rf value 0.45 on thin-layer silica gel plates with ether; [a] D0 = ¯ 350 + 10 (chloroform, c = 1.265).
Example 4 a) A suspension of 3.7 g of 4- (2,3-epoxypropoxy) phenyl-2,3,4,6-tetra-O-benzyl-ss-D-glucopyranoside in 50 ml of ethanol is mixed with 6 ml of isopropylamine added and heated to 700. After stirring for 2 hours, the clear solution is cooled and evaporated to dryness in a water jet vacuum. The residue on the flask is recrystallized from ether-petroleum ether, after which the 4- (3-isopropylamino-2-hy droxy-1-propoxy) -phenyl-2,3,4,6-tetra-O-benzyl-ss-D-gluco - pyranoside preserved, m.p. 95-980; [α] 20 D = -30 + 1 (chloroform, c = 1.045).
A solution of 4.1 g of 4- (3-isopropylamino-2-hydroxy-1-propoxy) -phenyl-2,3,4,6-tetra-O-benzyl-ss-D-glucopyranoside in 50 ml of methanol is with a solution of 400 mg of D tartaric acid in 10 ml of methanol, after which 4- (3-isopropylamino-2-hydroxy-1-propoxy) -phenyl-2,3,4,6-tetra-O-benzyl- ss-D- glucopyranoside-D-tartrate crystallized out; it is recrystallized from methanol, mp 189-1900; [a] 20 = = -110 + 10 (chloroform, c = 1.112).
The 4- (2,3-epoxypropoxy) -phenyl-2,3,4,6-tetra-O-benzyl-ss-D-glucopyranoside used as starting material is prepared as follows: b) A solution of 13.6 g 4-hydroxyphenyl-ss-D-glucopyranoside in 50 ml 1-n. Sodium hydroxide solution and 100 ml of methanol are mixed with 4.2 g of sodium hydrogen carbonate, then 6.2 g of allyl bromide are added dropwise with stirring at about 250 in the course of one hour. After stirring for a further 16 hours, the reaction mixture is diluted with 300 ml of methanol, filtered and the solvent is evaporated off in a water-jet vacuum.
The 4-allyloxyphenyl-B-D-glucopyranoside obtained is recrystallized from 100 ml of water, mp 144-1450; [zJ D0 = ¯ 540 + 10 (methanol, c = 1.101); Rf value 0.46 on thin-layer silica gel plates in the methylene chloride-methanol 3: 1 system.
c) A solution of 14 g of 4-allyloxyphenyl-B-D-glucopyranoside in 100 ml of dimethyl sulfoxide is mixed with 25 g of potassium hydroxide powder in a nitrogen atmosphere while stirring and with exclusion of moisture, and 24.7 g of benzyl chloride are added dropwise over 8 hours. After stirring for a further 5 hours, the reaction mixture is poured onto ice water, extracted with ether, the ether phase washed neutral with water, dried over magnesium sulfate and evaporated.
The 4-allyloxyphenyl-2,3,4,6-tetra O-benzyl-B-D-glucopyranoside crystallizes from ethanol and is recrystallized again from ethanol, mp 70-710; val 20 = -40 + 10
D (chloroform, c = 1,000); Rf value 0.42 on silica gel thin-layer plates in the ether-petroleum ether 1: 1 system.
d) A solution of 16.5 g of 4-allyloxyphenyl-2,3,4,6-tetra-O-benzyl-4-D-glucopyranoside in 250 ml of methylene chloride is refluxed with 8.2 g of m-chloroperbenzoic acid for 18 hours . The mixture is cooled, diluted with one liter of ether and washed neutral with dilute sodium carbonate solution, then with the above iron (II) sulfate solution, water, a dilute sodium carbonate solution and again with water, dried over magnesium sulfate and the solvent is evaporated off. The residue is purified by column chromatography on 500 g of silica gel (Merck) by eluting with methylene chloride.
The 4 (2,3-epoxypropoxy) -phenyl-2,3,4,6-tetra-O-benzyl-ss-D-glucopyranoside obtained in this way is recrystallized from ether-petroleum ether, F.
85870; [a] DO -40 + 10 (chloroform, c = 1.207); Rf value 0.1 on thin-layer silica gel plates with methylene chloride.
Example 5 a) 27.2 g (0.1 mol) of hydroquinone-ss-D-glucopyranoside and 250 ml of epichlorohydrin are dissolved in 1 liter of ethanol, 6.9 g (0.05 mol) of potassium carbonate are added and the mixture is refluxed for 6 hours heated. The deposited precipitate is filtered off with suction. The filtrate is evaporated in a water jet vacuum. The residue is dissolved in hot ethanol and filtered. The filtrate is cooled and ether is added until it starts to become cloudy. The reaction product which has precipitated in crystalline form is filtered off with suction and washed with ether.
This gives 4- (2,3-epoxy-propoxy) -phenyl-ss-D-glucopyranoside as a mixture of diastereomers, mp 154-1560, [al r, = -52.4 in methanol.
b) 25 g of the diastereomer mixture are dissolved in 300 ml of methanol and refluxed with 18.5 g of isopropylamine for 6 hours. The reaction mixture is then completely evaporated. The residue is dissolved in 35 ml of water and mixed with 11.6 g of cyclohexylsulfamic acid dissolved in acetone. Acetone is added to this solution until it begins to cloud. The reaction product begins to separate out in crystalline form. To complete the crystallization, the mixture is left to stand at 0 for 15 hours. The precipitated crystals are filtered off with suction and recrystallized three times from acetone-water.
10 g of the pure diastereomer of 1- [p- (ss-D-glucopyranosidyloxy) -phenoxy] -3-isopropylamino-2R-propanol-N-cyclohexylsulfaminate dihydrate, [20 16.5 + 0.50 (C. = 1.04% w / v in methanol).
The mother liquors obtained during the crystallization process are combined and concentrated in a water jet vacuum to form a syrup, which is dissolved in 50 ml of water. This solution is filtered through a column with 200 ml Amberlite IRA-400 R (strongly basic ion exchanger with trimethylammonium groups [grain size 0.38-0.45 mm]). The column is washed with water until the liberated base is completely eluted. The combined aqueous eluates are completely evaporated in a water jet vacuum and recrystallized three times from methanol isopropanol.
The second diastereomer is obtained, namely 1- [p- (ss-D-glucopyranosidyloxy) -phenoxy] -3-isopropylamino- (2S) -propanol, mp 157-1600 / 186-1890, [D20 = 400 + 1 , [cs] g = 138 + 1 138 f 1 (methanol, c = 1.008).
