Die Erfindung betrifft ein Verfahren zur Herstellung neuer 1,2,3,4-Tetrahydroisochinoline, welche eine relaxierende Wirkung auf die glatte Muskulatur besitzen. Die neuen 1,2,3,4 Tetrahydroisochinoline entsprechend der allgemeinen Formel:

in welcher Rl ein Wasserstoffatom oder einen Niedrigalkylrest, X ein Sauerstoff- oder Schwefelatom und R3 einen Phenylrest mit 1 bis 3 Substituenten aus der Gruppe der Niedrigalkoxy-, Hydroxy-, Amino-, Nitro-, Halogen(niedrig)alkyl -Reste und Halogenatome bedeuten.
Unter den hier verwendeten Ausdrücken < Niedrigalkyl Gruppen > und Niedrigalkoxy-Gruppen} sind, wenn nichts anderes angegeben ist, Alkyl- und Alkoxygruppen mit 1 bis 6 Kohlenstoffatomen zu verstehen.
Ein geeigneter Niedrigalkyl-Rest in der oben angegebenen Formel ist beispielsweise ein Alkyl-Rest mit 1 bis 6 Kohlenstoffatomen, z.B. ein Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, t-Butyl-, Pentyl- und Hexylrest, vorzugsweise ein Alkyl-Rest mit 1 bis 4 Kohlenstoffatomen, insbesondere ein Alkyl-Rest mit 1 bis 2 Kohlenstoffatomen.
Ein geeigneter Niedrigalkoxy-Rest in der obigen Formel ist z.B. ein Alkoxy-Rest mit 1 bis 6 Kohlenstoffatomen, beispielsweise ein Methoxy-, Äthoxy-, Propoxy-, Butoxy-, Isopropoxy-, Isobutoxy-, t-Butoxy-, Pentoxy-, Isopentoxy- oder Hexyloxyrest, vorzugsweise ein Alkoxy-Rest mit 1 bis 4 Kohlenstoffatomen, insbesondere ein Alkoxy-Rest mit 1 bis 2 Kohlenstoffatomen.
Ein geeigneter Halogen(niedrig)alkyl-Rest in der obigen Formel ist z.B. ein Halogenalkyl-Rest mit 1 bis 6 Kohlenstoffatomen, beispielsweise ein Chlormethyl-, Fluoräthyl-, Chlorpropyl-, Brompropyl-, Jodbutyl-, Chlorpentyl-, Bromchloräthyl-, Dichlormethyl-, Dichloräthyl-, Difluormethyl-, Dichlorpropyl-, Trichlormethyl-, Tribrommethyl-, Trifluormethyl-, Trichloräthyl- oder Tribromäthylrest, vorzugsweise ein Halogenalkyl-Rest mit 1 bis 4 Kohlenstoffatomen, insbesondere ein Halogenalkyl-Rest mit 1 bis 2 Kohlenstoffatomen.
Der in der obigen Formel verwendete Ausdruck zHalo- genatom > umfasst ein Chlor-, Brom-, Fluor- und Jodatom.
Die neuen 1,2,3,4-Tetrahydroisochinoline der Formel I und ihre pharmazeutisch verträglichen Salze weisen eine relaxierende Wirkung auf die glatte Muskulatur, insbesondere auf die glatten Vasculär- und Viszeralmuskeln auf. Dementsprechend haben sie eine vasodilatorische, intestinal-kontraktionsinhibierende und blasen-kontraktions-inhibierende Wirkung, weisen jedoch keine bronchien-dilatorische Wirkung auf und sind geeignet als vasodilatorische, intestinal-kontraktions-inhibierende und blasen-kontraktions-inhibierende Mittel.
Das erfindungsgemässe Verfahren besteht darin, dass man eine Verbindung der Formel:

in der R, und X die oben angegebene Bedeutung besitzen, R5 und R8 jeweils ein Wasserstoffatom oder einzeln oder gemeinsam Schutzgruppen von Hydroxyresten und R7 einen Phenylrest mit 1 bis 3 Substituenten aus der Gruppe der Niedrigalkoxy-, mit einer Schutzgruppe geschützten Hydroxy-, Nitro-, Halogen-(niedrig)-alkyl-, Aminoreste und Halogenatome bedeuten, oder ein Salz davon reduziert.

in der R1, R5 und R6 jeweils die oben angegebenen Bedeutungen besitzen, mit einer Verbindung der Formel: R7-XH2COOH (xI) in der R, und X wie oben definiert sind, oder einem reaktiven Derivat an dem Carboxylrest davon, und anschliessende Behandlung der erhaltenen Verbindung mit einem Dehydratisierungsmittel.
Beispiele für geeignete einzelne Schutzgruppen für einen Hydroxyrest der oben angegebenen Verbindungen VIII sind Acetyl-, Benzyloxycarbonyl-, 4- Nitrobenzyloxycarbonyl-, 4-Brombenzyloxycarbonyl-, 4-Methoxybenzyloxycarbonyl-, 3 ,4-Dimethoxybenzyloxycarbonyl-, 4- (Phenylazo)benzyloxy- carbonyl-, 4-(4- Methoxyphenylazo)benzyloxycarbonyl-, t-Butoxycarbonyl-, 1 ,l-Dimethylpropoxycarbonyl-, Isopropoxycarbonyl-, Diphenylmethoxycarbonyl-, 2-Pyridylmethoxycarbonyl-2,2,2-Trichloräthoxycarbonyl-, 2,2,2-Tribromäthoxycarbonyl-, 3-Jod-propoxycarbonyl-, 2-Furfuryloxycarbonyl-, I-Adamantyloxycarbonyl-, 3-Chinolyloxycarbonyl-, Trifluoracetyl-, Benzyl-, Trityl-, Methoxymethyl-, 2-Nitrophenylthio-, 2,4-Dinitrophenylthioreste und dergleichen.
Eine geeignete Schutzgruppe für einen Hydroxyrest, bei der es undR6 miteinander verbunden sind, ist z.B. ein Alkylenrest, wie ein Methylen-, Dimethylmethylen-, Äthylenrest und dergleichen.
Die Reduktion wird auf übliche Art und Weise durchgeführt, beispielsweise durch katalytische Hydrierung, durch Reduktion mit einem Reduktionsmittel, wie z.B. einem alkalischen Metallaluminiumhydrid oder einem alkalischen Metallborhydrid, und durch Reduktion mit einer Säure und einem Metall. Natürlich variiert die angewendete Reduktionsmethode je nach Art der Ausgangsverbindungen VIII und für den Fall, dass die Hydroxyreste in den Ausgangsverbindungen VIII durch Schutzgruppen geschützt sind, muss die Reduktion unter stark sauren Bedingungen durchgeführt werden. Bei einer katalytischen Reduktion wird die Umsetzung auf übliche Art und Weise unter Verwendung eines üblichen Katalysators, wie z.B. Palladiumkohle, Raney-Nickel, Platin(IV)oxyd und dgl., durchgeführt, und sie kann auch unter steigendem Druck durchgeführt werden.
Wenn ein alkalisches Metallaluminiumhydrid verwendet wird, wird die Umsetzung auf übliche Art und Weise in einem üblichen Lösungsmittel, z.B. Diäthyläther, Dibutyläther, Tetrahydrofuran, Dioxan und dgl., durchgeführt. Beispiele für geeignete alkalische Metallaluminiumhydride sind Alkalimetallaluminiumhydride (z.B. Lithiumaluminiumhydrid, Natriumaluminiumhydrid usw.) und Erdalkalimetallaluminiumhydride (z.B. Calciumaluminiumhydrid, Magnesiumaluminiumhydrid usw.).
Wenn eine Säure und ein Metall verwendet werden, wird die Umsetzung auf übliche Art und Weise unter Verwendung einer Säure, wie z.B. Chlorwasserstoffsäure, Schwefelsäure, Essigsäure usw., und eines Metalls, wie z.B. Eisen, Zink, Zinn usw., durchgeführt. Wenn ein alkalisches Metallborhydrid verwendet wird, wird die Umsetzung auf übliche Art und Weise in einem Lösungsmittel, wie z.B. Wasser, Methanol, Äthanol, Tetrahydrofuran, Dioxan usw., durchgeführt. Beispiele für geeignete alkalische Metallborhydride sind Alkalimetallborhydride (z.B. Natriumborhydrid, Lithiumborhydrid usw.) und Erdalkalimetallborhydride (z.B. Magnesiumborhydrid, Calciumborhydrid usw.).
Für den Fall, dass der Rest R7 in der obigen Formel VIII einen Phenylrest mit einem oder mehreren Nitroresten als Substituent(en) bedeutet, werden bei der erfindungsgemässen Umsetzung die Nitroreste in Aminoreste überführt, ausser bei Verwendung eines alkalischen Metallborhydrids als Reduktionsmittel, und es ist klar, dass die Reduktion von Nebenreaktionen begleitet ist, die ebenfalls in den Rahmen der vorliegenden Erfindung fallen.
In den nachfolgend beschriebenen Tests werden die relaxierenden Wirkungen bzw. Aktivitäten von typischen Verbindungen der Formel I auf die glatte Muskulatur erläutert, wobei es sich bei den einzelnen aktiven Bestandteilen um die nachfolgend numerierten Verbindungen handelt: Verbindung 1:

Test I
Intestinalmotilität bei Hunden
Verfahren: Bastardhunde mit einem Gewicht von 8 bis 16 kg, denen 24 Stunden lang kein Futter und kein Wasser gegeben worden war, wurden mit einer Kombination von Urethan (l,Sg/kg) und Morphin (15 mg/kg) anästhesiert. An dem Jejunum des Hundes wurde ein Ballon befestigt, auf einen Druck von 10 cm H2O komprimiert und mit einem Spannungsmessgerät verbunden.
Die Änderungen der Motilität wurden hauptsächlich in Form der Amplitude aufgezeichnet, wobei jedoch der Anzahl der Bewegungen Rechnung getragen wurde. Das Änderungsmaximum aller Messungen diente zur Berechnung der Dosis der 50%igen Inhibierung (ED50-Wert). Die für eine 80%ige Erholung erforderliche Zeit wurde als Wirkungsdauer angesehen. Die dabei erhaltenen Ergebnisse sind in der folgenden Tabelle angegeben.

Durchflussmenge des Blutes in den inneren Carotis und Femoralarterien
Verfahren: Bastardhunde mit einem Gewicht von 8 bis 15 kg wurden mit Pentobarbitalnatrium (35 mg/kg) anästhesiert. Die linke innere Carotisarterie und die rechte Femoralarterie wurden mit Sonden eines elektromagnetischen Strömungsmessgerätes versehen und die linke Femoralarterie wurde an ein Spannungsmessgerät angeschlossen, um den systemischen Blutdruck zu messen. Die Herzgeschwindigkeit (Herzfrequenz) wurde mit einem an einen Elektrokardiographen angeschlossenen Tachometer gemessen.
Die Blutdurchflussmenge, der Blutdruck und die Herzfrequenz wurden mit einem Polygraphen gleichzeitig aufgezeichnet. Die Testverbindung wurde intravenös verabreicht, wenn ein konstanter Wert bei jedem der untersuchten Parameter beobachtet wurde. Die Anderungen (in %) nach der Verabreichung der Testverbindung wurden errechnet. Die dabei erhaltenen Ergebnisse sind in der folgenden Tabelle angegeben.

Akute Toxizität
Verfahren:
Die Testverbindung wurde intravenös oder oral an Gruppen von männlichen Mäusen vom ICR-JCL Stamm, die ein Gewicht von 25 bis 35 g hatten, und wobei jede Gruppe aus 10 Tieren bestand, verabreicht. 7 Tage nach der Verabreichung wurden die toten Tiere gezählt und nach der Probit-Methode wurde die LD50 errechnet.

Beispiel I
Eine Lösung von 120 mg 1-(p-Hydroxyphenoxy)methyl- -6,7-dihydroxy-3,4-dihydroisochinolinhydrobromid und 40 mg Natriumborhydrid in 8 cm5 99%igem Athanol wurde 1,5 Stunden lang bei Raumtemperatur gerührt. Das Äthanol wur de aus der Lösung abdestilliert, und zu dem Rückstand wurde Wasser zugesetzt. Die wässrige Lösung wurde durch Zugabe von Chlorwasserstoffsäure neutralisiert, und die Niederschläge wurden durch Filtrieren gesammelt, mit Wasser gewaschen und getrocknet, wobei Kristalle erhalten wurden.
Die Kristalle wurden auf übliche Art und Weise in das entsprechende Hydrochlorid überführt, wobei 64 mg Kristalle von 1 -(p-Hydroxyphenoxy)methyl-6,7-dihydroxy- 1,2,3 ,4-tetrahy- droisochinolinhydrochlorid, F. 262 C, erhalten wurden.
Beispiel 2
5,0 g 1-(p-Benzyloxyphenoxy)methyl-6,7-dibenzyloxy-3,4- -dihydroisochinolinhydrochlorid wurden durch katalytische Reduktion reduziert, wobei die Umsetzung in einem äthanolischen Lösungsmittel bei 60 C unter Verwendung von 1,0 g 10%iger Palladiumkohle und 2 cm3 Chlorwasserstoffsäure unter Atmosphärendruck durchgeführt wurde. Die Reaktionsmischung wurde filtriert, und das Filtrat wurde unter vermindertem Druck eingeengt. Das Konzentrat wurde mit einem Methanoll Äther-Lösungsmittelgemisch behandelt, und die so erhaltenen Kristalle wurden aus dem gleichen Lösungsmittelgemisch umkristallisiert unter Bildung von 1-(p-Hy droxyphenoxy)methyl-6,7-dihydroxy- 1,2,3 ,4-tetrahydroisochi- nolinhydrochlorid, F. 2620C.
Beispiel 3
0,7 g 1-(p-Nitrophenoxy)methyl-6,7-dihydroxy-3,4-dihy- droisochinolin wurden durch katalytische Reduktion reduziert, wobei die Umsetzung in 100 ml Äthanol bei 60 C unter Verwendung von 200 mg 10%iger Palladiumkohle und 1 ml kenzentrierter Chlorwasserstoffsäure durchgeführt wurde. Die Reaktionsmischung wurde filtriert, und das Filtrat wurde unter vermindertem Druck eingeengt. Der Rückstand wurde aus einem Äthanol/Äther-Lösungsmittelgemisch umkristallisiert, und man erhielt 0,4 g Kristalle von l-(p-Aminophenoxy)methyl-6,7-dihydroxy-1 ,2,3,4-tetrahydroisochinolinhydrochlorid, F. 287-2880C.

Wegen ihrer bereits oben genannten Eigenschaften Aktivitäten) sind die neuen 1,2,3,4-Tetrahydroisochinoline der Formel I und ihre pharmazeutisch verträglichen Salze brauchbare Relaxiermittel für die glatte Muskulatur und insbesondere für glatte Vasculär- und Viszeralmuskeln.
Die neuen 1 ,2,3,4-Tetrahydroisochinoline der Formel I und ihre pharmazeutisch verträglichen Salze können nach den üblichen Methoden, den üblichen Typen von Dosierungseinheiten oder mit üblichen pharmazeutischen Trägern verabreicht werden zur Erzielung der relaxierenden Wirkung auf die glatte Muskulatur (glatte Muskeln) von Säugetieren.
Sie können in Form von pharmazeutischen Präparaten verwendet werden, in denen sie in Mischung mit einem für die enterale oder parenterale Verabreichung geeigneten organischen oder anorganischen pharmazeutischen Trägermaterial enthalten sind. Die orale Verabreichung unter Verwendung von Tabletten, Kapseln oder in flüssiger Form, wie z.B. in Form von Suspensionen, Lösungen oder Emulsionen ist besonders vorteilhaft. Wenn sie zu Tabletten verarbeitet werden, können die üblichen Bindemittel und Desintegriermittel, wie sie bei therapeutischen Dosierungseinheiten verwendet werden, eingesetzt werden. Beispiele für geeignete Bindemittel sind Glukose, Lactose, Akaziengummi, Gelatine, Mannit, Stärkepaste, Magnesiumtrisilicat und Talk. Beispiele für geeignete Desintegriermittel sind Maisstärke, Keratin, kolloidales Siliciumdioxyd und Kartoffelstärke.
Wenn sie in Form von Flüssigkeiten verabreicht werden, können die üblichen flüssigen Träger verwendet werden.
Die Einheitsdosierung oder die therapeutisch wirksame Menge der Verbindungen I und ihrer pharmazeutisch verträglichen Salze kann in der Humanmedizin innerhalb breiter Grenzen, beispielsweise von 0,01 bis etwa 100 mg, variieren.
Die obere Grenze ist nur durch den Grad des gewünschten Effektes und durch wirtschaftliche Erwägungen beschränkt.
Für die orale Verabreichung werden vorzugsweise etwa 1 bis etwa 100 mg des therapeutischen Mittels pro Dosierungseinheit verwendet. Tierexperimente haben gezeigt, dass Dosen von etwa 0,1 bis etwa 10 mg, die bei Bedarf täglich drei Mal orale verabreicht werden, eine bevorzugte Tagesdosis darstellen. Natürlich kann die Dosierung des jeweils verwendeten therapeutischen Mittels stark variieren, z.B. je nach Alter des Patienten und je nach Grad des gewünschten therapeutischen Effektes. Jede Einheitsdosierungsform der neuen therapeutischen Verbindungen kann etwa 0,5 bis etwa 99,5 Gew.-O/o der neuen therapeutischen Verbindungen, bezogen auf die Gesamtzusammensetzung, enthalten, wobei der Rest aus übli- chen pharmazeutischen Trägern besteht.
Unter den hier verwendeten Ausdruck pharmazeutische Träger, fallen auch nicht-therapeutische Materialien die üblicherweise in Einheitsdosen verwendet werden, dazu gehören z.B. Füllstoffe, Ver dünnungsmittel. Bindemittel, Gleitmittel, Desintegriermittel und Lösungsmittel. Natürlich ist es auch möglich, die neuen therapeutischen Verbindungen in Form der reinen Verbindungen ohne Verwendung eines pharmazeutischen Trägers zu verabreichen.
