Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von in 3-Stellung schwefelhaltigen Cephalosporinderivaten. Insbesondere bezieht sich die vorliegende Erfindung auf ein Verfahren zur Herstellung von 3-(5-Mercapto-1,3,4-thiadiazol-2-yl-thiomethyl)-7- (tetrazolylalkanamido)-3-cephem-4-carbonsäuren.
Diese Verbindungen können zu pharmazeutisch zulässigen, nicht toxischen Präparaten verarbeitet werden.
Die neuen Cephalosporinderivate besitzen ein breites Spektrum gegen Mikroorganismen, einschliesslich Proteus mirabilis.
Es sind verschiedene in den 3- und 7-Stellungen heterocyclisch substituierte Cephalosporinverbindungen geoffenbart worden. Ein Bedarf nach weiteren Cephalosporinen, welche ein breites antibakterielles Spektrum gegen Mikroorganismen aufweisen, liegt vor. Insbesondere trachtet man nach Cephalosporinen, welche gegen Mikroorganismen wirken, deren Wachstum mit den bekannten Cephalosporinen nicht hinreichend gehemmt wird. In der deutschen Offenlegungsschrift 1 953 861 wurde geoffenbart, dass Verbindungen mit einer 5 Methylthio-1,3 ,4-thiadiazol-2-ylthiomethylgruppe in der 3 Stellung des Cephalosporinringes gegen gewisse Mikroorganismen wirksam sind, doch war es erforderlich, weitere Cephalosporinverbindungen zu ergründen, welche den Erfordernissen gerecht werden.
Die erfindungsgemässen, in 3-Stellung schwefelhaltigen Cephalosporinderivate lassen sich durch die folgende allgemeine Strukturformel wiedergeben:

in welcher R einen gegebenenfalls substituierten, N-gebundenen Tetrazolylrest und A einen niederen Alkylenrest mit 1 bis 4 C-Atomen bedeuten.
Die Bezeichnung Tetrazolyl bedeutet einen einwertigen 5-gliedrigen heterocyclischen Ring, welcher aus 4 Stickstoffatomen und einem Kohlenstoffatom besteht, wie z.B. iH- Tetrazolyl und 'H-Tetrazolyl. wobei ein solcher Rest Substituenten tragen kann, wie z.B. einen niederen Alkylrest mit bis zu 4 Kohlenstoffatomen. wie z.B. Methyl, Äthyl, Propyl, Isopro pyl Butyl. Isobutyl. tert.-Butyl, 1-Methylpropyl oder 2 Methylpropyl.
Unter der Bezeichnung Niederalkylen versteht man zweiwertige aliphatische, geradlinige oder verzweigte Kohlenwasserstoffreste mit bis zu 4 Kohlenstoffatomen, wie z.B. Methylen, Äthylen, Propylen, Butylen, 1-Methylpropylen, 9-Methyl- propylen, 2-Äthyläthylen und 1-Propylmethylen.
Die nicht toxischen, pharmazeutisch zulässigen Salze der Verbindungen der Formel I sind in ähnlicher Weise anwendbar.
Bevorzugte Verbindungen sind jene, bei denen R die iH- Tetrazol-1-ylgruppe und A die Methylengruppe bedeuten.
Erfindungsgemäss lassen sich die neuen Verbindungen in verschiedener Weise aus 7-Aminocephalosporansäure oder Cephalosporin C gemäss USA-Patentschrift Nr. 3 516 997 und deutscher Offenlegungsschrift Nr. 2 055 796 herstellen.
Gemäss der erfindungsgemässen Ausführungsform werden die gewünschten Verbindungen dadurch erhalten, dass man eine in
7-Stellung acylierte Aminocephalosporansäure der Formel:

worin R und A die obigen Bedeutungen haben, oder ein Salz davon mit einem Dithiol der Formel:

umsetzt. Die Umsetzung kann in Gegenwart eines inerten Lösungsmittels. wie z.B. Aceton, Dioxan, Methanol, Äthanol, Tetrahydrofuran und dergleichen, oder mit einer Mischung solcher Lösungsmittel oder in Gegenwart einer wässrigen Lösung solcher Lösungsmittel oder von Wasser oder Pufferlösungen, wie z.B. Phosphatpuffer usw., oder in Gegenwart eines anderen beliebigen Verdünnungsmittels erfolgen.
Verwendet man solche Verdünnungsmittel, so werden Puffermittel vorzugsweise bei der Behandlung der Dithiole in ihrer freien Form verwendet, wodurch die Dithiole in eine reaktionsfähige Form übergeführt werden. Werden die Ausgangsverbindungen in ihrer freien Form verwendet, so kann die Umsetzung in Gegenwart einer Base, wie z.B. Natriumcarbonat, Triäthylamin oder dergleichen, erfolgen. Die Gegenwart einer solchen Base wird auch bei der Umsetzung zwischen den Ausgangsmaterialien, welche entweder in der freien Form oder in Form ihrer Salze vorliegen, und den Dithiolen bevorzugt. Die Umsetzung kann bei erhöhter Temperatur unter schwach sauren Bedingungen und nötigenfalls unter Druck und/oder in Gegenwart eines inerten Gases, wie z.B. Stickstoff, erfolgen.
Die Reaktionsbedingungen bezüglich Temperatur, Druck, pH Wert und dergleichen sind indessen nicht von besonderer Bedeutung.
Die erfindungsgemäss erhältlichen, gewünschten Verbindungen werden vorzugsweise für pharmazeutische Zwecke in die entsprechenden nicht toxischen, pharmazeutisch zulässigen Salze übergeführt. Dies kann durch Behandlung beispielsweise mit einem Alkalimetall, wie z.B. Natrium, Kalium oder dergleichen, einem Amin, wie z.B. Diphenylendiamin, Dicyclohexylamin, Dibenzyläthylendiamin, Triäthylamin oder dergleichen, geschehen.
Die erfindungsgemässen Verbindungen können in Form von pharmazeutischen Präparaten als Arzneimittel Verwendung finden. Diese Arzneimittel enthalten die in 3-Stellung schwefelhaltigen Cephalosporinderivate oder deren Salze nebst mindestens einem pharmazeutisch zulässigen organischen oder anorganischen, festen oder flüssigen Trägermittel, das sich für die orale oder parenterale Verabreichung eignet. Die pharmazeutischen Präparate können in fester Form, z.B. in Form von Kapseln, Tabletten oder Dragees, oder in flüssiger Form, z.B.
in Form von Lösungen, Suspensionen oder Emulsionen, vorliegen. Gewünschtenfalls können solche Präparate auch Hilfssubstanzen, wie z.B. Konservierungsmittel, Stabilisiermittel, Netzmittel oder Emulgiermittel, Salze zur Variierung des osmotischen Druckes und Puffermittel, enthalten.
Die Erfindung sei durch die folgenden Beispiele erläutert, ohne aber auf dieselben eingeschränkt zu sein.
Beispiel 1 7-( 1H-Tetrazol- 1-ylacetamido)-3-(5-mercapto- 1,3,4 thiadiazol-2-yl)-thiomethyl-3 -cephem -4-carbonsäure
Eine Lösung von 4,1 g des Natriumsalzes der 7-(1H Tetrazol- 1 -ylacetamido)-cephalosporansäure (l/2 H20), 1,48 g 1,3,4-Thiadiazol-2,5-dithiol und 1,68 g Natriumbicarbonat in 80 ml eines Phosphatpuffers (pH 6,4) wird während 3 Stunden bei 65'C gerührt. Dann wird das Reaktionsgemisch filtriert und das Filtrat mittels 10 %iger Salzsäure auf einen pH-Wert von 5 bis 6 eingestellt und mit Äthylacetat gewaschen. Die abgetrennte wässrige Schicht wird erneut mit 10 %iger Salzsäure auf einen pH-Wert von 2 bis 3 eingestellt und dann abermals mit Äthylacetat extrahiert.
Der Extrakt wird mit einer gesättigten Natriumchloridlösung gewaschen, über Magnesiumsulfat getrocknet und durch Aktivkohle entfärbt. Nach dem Abdestillieren des Lösungsmittels unter vermindertem Druck wird der Rückstand mit Äther trituriert, wobei man 2,78 g 7-(1H-Tetrazol-1-ylacetamido)-3-(5-mercapto- 1 ,3,4-thiadiazol-2-yl)-thiomethyl-3-cephem-4-carbonsäure vom Schmelzpunkt 88 bis 900C (Zersetzung) erhält.
Beispiel 2 7-(2H-Tetrazol-2-ylacetamido)-3-(5-mercapto- 1,3,4- thiadiazol-2-yl) -thiomethyl-3 -cephem-4-carbonsäure
Eine Lösung von 4,82 g des Triäthylaminsalzes der 7 (2H-Tetrazol-2-ylacetamido)-cephalosporansäure, 1,50 g 1,3,4-Thiadiazol-2,5-dithiol und 1,68 g Natriumbicarbonat in 60 ml Phosphatpuffer (pH 6,4) wird praktisch in der gleichen Weise wie in Beispiel 1 behandelt, wobei man 1,01 g pulverige 7-(2H-Tetrazol-2-ylacetamido)-3-(5-mercapto-1,3,4- thiadiazol-2-yl)-thiomethyl-3-cephem-4-carbonsäure vom Schmelzpunkt 115 bis 1200C (Zersetzung) erhält.
The present invention relates to a process for the preparation of 3-position sulfur-containing cephalosporin derivatives. In particular, the present invention relates to a process for the preparation of 3- (5-mercapto-1,3,4-thiadiazol-2-yl-thiomethyl) -7- (tetrazolylalkanamido) -3-cephem-4-carboxylic acids.
These compounds can be processed into pharmaceutically acceptable, non-toxic preparations.
The new cephalosporin derivatives have a broad spectrum against microorganisms, including Proteus mirabilis.
Various 3- and 7-position heterocyclic substituted cephalosporin compounds have been disclosed. There is a need for further cephalosporins which have a broad antibacterial spectrum against microorganisms. In particular, one looks for cephalosporins which act against microorganisms whose growth is not sufficiently inhibited with the known cephalosporins. Methyl, ethyl, propyl, isopropyl butyl. Isobutyl. tert-butyl, 1-methylpropyl or 2-methylpropyl.
The term lower alkylene is understood to mean divalent aliphatic, straight or branched hydrocarbon radicals with up to 4 carbon atoms, e.g. Methylene, ethylene, propylene, butylene, 1-methylpropylene, 9-methylpropylene, 2-ethylethylene and 1-propylmethylene.
The non-toxic, pharmaceutically acceptable salts of the compounds of the formula I can be used in a similar manner.
Preferred compounds are those in which R is the iH-tetrazol-1-yl group and A is the methylene group.

Example 1 7- (1H-Tetrazol-1-ylacetamido) -3- (5-mercapto-1,3,4-thiadiazol-2-yl) -thiomethyl-3-cephem -4-carboxylic acid
A solution of 4.1 g of the sodium salt of 7- (1H tetrazole-1-acetamido) -cephalosporanic acid (1/2 H20), 1.48 g of 1,3,4-thiadiazole-2,5-dithiol and 1.68 g of sodium bicarbonate in 80 ml of a phosphate buffer (pH 6.4) is stirred at 65 ° C. for 3 hours. The reaction mixture is then filtered and the filtrate is adjusted to a pH of 5 to 6 using 10% hydrochloric acid and washed with ethyl acetate. The separated aqueous layer is again adjusted to pH 2 to 3 with 10% hydrochloric acid and then extracted again with ethyl acetate.
