Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von neuen 18-Methylensteroiden der Androstanreihe und 19-Nor-androstanreihe und insbesondere solcher der Formel

worin X Wasserstoff oder eine Methylgruppe darstellt, R eine p-ständige veresterte Hydroxygruppe zusammen mit einem Wasserstoffatom oder einem gesättigten oder ungesättigten, unsubstituierten oder halogenierten niederaliphatischen Kohlenwasserstoffrest bedeutet und in welcher in 6,7-Stellung und bei Verbindungen, in denen X eine Methylgruppe bedeutet, in 6,7- und/oder in 1,2-Stellung weitere Doppelbindungen vorhanden sein können.
Veresterte Hydroxygruppen sind vor allem solche, die sich von organischen Carbonsäuren der aliphatischen, alicyclischen aromatischen oder heterocyclischen Reihe ableiten, insbesondere von solchen mit 1-18 Kohlenstoffatomen, z.B. der Ameisensäure, Essigsäure, Propionsäure, der Buttersäuren, Valeriansäuren, wie n-Valeriansäure, oder Trimethylessigsäure, der Capronsäuren, wie ,8-Trimethyl-propionsäure oder Diäthylessigsäure, der Oenanth-, Capryl-, Pelargon-, Capron-, Undecylsäuren, der Undecylensäure, der Laurin-, Myristin-, Palmitinoder Stearinsäuren, der Ölsäure, oder einer Cyclopropan-, -butan-, pentan- und -hexan Carbonsäure, einer Cycloalkylalkancarbonsäure, z.B.
Cyclopropylmethancarbonsäure, cy- clobutylmethancarbonsäure, Cyclopentyläthancarbonsäure, Cyclohexyläthancarbonsäure, der Benzoesäure, von Phenoxyalkansäuren, wie Phenoxyessigsäure, von Dicarbdnsäuren, wie Bernsteinsäure, Phthalsäure, Chinolinsäure, der Furan-2-carbonsäure, 5-tert.-Butyl-furan-2-carbonsäure, 5-Brom-furan-2 -carbonsäure, der Nicotinsäure oder der Isonicotinsäure, oder von Sulfonsäuren, wie Benzolsuifonsäuren, oder von anorganischen Säuren, wie z.B. Phosphor- oder Schwefelsäuren.
Insbesondere leiten sich die veresterten Hydroxylgruppen von niederaliphatischen Carbonsäuren, wie Essigsäure, Propionsäure, Trimethylessigsäure, Valeriansäure oder Capronsäure, ab.
Der in 17-Stellung eventuell vorhandene ungesättigte oder gesättigte, unsubstituierte oder halogenierte niederaliphatische Kohlenwasserstoffrest weist vorzugsweise 1-4 C-Atome auf und ist z.B. der Methyl-, Äthyl-, Propyl-, Isopropyl-, Allyl-, Vinyl-, Methallyl-, Äthinyl-, Propinyl-, Chlor- oder Fluorpropinylrest oder der Trichlor- oder Trifluorpropinyfrest.
Die neuen Verbindungen der Formel (1) weisen androgenanabole Wirkung und insbesondere eine antigonadotrope Wirkung auf. Verbindungen, in welchen in 17a-Stellung ein ungesättigter, unsubstituierter oder halogenierten Kohlenwasserstoffrest vorhanden ist, weisen zudem eine gestagene und antiovulatorische Wirkung auf.
Besonders zu nennen sind Ester des 1 8-Methylen-testo- sterons, des 18-Methylen-17a-methyl-testosterons, des 18-Me thylen-17a-äthinyl-testosterons sowie deren l-Dehydroderivaten und der entsprechenden 19-Norverbindungen, insbesondere die entsprechenden niederaliphatischen Carbonsäureester.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man in einer der obenstehenden Formel I entsprechenden Verbindung mit einer freien 17p-Hydroxylgruppe diese Hydroxylgruppe in an sich bekannter Weise verestert.
Wenn erwünscht, so kann man in erhaltenen A4-3-Oxo-Verbindungen in 6,7-Stellung, und, im Falle von 10-Methylverbindungen in 6,7- und/oder in 1 ,2-Stellung je eine Doppelbindung einführen.
Die Veresterung einer 170-Hydroxygruppe geschieht in an sich bekannter Weise, indem man die zu veresternden Verbindungen mit reaktionsfähigen Derivaten von Säuren, wie den Halogeniden oder Anhydriden, insbesondere jenen der oben genannten Säuren, in Gegenwart von tertiären Basen, wie Pyridin oder Chinolin, umsetzt.
Die wenn erwünscht durchführenden Dehydrierungen gemäss dem vorliegenden Verfahren können ebenfalls in an sich bekannter Weise ausgeführt werden, z.B. mikrobiologisch, insbesondere in 1,2-Stellung mittels des Mikroorganismus Corynebacterium simplex oder chemisch mit Selendioxyd in einem tertiären Alkohol, wie tert. Amylalkohol, oder mit einem Chinon, wie Dicyanodichlorbenzochinon, insbesondere zur Einführung auch der 6,7-Doppelbindung, z.B. mit Chloranil.
Die Ausgangsstoffe für das erfindungsgemässe Verfahren können z.B. so erhalten werden, dass man in Verbindungen der Formel

worin X Wasserstoff oder eine Methylgruppe bedeutet, oder in ihren entsprechenden Dehydroderivaten, die 17-Oxogruppe, gegebenenfalls unter vorübergehendem selektivem Schutz der 3-Oxogruppe, und eventuell unter Einführung eines gesättigten oder ungesättigten, unsubstituierten oder halogenierten Kohlenwasserstoffrestes in 17-Stellung, zur freien Hydroxygruppe reduziert.
Die Verbindungen der Formel (II) können ihrerseits aus 17, 18-Dioxoandrostan oder 19-Nor-androstan Verbindungen mit geschützter 17-Oxogruppe, und in welchen auch weitere gegebenenfalls vorhandene Oxogruppen, z.B. in 3-Stellung, geschützt sind, durch Umsetzung mit einem reaktiven Methylenphosphoran nach Wittig und, gegebenenfalls, durch Ausbildung der A4-3-Oxogruppe in an sich bekannter Weise hergestellt werden.
Man kann aber auch von 18-Cyano-Verbindungen der Androstanreihe oder 19-Norandrostanreihe ausgehen, in welcher Oxogruppen in geschützter Form vorliegen, sie durch Reduktion mit einem komplexen Leichtmetallhydrid, wie Lithiumaluminiumhyd, in die 1 8-Aminomethyl -steroide überführen und diese anschliessend in Form ihrer N-Oxide auf höhere Temperatur, z.B.in einem Lösungs- oder Suspensionsmittel, wie Tetralin, Decalin oder Collidin, z.B. bei 150-2500, erhitzen. Nach beiden Methoden werden am Schlusse die geschützte Oxogruppe in 17-Stellung und evtl. auch weitere geschützte Oxogruppen durch saure Hydrolyse in Freiheit gesetzt.
Die verfahrensgemäss erhaltenen Verbindungen können auch zur Herstellung von pharmazeutischen Präparaten zur Anwendung in der Human- oder Veterinärmedizin verwendet werden, welche die neuen, oben beschriebenen pharmakologisch wirksamen Stoffe als aktive Substanzen zusammen mit einem pharmazeutischen Trägermaterial enthalten.
Die erfindungsgemäss erhaltenen Verbindungen können auch als Futterzusatzmittel verwendet werden.
Die Erfindung wird im folgenden Beispiel näher beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel
Eine Suspension von 5,0 g 3,17-Dioxo-A4st8(lsa)-18-homo- -19-nor-androstadien in 25 ml absolutem Dioxan und 5,0 ml Äthylorthoformiat versetzt man mit 1 ml einer Lösung, hergestellt durch Vermischen von 0,488 g p-Toluensulphonsäure, 5,4 ml Dioxan und 1,1 ml absolutes Äther. Das Reaktionsgemisch wird 2 Stunden bei Zimmertemperatur gerührt und anschliessend mit 2,5 ml Pyridin versetzt. Das gelbe Reaktionsgemisch wird im Wasserstrahlvakuum eingedampft und der Rückstand auf einer 30fachen Gewichtsmenge Aluminiumoxyd (Aktivität III, basisch) adsorbiert. Durch Eluieren mit Petroläther-Toluol (50: 50)-Gemisch erhält man das reine 3-Äthoxy-l7-oxo-A325f l8 (18a) - I 8-homo- 1 9-nor-androstatrien, dessen IR-Spektrum unter anderen die folgenden Bande aufweist: 1740, 1655, 1622, 1235, 1185 und 1175cm-1.
