5H-Dibenzo[a,d]cycloheptene und 10,11 -Dihydro-5H-di- benzo[a,d]cycloheptene mit einer basischen Seitenkette in 5-Stellung sind bereits als antidepressiv wirkende Psychopharmaka bekannt, z. B. 10,11-Dihydro-5-(3-dimethylamino- propyliden)-5H-dibenzo[a,d]cyclohepten (Amitriptylin).
Es wurde nun überraschend gefunden, dass tricyclische Verbindungen der Formel

in der R ein Chlor- oder Fluoratom bedeutet, oder die entsprechenden 10,11-Dehydro-Derivate, sowie Isomere und Säureadditionssalze dieser Verbindungen, die sich von den bekannten Vertretern der betroffenen Substanzgruppe durch die Anwesenheit eines Chlor- oder Fluoratoms in 1-Stellung und eines am Stickstoff gebundenen Sauerstoffatoms unterscheiden, durch eine beträchtlich erhöhte antidepressive Wirkung und stark verminderte Toxizität auszeichnen. Als besonders vorteilhaft ist die fehlende oder nur geringfügige anticholinergische Wirkung zu erwähnen. Die Verbindungen der Formel I zeichnen sich ausserdem durch vielfältige Wirkungen auf das Nervensystem aus.
So sind narkosepotenzierende, adrenolytische, sedative, antihistaminartige und lokalanästhetische Wirkungen festgestellt worden.
In der obigen Formel I bedeutet R bevorzugt ein Chloratom. Interessante Vertreter von Verbindungen der Formel I sind z.
das 1 -Chlor-5 -(3 -dimethylaminopropyliden)-SH-dibenzo- [a,d]cyclohepten-N-oxid, das 1 -Chlor- 10,11 -dihydro-5-(3 -dimethylaminopropyliden)- 5H-dibenzo[a,d]cyclohepten-5 -oxid.
Besonders interessant ist das 1-Chlor-5-(3-dimethyl amino-propyliden)-5H-dibenzo[a,djcyclohepten-N-oxid.
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von tricyclischen Verbindungen der Formel 1 bzw.
von 10,11-Dehydro-Derivaten davon sowie von Isomeren und Säureadditionssalzen dieser Verbindungen, das dadurch gekennzeichnet ist, dass man eine Verbindung der Formel

worin eines der Symbole
Y1 und Y2 ein Wasserstoffatom und das andere eine Hydroxylgruppe bedeutet, oder ein 10,1 1-Dehydro-Derivat davon oder ein Säureadditionssalz dieser Verbindungen dehydratisiert. Anschliessend kann man in beliebiger Reihenfolge erwünschtenfalls die Isomeren aus einem erhaltenen Isomerengemisch isolieren und erXünschtenfalls eine erhaltene Base in ein Säureadditionssalz überführen.
Bevorzugte Ausgangsstoffe sind Verbindungen der Formel II, in denen Y1 eine Hydroxylgruppe und Y2 Wasserstoff darstellen.
Die erfindungsgemässe Dehydratisierung wird zweckmässig unter Verwendung von Mineralsäuren, wie Chlor- oder Bromwasserstoffsäure, durchgeführt, wobei in wasserfreiem oder wässrigem Medium gearbeitet werden kann. Vorzugsweise wird die Dehydratisierung in äthanolischer Salzsäure bei einer Temperatur zwischen Raumtemperatur und dem Siedepunkt des Reaktionsgemisches ausgeführt. Sie gelingt jedoch auch durch Erwärmen, z. B. bei 50 bis Rückflusstemperatur, vorzugsweise bei Rückflusstemperatur, mit einem hochsiedenden, wasserfreien Lösungsmittel, wie Dimethylsulfoxid. Andere übliche Wasserabspaltungsmittel können ebenfalls eingesetzt werden, z. B. Schwefelsäure, Phosphoroxychlorid, Zinkchlorid oder Kaliumbisulfat, z.
B. in einem inerten organischen Lösungsmittel, beispielsweise in Chloroform oder Methylenchlorid, und bei einer Temperatur zwischen Raumtemperatur und dem Siedepunkt des Reaktionsgemisches.
Ausgangscarbinole der Formel II, in denen Y1 Hydroxyl und Y2 Wasserstoff darstellen, sind z. B. in der Weise erhältlich, dass man 1 -Chlor(bzw. Fluor)-5H-dibenzo[a,djcyclo- hepten-5-on oder die entsprechende 10,1 1-Dihydroverbin- dung mit einem Dimethylaminopropylmagnesiumhalogenid umsetzt. Nach Hydrolyse des entstandenen Additionsproduktes, z. B. mit gesättigter Ammoniumchloridlösung, wird das erhaltene 1 -Chlor(bzw. Fluor) -5-(3 -dimethylaminopropyl)- 5-hydroxy-5H-dibenzo[a,d]cyclohepten bzw. die erhaltene, entsprechende 10,11 -Dihydroverbindung oxydiert. Die Oxydation erfolgt bevorzugt durch Behandeln mit Wasserstoffperoxid in einem Lösungsmittel, z. B.
Methanol, Äthanol, Äther, Benzol oder Chloroform, bei etwa Raumtemperatur. Nach Zerstören des überschüssigen Wasserstoffperoxids mit z. B.
Platinschwarz kann die gewünschte Verbindung der Formel II nach Abdampfen des Lösungsmittels gewonnen werden.
Manchmal kann man das Produkt durch kräftiges Rühren, z. B. bis zu einer Temperatur zwischen etwa -20 und 0 C, zum Kristallisieren bringen und anschliessend durch einfaches Filtrieren und Nachwaschen gewinnen.
Ausgangscarbinole der Formel II, in denen umgekehrt Y2 Wasserstoff und Y2 Hydroxyl bedeuten, erhält man beispielsweise dadurch, dass man ein entsprechendes tricyclisches 5-Keton mit Äthylmagnesiumbromid umsetzt und das Reaktionsprodukt hydrolysiert. Die entstandene 5-Hydroxy5-äthyl-verbindung wird mit Acetylchlorid dehydratisiert und anschliessend mit Ameisensäure und Wasserstoffperoxid behandelt. Es entsteht eine 5-Hydroxy-5-(1-hydroxyäthyl7-ver- bindung, welche mit wässriger Schwefelsäure zu der entsprechenden 5-Acetyl-verbindung dehydratisiert wird. Durch Behandeln mit Formaldehyd und Dimethylamin-hydrochlorid erhält man eine 5-Dimethylaminopropionyl-verbindung, die nach Reduktion mit Natriumborhydrid in das entsprechende Carbinol übergeht.
Dieses wird anschliessend, wie oben für die Herstellung der Carbinole der Formel II, in denen Y1 Hydroxyl und Y2 Wasserstoff darstellen, beschrieben, oxydiert.
Die oben gegebenen Methoden zur Herstellung von Ausgangsverbindungen der Formel II sind nur beispielsweise angeführt. Andere, dem Fachmann offensichtliche Methoden kommen selbstverständlich auch in Frage.
Erhaltene Verbindungen der Formel I und deren Salze können in ihre geometrischen Isomeren, d. h. a- bzw. ss-Isomeren, getrennt werden Die Trennungsmethoden sind an sich bekannt. Bevorzugt trennt man die geometrischen Isomeren durch fraktionierte Kristallisation der Säureadditionssalze aus einem Lösungsmittel, z. B. Aceton oder aus einem Lösungsmittelgemisch, z. B. Methanol/Diäthyläther.
Erhaltene Verbindungen der Formel I und deren Salze liegen als Racemate vor. Ein Racemat kann in seine optischen Isomeren in an sich bekannter Weise, z. B. durch Umsetzung mit optisch aktiven Säuren, wie Weinsäure oder Camphersulfosäure, und anschliessende Kristallisation, aufgetrennt werden.
Die Trennung der geometrischen und/oder optischen Isomeren kann auch auf einer Zwischenproduktstufe vorgenommen werden, so dass in dieser Weise das erfindungsgemässe Verfahren mit geometrisch bzw. optisch einheitlichen Ausgangsmaterialien der Formel II ausgeführt wird.
Die Verbindungen der Formel I haben basischen Charakter und können in ihre Säureadditionssalze übergeführt werden. Solche Salze sind beispielsweise diejenigen mit organischen Säuren, wie Oxalsäure, Zitronensäure, Essigsäure, Milchsäure, Maleinsäure und Weinsäure, oder mit anorganischen Säuren, wie Chlorwasserstoffsäure, Bromwasserstoffsäure oder Schwefelsäure. Die Säureadditionssalze sind kristalline, feste Substanzen, die in Wasser löslich, in polaren Lösungsmitteln, wie Methanol, Äthanol usw., etwas weniger löslich und in nichtpolaren Lösungsmitteln, wie Benzol, Äther und Petroläther, relativ unlöslich sind.
Wie vorstehend erwähnt, besitzen die Verbindungen der Formel I eine überraschende antidepressive Wirkung. Zum Beispiel wurden Gruppen von je 10 Mäusen die zu prüfenden Verbindungen der Formel I in den nachstehend aufgeführten Dosen subcutan injiziert. 16 Stunden später erhielten die Tiere 5 mg/kg 2-Hydroxy-2-äthyl-2-isobutyl-9,10-dimeth oxy-1,2,3,4,6,7-hexahydro-1 lbH-benzo[a]chinolizin-hydro- chlorid (Substanz A) subcutan injiziert. Die gleiche Dosis wurde einer Gruppe von 10 nicht vorbehandelten Mäusen verabreicht. Nach weiteren 30 Minuten wurden allen Tieren sowie einer zusätzlichen Gruppe von 10 Mäusen, Äthanol in der Dosis von 3,75 g/kg i. p. appliziert. In jeder Gruppe der so behandelten Tiere wurde die durchschnittliche Schlafdauer ermittelt.
Die prozentuale Abschwächung der Schlafdauer im Vergleich zu dem durch die Substanz A verstärkten Äthanol-Schlaf dient als Mass für die Stärke der antidepressiven Wirkung.
In der nachstehenden Tabelle wird für verschiedene Verbindungen der Formel I die prozentuale Abschwächung der Schlafdauer angegeben.

Die geringe Toxizität der Verbindungen der Formel I kann durch die folgenden Werte für die akute Toxizität an Mäusen (24-Stunden-Werte) illustriert werden: Wirkstoff DL50 i.V. p.O.

Die fehlende bzw. geringe anticholinergische Wirkung kann durch das Ausbleiben der Salivationshemmung an Kaninchen festgestellt werden: Bei Kaninchen in leichter Urethannarkose wurde durch Pilocarpin-Injektion die Salivation gesteigert und die Speichelmenge in 5-Minuten-Intervallen gemessen. 1 -Chlor-5-(3-dimethylaminopropyliden) -SH-di- benzo[a,d]cyclohepten-N-oxid-hydrochlorid hemmte in 1 und 3 mg/kg i. v. die Salivation nicht und in 6 mg/kg nur noch gering.
Bei Verabreichung dieser Verbindung sowie auch bei Verabreichung von 1 -Chlor-1 0,11 -dihydrn-5-(3-dimethyl- aminopropyliden) -5H-dibenzo[a,d]cyclohepten-N-oxid-hy- drochlorid oder 1-Fluor- 10,11 -dihydro-5-(3 -dimethylamino- propyliden)-5H-dibenzo[a,djcyclohepten-N-oxid-hydrochlo- rid konnte eine geringere Hemmung der Salivation beobachtet werden als bei Verabreichung von entsprechenden Mengen von Amitriptylin.
Die erfindungsgemäss erhältlichen Verbindungen können als Heilmittel z. B. in Form pharmazeutischer Präparate Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale, z. B. orale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen inerten Trägermaterial, wie z. B. Wasser, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Gummiarabicum, Polyalkylenglykole, Vaseline usw. enthalten. Die pharmazeutischen Präparate können in fester Form oder in flüssiger Form vorliegen. Gegebenenfalls enthalten sie Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Zweckmässige pharmazeutische Dosierungsformen enthalten etwa 1 bis 200 mg einer Verbindung der Formel I. Zweckmässige orale Dosierungsbereiche liegen bei etwa 0,1 mg/kg pro Tag bis etwa 5 mg/kg pro Tag. Zweckmässige parenterale Dosierungsbereiche liegen bei etwa 0,01 mg/kg pro Tag bis etwa 1 mg/kg pro Tag.
In einem klinischen Versuch wurde 41 an Depression leidende, hospitalisierte Patienten mit 1-Chlor-5-(3-dimethyl- aminopropyliden) -SH-dibenzo[a,d]cyclohepten-N-oxid- hydrochlorid wie folgt behandelt: Tagesdosis, mg 150 p.o. 75 p.o.

Das Präparat entfaltete bei allen Patienten eine deutliche antidepressive Wirkung. Die Verträglichkeit des Präparates war sehr gut. Insbesondere fiel die Abwesenheit verschiede ner nachteiliger Nebenwirkungen, wie anticholinergischer Wirkung (z. B. Mundtrockenheit, Obstipation und Miktionsbeschwerden), sedierender Wirkung, Vitalstörungen (Störung des Schlafs und/oder Appetits), auf. In dieser Hinsicht konnte ein günstigeres Ergebnis verzeichnet werden als bei Verwendung von entsprechenden Mengen Amitriptylin.
Des weiteren wurde, im Vergleich mit Amitriptylin, ein rascherer Wirkungseintritt beobachtet. Nach erfolgter Behandlung mit 1-Chlor-5-(3-dimethylaminopropyliden)-SH- dibenzo[a,d]cyclohepten-N-oxid-hydrochlorid konnte eine Rückbildung der Krankheitssymptome verzeichnet werden.
Aufgrund dieser Tatsachen war in der Mehrzahl der Fälle eine Entlassung der Patienten aus der stationären Behandlung möglich.
Beispiel 1
5 g 1-Chlor- 10,1 1-dihydro-5-(3-dimethylaminopropyl)-5 hydroxy-5H -dibenzo[a ,d]cyclohepten-N -oxid-hydrochlond werden in 20 ml absolutem Methanol gelöst und mit 2 ml 24%iger (Gew. pro Volumen) methanolischer Salzsäure 2 Stunden unter Rückflussbedingungen erhitzt. Nach dem Erkalten wird das Reaktionsgemisch mit Äther versetzt. Es kristallisiert das 1-Chlor-10,1 1-dihydro-5-(3-dimethylamino- propyliden) -5H-dibenzo[a,d]cyclohepten-N-oxid-hydrochlo- rid, das bei 163-165 C schmilzt.
Das als Ausgangsverbindung verwendete 1-Chlor-10,11- dihydro-5-(3-dimethylaminopropyl)-5-hydroxy-5H-dibenzo- [a,d]cyclohepten-N-oxid-hydrochlorid kann wie folgt hergestellt werden:
86,6 g Gilman-Legierung oder Magnesium-Späne werden in 800 ml absolutem Äther mit einer Spur Jod unter Rückflussbedingungen erhitzt. Anschliessend wird innerhalb 2 Stunden eine Lösung von 390 g Dimethylaminopropylchlorid in 500 ml absolutem Tetrahydrofuran eingetropft. Es wird weitere 3 Stunden unter Rückflussbedingungen erhitzt.
Zu der erhaltenen, auf 15 C abgekühlten Suspension wird innerhalb 15 Minuten eine Lösung von 242,7 g 1-Chlor-10,11-di hydro-5H-dibenzo[a,djcyclohepten-5-on in 500 ml absolutem Tetrahydrofuran zugetropft und das Ganze noch 12 Stunden unter Rückflussbedingungen erhitzt. Das Reaktionsgemisch wird anschliessend auf 15 abgekühlt und mit 500 ml gesättigter Ammoniumchloridlösung hydrolysiert. Das Ganze wird filtriert und mit Methylenchlorid nachgewaschen. Das Filtrat wird mit Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Der zurückbleibende gelbeRohstoff wird aus 6000 ml hochsiedendem Petroläther umkristallisiert.
Man erhält 1-Chlor- 10,1 1-dihydro-5-(3-dimethylaminopropyl)-5- hydroxy-5H-dibenzo[a,d]cyclohepten , das bei 131-132 C schmilzt.
40 g 1-Chlor- 10,1 1-dihydro-5-(3-dimethylaminopropyl)- 5-hydroxy-SH-dibenzo[a,d]cyclohepten werden in 1000 ml Methanol gelöst, mit 61 ml 30 3'Oiger wässriger Wasserstoffperoxidlösung versetzt und 160 Stunden bei Zimmertemperatur gerührt. Durch vorsichtige Zugabe von Platinschwarz wird anschliessend der Überschuss an Wasserstoffperoxid zerstört.
Das Reaktionsgemisch wird dann filtriert und unter vermindertem Druck eingedampft. Der Rückstand wird mit methanolischer Salzsäure angesäuert. Nach dem Zusatz von Äther fällt das Hydrochlorid von 1-Chlor-10,11-dihydro-5-(3-dimethylaminopropyl) -5-hydroxy-5H-dibenzo[a,djcyclohep- ten-N-oxid kristallin aus, Schmelzpunkt 164-166" C.
Beispiel 2
Wird im Verfahren nach Beispiel 1 als Ausgangsmaterial anstatt 1-Chlor-10,1 1-dihydro-5-(3-dimethylaminopropyl)- 5-hydroxy-SH -dibenzo[a,d]cyclohepten-N -oxid-hydrochlorid das 1 -Chlor-5-(3 -dimethylaminopropyl)-5-hydroxy-5H-di- benzo[a,d]cyclohepten-N-oxid-hydrochlorid eingesetzt, so erhält man unter sonst gleichen Bedingungen das Hydrochlorid von 1 -Chlor-5-(3 -dimethylaminopropyliden)-5H-dibenzo- [a,d]cyclohepten-N-oxid, das nach dem Umkristallisieren aus Methanol/Äther bei 172-174 C schmilzt.
Das in der obigen Reaktion als Ausgangsmaterial eingesetzte 1-Chlor-5-(3-dimethylaminopropyl)-5-hydroxy-SH-di- benzo[a,d]cyclohepten-N-oxid-hydrochlorid kann wie folgt hergestellt werden:
17 g Magnesium und 85 g Dimethylaminopropylchlorid werden miteinander umgesetzt. Die entstandene Grignardverbindung wird dann mit einer Lösung von 52 g 1-Chlor-SHdibenzo[a,d]cyclohepten-5-on in 500 ml absolutem Tetrahydrofuran versetzt. Nach Aufarbeitung erhält man 1-Chlor5 -(3-dimethylaminopropyl) -5-hydroxy-5H-dibenzo[a,d] cyclohepten vom Schmelzpunkt 134-136" C. Das daraus hergestellte Hydrochlorid schmilzt nach dem Umkristallisieren aus Methanol/Äther bei 226-227 C.
2gl-Chlor-5-(3 -dimethylaminopropyl) -5-hydroxy-SH- dibenzo[a,d]cyclohepten werden in 60 ml Methanol gelöst und mit 2,1 g 30%iger wässriger Wasserstoffperoxidlösung versetzt. Das Ganze wird 35 Stunden bei 500 C gerührt. Nach Abkühlen auf 100 C scheiden sich Kristalle aus. Diese werden filtriert und mit Äther gewaschen. Man erhält 1-Chlor 5-(3 -dimethylaminopropyl) -5-hydroxy-SH-dibenzo[a,d]cy- clohepten-N-oxid, das bei 161-165 C unter Zersetzung schmilzt. Diese Verbindung wird in Äther gelöst und mit methanolischer Salzsäure versetzt. Man erhält 1-Chlor-5-(3dimethylaminopropyl)-5-hydroxy-SH-dibenzo[a,d]cyclohepten-N-oxid-hydrochlorid, das nach dem Umkristallisieren aus Methanol/Äther bei 174-177" C schmilzt.
5H-dibenzo [a, d] cycloheptenes and 10,11-dihydro-5H-dibenzo [a, d] cycloheptenes with a basic side chain in the 5-position are already known as antidepressant psychotropic drugs, z. B. 10,11-dihydro-5- (3-dimethylaminopropylidene) -5H-dibenzo [a, d] cycloheptene (amitriptyline).

Y1 and Y2 represent a hydrogen atom and the other represents a hydroxyl group, or a 10,1 1-dehydro derivative thereof or an acid addition salt of these compounds is dehydrated. The isomers can then be isolated in any order, if desired, from a mixture of isomers obtained and, if desired, a base obtained can be converted into an acid addition salt.
Preferred starting materials are compounds of the formula II in which Y1 is a hydroxyl group and Y2 is hydrogen.
The dehydration according to the invention is expediently carried out using mineral acids, such as hydrochloric or hydrobromic acid, it being possible to work in an anhydrous or aqueous medium. The dehydration is preferably carried out in ethanolic hydrochloric acid at a temperature between room temperature and the boiling point of the reaction mixture. However, it can also be achieved by heating, e.g. B. at 50 to reflux temperature, preferably at reflux temperature, with a high-boiling, anhydrous solvent such as dimethyl sulfoxide. Other common dehydrating agents can also be used, e.g. B. sulfuric acid, phosphorus oxychloride, zinc chloride or potassium bisulfate, e.g.
