Es ist bekannt, dass zwischen dem Vorhandensein von Zahnbelag auf menschlichen Zähnen und dem Auftreten von Karies ein enger Zusammenhang besteht. Je länger Zahnbelag auf den Zähnen lagert, desto wahrscheinlicher ist die Bildung von Kariesläsionen im unter dem Zahnbelag befindlichen Zahnschmelz. Das Entfernen von Zahnbelag oder das Verhinden der Entstehung desselben ist deshalb nicht nur aus hygienischen und kosmetischen Gründen von Bedeutung, sondern trägt auch zur Verminderung des Auftretens von Karies bei.
Es wurden deshalb bereits zahlreiche Mittel zum Entfernen von Zahnbelag oder zum Verhindern der Bildung desselben auf menschlichen Zähnen vorgeschlagen. Neben der Entfernung des verkalkten Belags oder Zahnsteins durch Abkratzen mit scharfkantigen Instrumenten werden auch starke Schleifmittel zur Entfernung des Belags benutzt. Derartige Massnahmen haben sich aufgrund der am Zahnschmelz auftretenden starken Abrasion als unzweckmässig erwiesen.
Neben der Anwendung von komplexbildenden Substanzen wie beispielsweise Äthylendiaminotetraessigsäure oder deren wasserlöslichen Salzen oder Phosphonsäuren, die den Zahnbelag mittels Komplexbildung in Lösung bringen sollen, wurde auch bereits die Anwendung von Enzymen für diesen Zweck vorgeschlagen. Die bisher zum Einsatz gekommenen Enzyme brachten jedoch keinen zufriedenstellenden und vollständigen Abbau des menschlichen Zahnbelags.
Durch A. J. Gibbons und S. B. Banghart wurde in Arch.
Oral. Biol. Vol. 12, 11-25 (1967) die Zusammensetzung von Zahnbelag untersucht und extracelluläre Dextrane als wichtiger Bestandteil des kariogenen menschlichen Zahnbelags identifiziert. Diese Erkenntnis wurde durch die Synthese des gleichen extracellulären Dextrans durch in der menschlichen Mundhöhle vorkommende kariogene Streptococcusstämme erhärtet. Diese Autoren fanden weiterhin, dass sich die im menschlichen Zahnbelag vorkommenden extracellulären Polysaccharide mit einem hohen Dextrangehalt durch das Enzym Dextranase aus Penicillium funiculosum (N. R. R. L. 1768) weitgehend abbauen lassen.
Die gleiche Erkenntnis liegt auch der offengelegten niederländischen Patentanmeldung 6 809 687 zugrunde, die sich auf orale Präparate und Mittel zum Entfernen und Bekämpfen der Bildung von Zahnbelag auf Zähnen und Zahnfleisch bezieht und durch den Einsatz von Dextranase, insbesondere solcher aus verschiedenen Penicilliumarten, gekennzeichnet ist.
Es wurde nun gefunden, dass eine erhöhte Schnelligkeit des Abbaus von menschlichem Zahnbelag in vitro und auch in vivo und damit eine erhöhte Kariesprophylaxe dadurch erreicht werden kann, wenn man anstelle des für diesen Zweck bereits empfohlenen Enzyms Dextranase Enzymgemische und/oder Enzymkomplexe aus Dextranasen und weiteren Carbohydrasen oder Dextranasen und Proteasen oder Dextranasen, sonstigen Carbohydrasen und Proteasen verwendet.
Die verstärkte Wirksamkeit solcher Enzymkomplexe oder -gemische beruht wohl auf der Tatsache, dass die kariogenen, extracellulären Polysaccharide im Zahnbelag neben Dextranasen noch andere Polysaccharide enthalten und deshalb durch die Mitverwendung von weiteren Carbohydrasen ein schnellerer und vollständigerer Abbau zu wasserlöslichen Spaltprodukten erzielt wird. Das ist insbesondere bei der Einarbeitung solcher Enzyme in die üblichen Zahnpfiegemittel, die ja nur verhältnismässig kurze Zeit im Zahn- und Mundbereich verbleiben, von erhöhter Bedeutung.
Die Mitverwendung proteolytischer Enzyme ist einmal dann zweckmässig, wenn zusätzlich zur Entfernung des Zahnbelags noch ein Abbau von eiweisshaltigen Speiseresten im Zahn- und Mundbereich erreicht werden soll, zum anderen ist bekannt, dass die Plaquematrix neben Polysacchariden Proteine enthält, die durch Einwirkung von Proteasen in niedermolekulare wasserlösliche Bruchstücke abgebaut werden und dadurch die kariogenen Substrateinlagerungen für die Einwirkung von Dextranase und den anderen Carbohydrasen freigemacht werden.
Die erfindungsgemäss in Mund- und Zahnpflegemitteln zum Einsatz gelangenden Dextranasen, sonstigen Carbohydrasen und/oder Proteasen können aus den für diese Zwecke beschriebenen Mikroorganismen gewonnen werden.
Mit Vorzug einsetzbare Carbohydrasen sind a-Amylase, ss-Amylase, Glucoamylase, Cellulase, Laminarinase, Dextrin1,6-glucosidase sowie die verschiedenen Glucosidasen wie aund ss-Glucosidase, a- und ,B-Galactosidase und a- und ss Fructosidase. Besonders geeignet sind dextranasehaltige Enzymkomplexe und/oder Enzymgemische, die Amylasen wie a-Amylase und ss-Amylase sowie Glucoamylase (a-1,4 Glucanglucohydrolase) enthalten. Als beispielhaft zur Herstellung von Dextranasen allein oder Dextranasen, weitere Carbohydrasen und/oder Proteasen enthaltenden Enzymkomplexen seien hier die Gattungen Aspergillus, beispielsweise A. niger, Penicillium, wie P. funiculosum, P. lilacinum, p. verryculosum, Cellvibrio, Cytophaga und Spicaria, wie Sp.
violacea, genannt. Wenn Enzymgemische aus Dextranasen, weiteren Carbohydrasen und Proteasen Einsatz finden sollen, so sind neben den obenerwähnten Dextranasen und/oder sonstigen Carbohydrasen liefernden Mikroorganismen zur Gewinnung von sonstigen Carbohydrasen und/oder Proteasen beispielsweise noch Pilze der Gattungen Aspergillus, ins besorlliere A. oryzae, A. aureus, A flavus, A. awamori oder A. ne zili, Alternaria Rhizopus, Mucor, Monilia, Gliocladium, Neui pora, Trichoderma oder Penicillium, verschiedene Hefesl.,mme, sowie Bakterienstämme, beispielsweise der Gattungen Streptomyces und Bacillus wie B. subtilis, B. dia staticuE B. amyloliquefaciens, B. stearothermophilus, B. mo riguchiensis oder B.
mesentericus, geeignet. Weitere Proteasen sind auch noch solche tierischen oder pflanzlichen Ursprungs, beispielsweise Pepsin, Trypsin, Papain, Bromelin oder Ficin.
Die überlegene Wirkung der erfindungsgemäss eingesetzten Enzymkomplexe oder -gemische hinsichtlich des Abbaus der im menschlichen Zahnbelag vorhandenen hochmolekularen Bestandteile wurde durch den folgenden Vergleichsversuch bewiesen:
Kariogene Streptokokkenstämme, die aus menschlichem Zahnbelag isoliert worden waren, wurden in einem Saccharosenährmedium kultiviert und die gebildeten extracellulären Polysaccharide isoliert.
Eine genaue Beschreibung der Arbeitsmethode findet sich bei R. J. Gibbons und S. B. Bangart in Archives of Oral Biology , Vol. 12, Nr. 1, Seite 11-24 (1967).
Jeweils 4 mg der so gewonnenen Substanz, die, wie aus der Literatur bekannt, hinsichtlich ihrer Zusammensetzung den im menschlichen Zahnbelag vorhandenen Substanzen entspricht, wurden mit jeweils 1 mg/ml Enzym 2 Stunden bei 37 C und pH 5,6 inkubiert, wobei das Gemisch alle 15 Minu- ten für 30 Sekunden durchgerührt wurde. Der Abbau wurde chromatographisch verfolgt, wobei folgendes Ergebnis erzielt wurde:
Dextranase, die durch Züchtung eines Hefestammes der Gattung Lipomyces starkeyi erhalten wurde und keine Nebenaktivitäten aufwies, zeigte nur einen ganz geringen Abbau der in vitro hergestellten Modellsubstanz für menschlichen Zahnbelag.
Mit einem aus dem gleichen Hefestamm gewonnen Enzymkomplex, der neben Dextranaseaktivität noch Levanaseund a-Amylaseaktivität aufwies, konnte hingegen ein vollständiger Abbau zu wasserlöslichen, niedermolekularen Produkten erreicht werden.
Mit a-Amylase der gleichen Herkunft ohne Nebenaktivi tät wurde erwartungsgemäss überhaupt keine Hydrolyse der durch kariogene Streptokokken erzeugten extracellulären Polysaccharide erreicht.
Die Zahn- und Mundpflegemittel nach der Erfindung mit einem Gehalt an Dextranasen, sonstigen Carbohydrasen und/ oder Proteasen können in Form von Zahnpasten, Mundwässern, Zahnpulvern, Zahnlotionen, Zahnreinigungs-, Kau- und Lutschtabletten oder -dragees oder als Kaugummi zur Anwendung kommen, also praktisch in jeder mit den Zähnen in Berührung kommender Applikationsform.
Die Zahn- und Mundpflegemittel nach der vorliegenden Erfindung können die an sich bekannten und üblichen Aufbau- und Zusatzstoffe enthalten. Als Putzkörper in Zahnpasten und Zahnpulvern haben sich beispielsweise Calciumcarbonat, kolloidale Kieselsäure, Dicalciumphosphat, unlösliche Alkalimetaphosphate, Aluminiumoxid und/oder dessen Trihydrat, Kaolin, Bentonit und pulverförmige Kunststoffe wie Polyamide, Polyvinylchlorid, Polymethylmethacrylat, pulverförmige Epoxyharze, Polyäthylen oder Polypropylen als geeignet erwiesen.
Als Binde- und Verdickungsmittel in Zahnpasten sind Stärke, Tragant Methylcellulose, Alginate, Polyacrylsäure, Polyvinylpyrrolidon, Pektin und Carboxymethylcellulose zu nennen.
Geeignete Feuchthaltemittel sind Glycerin, 1,3-Propandiol, Sorbit und andere Zuckeralkohole sowie Polyäthylenoxide verschiedener Kettenlängen.
Weiterhin können oberflächenaktive Substanzen wie Al kylarylsulfon ate, Fettalkoholsulfate, Olefinsulfonate, Fettsäurekondensationsprodukte, Äthylenoxidkondensate mit Alkoholen oder Aminen, Aminoxide sowie quaternäre Ammoniumverbindungen mitverwendet werden. Auch Amphotenside können, gegebenenfalls im Gemisch mit den anderen aufgezählten oberflächenaktiven Substanzen, in den erfindungsgemässen Zahn- und Mundpflegemitteln Verwendung finden.
Zweckmässig ist weiterhin der Zusatz eines Aromastoffes, wobei verschiedene ätherische Öle wie Pfefferminzöl. Eukalyptusöl, Wintergrün-, Zimt- oder Nelkenöl verwendet werden können. Besonders zweckmässig ist der Einsatz terpenfreier oder zumindest terpenarmer Aromastoffe. Die Aromastoffe können auch an feingepulverten Kunststoffen absorbiert sein, die gleichzeitig als Poliermittel Gebrauch finden können. Dadurch wird ein verhältnismässig lang andauernder Nachgeschmack im Mund nach dem Gebrauch des Zahnund Mundpf'lcgemittels erhalten.
Zur Konse r vierung der erfindungsgemässen Zahn- und Mundpflegenettel dienen bevorzugt Ester der p-Hydroxybenzoesäure. doch können auch alle anderen bisher für diesen Zweck X orgeschlagenen Substanzen Verwendung finden.
Neben den erfindungsgemäss in den Zahn- und Mund pflegemittehl enthaltenen Enzymkomplexen und/odel -gemischen aus lJextranasen, weiteren Carbohydrasen und/oder Proteasen können die Zahn- und Mundpflegemittel noch weitere Stoffe enthalten. Als Beispiele seien verschiedene Vitamine sowie inbesondere Fluorverbindungen genannt die geeignet sind. d kariesprotektive Wirkung der Enzymkolnbina- tion noch zu l. erstützen. Geeignete Fluorverbindungen sind beispielswx cise Alkalifluoride, wie Natriumfluorid.
Kaliumfluorid, Lithiumfluorid und/oder Ammoniumfluorid, Zinnfluorid, Indiumfluorid, Zirkonfluorid, Alkalimonofluorphosphate, wie Ammonium-, Natrium-, Kalium- und/oder Lithiummonofluorphosphat, Fluorsilikate oder Fluorstannite.
Es können auch organische Fluorverbindungen, wie Aminhydrofluoride, Alkanolaminhydrofluoride, quaternäre Ammoniumfluoride oder Additionsverbindungen von Aminosäuren mit Fluorwasserstoff oder Fluoriden eingesetzt werden, wobei der Mengenanteil bis zu einem Fluorgehalt von 0,15 Gew. %, bezogen auf das Gesamtgewicht des Mittels, betragen kann.
Möglich und unter gewissen Bedingungen auch zweck mässig ist weiterhin die Mitverwendung organischer wasser löslicher Phosphate, beispielsweise Zuckerphosphaten, sowie von anorganischen wasserlöslichen Phosphaten. Grundvoraus setzung für die Mitverwendung aller aufgezählten Zusatz stoffe ist selbstverständlich ihre Eigenschaft, auf die Enzym gemische und/oder -komplexe aus Dextranasen, weiteren 3 Carbohydrasen und/oder Proteasen nicht inaktivierend zu wirken. Auch der Zusatz weiterer zur Verwendung in Zahn pflegemitteln an sich bekannter Enzyme, beispielsweise Lipa sen, Oxidasen oder Oxidoreductasen, ist möglich.
Der optimale pH-Bereich für die erfindungsgemässen
Zahn- und Mundpflegemittel liegt bei etwa 4,5 bis etwa 7,0.

The use of proteolytic enzymes is advisable if, in addition to removing plaque, a breakdown of protein-containing food residues in the tooth and mouth area is to be achieved; on the other hand, it is known that the plaque matrix contains, in addition to polysaccharides, proteins that are converted into low-molecular proteins by the action of proteases water-soluble fragments are broken down and thereby the cariogenic substrate deposits are freed for the action of dextranase and the other carbohydrases.
The dextranases, other carbohydrases and / or proteases used in oral and dental care products according to the invention can be obtained from the microorganisms described for these purposes.

Dextranase, which was obtained by cultivating a yeast strain of the genus Lipomyces starkeyi and which had no side activities, showed only very little degradation of the model substance for human dental plaque produced in vitro.
With an enzyme complex obtained from the same yeast strain, which, in addition to dextranase activity, also had levanase and α-amylase activity, on the other hand, it was possible to achieve complete degradation to water-soluble, low-molecular-weight products.
As expected, no hydrolysis of the extracellular polysaccharides produced by cariogenic streptococci was achieved with α-amylase of the same origin without any secondary activity.

The dentifrice and oral care products according to the present invention can contain the usual build-up substances and additives which are known per se. Calcium carbonate, colloidal silicic acid, dicalcium phosphate, insoluble alkali metaphosphates, aluminum oxide and / or its trihydrate, kaolin, bentonite and powdered plastics such as polyamides, polyvinyl chloride, polymethyl methacrylate, powdered epoxy resins, polyethylene or polypropylene have proven to be suitable as cleaning agents in toothpastes and tooth powders.
The binding and thickening agents in toothpastes include starch, tragacanth, methylcellulose, alginates, polyacrylic acid, polyvinylpyrrolidone, pectin and carboxymethylcellulose.
Suitable humectants are glycerine, 1,3-propanediol, sorbitol and other sugar alcohols and polyethylene oxides of various chain lengths.
Surface-active substances such as alkylarylsulfonates, fatty alcohol sulfates, olefin sulfonates, fatty acid condensation products, ethylene oxide condensates with alcohols or amines, amine oxides and quaternary ammonium compounds can also be used. Amphoteric surfactants can also be used in the dental and oral care agents according to the invention, if appropriate in a mixture with the other surface-active substances listed.
It is also advisable to add a flavoring substance, with various essential oils such as peppermint oil. Eucalyptus oil, wintergreen, cinnamon or clove oil can be used.
