Gegenstand der Erfindung ist ein Verfahren zur Herstellung neucr Cycloadditions-Verbindungen, welches dadurch gekennzeichnet ist, dass man ein am Kern gegebenenfalls substituiertes Vinylpyridin mit einer dienophilen Verbindung umsetzt. Die Erfindung umfasst ferner die nach dem Verfahren erhaltenen Cycloadditions-Verbindungen sowie deren Verwendung zur Herstellung ihrer Spaltprodukte.
Vinylpyridine sind als Stickstoff-Analoga des Styrols polymerisationsfreudige Verbindungen. Mit andern ungesattigten Substanzen reagieren Sie unter Bildung von
Mischpolymerisaten Überraschenderweise hat sich nun gezeigt, dass Umsetzungen von Vinylpyridinen mit Dienophilen, namentlich in Gegenwart eines Polymerisationsinhibitors, nicht zu Polymeren. sondern zu Cycloadditionsprodukten, heispielsweise zu 1:2- oder 1: :3-Additionsverbindungen führen.
So liefert z.B. 4-Vinylpyridin mit N-substituierten Maleinimiden 1 :3-Addukte der im nachfolgenden Formelschema wiedergegehenen Struktur A. 2-Vinvlpyridine neben 1:3
Addukten auch 1 :2-Addukte der Struktur B.
In den Formeln bedeutet R aliphatische. carbo- oder heterocyclische Reste. insbesondere Alkyl oder Phenyl.

zu deren Bildung die Vinylgruppe des 4-Vinylpyridins gemeinsam mit der dazu konjugierten Doppelbindung des Pyridinkerns mit dem Dienophil reagiert, in ähnlicher Weise, wie dies zum ersten Mal am Beispiel des asymetrischen Diphenyläthylens gezeigt wurde (vgl. Wagner-Jauregg, Ber.

Organischen Chemie, 3. Aufl., Heidelberg 1966, p. 620). Man würde erwarten, dass A' unter Verschiebung einer Dopuelbin- dung in das stabile (aromatische) Pyridinderivat übergeht.
Dies ist aber nicht der Fall, sondern A' reagiert als zyklische
Schiff'sche Base mit 2 Molen eines Dienophils unter Bildung von Spiroverbindungen weiter, ähnlich wie dies in der schweiz.
Patentschrift Nr. 529 152 aufgezeigt ist.
Die Verbindungen der Struktur A lassen sich durch Säuren in Substanzen vom Typus C plus D1 (bzw. D2) spalten. Alle angegebenen Strukturformeln sind mit den NMR-Spektren vereinbar.
Die verfahrensgemäss erhaltenen Verbindungen und ihre
Spaltprodukte kommen entweder als solche oder nach weiterer
Umwandlung, beispielsweise nach Reduktion von Carbonyl gruppen zu Methylengruppen, für die Verwendung als Arznei mittel in Betracht.
Beispiel I Atltlition von 3 Mol N-n-Butylmaleinimicl an I Mol 4 Vinylpyriclin:
Eine Lösung von 3,5 g 4-Vinylpyridin, 15 g N-n
Butylmaleinimid und 0,2 g Hydrochinon in 45 ml Acetonitril wurde 65 Stunden lang unter Rühren auf Rückfluss
Temperatur erhitzt. Nach Entfernen des Lösungsmittels im
Vakuum und Umkristallisieren des braunen Rückstandes aus einem Gemisch von Methylendichlorid und Methanol erhielt man 7 g farblose Nadeln vom Smp. 202-203 C.

Säure-Spaltung des so erhaltenen Additionsproduktes A:
Eine Mischung von 2 g des gemäss Beispiel 1 dargestellten
Produktes mit 30 ml konzentrierter Salzsäure (37 %ig) und
60 ml Methanol wurde mit Rühren unter Rückfluss 7 Std.
gekocht. Die beim Abkühlen ausgefallene und nach Absaugen mit Wasser gewaschene Substanz kristallisierte man aus einer
Mischung von Methylendichlorid und Methanol um und erhielt
0,9 glänzende farblose Kristalle vom Smp. 177-180 C.

Die wässrigen sauren Filtrate wurden im Vakuum zur
Trockene eingedampft und der Rückstand mit 30 ml Wasser versetzt. Die Filtration durch eine Celitschicht ergab eine klare
Lösung. die man in einem Eis-Wasser-Bad abkühlte und mit
25%igem Natriumkarbonat stark basisch machte. Nach zweimaliger Extraktion mit je 25 ml Aether, Waschen der
Extrakte mit Wasser, Trocknen über wasserfreiem Magnesium sulfat und Abkühlung auf 0 wurde trockenes Chlorwasser stoff-Gas in die ätherische Lösung eingeleitet und das erhaltene
Hydrochlorid abgesaugt. Umkristallisation aus einem Gemisch von Methanol und Aether lieferte ein farbloses Hydrochlorid vom Smp. 251-255 C.

Beispiel 2 Aclblitionsproclukt von 3 Mol N-n-Propylmaleinimitl an I Mol 4- Vinylpyflclin
4 g 4-Vinylpyridin, 15 g N-Propylmaleinimid und 1 g Hydrochinon erhitzte man in 45 ml Acetonitril mit Rühren unter Rückfluss während 62 Std. Das Lösungsmittel wurde im Vakuum entfernt und die verbleibende braune, gummiartige Masse an 300 g neutralem Aluminiumoxyd chromatographiert.
Die Elution mit Benzol-Methylendichlorid (1:1) ergab eine klebrige Masse, die nach Durchrühren mit Aether abgesaugt werden konnte und bei Umkristallisieren aus Methylendichlorid-Methanol 2,5 g eines farblosen mikrokristallinen Produktes vom Smp. 216-218 C ergab.

3 g 4-Vinylpyridin, 5 g N-Phenylmaleinimid und 0,5 g
Hydrochinon wurden in gleicher Weise wie im Beispiel 2 erhitzt. Nach Entfernen des Lösungsmittels im Vakuum wurde der rötliche Rückstand mit 80 ml Methanol eine Stunde lang gerührt und dann abgesaugt. Den rötlichen Rückstand (4 g) kristallisierte man aus einem Gemisch von Acetonitril mit
Methanol um und erhielt 2 g eines mikrokristallinen Adduktes vom Smp. 290-293 "C.

Beispiel 4 Umsetzung von 2 Mol N-n-Bzltylmaleinimitl mit I Mol 2 Vinylpyri(lin:
Eine Lösung von 1,7 g (16,3 mmol) 2-Vinylpyridin, 5 g N
Butylmaleinimid (32,7 mmol) und 0,35 g 4-tert.-Butyl brenzcatechin in 20 ml n-Butanol wurde unter Rühren am
Rückfluss 40 Stunden lang erhitzt. Die nach Abdampfen des
Lösungsmittels im Vakuum verbleibende gummiartige Masse chromatographierte man über 75 g neutralem Aluminiumoxyd.
Die Elution mit Methylendichlorid ergab ein dunkles gummi artiges Produkt, das beim Durchkneten mit einem Gemisch von
Hexan-Aether eine weisse Masse ergab. Die Umkristallisation aus Methylendichlorid-Hexan lieferte 0,4 g farblose flockige
Kristalle vom Smp. 186-188 .

Beispiel 5 Ablblitionsprodukt von 2 Mol N-n-Butyl-maleinimid an I Mol 2 Vinyl-5-ärhylpyri < fin:
2,2 g 2-Vinyl-5-äthylpyridin, 5 g N-Butylmaleinimid und 0,35 g 4-tert.-Butyl-brenzcatechin wurden in 20 ml n-Butanol während 25 Stunden unter Rühren auf Rückflusstemperatur erhitzt. Nach Abdampfen des Lösungsmittels im Vakuum und Aufnehmen des dunklen Rückstandes in Aether fielen weisse Kristalle aus, die nach Umkristallisieren aus einem Methy lenchlond-Hexan-Gemisch 0,6 g Addukt in Form glänzend weisser Kristalle vom Smp. 182-184 C lieferten.

Elution with benzene-methylene dichloride (1: 1) gave a sticky mass which, after stirring through with ether, could be suctioned off and, on recrystallization from methylene dichloride-methanol, gave 2.5 g of a colorless microcrystalline product with a melting point of 216-218 ° C.
