Es ist bekannt, dass gewisse partiell hydrierte Phenanthridine. welche in 9-Stellung mit einer freien oder einer verätherten Hidroxygruppe substituiert sind, eine pharmakologische, und zwar insbesondere eine analgetische Wirkung, besitzen.
Es wurde nun gefunden, dass die pharmakologische Aktivität derartiger Phenanthridine wesentlich erhöht ist, wenn das Kohlenstoffatom in Stellung lOb mit einer niederen Alkylgruppe. einer niederen Cycloalkylalkylgruppe oder einer niederen Alkenylgruppe verknüpft ist.
Diese Aktivitätserhöhung sei anhand der folgenden Beispiele veranschaulicht:
Es wurde die analgetische Wirkung mittels des Writhing Tests [modifizierte Methode von E. Siegismund et al., Proc.
Soc. Exp. Biol. Med. 95, 729-31(1957)] und die Toxizität der folgenden Substanzen an Mäusen bestimmt:
Substanz A = 9-Methoxy-1,2,3,4,4a,5,6,10b- ctahdrophenanthridin-hydrochlorid.
Substanz B = 9-Methoxy-1 Ob-methyl- 1 ,2,3,4,4a,5,6, lOb- octahvdrophenanthridin-hydrochlorid.
Substanz C = 5-Methyl-9-methoxy- 1,2,3,4,4a,5,6,10b- octahydrophenanthridin-hydrochlorid.
Substanz D = 5,1 Ob-Dimethyl-9-methoxy- 1,2,3 l 4a,5,6,1 Ob-octahydrophenanthridin-hydrochlorid .

Aus den obigen Zahlenwerten ersieht man, dass die Aktivitätserhöhung nicht von einem proportionalen Anstieg der Toxizität begleitet ist. Dies heisst mit anderen Worten, dass der therapeutische Index einer Verbindung mit einem quart ären Kohlenstoffatom in Stellung 10b wesentlich günstiger liegt als der therapeutische Index der entsprechenden am Kohlenstoffatom 10b nicht substituierten Verbindung.

Erwünschtenfalls wird eine im aromatischen Ring einer solchen Verbindung vorhandene Alkoxygruppe R40 in die Hydroxygruppe überführt, eine so erhaltene Octahydrophenanthridinverbindung erwünschtenfalls am Stickstoff mit einer Alkylgruppe oder Cycloalkylalkylgruppe substituiert und eine so erhaltene Octahydrophenanthridinverbindung gegebenenfalls in ein Salz übergeführt und bzw. oder, im Falle eines Racemates, dieses in die optischen Antipoden aufgespalten.
Die erfindungsgemäss hergestellten Octahydrophenanthridine können, falls R2 Wasserstoff ist, am Stickstoff in an sich bekannter Weise mit einer niederen Alkyl- oder Cycloalkylalkylgruppe substituiert werden.
Auch kann, falls erwünscht, eine im aromatischen Ring einer so erhaltenen Octahydrophenanthridinverbindung enthaltene Alkoxygruppe oder Aralkyloxygruppe in an sich bekannter Weise in die Hydroxygruppe übergeführt werden.
Ferner kann ein so erhaltenes Racemat in an sich bekannter Weise in die optischen Antipoden aufgespalten werden.
Schliesslich können die erhaltenen Octahydrophenanthridinbasen in Säureadditionssalze übergeführt werden.
Durch Cyclisierung von Verbindungen der Formel II sowie durch allfällige Nachfolgeoperationen kann man somit racemische oder optisch aktive Octahydrophenanthridinderivate der Formel I bzw. Salze dieser Verbindungen, erhalten.
Durch Cyclisierung von Verbindungen der obigen Formel II und eventuelle nachfolgende Überführungen einer im aromatischen Ring des Cyclisierungsproduktes enthaltenen Alkoxy- gruppe oder Aralkyloxygruppe in die Hydroxygruppe gelangt man direkt zu Octahydrophenanthridinverbindungen der Formel I, worin R4 die oben angegebene Bedeutung besitzt bzw. nach erfolgter Ätherspaltung durch Wasserstoff ersetzt ist.
Die auf diese Weise erhaltenen Octahydrophenanthridine können gewünschtenfalls den folgenden Operationen unterworfen werden: Substitution am Stickstoff, Aufspaltung eines Racemates in die optischen Antipoden und Salzbildung.
Die Cyclisierung der Verbindungen der Formel II kann in einfacher Weise mittels eines sauren Agens durchgeführt werden. Als saures Agens können die üblichen Mineralsäuren oder organischen Säuren verwendet werden. Vorzugsweise wird als saures Agens Salzsäure verwendet. Bei Durchführung der Cyclisierung mit einem von Ameisensäure verschiedenen sauren Agens wird die Bedeutung des Symbols R2 nicht verän dert, d. h. R2 hat im Cyclisierungsprodukt dieselbe Bedeutung wie im Ausgangsmaterial der Formel II. Wenn man hingegen die Cyclisierung mittels Ameisensäure ausführt und hierbei ein Ausgangsmaterial der Formel II verwendet, in welchem R2 Wasserstoff bedeutet, so erhält man ein Cyclisierungsprodukt der Formel I, in welchem R2 die Methylgruppe bedeutet.
Wenn die Cyclisierung in Abwesenheit eines sauren Mittels durchgeführt wird, so können bis-Verbindungen der Formel

als Nebenprodukte isoliert werden. Die Säurebehandlung dieser bis-Verbindungen der Formel III führt zu deren Spaltung.
Die bei der Definition der Substituenten Rl, R2 und R4 verwendeten Begriffe Alkyl-, Alkoxy- und Cycloalkylalkylgruppen beziehen sich, wie gesagt, auf solche Gruppen, welche bis zu sieben Kohlenstoffatome enthalten, wie die Methyl-, Äthyl-, Propyl-, Isopropyl- und Butylgruppe; die Cyclopropylmethyl-, die Cyclobutylmethyl-, die Cyclopentylmethyl-, die Cyclopropyläthyl- und die Cyclobutyläthylgruppe sowie die Methoxy- Äthoxy- und Isopropyloxygruppe. Bevorzugte Vertreter hiervon sind Methyl, Äthyl und Cyclopropylmethyl sowie Methoxy und Äthoxy.
Unter Alkenylgruppen sind Alkenylgruppen mit bis zu vier Kohlenstoffatomen zu verstehen, insbesondere die Allylgruppe.
Unter Hydroxyalkyl- und Alkoxyalkylgruppen sind Gruppen zu verstehen, deren Alkyl- bzw. Alkoxyreste bis zu vier Kohlenstoffatome enthalten, beispielsweise Hydroxymethyl, Hydroxyäthyl, Hydroxypropyl und Methoxymethyl, Methoxy äthyl, Methoxypropyl, Athoxymethyl, Äthoxyäthyl, Äthoxypropyl und dergleichen.

Bei Verwendung dieser Ausgangsmaterialien erhält man das 9-Methoxy-1Ob-methyl-1,2,3,4,4a,5,6, 1 Ob- octahydro-phenanthridin bzw. Salze davon. Diese Base und deren Salze, insbesondere die das polarisierte Licht in methanolischer Lösung linksdrehenden Antipoden, zeichnen sich durch besonders vorteilhafte analgetische Wirkung aus.

zum entsprechenden Amin der Formel IV bewerkstelligt werden. Die Reduktion des Oxims der Formel VI erfolgt hierbei zweckmässig katalytisch, unter Verwendung eines Raney Nickel- oder Palladium-Katalysators oder mittels Lithiumaluminiumhydrid.
Die Amine der Formel IV können aber auch durch Umsetzung der entsprechenden Ketone der Formel V mit Ammoniak und Reduktion der dabei gebildeten Iminoverbindungen erhalten werden. Diese Reduktion wird zweckmässig katalytisch, unter Verwendung eines Platin- oder Palladiumkatalysators, oder mittels Natriumborhydrid durchgeführt.
Die Herstellung der Ketone der Formel V wird im folgenden anhand eines Beispiels, nämlich der Herstellung von 2 Methyl-2-(3'-methoxy-phenyl)-cyclohexanon beschrieben:
Es wird m-Methoxy-ss-nitro-styrol mit 1,3-Butadien zum 4 (3 -Methoxy-phenyl)-5-nitro-cyclohexen umgesetzt. Diese Verbindung wird mit Natriumalkoholat zur Reaktion gebracht, wobei 2-(3 -Methoxy-phenyl)-cyclohexanon gebildet wird, welches unter Verwendung eines Palladium-Kohle-Katalysators zum 2-(3 -Methoxy-phenyl)-cyclohexanon hydriert wird.
Diese Verbindung wird mit Methyljodid umgesetzt, wobei man 2-Methyl-2-(3 -methoxy-phenyl)-cyclohexanon erhält. Die übrigen Ketone der Formel V können in analoger Weise erhalten werden.
Die erfindungsgemäss erhältlichen in 9-Stellung mit der Hydroxygruppe, einer niederen Alkoxygruppe, einer niederen Hydroxyalkoxygruppe oder einer niederen Alkoxyalkyloxygruppe substituierten, partiell hydrierten Phenanthridinderivate. deren Kohlenstoffatom in Stellung 10b durch Verknüpfung mit einem monovalenten Substituenten quartär gemacht isi, besitzen, wie bereits angeführt wurde, eine analgetische Wirkung. Ferner wurde bei derartigen Verbindungen eine hustenhemmende, entzündungshemmende und antipyretische Wirkung festgestellt.
Es wurde festgestellt. dass bei der Aufspaltung einer racemischen Verbindung in die optischen Antipoden eine Disproportionierung des Wirkungsgrades eintritt. So wurde beispielsweise gefunden, dass das in methanolischer Lösung das polarisierte Licht linksdrehende 9-Methoxy- 1 Ob-methyl-1,2,3,4,4a,5,6,1 Ob- octahydro-phenanthridin eine bedeutend höhere Wirksamkeit aufweist als der entsprechende rechtsdrehende Antipode und das racemische Gemisch.
Die Verfahrensprodukte können als Heilmittel in Form pharmazeutischer Präparate Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen inerten Trägermaterial enthalten.
Die pharmazeutischen Präparate können in fester Form oder in flüssiger Form vorliegen. Gegebenenfalls enthalten sie Hilfs- stoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Beispiel I
Es werden 14,5 g 2-Methyl-2-(3 -methoxy-phenyl)-cyclohexylamin in 73 ml Methanol gelöst, worauf der Lösung 18 ml einer 38obigen wässrigen Formaldehydlösung zugesetzt werden und das Reaktionsgemisch 14 Stunden stehen gelassen wird. Der nach Eindampfen unter vermindertem Druck erhaltene Rückstand wird in Alkohol gelöst und die Lösung wird mit alkoholischer Salzsäure bis zur kongosauren Reaktion versetzt. Beim Einengen unter vermindertem Druck erhält man 9-Methoxy-1 Ob-methyl-l ,2,3,4,4a,5,6,10b- octahydro-phenanthridin-hydrochlorid, welches nach Umkristallisation aus Alkohol-Äther bei 219 2200C schmilzt.
Das hierbei als Ausgangsmaterial verwendete 2-Methyl-2 (3 -methoxy-phenyl)-cyclohexylamin kann wie folgt hergestellt werden:
Es werden 13 g 2-Methyl-2-(3 -methoxy-phenyl)-cyclohexanonoxim in 150 ml Methanol gelöst, worauf der Lösung eine Lösung von 3 g Kaliumhydroxyd in 5ml Wasser zugesetzt wird. Dann wird bei etwa 40OC in Gegenwart von 20 g Raney-Nickel hydriert. Die Wasserstoffaufnahme erfolgt hierbei sehr rasch.
Nach Abtrennung des Katalysators und Abdampfen des Methanols unter vermindertem Druck wird der Rückstand in Äther aufgenommen und die ätherische Lösung mit Wasser gewaschen und zur Trockene eingedampft. Der Rückstand wird über das 2-Methyl-2-(3'-methoxy-phenyl)-cyclohexyl- amin-hydrochlorid, welches spontan aus Äthanol auskristalli siert und bei 245-2460C schmilzt, gereinigt. Durch Alkalisch stellen dieses Salzes wird die freie Base erhalten.
Das hierbei als Ausgangsmaterial verwendete
2-Methyl-2-(3'-methoxy-phenyl)-cyclohexanonoxim kann wie folgt erhalten werden:
150 g m-Methoxy-ss-nitro-styrol werden zusammen mit einer Lösung von 150 g 1,3-Butadien in 1500 ml Chloroform nach Zugabe von 750 ml Hydrochinon in einem Autoklaven in einer Stickstoffatmosphäre unter einem Druck von 20 Atmosphären 24 Stunden auf 14( > C erhitzt. Das Reaktionsprodukt wird filtriert und das klare Filtrat wird unter vermindertem Druck zur Trockne eingedampft. Der Eindampfrückstand wird in Äther aufgenommen und die ätherische Lösung wird nacheinander mit verdünnter Natronlauge, Salzsäure und Wasser gewaschen.
Nach Trocknen über Natriumsulfat und Abdampfen des Äthers kristallisiert man den erhaltenen Rückstand aus einem Gemisch von Isopropyläther und tiefsiedendem Petrol äther um, wobei man 4-(3 '-Methoxy-phenyl)-5 -nitro -cyclohe- xen-(l) erhält, das bei 74-75oC schmilzt.
Zu einer Lösung von 282 g des nach den obigen Angaben erhaltenen 4-(3'-Methoxy-phenyl)-5-nitro-cyclohexen-(1) in 1500 ml wasserfreiem Äthanol wird, in einer Stickstoffatmosphäre, eine äthanolische Natriumäthylatlösung zugetropft, welche aus 51 8 Natrium und 1200 ml absolutem Athanol hergestellt wurde. Bei der Zugabe der Natriumäthylatlösung scheidet sich ein dicker Brei ab, der während 15 Stunden gut gerührt wird. Hierauf wird das Reaktionsgemisch unter Stickstoff langsam einem auf -50C gekühlten Gemisch von 15 kg Eis, 12 Liter Äthanol und 1500 ml Salzsäure zugegeben, wobei kräftig gerührt wird.
Nach weiterem Rühren bei O und während einer Stunde und während des Ansteigens der Temperatur des Reaktionsgemisches auf Raumtemperatur wird dieses mit Wasser verdünnt und mit Methylenchlorid extrahiert. Der Methylenchloridextrakt wird getrocknet und unter vermindertem Druck eingedampft. Der Eindampfrückstand wird in Methanol aufgenommen und in Gegenwart von 50 g eines Palladium-Kohle-Katalysators bei etwa 40C hydriert. Nach Aufnahme der berechneten Wasserstoffmenge wird der Katalysator abfiltiert und das Filtrat durch Eindampfen vom Methanol befreit. Der Rückstand wird im Hochvakuum destilliert, wobei man 2-(3'-Methoxyphenyl)-cyclohexanon erhält; Kpol = 125-1300C.
11,6 g einer 50%gen Suspension von Natriumamid in Benzol werden in einer Stickstoffatmosphäre in 100 ml abs. Benzol eingebracht. Hierauf werden 30,6 g des nach den obigen Angaben erhaltenen 2-(3'-Methoxy-phenyl)-cyclohexanon in 100 ml absolutem Benzol zugesetzt und das Reaktionsgemisch wird 5 Stunden am Rückfluss gekocht. Nach dem Abkühlen wird das Reaktionsgemisch mit 40 g Methyljodid versetzt und 15 Stunden auf 50 C erwärmt. Nach langsamer Zugabe von Wasser wird die benzolische Lösung nacheinander mit verdünnter Natronlauge, Wasser, Salzsäure und nochmals mit Wasser gewaschen. Nach Abdampfen des Benzols wird der Rückstand im Hochvakuum destilliert, wobei man 2-Methyl-2 (3'-methoxy-phenyl)-cyclohexanon (Kpo,l= 116-117 > ) erhält.
11 g des so erhaltenen 2-Methyl-2-(3 -methoxy-phenyl)cyclohexanons in 15 ml Äthanol werden zusammen mit 6 g Hydroxylamin-hydrochlorid, 7 g Natriumacetat und 14 ml Wasser 30 Minuten auf 400C erwärmt. Es scheidet sich hierbei eine ölige Substanz ab, die langsam erstarrt. Durch Umkristallisieren dieser Substanz aus wässrigem Äthanol erhält man 2 Methyl-2-(3'-methoxy-phenyl)-cyclohexanon-oxim vom Schmelzpunkt 89-90 .
Beispiel 2
Es werden 2 g 9-Methoxy-IOb-methyl-1 ,2,3,4,4a,5,6,1Ob- octahydro-phenanthridin (erhalten aus dem gemäss Beispiel 1 gewonnenen Hydrochlorid) mit 60 ml Bromwasserstoffsäure (konstantsiedend) während vier Stunden am Rückfluss erhitzt. Hierauf wird das Reaktionsgemisch unter vermindertem Druck eingedampft, der Rückstand mit verdünnter Natronlauge versetzt, mit Äther extrahiert und mehrmals mit verdünnter Natronlauge und zuletzt mit Wasser gewaschen. Nach dem Abdampfen des Äthers bleibt 9-Hydroxy-lOb-methyl-1,2,3,4,4a,5,6,10b- octahydrophenanthridin zurück, dessen Hydrochlorid bei 297-2980C schmilzt.
Beispiel 3
In Analogie zu Beispiel 2 erhält man aus 6,1 Ob-Dimethyl-9-methoxy-octahydro-phenanthridin das 6,1 Ob-Dimethyl-9-hydroxy-octahydro-phenanthridin, dessen Hydrochlorid bei 302-3030C schmilzt.
Beispiel 4
6,5 g 9-Methoxy-lOb-methyl-1,2,3,4,4a,5,6,10b- octahydro-phenanthridin werden zusammen mit 25 ml Formaldehyd (38%ige wässrige Lösung) und 25 ml Ameisensäure (100%mg) zwei Stunden am Rückfluss gekocht. Das Reaktionsgemisch wird hierauf unter vermindertem Druck eingedampft, der Eindampfrückstand wird mit verdünnter Natronlauge versetzt, in einem Gemisch von Methylenchlorid und Äther aufgenommen und mit Wasser gewaschen. Nach dem Abdampfen des Äthers bleibt 5,10b-Dimethyl-9-methoxy- 1,2,3,4,4a,5,6,10b- octahydro-phenanthridin zurück.
Das Hydrochlorid dieser Base schmilzt bei 221-222 C
Beispiel 5
12 g (+)-2-Methyl-2-(3 > -methoxy-phenyl)-cyclohexylamin werden mit 20 ml Methanol und 6 ml einer 38%igen wässerigen Formaldehydlösung versetzt, worauf das Reaktionsgemisch über Nacht bei Raumtemperatur stehen gelassen wird.
Hierauf wird unter vermindertem Druck eingeengt, das restliche Wasser nach Zugabe von Äthanol azeotrop abdestilliert, der Rückstand mit alkoholischer Salzsäure versetzt und eingedampft. Der Eindampfrückstand wird aus einem Gemisch von Alkohol und Äther auskristallisiert, wobei man ()-9-Methoxy-lOb-methyl-1,2,3,4,4a,5,6,10b- octahydro-phenanthridin-hydrochlorid erhält. Smp. = 219-2200C, [a]25 = -24,920 (c = 1,2 in Methanol).
Durch Chromatographie der Mutterlaugen erhält man das isomere ( - )-9-Methoxy-lOb-methyl-1,2,3,4,4a,5,6,10b- octahydro-phenanthridin-hydrochlorid mit axialer Lage des Wasserstoffatoms in Stellung 4a, welches einen Schmelzpunkt von 268-2690C aufweist, sowie das (-)-S,lOb-Dimethyl-9-methoxy- 1,2,3,4,4a,5,6,10b- octahydro-phenanthridin-hydrochlorid mit einem Schmelzpunkt von 24O2410C.
In analoger Weise kann, ausgehend vom (- )-2-Methyl-2.
(3 -methoxy-phenyl)-cyclohexylamin, das ( + )-9-Methoxy- 10b-methyl-1,2,3,4,4a,5,6,10b- octahydro-phenanthridin-hydrochlorid erhalten werden; Smp. = 2l62l70C, [a]D5 = +21,140 (c = 1,01 in Methanol).
Die hierbei als Ausgangsmaterialien verwendeten optischen Antipoden von 2-Methyl-2-(3/-methoxy-phenyl)- cyclohexylamin können wie folgt gewonnen werden:
Eine Lösung von 4,4 g racemischem 2-Methyl-2-(3/-methoxy-phenyl)-cyclohexylamin (erhalten nach den Angaben in Beispiel 1) in 80 ml Äthanol wird mit einer Lösung von 3 g D-Weinsäure in 20 ml Alkohol versetzt. Aus dem Gemisch kristallisiert das (+ )-2-Methyl-2- (3 -methoxy-phenyl)- cyclohexylamin-tartrat (das in methanolischer Lösung rechtsdrehende Tartrat) aus; Schmelzpunkt 173-l75'C, [a]D = +46,10 (c = 1,26 in Methanol).
Das nach Umkristallisation aus Äthanol erhaltene Salz schmilzt bei 184'C; [a]25 = +59,1' (c = 1,22 in Methanol).
Beim Einengen der Mutterlaugen kristallisiert der (-) Antipode aus; Smp. = 120-130 , [a]Dl = -24,80 (c = 1,39 in Methanol).
Beispiel 6
In Analogie zu Beispiel 1 wird das mit Formaldehyd behandelte 2-Allyl-2-(3!-methoxy-phenyl)-cyclohexylamin zum 9-Methoxy-lOb-allyl- 1,2,3,4,4a,5,6,10b- octahydrophenanthridin cyclisiert, dessen Hydrochlorid bei 214-2150C schmilzt.
Das hierbei als Ausgangsmaterial verwendete 2-Allyl-2-(3/methoxy-phenyl)-cyclohexylamin kann wie folgt erhalten werden:
Es werden 21 g Lithiumaluminiumhydrid in einem Gemisch von 100 ml wasserfreiem Äther und 200 ml wasserfreiem Tetrahydrofuran suspendiert, worauf die erhaltene Suspension tropfenweise mit einer Lösung von 39 g 2-Allyl-2-(3'-methoxy-phenyl)-cyclohexanon-oxim in 150 ml wasserfreiem Tetrahydrofuran versetzt wird. Nach 24-stündigem Kochen am Rückfluss, Zugabe von Äther und Zersetzung des überschüssigen Lithiumaluminiumhydrids mit Wasser, werden die anorganischen Salze durch Filtrieren abgetrennt. Das Filtrat wird hierauf unter vermindertem Druck eingedampft und der Rückstand im Hochvakuum destilliert, wobei bei einem Druck von 0,01 mm 2-Allyl-2-(3'-methoxy-phenyl) -cyclohexylamin bei 120-1220C überdestilliert.
Das Hydrochlorid dieser Base schmilzt bei 210-2110C.
Das hierbei als Ausgangsmaterial verwendete 2-Allyl-2-(3l- methoxy-phenyl)-cyclohexanonoxim kann in Analogie zu den entsprechenden Angaben in Beispiel 1 wie folgt erhalten werden:
3-Methoxy-phenyl-cyclohexanon wird in Gegenwart von Natriumamid mit Allylbromid zum 2-Allyl-2-(3'-methoxyphenyl)-cyclohexanon umgesetzt; Kpo.oos = 109oC. Diese Verbindung wird mit Hydroxylamin zum 2-Allyl-2-(31-meth- oxy-phenyl)-cyclohexanon-oxim umgesetzt; Smp. = 103 1040C.
Beispiel 7
In Analogie zu Beispiel 1 wird das mit Formaldehyd behandelte 2-Propyl-2-(3/-methoxy-phenyl)-cyclohexylamin zum 9-Methoxy-l Ob-propyl-1,2,3,4,4a,5,6, lOb- octahydrophenanthridin cyclisiert, dessen Hydrochlorid bei 239-2400C schmilzt.
Das hierbei als Ausgangsmaterial verwendete 2-Propyl-2 (3;-methoxy-phenyl)-cyclohexylamin kann wie folgt erhalten werden:
52 g 2-Allyl-2-(3 -methoxy-phenyl)-cyclohexanon-oxim werden in Methanol in Gegenwart von Raney-Nickel und einer Lösung von 11,2 g Kaliumhydroxyd in 11 ml Wasser hydriert. Nach Aufnahme der berechneten Wasserstoffmenge wird der Katalysator durch Filtrieren abgetrennt und das Filtrat unter vermindertem Druck eingedampft. Der Rückstand wird in Äther aufgenommen und die ätherische Lösung wird mit Wasser neutral gewaschen. Der nach dem Abdampfen des Äthers verbleibende ölige Rückstand wird mit alkoholischer Salzsäure behandelt.
Man erhält 2-Propyl-2-(3 -methoxy phenyl)-cyclohexylamin-hydrochlorid mit einem Schmelzpunkt von 239-240'C. Durch Alkalischstellen dieses Salzes erhält man die freie Base.
Beispiel 8
In zu Beispiel 4 analoger Weise erhält man durch Behandlung von 6.1Ob-Dimethyl-9-methoxy-1,2,3,4,4a,5,6,10b- octahydrophenanthridin mit Formaldehyd und Ameisensäure das 5.6. lüb-Tn.methyl-9-methoxy-1,2,3,4,4a,5,6,10b- octahydrophenanthridin.
Schmelzpunkt des Sulfosalicylates = 245-246oC; Schmelzpunkt des Hydrochlorids 186-1870C.
Beispiel 9
In zu Beispiel 4 analoger Weise erhält man durch Behandlung von .10b-Dimethyl-9-hydroxy-1,2,3,4.4a,5,6,10b- octahydro-phenanthridin (hergestellt gemäss Beispiel 3) mit Formaldehyd und Ameisen säure das 5,6.lr)b-Trimethyl -9-hydroxy- 1 ,2,3.4,4a.5,6. 1Ob-octahydro-phenanthridin, dessen Hydrochlorid bei 250-251oC schmilzt.
Beispiel 10
In zu Beispiel 2 analoger Weise erhält man durch Behandlung von 5,6.1 Ob-Trimethyl-9-methoxy-1,2,3,4,4a,5,6,10b- octahydro-phenanthridin (hergestellt gemäss Beispiel 8) mit Bromwasserstoffsäure das 5.6.1 0b-Trimethyl-9-hydroxy- 1 .2.3.4,4a,5,6,10b-octahydro-phenanthridin, dessen Hydrochlorid bei 250-2510C schmilzt. Diese Verbindung ist identisch mit der nach Beispiel 9 hergestellten Verbindung.
Beispiel 11
In zu Beispiel 2 analoger Weise erhält man durch Behandlung von 5.lOb-Dimethyl-9-methoxy-1,2,3,4,4a,5,6,10b- octahydro-phenanthridin (hergestellt nach Beispiel 4) mit Bromwasserstoffsäure das 5,1 Ob-Dimethyl-9-hydroxy 1 .2.3,4.4a.5,6,10b-octahydro-phenanthn.din, dessen Hydrochlorid bei 289-290'C schmilzt.
Beispiel 12
Das 5,1 Ob-Dimethyl-9-hydroxy- 1,2,3,4,4a,5,6,1 Ob- octahydro-phenanthridin kann auch durch Umsetzung von 9-Hydroxy-lOb-Methyl-
1 .2.3.4,4a.5,6,10b-octahydro-phenanthridin (hergestellt gemäss Beispiel 2) mit Formaldehyd und Ameisen säure in Analogie zu Beispiel 4 erhalten werden.
Beispiel 13
Durch Umsetzung von 6,8 g 2-Methyl-2-(3 -hydroxyäthyloxy-phenyl)-cyclohexylamin mit 3,3 g Formaldehyd, Eindampfen des Reaktionsgemisches, Zusatz von 50 ml konzentrierter Salzsäure. Eindampfen und Kristallisation aus Aceton, analog den Angaben in Beispiel 1, erhält man 2,8 g 9-Hydroxyäthyloxy-lOb-methyl- 1,2.3,4,4a.5,6,1 Ob-octahydro-phenanthridin, welches bei 136-1370C schmilzt.
Das hierbei als Ausgangsmaterial verwendete 2-Methyl-2-(3'-hydroxyäthyloxy-phenyl)-cycloheXylamin kann wie folgt erhalten werden:
76,7 g 2-Methyl-2-(3'-methoxy-phenyl)-cyclohexanon werden zusammen mit 203 g Pyridin-hydrochlorid sechs Stunden auf 190'C erhitzt. Man erhält hierbei 57,8 g rohes 2 Methyl-2-(3 '-hydroxy-phenyl)-cyclohexanon.
Einer Lösung von 7,3 g Natrium in 250 ml wasserfreiem Äthanol werden 64,7 g des so erhaltenen Hydroxyketons und 59,4 g Äthylenbromhydrin in 50 ml wasserfreiem Äthanol zugesetzt. Man erhält 72,5 g einer öligen Substanz, welche nach Destillation bei 164-165 C/0,2 mm 56 g 2-Methyl-2-(3 hydroxyäthoxy-phenyl)-cyclohexanon gibt. Dieses Keton wird in 300 ml Äthanol gelöst und die Lösung wird zusammen mit 23,5 g Hydroxylamin-hydrochlorid, 25 ml Wasser und 45 g Natriumhydroxyd 30 Minuten auf 400C erwärmt. Der nach dem Eindampfen erhaltene Rückstand (62,5 g) wird aus einem Gemisch von Aceton und Petroläther kristallisiert, wobei man 2-Methyl-2-(31-hydroxyäthoxy-phenyl)-cydohexanonoxim vom Schmelzpunkt 108-109oC erhält.
2,6 g dieses Oxims werden in 100 ml Methanol gelöst, worauf der so erhaltenen Lösung eine Lösung von 0,56 g Kaliumhydroxyd in 6 ml Wasser zugesetzt wird. Dann wird bei 400C in Gegenwart von 3 g Raney-Nickel hydriert. Nach Abtrennen des Katalysators und Abdampfen des Methanols unter vermindertem Druck wird der Rückstand in Äther aufgenommen und die ätherische Lösung mit Wasser gewaschen und zur Trokkene eingedampft, wobei man 2-Methyl-2-(3 -hydroxyäthoxyphenyl)-cyclohexylamin erhält. Das Hydrochlorid dieser Base schmilzt bei 251-2520.
Beispiel 14
6 g 6-tert.Butyl-9-methoxy-1 Ob-methyl- 1 ,2,3,4,4a,5,6, 1 Ob-octahydro-phenanthridin werden analog der in Beispiel 4 beschriebenen Methode mit 30 ml Formaldehydlösung und 30 ml 100%iger Ameisensäure behandelt, wobei man 5,7 g rohes 5,10b-Dimethyl-6-tert.butyl-9-methoxy- 1,2,3 ,4,4a,5,6, 1 Ob-octahydro-phenanthridin erhält. Durch Behandlung dieser Base mit Oxalsäure erhält man das entsprechende Oxalat mit einem Schmelzpunkt von 207-2080C.
Beispiel 15
Aus den bei der Herstellung von 2-(3 -methoxy-phenyl)- cyclohexylamin gemäss den Angaben in Beispiel 1 zurückbleibenden Mutterlaugen wird durch Chromatographie an Silicagel das isomere 2-(3'-methoxy-phenyl)-cyclohexylamin mit axialer Lage des Wasserstoffatoms in Stellung 1 gewonnen. Das Hydrochlorid dieser Base schmilzt bei 216-2170C.
4,4 g des so erhaltenen 2-Methyl-2-(3 -Methoxy-phenyl)cyclohexylamins (mit axialer Lage des Wasserstoffatoms in Stellung 1) werden in 2 ml Methanol gelöst, worauf der Lösung 2 ml Formaldehydlösung zugesetzt werden und das Reaktionsgemisch 30 Minuten auf 60C erhitzt wird. Nach Behandlung mit 10 ml verdünnter Salzsäure und Eindampfen erhält man das 9-Methoxy- 1 Ob-methyl- 1 ,2,3,4,4a,5,6, Ob- octahydro-phenanthridin-hydrochlorid (mit axialer Lage des Wasserstoffatoms in Stellung 4a) mit einem Schmelzpunkt von 188-1890C. Diese Verbindung ist mit dem gemäss Beispiel 1 erhaltenen Hydrochlorid (Schmelzpunkt 219-220'C) isomer.
Beispiel 16
In Analogie zu Beispiel 1 wird 9-Methoxy- 1 Ob-cyclopropylmethyl- 1 ,2,3,4,4a,5,6, 1 Ob- octahydro-phenanthridin-hydrochlorid hergestellt, welches einen Schmelzpunkt von 1570-1590C aufweist.
Beispiel 17
In Analogie zu Beispiel 1 wird 9-Methoxy-lOb-äthyl-1,2,3,4,4a,5,6,10b- octahydro-phenanthridin-hydrochlorid hergestellt, welches einen Schmelzpunkt von 231-2320C aufweist. Aus den Mutterlaugen erhält man hierbei ein Isomeres mit einem Schmelzpunkt von 19O1940C.
Beispiel 18
Es werden, entsprechend den Angaben in Beispiel 5, 12 g (+ )-2-Methyl-2-(3 -methoxy-phenyl)-cyclohexylamin mit 20 ml Methanol und 6 ml einer 38obigen wässrigen Formaldehydlösung versetzt, worauf das Reaktionsgemisch über Nacht bei Raumtemperatur stehen gelassen wird. Hierauf wird unter vermindertem Druck eingeengt und das restliche Wasser nach Zugabe von Äthanol azeotrop abdestilliert. Man erhält hierbei (-)-5,5 -Methylen-bis-(9-methoxy- 1 Ob-methyl 1,2,3,4,4a,5,6,10b-octahydro-phenanthridin) mit einem Schmelzpunkt von 190-1920C; [alos = - 50,60 (c = 1% in Methanol).
Diese Bis-Verbindung kann durch Behandlung mit alkoholischer Salzsäure, entsprechend den Angaben in Beispiel 5, in das ( -)-9-Methoxy-lOb-methyl-1,2.3,4,4a,5,6,1 octahydro-phenanthridin-hydrochlorid übergeführt werden.
