Umesterung abgespalten und die Verfahrensprodukte gewünschtenfalls in Salze überführt werden.
Die Verfahrensprodukte zeichnen sich durch eine gute und langandauernde broncholytische Wirksamkeit aus.
Als besonders bevorzugte Methode zur Durchführung dieser Reduktion wurde die katalytische Hydrierung der Verbindungen der Formel II, insbesondere unter Verwendung von Palladiumkohle als Katalysator, angegeben.
Bei der praktischen Durchführung des Verfahrens zeigte sich jedoch, dass die Reinheit der Verbindungen der Formel I nicht befriedigt. Es konnten, vor allem bei der chromatographischen Kontrolle, Nebenprodukte beobachtet werden, die entweder auf eine unvollständige Reduktion der Ausgangssubstanzen oder auf eine weitergehende Reduktion zurückzuführen sind, so dass ein Sauerstoffatom vollständig abhydriert wird.
Überraschenderweise konnte nun gefunden werden, dass eine einheitliche Reaktionslenkung der Reduktion der Verbindungen der Formel II zu den Bisalkanolaminen ohne Bildung dieser unangenehmen Nebenprodukte möglich ist, wenn die Reduktion mit komplexen Hydriden des Bors durchgeführt wird, wobei im Gegensatz zu der sonst üblichen Durchführung der Reduktion mit solchen Borhydriden unter Bedingungen gearbeitet wird, bei denen eine merkliche Zersetzung des Borhydrids stattfindet. Unter diesen Bedingungen können die Verbindungen der Formel I in nahezu quantitativer Ausbeute völlig rein gewonnen werden.
Gegenstand der vorliegenden Erfindung ist demnach ein Verfahren gemäss Hauptpatent Nr. 451 200, das dadurch gekennzeichnet ist, dass die Reduktion der freien Bis-alkanonaminderivate der Formel II oder von deren Salzen in wässrigem Medium mit im Überschuss vorliegenden komplexen Hydriden des Bors, ohne Zusatz von oder in Gegenwart von weniger als jener Menge am Alkalihydroxyd, die zur Freisetzung der Base und/oder zur Absättigung der phenolischen Hydroxylgruppen erforderlich ist, durchgeführt wird. Noch vorhandene Reste R' und R" mit einer Bedeutung ungleich Wasserstoff werden anschliessend, wie beim Verfahren des Hauptpatentes, durch Hydrogenolyse oder Verseifung bzw.
Umesterung abgespalten und die Verfahrensprodukte gewünschtenfalls in Salze übergeführt.
Die für die erfolgreiche Reduktion nötigen Bedingungen, bei denen sich die komplexen Borhydride zersetzen, können einfach dadurch erreicht werden, dass die Salze der Bis-alkanonaminderivate der Formel II in wässriger Suspension mit dem komplexen Borhydrid umgesetzt werden, wobei durch die durch die Hydrolyse des Salzes und die Acidität der phenolischen OH-Gruppen bewirkte saure Reaktion der Reaktionsmischung das Borhydrid teilweise zersetzt wird. Da dadurch ein Teil des Borhydrids der Reaktion entzogen wird, muss zweckmässig ein entsprechender Überschuss des Borhydrids eingesetzt werden, der bei Arbeiten mit wässrigen Suspensionen des Salzes dann, wenn R' Wasserstoff bedeutet, zweckmässig 4-5 Äquivalente komplexes Borhydrid pro Mol Bis-alkanonaminderivat betragen soll. Unter diesen Bedingungen ist die Reaktion bei Raumtemperatur nach 15-30 Minuten beendet.
Diese Verfahrensvariante hat jedoch den Nachteil, dass sie wegen der stürmischen Reaktion besonders bei grösseren Ansätzen nur schwer beherrscht werden kann.
Gemäss einer besonders bevorzugten Ausführungsform des erfindungsgemässen Verfahrens ist es möglich, die Reaktion, vor allem bei grösseren Ansätzen, besser beherrschbar zu machen. Es konnte nämlich festgestellt werden, dass ein befriedigender Verlauf der Reduktion dann erzielt wird, wenn die Zersetzung des komplexen Borhydrids bei etwa 20% der eingesetzten Menge oder darüber liegt. Diese Forderung ist auch dann noch erfüllt, wenn ein Teil der sauren Funktionen des Moleküls durch Zusatz von Alkalilauge neutralisiert wird.
Geht man vom Salz des Bisalkanonamines der Formel II aus, bei dem R' gleich H ist, so kann bereits durch Zusatz von 1 Mol Lauge die Reaktionsgeschwindigkeit soweit gemindert werden, dass auch grössere Ansätze leicht unter Kontrolle gehalten werden können. Die zugesetzte Laugemenge sollte dann, wenn von den Bisalkanonaminsalzen der Formel II, bei denen R' = H ausgegangen wird, 3 Mol nicht überschreiten; setzt man die freie Base der Formel II, bei der R' gleich Wasserstoff ist, ein, so liegt die Grenze des tolerierbaren Alkalihydroxydzusatzes bei etwa 1 Mo pro Mol Bis-alkanonaminderivat. Die damit verbundene geringere Reaktionsgeschwindigkeit kann, falls erwünscht, durch eine mässige Temperaturerhöhung auf maximal 70" C, vorzugsweise bis 50 C, kompensiert werden.
Nach Massgabe der innerhalb der angegebenen Grenzen zugesetzten Alkalihydroxydmenge kann die notwendige Borhydridmenge verringert und dadurch das Ver fahren wirtschaftlicher gemacht werden. So sind z. B. bei der Reduktion der Bisalkanonaminsalze der Formel II, in der R' Wasserstoff bedeutet, bei Anwesenheit von 3 Mol Alkalihydroxyd maximal 3 Äquivalente Borhydrid notwendig.
Als komplexe Borhydride sind neben Natrium- und Kaliumborhydrid auch jene von quartären organischen Basen wie Tetramethylammoniumborhydrid geeignet.
Zur praktischen Durchführung der Reaktion wird zunächst das Bis-alkanonaminderivat der Formel II oder dessen Salz in Wasser suspendiert, gewünschtenfalls die nötige Laugemenge zugesetzt und die wässerige Lösung des komplexen Borhydrids unter Rühren rasch zugegeben. Der durch die Zersetzung des Hydrids frei werdende Wasserstoff verdrängt die Luft aus der Apparatur und dient als Schutzgas, so dass sich eine gesonderte Schutzgasatmosphäre erübrigt, sofern keine allzu langen Reaktionszeiten benötigt werden.
Die Aufarbeitung gestaltet sich besonders günstig, wenn das Reaktionsgemisch nach beendeter Reduktion auf einen pH-Wert von etwa 3 angesäuert und das Bis-alkanolaminderivat als Salz gewonnen wird. Am günstigsten ist eine Isolierung über ein in Wasser schwer lösliches Salz, wie beispielsweise das Sulfat oder das 4' ,4'-Diamino-stilben-2,2' -disulfonat, die durch Zusatz der entsprechenden Säuren oder eines leichtlöslichen Salzes dieser Säuren, wie z. B. Natriumsulfat, zum Reaktionsgemisch ausgefällt werden. Diese schwer löslichen Salze können nach Isolierung auf übliche Weise in andere Salze übergeführt werden oder die Basen daraus in Freiheit gesetzt werden. Sind noch hydrogenolytisch oder hydrolytisch abspaltbare Reste an den phenolischen Sauerstoffatomen oder an den Stickstoffatolr.en, so werden sie auf übliche Weise abgespalten.
Die dabei erhaltenen Substanzen der Formel I sind schon als Rohprodukt farblos und chromatographisch völlig einheitlich. Sie bedürfen keiner weiteren Reinigung, was als besonderer Vorteil des erfindungsgemässen Verfahrens anzusehen ist.
Die Schmelzpunkte der Verbindungen der Formel I sind wegen der dabei auftretenden Zersetzung nur wenig charakteristisch, die Reinheitskontrolle wurde daher mittels Dünnschichtchromatographie durchgeführt.
Die in den Beispielen angegebenen Teile sind Gewichtsteile.
Beispiel 1
50,0 Teile N,N'-Bis-[2-(3' ,4'-dihydroxy-phenyl)-2- oxoäthyl]-hexamethylendiamin-sulfaminat (0,082 Mol) werden in 750 Teilen H2O suspendiert, 245,5 Volumteile 1n
NaOH (0,245 Mol) unter H2 als Schutzgas werden zugege ben, eine Lösung von 5 Teilen NaBH4 (93 %mg) (0,246 Äquivalente) in 250 Teilen H2O zugesetzt und 7 Tage bei Raum temperatur gerührt. Nach beendeter Reaktion wird die klare, gelb gefärbte Lösung unter CO2 langsam mit 4n HC1 auf ein pH von 2-3 eingestellt und mit 0,05 Teilen Na-Salz der Äthy lendiamintetraessigsäure und 2,5 Teilen Tierkohle versetzt.
Nach 30 Minuten langem Rühren wird abfiltriert Man erhält ein klares und farbloses Filtrat, aus dem durch Zusatz einer
Lösung von 23,5 Teilen Na2SO4 in 100 Teilen H2O das Sul fat ausgefällt wird. Nach einstündigem Rühren bei Raum temperatur wird das ausgefallene Kristallisat abgesaugt, mit
Wasser gewaschen und getrocknet. Man erhält 41,3 Teile
N,N' -Bis- [2-(3' ,4'-dihydroxy-phenyl)-2-hydroxy-äthyl] hexamethylendiamin-sulfat mit einem Mikro-FP ab 205 C unter Zersetzung und einem Wassergehalt von 2,9%. Berech net auf die Trockensubstanz entspricht diese Ausbeute 94,3 % der Theorie.

Beispiel 2
2,0 Teile N,N' -Bis- [2-(3' ,4'-dihydroxy-phenyl)-2-oxo- äthyl]-hexamethylendiamin-sulfaminat (0,0033 Mol) werden unter H2 als Schutzgas in 30 Teilen H 20 suspendiert und mit 8,2 ml 1 n NaOH (0,0082 Mol) und anschliessend mit 0,2 Teilen NaBH4 (93 ifoig) (0,0098 Äquiv.) versetzt. Das Reaktionsgemisch wird unter Rühren auf 50-55" C erwärmt, worauf nach 6 Stunden die Reaktion beendet ist.
Zur Zerstörung der Komplexverbindungen wird mit 1,4 ml 1n HCI auf pH 2,8 gestellt, eine Lösung von 1 Teil Na2SO4 in wenig Wasser zugegeben und das ausgefallene Kristallisat abgesaugt. Ausbeute an N,N' -Bis- [2-(3 4' -dihydroxy-phenyl)-2-hydroxyäthyl]hexamethylendiamin-sulfat 1,65 Teile, das sind 95,5% der Theorie (berechnet auf Trockensubstanz).
H2O-Gehalt: 1,6% Mikroschmelzpunkt ab 205 C unter Zersetzung.
Das wie in Beispiel 1 durchgeführte Dünnschichtchromatogramm zeigt, dass die Substanz einheitlich ist.
Beispiel 3
173 Teile N,N'-Bis-[-(3',4'-dihydroxy-phenyl)-2-oxo- äthyl]-hexamethylendiamin-sulfaminat (0,283 Mol) werden in 2300 Teilen Wasser suspendiert und mit 284 ml n/l NaOH (0,284 Mol) sowie mit einer Lösung von 28,7 Teilen NaBH4 (93%mg) (1,4 Äquiv.) in 300 Teilen Wasser unter Rühren versetzt und bei einer Temperatur von 28 C 3,5 Stunden gerührt. Nach 1,5 Stunden war die Ausgangssubstanz in Lösung gegangen. Nach Aufarbeitung (erfolgte wie in Beispiel 1 beschrieben) erhält man 145,9 Teile N,N'-Bis-[2-(3',4'-di hydroxy-phenyl-2-hydroxyäthyl]-hexamethylendiamin-sulfat mit einem Wassergehalt von 3,9%. Diese Ausbeute entspricht 95,4% der Theorie, berechnet für die wasserfreie Substanz.
Die chromatographische Reinheitskontrolle wurde wie in Beispiel 1 beschrieben durchgeführt.
Beispiel 4
5,0 Teile N,N' -Bis- [2-(3 ' ,4' -dihydroxy-phenyl)-2-oxo äthyl]-hexamethylendiamin-sulfaminat (0,0082 Mol) werden in 125 Teilen H2O suspendiert und unter intensivem Rühren rasch mit einer Lösung von 0,8 Teilen NaBH4 97%mg (0,041 Äquiv.) in 25 Teilen H 2O versetzt. Die Temperatur des Reaktionsgemisches steigt dabei auf 30 C an. Schon nach wenigen Minuten geht das Einsatzprodukt in Lösung, die Reaktion ist nach 30 Minuten beendet.
Das Reaktionsgemisch wird nun unter CO2 mit 23 ml In HCI angesäuert, mit einer Lösung von 2,5 Teilen Na2SO4 in 10 Teilen Wasser versetzt und das schwer lösliche, auskristallisierte N,N'-Bis-[2-(3',4'dihydroxy-phenyl) -2-hydroxy-äthyl] -hexamethylendiaminsulfat nach 1 Stunde abgesaugt, mit Wasser gewaschen und getrocknet.
Ausbeute: 4,2 Teile, das sind 96,2% der Theorie (berechnet auf Trockensubstanz) H 2O-Gehalt: 2,6%.
Dünnschichtchromatogramm, durchgeführt wie in Beispiel 1 einheitlich.
Beispiel 5
0,58 Teile Tetramethylammoniumborhydrid (0,013 Äquiv.) werden in 50 Teilen Wasser gelöst. Hierauf werden bei guter Rührung 2,0 Teile N,N'-Bis-[2-(3',4'-dihydroxy phenyl)-2-oxoäthyl]-hexamethylendiamin-sulfaminat (0,0033 Mol) zugegeben und insgesamt 30 Minuten gerührt. Die klare Lösung wird dann durch Zusatz von 1n HCI auf ein pH zwischen 2-3 eingestellt, mit einer Lösung von 1 Teil Na2SO4 in wenig Wasser versetzt und das ausgefallene Kristallisat nach 1 Stunde abgesaugt. Ausbeute: 1,65 Teile N,N'-Bis-[2 (3 ' ,4'-dihydroxy-phenyl) -2-hydroxyäthyl]-hexamethylen- diamin-sulfat, das sind 95,3% der Theorie (berechnet auf Trockensubstanz) H 2O-Gehalt: 1,7% Dünnschichtchromatogramm, durchgeführt wie in Beispiel 1 beschrieben, einheitlich.
Beispiel 6
Bei analoger Arbeitsweise wie in Beispiel 5 werden bei Verwendung von 0,36 Teilen Kaliumborhydrid (0,013 Äquiv.) auf 2 Teile N,N'-Bis-[2-(3' ,4'-dihydroxyphenyl)-2-oxo- äthyl]-hexamethylendiamin-sulfaminat 1,7 Teile N,N'-Bis [2-(3' ,4'-dihydroxy-phenyl)-2-hydroxyäthyl]-hexamethy- lendiamin-sulfat mit einem H 2O-Gehalt von 3,0% erhalten.
Diese Menge entspricht 97% der theoretisch möglichen Ausbeute (berechnet auf Trockensubstanz).
Dünnschichtchromatogramm einheitlich.
Beispiel 7
2,2 Teile N,N'-Bis-[2-(3' ,4'-dihydroxy-phenyl)-2-oxo- äthyl]-tetramethylendiamin-dihydrochlorid (0,0047 Mol) werden in 25 Teilen Wasser suspendiert und unter guter Rührung mit einer Lösung von 0,5 Teilen NaBH 4 (93 %ig) (0,025 Äquiv.) in 5 Teilen Wasser versetzt. Nach einer Reaktionsdauer von 45 Minuten wurde durch Zusatz von 14 ml in HCI auf ein pH von 3 eingestellt und sodann eine Lösung von 3,5 Teilen 4,4'-Diamino-stilben-2,2'-disulfonsäure in 19 ml 1n NaOH unter Rühren langsam zugetropft und 30 Minuten nachgerührt. Das ausgefallene Kristallisat wurde abgesaugt, mit Wasser nachgewaschen und getrocknet.
Ausbeute 3,3 Teile an N,N'-Bis-[2-(3',4'-dihydroxy phenyl)-2 -hydroxyäthyl] -tetramethylendiamin-4 -diamino- stilben-2,2'-disulfonat, das entspricht der theoretischen Ausbeute. Die Substanz schmilzt bis 3500 C nicht.
Zur Umwandlung in das Hydrochlorid werden 2,5 Teile des Diaminostilbendisulfonates von N,N' -Bis-[2-(3' ,4'-di- hydroxy-phenyl)-2-hydroxy-äthyl]-tetramethylendiamin in 100 Teilen heissem Wasser suspendiert, 7,3 ml in HCI zugegeben und 15 Minuten gerührt. Nach dem Erkalten wird die ausgeschiedene 4,4'-Diamino-stilben-2,2'-disulfonsäure abgesaugt (1,2 Teile) und das Filtrat im Vakuum eingeengt.
Der Rückstand wird mit 100 ml Isopropanol versetzt und das ausgeschiedene Kristallisat nach einier Zeit abgesaugt.
Ausbeute: 1,4 Teile Hydrochlorid, das sind 91,85% der Theorie. Nach der Umkristallisation aus Methanol-Isopropanol besitzt die Substanz einen MFp von 193-196 C unter Zersetzung.

When the process was carried out in practice, however, it was found that the purity of the compounds of the formula I is unsatisfactory. By-products could be observed, especially in the chromatographic control, which are either due to an incomplete reduction of the starting substances or to a further reduction so that one oxygen atom is completely hydrated.
Surprisingly, it has now been found that a uniform reaction control of the reduction of the compounds of the formula II to the bisalkanolamines without the formation of these unpleasant by-products is possible if the reduction is carried out with complex hydrides of boron, in contrast to the usual way of carrying out the reduction with such borohydrides are operated under conditions in which a noticeable decomposition of the borohydride takes place. Under these conditions the compounds of the formula I can be obtained completely pure in almost quantitative yield.
The present invention accordingly relates to a process according to main patent no. 451 200, which is characterized in that the reduction of the free bis-alkanonamine derivatives of the formula II or of their salts in an aqueous medium with complex hydrides of boron present in excess, without the addition of or in the presence of less than that amount of alkali metal hydroxide which is required to liberate the base and / or to saturate the phenolic hydroxyl groups. Cleaved transesterification and, if desired, converted the process products into salts.
The conditions necessary for the successful reduction, under which the complex borohydrides decompose, can be achieved simply by reacting the salts of the bis-alkanone amine derivatives of the formula II in aqueous suspension with the complex borohydride, whereby by the hydrolysis of the salt and the acidity of the phenolic OH groups caused acidic reaction of the reaction mixture, the borohydride is partially decomposed. Since this removes part of the borohydride from the reaction, it is advisable to use a corresponding excess of the borohydride, which when working with aqueous suspensions of the salt, if R 'is hydrogen, expediently amount to 4-5 equivalents of complex borohydride per mole of bis-alkanone amine derivative should. Under these conditions, the reaction is complete after 15-30 minutes at room temperature.
However, this variant of the process has the disadvantage that it is difficult to control because of the stormy reaction, especially in the case of larger batches.
According to a particularly preferred embodiment of the process according to the invention, it is possible to make the reaction more controllable, especially in the case of larger batches. This is because it was found that a satisfactory course of the reduction is achieved when the decomposition of the complex borohydride is about 20% of the amount used or more. This requirement is also still met if some of the acidic functions of the molecule are neutralized by adding alkali.
If one starts from the salt of the bisalkanonamines of the formula II, in which R 'is equal to H, the reaction rate can be reduced by adding 1 mol of alkali to such an extent that even larger batches can easily be kept under control. The amount of caustic added should then, if the bisalkanonamine salts of the formula II, in which R '= H is assumed, not exceed 3 mol; if the free base of the formula II, in which R 'is hydrogen, is used, the limit of the tolerable addition of alkali metal hydroxide is about 1 Mo per mole of bis-alkanone amine derivative. The associated lower reaction rate can, if desired, be compensated for by a moderate increase in temperature to a maximum of 70.degree. C., preferably up to 50.degree.
Depending on the amount of alkali metal hydroxide added within the specified limits, the amount of borohydride required can be reduced, thereby making the process more economical. So are z. B. in the reduction of the bisalkanonamine salts of the formula II, in which R 'is hydrogen, in the presence of 3 moles of alkali metal hydroxide a maximum of 3 equivalents of borohydride is necessary.
In addition to sodium and potassium borohydrides, those of quaternary organic bases such as tetramethylammonium borohydride are also suitable as complex borohydrides.
To carry out the reaction in practice, the bis-alkanone amine derivative of the formula II or its salt is first suspended in water, the required amount of alkali is added if desired and the aqueous solution of the complex borohydride is added rapidly with stirring. The hydrogen released by the decomposition of the hydride displaces the air from the apparatus and serves as a protective gas, so that a separate protective gas atmosphere is unnecessary if the reaction times are not too long.
The work-up is particularly advantageous if the reaction mixture is acidified to a pH of about 3 after the reduction has ended and the bis-alkanolamine derivative is obtained as a salt. It is best to isolate it using a salt that is sparingly soluble in water, such as sulfate or 4 ', 4'-diamino-stilbene-2,2'-disulfonate, which can be obtained by adding the corresponding acids or a readily soluble salt of these acids, such as z. B. sodium sulfate, to be precipitated to the reaction mixture. After isolation, these sparingly soluble salts can be converted into other salts in the usual way or the bases can be released from them. If there are still residues on the phenolic oxygen atoms or on the nitrogen atoms that can be split off hydrogenolytically or hydrolytically, they are split off in the usual way.
The substances of the formula I obtained in this way are colorless even as the crude product and completely chromatographically uniform. They do not require any further purification, which is to be regarded as a particular advantage of the process according to the invention.
The melting points of the compounds of the formula I are only slightly characteristic because of the decomposition which occurs in the process; the purity control was therefore carried out by means of thin-layer chromatography.
The parts given in the examples are parts by weight.
example 1
50.0 parts of N, N'-bis [2- (3 ', 4'-dihydroxyphenyl) -2-oxoethyl] -hexamethylenediamine sulfaminate (0.082 mol) are suspended in 750 parts of H2O, 245.5 parts by volume of 1N
NaOH (0.245 mol) under H2 as protective gas are added, a solution of 5 parts of NaBH4 (93% mg) (0.246 equivalents) in 250 parts of H2O is added and the mixture is stirred at room temperature for 7 days.
