Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung substituierter Isochinoline. Die neuen Ver bindungen können zur Bronchienerweiterung verwendet werden.
Bei der Behandlung der Bronchienverengung ist es notwendig, dass das therapeutische Mittel eine Bron chienerweiterung bereits bei Dosen bewirkt, die keine unerwünschten Nebenwirkungen zur Folge haben. Die erfindungsgemäss erhältlichen Verbindungen bewirken Bronchienerweiterung bereits bei Dosen, bei welchen keine sonstigen nachteiligen Wirkungen auftreten.

Bevorzugte bronehienerweiternde Verbindungen sind: der Isobutylester der 4-(6,7-Dimethoxyisochinolin-1-yl)- -piperazin-1-carbonsäure, der Äthylester der 4-(6,7- -Dimethoxyisochinolin-1-yl)-piperazin-l-carbonsäure, der 2-Methyl-2-hydroxypropylester der 4-(6,7-Dimethoxyiso- chinolin-1-yl)-piperazin-l-carbonsäure und 4-Äthylami- no-6,7-dimethox'yisochinolin.

Wie aus obigem Reaktionsschema zu ersehen ist, wer den die Substituenten in 3-,6- und 7-Stellung (R" A und B) durch die Substituenten im als Ausgangsmaterial ver wendeten Phenäthylamin vorgegeben, während der Ami- nosubstituent in 1-Stellung durch das in der letzten Re aktionsstufe verwendete Amin bestimmt wird.
In den Verbindungen der Formel 1, in welchen die Substituen- ten R5, R13, R14, R15 und X komplex sind, kann es not wendig bis bevorzugt sein, das Endprodukt aus der 1- Chlor-Verbindung in zwei oder mehreren Stufen, wie nachstehend beschrieben, herzustellen.
Die Aminierung des 1-Chlor-isochinolins erfolgt ge wöhnlich in wässrigen oder organischen Lösungsmitteln. Obgleich Äthanol als Lösungsmittel bevorzugt wird, kön nen andere polare Lösungsmittel, wie Dimethylformamid, Dioxan, Tetrahydrofuran oder Methanol, ebenfalls ver wendet werden. Vorzugsweise arbeitet man mit einem Überschuss an Amin oder Base. Die Aminierungsreak- tion erfolgt in der Regel bei Temperaturen zwischen <B>100</B> und 200' und die Reaktionszeit kann<B>10</B> bis<B>36</B> Stun den betragen. Bei Verwendung von Äthanol als Lösungs mittel liegen bevorzugte Reaktionstemperaturen zwi schen 120 und 1400 und bevorzugte Reaktionszeiten zwi schen<B>16</B> und 24 Stunden vor.
Die neuen Verbindungen der Formel<B>1</B> sowie auch VII, VIII und IX können ebenfalls unter Verwendung des 1-Brom-isochinolins anstelle der 1-Chlor-Verbindung hergestellt werden. Bei Verwendung dieser Ausgangsma terialien können die Reaktionsbedingungen von den für die 1-Chlor-Verbindung geeigneten Bedingungen etwas abweichen. Die geeigneten Bedingungen sind jedoch für den Fachmann leicht zu ermitteln.
Es empfiehlt sich häufig, das Endprodukt in zwei oder mehreren Stufen aus der entsprechenden Chlor- Verbindung herzustellen. Trotzdem wird das Endprodukt erfindungsgemäss leichter in einer Stufe durch direkte Aminierung der Chlorverbindung bevorzugt erhalten. Die zur Herstellung der neuen Verbindungen in einer oder mehreren Stufen aus den Chlorderivaten geeigne ten Aminoverbindungen können nach folgendem Reak tionsschema hergestellt werden:

In den obigen Formeln bedeutet X, Chlor oder Brom. Die letzte Verbindung kann direkt mit dem 1-Chlor-iso- chinolin unter Bildung von neuen Verbindungen, in wel chen X Chlor oder Brom ist umgesetzt werden.
Die Verbindungen, in denen X Chlor oder Brom ist, können dann in die entsprechenden Verbindungen mit X = Hydroxyl überführt werden durch Behandlung mit 0,1 n Salzsäure. Diese Verbindungen mit X = Hydroxyl können sodann in Verbindungen mit einer Acyloxy- oder Aroyloxygruppe überführt werden unter Verwendung des entsprechenden Säurechlorids.

Verbindungen, in denen X eine Alkanoylamino-, Arylamino- oder Formamidogruppe ist, können aus Ver bindungen mit unsubstituierter Aminogruppe durch Be handlung mit dem entsprechenden Säurechlorid herge stellt werden.
Abwandlungen der obigen Verfahren zur Herstellung der neuen Verbindungen aus ähnlichen Ausgangsmate rialien liegen für den Fachmann auf der Hand.
Bekannte Verfahren zur Herstellung von Salzen ba sischer Verbindungen lassen sich auch auf die Herstel lung der neuen Verbindungen anwenden. Derartige Salze können sowohl mit pharmazeutisch verträglichen wie auch nicht verträglichen Säuren gebildet werden. Unter pharmazeutisch verträglich werden salzbildende Säuren verstanden, die die Toxizität der basischen Verbindung nicht wesentlich erhöhen. Bevorzugte Salze, die beson dere therapeutische Bedeutung besitzen, sind die Säure additionssalze. Hierzu gehören die Salze von Mineral säuren, wie Salzsäure, Jodwasserstoffsäure, Bromwasser stoffsäure, Phosphorsäure, Metaphosphorsäure u.
Schwe felsäuren, wie auch die Salze von organischen Säuren, wie Weinsäure, Essigsäure, Zitronensäure, Äpfelsäure, Maleinsäure, Methylsulfonsäure, Äthansulfonsäure, Ben- zolsulfonsäure, p-Toluolsulfonsäure und Gluconsäure u. dergleichen.
Pharmazeutisch nicht verträgliche Säureadditionssalze können bei der Isolierung und Reinigung der neuen Ver bindungen wertvoll sein. Sie können ferner bei der Her stellung der therapeutisch wertvollen pharmazeutisch ver träglichen Salze eingesetzt werden. Salze dieser Art sind z.B. solche mit Fluorwasserstoffsäure und Perchlorsäure. Hydrofluoride eignen sich speziell zur Herstellung von pharmazeutisch verträglichen Salzen.
Die erfindungsgemäss herstellbaren Verbindungen können an Patienten mit Bronchienverengung verabreicht werden. Die Bronchienverengung kann funktionell be dingt oder durch allergische oder asthmatische Reaktio nen oder durch eine mikrobielle Infektion verursacht sein. Die neuen Verbindungen können allein oder in Kom bination mit pharmazeutisch verträglichen Trägern ver abreicht werden. Das Verhältnis von aktivem Wirkstoff zum Träger wird durch die Löslichkeit und chemische Struktur des Wirkstoffs, die Art der Verabreichung und die Bedürfnisse der pharmazeutischen Praxis bestimmt. Bei Verabreichung in Form von Tabletten werden bei spielsweise Streckmittel, wie Laktose, Natriumzitrat, Cal- ciumcarbonat, Dicalciumphosphat oder dergleichen ver wendet.
Bei der Herstellung von Tabletten zur oralen Verabreichung können ferner den Zerfall begünstigende Mittel, wie Stärke, Alginsäuren und bestimmte komplexe Silicate, zusammen mit Gleitmitteln, wie Magnesium- stearat, Natriumlaurylsulfat und Talk, verwendet werden. Bei der Herstellung von Kapseln zur oralen Verabrei chung werden Laktose und hochmolekulare Polyäthylen- glykole als Träger bevorzugt. Zur Herstellung wässriger Suspensionen zur oralen Verabreichung werden die er- findungsgemässen Verbindungen mit Emulgatoren oder Suspendiermitteln vereinigt.
Verdünnungsmittel, wie Äthanol, Propylenglykol, Glycerin oder Gemischen davon und ähnliche Stoffe, können ebenfalls verwendet wer den. Zur parenteralen Verabreichung werden Lösungen der erfindungsgemässen Wirkstoffe zusammen mit ande ren löslichen Stoffen, wie Glukose oder Kochsalz, her gestellt. Diese wässrigen Lösungen werden zweckmässig gepuffert, so dass isotechnische Lösungen entstehen.
Zur Behandlung der Bronchienverengung können die erfindungsgemäss erhältlichen Verbindungen mittels In- halatoren oder anderen Vorrichtungen verabreicht wer den, die die aktive Verbindung in direkten Kontakt mit den verengten Bereichen<U>bringen.</U>
Die zur Bronchienerweiterung benötigte Wirkstoff menge hängt von der Art und dem Ausmass der Bron chienverengung ab. Gewöhnlich werden zunächst kleine ,Dosen verabreicht unter allmählicher Steigerung, bis die optimale Dosis bestimmt ist. Zur oralen Verabreichung werden grössere Mengen als bei parenteraler Verabrei chung benötigt. Im allgemeinen liegen die verabreichten Dosen zwischen etwa 0,02 und 200 mg Wirkstoff pro Kilogramm Körpergewicht, wobei auf einmal oder in Portionen verabreicht werden kann. Tabletten mit<B>0,1</B> bis<B><I>50</I></B><I> mg</I> Wirkstoff zeigten sich als besonders brauch bar.
Die therapeutische Wirkung der erfindungsgemäss er hältlichen Verbindungen wurde an Meerschweinchen u. Hunden festgestellt. Es wurde gefunden, dass die Verbin dungen über längere Zeiträume wirksam blieben, nach der Verabreichung gut absorbiert wurden und relativ ge <U>ringe</U> Nebenwirkungen auf das Zentralnervensystem aus übten.
Es wurde auch festgestellt, dass die erfindungsge mäss erhältlichen Verbindungen die Wirkung des En zyms Phosphodiesterase, das die Umwandlung von Ade- nosin-3',5'-monophosphat in Adenosin-5'-monophosphat katalysiert, inhibieren. In Phosphodiesterase enthalten den Systemen, in denen ein hoher Spiegel an Adenosin- -3',5'-monophosphat aufrechterhalten werden soll, kön nen daher die neuen Verbindungen mit Erfolg eingesetzt werden.
Die Inhibitorwirkung ist von Bedeutung, da be kanntlich das Mononukleotid Adenosin-3',5'-monophos- phat ein wichtiger Regulator zahlreicher Zell- und Ge webevorgänge ist, beispielsweise der Entspannung der glatten Muskulatur, der Lipolyse und der Glycolyse. Die vorliegenden Verbindungen sind gewebs-spezifische In- hibitoren des Enzyms, d.h. sie inhibieren das Enzym in gewissen Geweben, in anderen hingegen nicht; soll der Spiegel an Adenosin-3,5-monophosphat daher nur in be stimmten Geweben erhöht werden, so ist die Verwendung der erfindungsgemässen Verbindungen besonders vor teilhaft.
Wie bekannt, hängt die Inhibierung der Phosphodi- esterase mit der bronchienerweiternden Wirkung zusam men. Zahlreiche neue Verbindungen wurden daher auf ihre inhibierende Wirkung untersucht in Hinblick auf eine potentielle Wirkung als Bronchiendilatoren.
Die folgenden Beispiele sollen die Erfindung nur nä her erläutern, jedoch nicht begrenzen. <I>Beispiel<B>1</B></I> Herstellung von 4-(6,7-Dimethoxyisochinolin-1-yl)- -piperazin-l-carbonsäure, Isobutylester Teil<B>A:</B> Herstellung von N-carbäthoxyhomoverathylamin Einer Lösung von 362 g (2,0 Mol) Homoveratrylamin in<B>50</B> cm' wasserfreiem Benzol wurde eine Lösung von 109 g (1,0 Mol) Äthylchlorformiat in 200 cm Benzol tropfenweise zugesetzt, wobei die Temperatur auf 50- 550C gehalten wurde.
Die dabei entstehende Aufschlärn- mung wurde<B>16</B> Std. lang bei Raumtemperatur gerührt und in<B>500</B> cm3 Wasser gegossen. Die Benzolschicht wurde abgetrennt, mit<B>500</B> cm:
' gesättigter Natriumbicar- bonatlösung gewaschen und über Natriumsulfat getrock net. NachVerdampfen des Lösungsmittels und Destillie ren des rückständigen öles erhielt man<B>186 g</B> (74%) einer klaren farblosen Flüssigkeit mit einem Schmelz punkt von 174-177'C (0,2 mm), die beim Stehenlassen kristallisierte, wobei sich ein weisser kristalliner Fest stoff bildete. Schmelzpunkt 61-62-C.
Teil B: Herstellung von 6,7-Dimethoxy-3,4-dihydro-1(2H)- -isochinolinon Einem Liter Polyphosphorsäure wurden unter Rüh ren 330 g (1,3 Mol) N-carbäthoxyhomoveratrylamin zu gesetzt Das dabei entstehende Gemisch wurde<B>30</B> Minu ten lang bei 140 C gerührt und in 2 Liter Eiswasser ge gossen. Die wässrige Lösung wurde mit Ammoniumhy droxyd alkalisch gemacht und mehrere Male mit Chloro form extrahiert. Die vereinigten Extrakte wurden über Natriumsulfat getrocknet und zur Trockne eingedampft. Der Rückstand wurde aus Methylenchlorid-Äthylacetat umkristallisiert, wobei sich 120<B>g</B> (44,50/0) weisser Plätt chen bildeten. Schmelzpunkt 159-161 C.
Teil C: Herstellung von 6,7-Dimethoxy-1(2H)-isochinolinon Ein Gemisch aus 46,0 g (0,222 Mol) 6,7-Dimethoxy- -3,4-dihydro-1(2H)-isochinolinon und 7,0 g 25%iges Pal ladium auf Kohle wurde<B>30</B> Minuten lang bei 2400C un ter einer Stickstoffatmosphäre gerührt, wonach die Was serstoffentwicklung aufhört. Das Gemisch wurde auf Raumtemperatur abgekühlt und mehrere Male mit heis- sem Chloroform gewaschen. Die vereinigten Chloroform gemische wurden zur Trockne eingedampft und ergaben 39,0 g (860/,) eines weissen kristallinen Produktes. Smp.
<B>228-2320C.</B> Teil<B>D:</B> Herstellung von 1-Chlor-6,7-dimethoxyisochinolin Ein Gemisch aus 39 g (0,191 Mol) 6,7-Dimethoxy- -1(2H)-isochinolinon, und 220 cm Phosphoroxychlorid wurden 1 Std. lang unter Rückfluss gerührt. Die dabei entstehende bernsteinfarbige Lösung wurde zur Trockne eingedampft und derRückstand in Methylenchlorid ge löst und langsam 100 cm einer konzentrierten Ammo niumhydroxydlösung zugegebem Die Methylenchlorid- schicht wurde abgetrennt und die wässrige Schicht mit 3 Portionen von je 100 cm Methylenchlorid extrahiert. Die vereinigten organischen Extrakte wurden über Na triumsulfat getrocknet und eingedampft, wobei sich<B>35,0 g</B> (820%) eines weissen kristallinen Produktes bildeten.

Die dabei entstehende bernsteinfarbige Lösung wurde eingedampft, in Wasser aufgeschlämmt und fil triert, wobei sich 4,3<B>g</B> des rohen Produktes bildeten. Dieses wurde in 30 cm Methanol umkristallisiert und ergab 2,58 g (38,5%) des Produktes als blassrosa Na deln.

Äthylester der 4-(6,7-Dimethoxy- isochinolin-1-yl)piperazin-l-carbonsäure, Schmelzpunkt 130-1311Q umkristallisiert aus Isopropyläther, Schmelz punkt des Hydrochlorids 107-1080C (Zers.).
Der 2-,Methyl-2-hydroxypropylester der 4-(6,7-Dimeth- oxy-isochinolin-1-yl)-piperazin-1-carbonsäure, Schmelz punkt 133-1340C; umkristallisiert aus Äthylacetat-Hexan; Schmelzpunkt des Hydrochlorids 1100C (Zers.) konnte folgendermassen hergestellt werden-.
Zu 200 cm eines 50%igen Gemischs aus Schwefel säure und Wasser wurden 25 g (0,0615 Mol) 4-(6,7-Di- methoxychinazolin-4-yl)-piperazin-1-carbonsäure, 2-Me- thyl-2-propenylesterhydrochlorid zugesetzt.Die dabei ent stehende gelbe Lösung wurde<B>1</B> Stunde lang bei Raum temperatur gerührt, in 200 g Eis gegossen und mit 400%- iger Natriumhydroxydlösung alkalisch gemacht, wobei die Temperatur unter 400C gehalten wurde.
Die dabei entstehende Lösung wurde mit 4 Portionen von je 200 cm Methylenchlorid extrahiert, die vereinigten organi schen Extrakte mit 0,5 n Salzsäure extrahiert und schliess- lich mit 3 Portionen von je 100 cm Wasser extrahiert. Die vereinigten wässrigen Extrakte wurden mit 400%iger Natriumhydroxydlösung alkalisch gemacht und mit 3 Portionen von je 200 cm Methylenchlorid extrahiert. Die letzteren Methylenchloridextrakte wurden vereinigt, über Natriumsulfat getrocknet und eingedampft.
<I>Beispiel<B>3</B></I> Herstellung von 1-Äthylamino-6,7-dimethoxyisochinolin Das nach dem Verfahren des Teiles D des Beispiels 1 hergestellte 1-Chlor-6,7-dimethoxyisochinolin wurde nach den Verfahren des Teiles E des Beispiels 1 mit Äthyl- amin umgesetzt, wobei sich 1-Äthylamino-6,7-dimethoxy- isochinolin mit einem Schmelzpunkt von 194-1950C bil dete und wurde aus Methanol-Wasser umkristallisiert. Das Hydrochlorid besass -einen Schmelzpunkt von 224 <B>225-C</B> (Zersetzung).
<I>Beispiel 4</I> Herstellung von 1-Piperazinyl-6,7-dimethoxyisochinolin Eine Lösung von 84,0 g (0,243 Mol) des nach dem Verfahren des Beispiels 2 hergestellten 4-(6,7-,Dimethoxy- isochinolin-1-yl)-piperazin-l-carbonsäure, Methylesters in einem Liter Methanol und<B>250</B> cm# 300/,iges Natrium- hydroxyd wurden<B>18</B> Std. unter Rückfluss erhitzt. Die dabei entstehende Suspension wurde eingedampft, um das Methanol zu entfernen, mit<B>500</B> cm3 Wasser ver dünnt und mit<B>3</B> Portionen von<B>je 25</B> cräs Methylenchlo- rid extrahiert.
Die vereinigten Extrakte wurden über Natriumsulfat getrocknet und auf 250 cm eingedampft. Zu diesem Produkt wurden 700 cm' Isopropyläther zu gesetzt, die Lösung auf<B>300</B> cm-' eingedampft, in einem Eisbad abgeschreckt und filtriert, wobei<B><I>51 g</I></B> (670/c,) eines weissen kristallinen Produktes erhalten wurden. <I>Beispiel<B>5</B></I> Herstellung des 2-Dimethylaminoäthylesters der 4-(6,7- -Dimethoxyisochinolin-1-yl)-piperazin-1-carbonsäure Teil<B>A:</B> Herstellung des Phenylesters der 4-(6,7-Dimethoxyiso- chinolin-1-yl)-piperazin-l-thiolcarbonsäure Einer kalten Lösung (O C) von 19 g (0,0695 Mol) des nach dem Verfahren des Beispiels 4 hergestellten 1- -Piperazinyl-6,7-dimethoxyisochinolins und 7,65 g (0,076 Mol) Triäthylamin in 100 cm Methylenchlorid wurden tropfenweise 12,0 g (0,0695 Mol) Phenylchlorthiolfor- miat zugesetzt.
Die dabei entstehende Suspension wurde <B>15</B> Minuten bei Raumtemperatur gerührt und mit<B>100</B> cm-' Methylenchlorid verdünnt. Diese Lösung wurde mit 2 Portionen von je 50 cm Wasser gewaschen, über Na triumsulfat getrocknet und unter Bildung eines kristalli nen Rückstandes eingedampft. Der Rückstand wurde aus <B>100</B> cm,' Methanol umkristallisiert, wobei sich<B>18,7 g</B> <B>(66%)</B> des Produktes als blassgelber kristalliner Fest stoff abschieden. Schmelzpunkt 137-1381C.

Nach Abkühlung auf Raumtemperatur wurden 4,1<B>g</B> (0,01 Mol) 4-(6,7-Dimethoxyisochinolin-1-yl)-piperazin-l- -thiol-carbonsäure, Phenylester zugesetzt und die Lösung <B>30</B> Minuten lang bei Raumtemperatur gerührt. Das Pro dukt wurde mit 70 cm Wasser gerührt und eingedampft, um das Tetrahydrofuran zu entfernen und die dabei ent stehende Lösung mit Portionen von je 100 cm Methy- lenchlorid extrahiert. Die vereinigten Extrakte wurden über Natriumsulfat getrocknet und abgedampft, wobei ein viskoses öl zurück blieb, das mit Isopropyläther zer rieben wurde, wobei sich<B>3,1 g</B> (80%) eines weissen kri stallinen Produktes abschieden.

<I>Beispiel<B>8</B></I> Bronchienerweiternde Wirkung Unbetäubte, weibliche Meerschweinchen, welche 12 Stunden gehungert hatten, erhielten orale und parenter- ale Gaben der Verbindung, die auf ihre Wirksamkeit untersucht werden sollte.
Kontrolltiere erhielten Gaben einer Salzlösung, welche nicht die zu untersuchende Ver bindung enthielt. Im Anschluss an die Verabreichung wurde jedes Tier mit Histamin-Aerosol erregt.
Die Methode der Erregung bestand darin, dass man eine 0,4%ige wässrige Lösung von Histamin bei einem Druck von 0,35 kg/qcm eine Minute in einen 20 X 20 X<B>30</B> cm Plastikbehälter sprühte. Unmittelbar nachdem der Behälter mit dem Histamin besprüht worden war, wurden die Tiere hineingesetzt. Nach Verlauf einer Mi nute wurde der Atmungszustand, der die Bronchialveren- gung wiederspiegelt, bestimmt. Die ermittelten Werte wurden als normale Atmung<B>(0),</B> leicht vertiefte Atmung <B>(1),</B> schwere Atmung (2), sehr schwere Atmung und Ataxie (3) und Bewusstlosigkeit (4) eingestuft. As is known, the inhibition of phosphodiesterase is related to the bronchodilating effect. Numerous new compounds have therefore been examined for their inhibitory effect with regard to a potential effect as bronchodilators.
The following examples are only intended to explain the invention in more detail, but not to limit it. <I> Example<B>1</B> </I> Production of 4- (6,7-dimethoxyisoquinolin-1-yl) - -piperazine-1-carboxylic acid, isobutyl ester part <B> A: </B> Preparation of N-carbäthoxyhomoverethylamin A solution of 109 g (1.0 mol) ethyl chloroformate in 200 cm benzene was added dropwise to a solution of 362 g (2.0 mol) homoveratrylamine in 50 cm 'anhydrous benzene. the temperature was kept at 50-550C.
