EMI0001.0008
dans laquelle X est un atome d'halogène; M est un ato me de métal choisi dans le groupe comprenant le magné sium et l'aluminium ; m est la valence du métal M ;pour former l'alcool P-acétylénique correspondant de formule R1R2C(OH) - CH2 - C - CH (IV) dans laquelle RI, R2 sont définis comme ci-dessus;
b) la transformation de cet alcool p-acétylénique IV en son dérivé organo-magnésien par action d'un halogé- nure d'alcoylmagnésium, suivie du traitement du dérivé organo-magnésien ainsi obtenu par de l'anhydride carbo nique, dans un solvant organique inerte tel que le tétra- hydrofuranne ou le benzène, pour former l'acide α-acé- tylénique correspondant de formule R1R2 - C(OH) - CH2 - C = C - COOH (V) dans laquelle R1 et R2 sont définis comme ci-dessus;

Preferably, the alkyl part of the above radicals contains 1 to 3 carbon atoms.
The process of the invention comprises the following steps a) the condensation of a carbonyl derivative of formula R1R2CO (III) in which R1 and R2 are defined as above with a metal-propargyl halide of formula
EMI0001.0008
wherein X is a halogen atom; M is a metal atom selected from the group consisting of magnesium and aluminum; m is the valency of the metal M; to form the corresponding P-acetylenic alcohol of formula R1R2C (OH) - CH2 - C - CH (IV) in which R1, R2 are defined as above;
b) the transformation of this p-acetylenic alcohol IV into its organomagnesium derivative by the action of an alkylmagnesium halide, followed by the treatment of the organomagnesium derivative thus obtained with carbonic anhydride, in a solvent inert organic such as tetrahydrofuran or benzene, to form the corresponding α -acetylenic acid of formula R1R2 - C (OH) - CH2 - C = C - COOH (V) in which R1 and R2 are defined as above;
c) the hydrogenation of this α -acetylenic acid V at ordinary pressure, in a lower alkanol such as methanol or ethanol in the presence of a catalyst such as 5% palladium on a barium sulfate support, to the corresponding ethylenic acid followed by cyclization of this ethylenic acid to the lactone of formula I by heating under vacuum.
Preferably, it is heated to 120 ° C. under a vacuum of 20 Torr.
The 8-lactones- α, α-unsaturated of formula I are new compounds.
The new 8-lactones- α, P-unsaturated of formula I are pharmacologically active and exhibit the anticonvulsant, local anesthetic activities of di-hydrokawain, dihydromethylsticin or di-hydroyangonin, as shown by the test of electrically induced convulsions in mice (anticonvulsant activity) or the intracutaneous injection test in guinea pigs (local anesthetic activity).
The new 8-lactones- α, p-unsaturated are useful therapeutically in the same applications as the cited products, in particular as sedatives, anticonvulsants, tranquilizers and local anesthetics.
The p-acetylenic alcohols of formula IV and the α-acetylenic acids of formula V are also new compounds. The invention is illustrated by the following examples: Example 1: 6- (p-phenethyl) -5,6-dihydro-2-pyrone 1) 6-Phenyl-hexyn-4-ol.
