Verfahren zur Herstellung neuer tricyclischer Aminoketone
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer tricyclischer Aminoketone, die wertvolle pharmakologische Eigenschaften aufweisen und chemotherapeutisch wirksam sind.
Die erfindungsgemäss herstellbaren Verbindungen der vorliegenden Erfindung entsprechen der Formel

Hierzu gehören auch die pharmakologisch verträglichen Säureadditionssalze dieser Verbindungen
Die erfindungsgemäss herstellbaren Verbindungen weisen ein interessantes pharmakologisches Wirkungsspektrum auf, wie an der hohen Aktivität bei den folgenden Testen zu erkennen ist: Test Bezugsliteratur Antagonismus der r. Domenjoz, W. Theobald, Reserpin-Ptosis Arch. int. Pharmacodyn.

Diese pharmakologische Wirkungsiihersicht lässt erken- nen, dass die Verbindungen eine Wirkung auf das Zentralnervensystem austiben und daher zur Behandlung von Stö- rungen dieses Systems verwendet werden können. Die Wir kungsübersicht veranschaulicht insbesondere die Brauchbarkeit dieser Verbindungen zur Behandlung von psychischen depressiven Störungen und pathologischen Zuständen, die auf eine Behandlung mit Antiparkinson-Priparaten ansprechen.
Wenn auch die oben angegebene Formel Verbindungen umfasst, die durch ein verhältnismässig breites Spektrum von pharmakologischen Eigenschaften ausgezeichnet sind, die ihrerseits von der Stellung und der Art der einzelnen Substituenten abhängig sind, so ist doch eine gemeinsame Eigenschaft all dieser Verbindungen, dass sie auf das Zentralvervensystem einwirken und bemerkenswert niedrige Toxizität aufweisen.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man eine tricyclische Verbindung der Formel

Diese Umsetzung wird insbesondere in einem inerten Lösungsmittel bei einer Temperatur zwischen 0 C und dem
Siedepunkt des Reaktionsgemisches durchgeführt, und zwar in
Anwesenheit oder Abwesenheit eines säurebindenden Mit tels, wie eines Alkalihydroxyds, -amids, -carbonats oder -hydrogencarbonats. Anstelle eines säurebindenden Mittels kann bei dem Verfahren auch ein Überschuss an der Amin komponente verwendet werden.

HO(CH2)nCOR (IIIb) umgesetzt.
Die Chlorwasserstoff- und Bromwasserstoffsäureester des Alkohols derFormel (IIIb) sind besonders gut geeignet.
Die Umsetzung wird insbesondere in Gegenwart eines säure- bindenden Mittels, wie eines Hydroxyds, Carbonats oder Hydrogencarbonats eines Alkali- oder Erdalkalimetalls, oder eines tertlären Amins oder in Gegenwart eines flberschusses an dem sekundären Amin der Formel (IIb) durchgefiihrt.
Die Umsetzung kann in einem Lösungsmittel oder in mehreren verschiedenen Ldsungsmitteln, wie Kohlenwasserstoffen, Athern, Estern, Ketonen oder in Gemischen derselben mit Wasser bei einer Temperatur zwischen 0 C und dem Siedepunkt des Reaktionsgemisches durchgefihrt werden.
Es sind verschiedene stereoisomere Formen der Verbindungen der Formeln (I) und (Ia) möglich. sofern R1 eine andere Gruppe als ein Wasserstoffatom ist. Es können Ge- mische stereoisomerer Formen als solche verwendet oder gewünschtenfalls nach lichen Methoden getrennt werden.
In der Regel zeigen die trans-Formen der Verbindungen der Formeln I und Ia, in denen Y einen ungesättigten Rest bedeutet, eine stärkere pharmakologische Wirksamkeit als die cic-Formen. Reine Stereoisomere kdnnen auch erhalten werden, wenn man reine Stereoisomere als Ausgangsmaterialien verwendet, vorausgesetzt, dass das asymmetrische Element (ein Koblenstoffatom oder eine äthylenische Bindung) durch den Prozess nicht beeinflusst wird.
Für therapeutische Zwecke können die Basen der Formeln (I) und (Ia) als solehe verwendet werden oder in Form der Säureadditionssalze, die Anionen enthalten, welche pharmakologisch verträglich sind, z. B. als Hydrochloride, Hydrobromide, Phosphate, Nitrate, Sulfate, Hydrogenoxalate, Oxalate, Succinate, Tartrate, Methansulfonate und Äthan- sulfonate, so dass die vorteilhaften phsysiologischen Eigenschaften nicht durch Nebenwirkungen, die den Anionen zugeschrieben werden müssten, aufgehoben 'verden. FOr gewisse Zwecke können auch die von Verbindungen der For meln I und Ia abgeleiteten quaternären Ammoniumsalze verwendet werden, die gewöhnlich durch Umsetzung mit organischen Halogeniden oder anderen reaktionsfähigen organischen Esters (z.
B. einen niedermolekularen Alkyl- oder Benzylchlorid, -bromid, -jodid oder einem Alkyl- oder Arylsulfonat) erhalten werden können.
Die beschriebenen Verbindungen können in an sich üb- licher Weise verabfolgt werden, beispielsweise oral in Form von Tabletten oder Kapsein oder parenteral in Form von Lösungen oder Suspensionen in pharmakologisch verträg- lichen Trägern.
Die folgenden Beispiele sollen die Erfindung näher veranschaulichen, deren Umfang jedoch in keiner Weise ein schriinken.
Beispiel 1
2,5 Teile 2-Methylaminomethyl-2-(4-methoxyphenyl)- 1,3-dioxolan werden in 25 Teilen Acetonitril gelöst, und es werden 5 Teile 5-(3-Bromopropyl)-10,11-dihydro-5H- dibenzo(a,d)cyclohepten zugegeben. Das Reaktionsgemisch wird 5 Stunden unter Riickfluss erhitzt. Nach dem Abktihlen auf Zimmertemperatur werden 1,35 Teile Chlorwasserstoff in 10 Teilen Wasser zugegeben. Die Lösung wird 16 Stunden bei Zimmertemperatur stehengelassen und dann in eine Lösung von 10 Teilen Kaliumcarbonat in Wasser gegossen. Das anfallende Gemisch wird mit anther extrahiert. Der Äther- extrakt wird im Vakuum zur Trockne eingedampft. Der Riickstand wird in Butanon gelöst und eine Lösung von Oxalsäure in Butanon zugegeben.
Beim Abkahlen scheiden sich Kristalle des Hydrogenoxalats des 5-[N-(4-Methoxy-phenacy)-N-methyl-3-aminopropyl]-5H-dibenzo(a,d)cycloheptens aus, und sie werden abfiltriert. Der Schmelzpunkt liegt bei 187,5 bis 188,5 C.
Beispiel 2
26,7 Teile 9-(3-Methyl-aminopropyliden)-thioxanthen und 25,2 Teile cv-Brom-4-methoxyacetophenon werden in 300 Teilen Benzol gelöst, und es wird eine Lösung von 13,6 Teilen Natriumhydrogencarbonat in 150 Teilen Wasser zugegeben. Das Reaktionsgemisch wird dann 3 Stunden bei Zimmertemperatur gerührt. Die wässrige Phase wird abgetrennt und verworfen. Die organische Phase wird mit einer wässrigen Lösung von Salzsäure extrahiert, bis ein pH von 3,2 erhalten wird. Die organische Phase wird dann iiber wasserfreiem Natriumsulfat getrocknet und in eine Lösung der äquimolaren Menge Oxalsäure in Butanon gegossen.
Die so erhaltenen Kristalle des Hydrogenoxalats des 9 [N-(4-Methoxyphenacyl)-N-methyl-3-aminopropyliden]- thioxanthens weisen einen Schmelzpunkt von 194 bis 195 C out'.
In der gleichen Weise wie in dem obenstehenden Beispiel werden die folgenden Verbindungen hergestellt: 5-(N-Phenacyl-N-methyl-3-aminopropyliden)-10,11-di hydro-5H-dibenzo(a,d)cyclohepten-hydrogenoxalat,

Das 5-[N-(4-Methoxyphenacyl) -N-methyl3-amino- propyl]-5H-dibenzo(a,d)cyclohepten-hydrochlorid, der Milchzucker und die Stirke werden miteinander vermischt und gesiebt. Das Polyvinylpyrrolidon wird in einer geeigneten Menge Wasser gelöst und zu dem erwähnten Gemisch zugegeben, welches danach granuliert wird. Das so erhaltene Granulat wird dann getrocknet und mit dem Magnesiumstearat und dem Talkum gemischt, und hierauf werden Tabletten gepresst.
Beispiel 4 Wässrige Suspension für Injektionszwecke 5-(N-Phenocyl-N-methyl-3 -aminopropyliden)
10,11-dihydro-5H-dibenzo(a,d)cyclohepten hydrochlorid 250 mg
Methylcellulose 15 mg Polyoxylthyl ensorbita n-mo nostearat (Tween 80 [TM] Atlas) 20 mg destilliertes Wasser zum AuffOllen auf 5 ml
Wenn auch in den vorhergehenden Beispielen die Herstellung von Verbindungen beschrieben ist, die eine Methylgruppe oder eine andere niedermolekulare Alkylgruppe enthalten, so versteht es sich doch von selbst, dass Verbindungen, welche andere niedermolekulare Alkylgruppen geradkettiger oder verzweigter Natur mit bis zu 4 Kohlenstoffatomen eiaschliesslich enthalten, wie Methyl-, Athyl-, Propyl, Isopropyl-, Butyl-, sek.-Butyl-, tert.-Butyl-Gruppen,
in völlig analoger Weise durch Einsetzen des diesen andersartigen niedermolekularen Alkylrest aufweisenden Ausgangsmaterials hergestellt werden können. In entsprechender Weise werden in den Fallen, in denen Methoxygruppen oder andere niedermolekulare Alkoxygruppen vorhanden sind, Verbindungen mit einem Gehalt an anderen niedermolekularen Alkoxygruppen, die verschiedene niedermolekulare Alkylgruppen mit bis zu 4 Kohlenstoffatomen einschliesslich enthalten, in analoger Weise aus den geeigneten, niedermolekularen Alkoxygruppen aufweisenden Materialien hergestellt. Schliesslich können ebenfalls in analoger Weise andere Molekülabwandlungen nach dem hier offenbarten erfinderischen Prinzip leicht vorgenommen werden.
Die erfindungsgemäss erhältlichen Verbindungen sind ganz allgemein durch die weiter oben beschriebenen pharmakologische Wirksamkeit gekennzeichnet, die sie befähigt, gewissen physiologischen Abnormitäten im Körper eines Lebewesens entgegenzuwirken Wirksome Mengen der erfindungsgemäss herstellbaren pharmakologisch-aktiven Verbindungen können einem Lcbewcsen in cincr jeden der zahl- reichen Anwendungsformen verabfolgt werden, z. B. in oraler Form als Kapseln oder Tabletten oder parenteral in Form von sterilen Lösungen oder Suspensionen oder durch Pillen Implantation, und in gewissen Fallen auch durch interve- nöse Applikation steriler Lösungen. Schliesslich können sie auch kutan, subkutan, bukal, intramuskulär und intraperitoneal verabfolgt werden.
Zu den Lebewesen, die mit den erfindungsgemäss erhält- lichen Verbindungen und Präpararten und nach den neuen Methoden zur Linderung der bcschriebenen und bzw. oder ähnlicher Leiden behandelt werden können, gehören ausser den Menschen auch Haustiere, wie Hunde und Katzen, landwirtschaftliche Nutztiere, wie Pferde, Kühe, Schafe und Ziegen.
Pharmazeutische Priparate werden in der Regel aus einer vorbestimmten Menge einer oder mehrerer erfindungsgemäss herstellbarer Verbindungen vorzugsweise in fester Form hergestellt. Solche Formulierungen können in Form von Pulvern, Elixieren, Lösungen, Pillen, Kapseln, Pellets oder Tabletten vorliegen, und zwar mit oder ohne - besser jedoch mit - Zusatz irgendeines der zahlreichen pharmakologisch vertriglichen Begleit- oder Trigerstoffe. Liegt der Wirkstoff im Gemisch mit einem pharmazeutischen Begleit- oder Trigerstoff vor, so soll der Wirkstoff etwa 0,01 bis etwa 75%, normalerweise etwa 0,05 bis etwa 15% des Gewichts des Prä- parates ausmachen.
In solchen Präparaten können als Tri- germaterial Stärke, Zucker, Talkum, die gemeinhin verwendeten synthetischen und natürlichen Gummen, Wasser und dergleichen mitverwendet werden. Zum Formen von Tabletten können Bindemittel, wie Gelatine, und Gleitmittel, wie Natriumstearat, verwendet werden. Den Tabletten können den Zerfall begünstigende Mittel, wie Natriumbicarbonat, einverleibt werden.
Wenn auch verhältnismässig kleine Mengen der erfin dungsgemiss herstellbaren Verbindungen sogar solche bis herab zu 5,0 mg, an Menschen, die ein verhältnismässig geringes Körpergewicht aufweisen, verabfolgt werden kijnnen, so betragen doch die Dosierungseinheiten 5 mg oder mehr, und besser noch 25, 50 oder 100 mg oder mehr, und zwar je nach dem zu behandelnden Patienten und dem besonderen Heilergebnis, das man zu erzielen wünscht, wie es im übrigen dem Fachmann geliufig ist. Als noch breitere Bereiche sind Mengen von 1 bis 3000 mg pro Dosierungseinheit zu nennen.
Die neuen Verbindungen kinnen mit anderen pharmakologisch wirksamen Substanzen, wie analgetischen Mitteln, Tranquilizern, Steroiden oder Hormonen und dergleichen, oder mit Puffern oder säureabstumfenden Mitteln kombiniert werden, und die in den Präparatmischungen vorhandenen Mengen dieser Wirkstoffe können in weiten Grenzen schwanken. Es ist lediglich erforderlich, dass die erfindungsgemäss erhiltliche Verbindung in einer wirksamen Menge vorhanden ist, d. h. in einer so grossen Menge, dass man eine geeignete wirksame Dosierung, die der angewendeten Dosierungsform angepasst ist, erreicht. Es ist offensichtlich, dass man mehrere Dosierungsformen gleichzeitig verabfolgen kann.
Die genauen Einzeldosierungen und Höhen der Tagesdosen hängen in jedem besonderen Fall natürlich von den wohlfundierten medizinischen und bzw. oder tiermedizinischen Verordnungsgrundsätzen ab. Bei der Verabfolgung an Menschen kinnen die erfindungsgemäss herstellbaren Verbindungen in der Regel in einer Menge von 25 bis 1500 mg pro Tag und Patient, aufgeteilt in 1 bis 4 Dosierungen, innerhalb einer Zeitspanne von einem Tag bis zu einem Jahr ver abfolgt werden.
