Atomen bedeuten, worin ferner, falls Rw und R5 zusammen -CH= CH-CH=CH- bedeuten, R5 = H sein kann sowie ihre Salze mit Säuren oder gegebenenfalls Basen und ihre quartären Ammoniumsalze bei guter Verträglichkeit eine hervorragende antiphlogistische Wirkung besitzen, die von einer guten analgetischen und antipyretischen Wirkung begleitet ist.
Ferner treten bakteriostatische, bakterizide, antiprotozoäre, diuretische, blutzuckersenkende, choleretische, cholesterinspiegelsenkende und Strahlenschutz-Wirkungen auf.
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Aminophenylessigsäure-Derivaten der Formel 1, das dadurch gekennzeichnet ist, dass man eine Verbindung der Formel 2

Aminogruppe, einen Schwefelsäure-Alkylschwefel- säure oder Sulfonsäurerest bedeutet mit einer Verbindung der Formel 3 R7R8NH 3
Die Reaktionsbedingungen für eine solche Umaminierung sind in der Literatur beschrieben. Vorteilhaft setzt man die Amine der Formel 2 bzw. 5 im Überschuss ein und arbeitet in Anwesenheit von Katalysatoren wie Säuren, Metallsalzen, Jod, Dehydratisierungsmitteln, Hy driemngs-Dehydrierungskatalysatoren oder Natriumhydrogensulfid.
In einzelnen Fällen verlaufen Umaminierungen jedoch auch in Abwesenheit eines Katalysators.
Es ist zweckmässig, die Umsetzungen in Gegenwart eines inerten Lösungsmittels durchzuführen, ferner ist es vorteilhaft, die Umsetzung unter erhöhtem Druck sowie bei höheren Temperaturen vorzunehmen.
Als Verbindungen 2 setzt man bevorzugt pJod-, p Brom- oder p-Chlor-phenylessigsäurederivate ein, wobei durch den Effekt von stark elektronenanziehenden Gruppen, wie beispielsweise NO5, Acylamino oder C:F8, in 3-Stellung und/oder gegebenenfalls auch in 2-, 5- oder 6-Stellung das Halogen-Atom gelockert und daher die Umsetzung mit den sekundären Aminen der Formel 3 wesentlich erleichtert wird. Man führt die Reaktion je nach der Reaktionsfähigkeit der pJlalogenphenylessig- säurederivate bevorzugt zwischen Raumtemperatur und 3200, gegebenenfalls im Autoklaven unter Druck (bis zu 200 at) durch. Dabei kann man die jeweiligen umzusetzenden Amine der Formel 3, wie Piperidin oder Pyrrolidin in grossem Überschuss gleichzeitig als Lösungsmittel einsetzen.
Sonst arbeitet man bevorzugt in inerten Lösungsmitteln wie Chloroform, Benzol, Dioxan, Tetrahydrofuran, Dimethylformamid, Dichlorbenzol oder Äthylenglykol unter säurebindenden Zusätzen, wie Natrium- oder Kaliumcarbonat.
So gelingt es beispielsweise glatt, aus 3-Nitro-4-brom -z-methylphenylessigsäure oder aus 3 -Nitro-4-brom-a- -methylnaphthyl-l-essigsäure durch Umsetzung mit Piperidin, Homopiperidin oder Pyrrolidin die entsprechenden 4-Piperidino-, 4-Homopiperidino- bzw. 4-Pyrrolidino-phenylessigsäure- bzw. -naphthyl-l-essigsäurederivate herzustellen.
Als Verbindungen der Formel 5 eignen sich z.B. o,w'- -Dihalogenalkane, wie 1,4-Dibrombutan, 1,5-Dichlorpen- tan, 1,6-Dijodhexan, aber auch Schwefel- oder Sauerstoffringverbindungen, wie Tetrahydrofuran, -pyran, -thiophen oder thiopyran. Die Umsetzung solcher Schwefel- oder Sauerstoffringverbindungen mit Aminen der Formel 4 erfolgt im allgemeinen unter drastischen Bedingungen, wie vorzugsweise bei Temperaturen über 1500 im Autoklaven.
Eine Verbindung der Formel 1 kann mit einer Säure in üblicher Weise in das zugehörige Säureadditionssalz übergeführt werden. Für diese Umsetzung kommen solche Säuren in Frage, die physiologisch unbedenkliche Salze liefern. So können organische und anorganische Säuren, wie z.B. aliphatische, alicyclische, araliphatische, aromatische oder heterocyclische ein- oder mehrbasige Carbon- oder Sulfonsäuren, wie Ameisensäure, Essigsäure, Propionsäure, Pivalinsäure, Diäthylessigsäure, Oxalsäure, Malonsäure0, Bernsteinsäure, Pimelinsäure, Fumarsäure, Maleinsäure, Milchsäure, Weinsäure, Äpfelsäure Aminocarbonsäuren, Sulfaminsäure, Benzoesäure, Salicylsäure, Phenylpropionsäure, Citronensäure, Gluconsäure, Ascorbinsäure, Nicotinsäure, Isonicotinsäure, Methansulfonsäure,
Äthandisulfonsäure, p - Hydroxy äthansulfonsäure, p-Toluolsulfonsäure, Naphthalinrnonound -disulfonsäuren, Schwefelsäure, Salpetersäure, Halogenwasserstoffsäuren, wie Chlorwasserstoffsäure oder Bromwasserstoffsäure, oder Phosphorsäuren, wie Orthophosphorsäure, usw. verwendet werden.

Die Umsetzung von Aminen der Formel 3 bzw. 4 mit Verbindungen der Formel 2 bzw. 5 erfolgt grundsätzlich nach den in der Literatur beschriebenen bzw.
zitierten üblichen Austauschreaktionen. Wählt man als Reaktionskomponente der Formel 2 bzw. 5 ein Halogenid so kann die Umsetzung in Gegenwart oder Abwesenheit eines Lösungsmittels durchgeführt werden. Als Lösungsmittel kommen beispielsweise niedere aliphatische Alkohole, Benzol oder Toluol in Frage.
Je nachdem, welches Halogenid als Ausgangsmaterial verwendet wird, arbeitet man bei niedrigen Temperaturen, z.B. Raumtemperatur, oder bei höheren Temperaturen bis zu der Siedetemperatur des verwendeten Lösungsmittels.
In einzelnen Fällen kann es erforderlich sein, die Umsetzung unter Druck oder bei erhöhter Temperatur durchzuführen. Die Verwendung eines Katalysators, z.B.
einer Base, ist möglich, aber in der Regel nicht erforderlich. Vorteilhaft wendet man die Amine im über schuss an.
Verwendet man als Ausgangsmaterial der Formel 2 ein Phenol (X1 = OH) bzw. als Ausgangsmaterial der Formel 5 einen Alkohol (mindestens einer der Reste X1 = OH) oder einen vorzugsweise niederen aliphatischen Carbonsäureester eines solchen Phenols oder Alkohols, so wird die Umsetzung mit den Aminen zweckmässig in Anwesenheit eines Katalysators durchgeführt.
Als Katalysator kommt entweder ein Dehydratisierungsmittel oder ein Dehydrierungsmittel oder ein Gemisch dieser Katalysatoren in Frage. Als Dehydratisierungskatalysator sei beispielsweise Aluminiumoxid genannt das gegebenenfalls noch mit anderen geeigneten Oxiden aktiviert sein kann. Als De,hydrierungsmittel können zB. Raney-Nickel oder die üblichen Edelmetallkatalysatoren, wie beispielsweise Palladiumoxid oder Palladiumkohle, verwendet werden.
Man kann ferner als Ausgangsmaterial für diese Reaktion eine Verbindung der Formel 2 bzw. 5 verwenden, worin X1 eine sekundäre oder tertiäre Aminogruppe bedeutet Unter den Reaktionsbedingungen einer Umaminierung wird eine solche sekundäre oder tertiäre Aminogruppe in Verbindung der Formel 2 durch Behandlung mit Aminen der Formel 3 durch die Gruppe R,RsN- ausgetauscht. Völlig analog kann man durch eine Umami nierungsreaktion sekundäre oder tertiäre Aminogruppen in Verbindungen der Formel 5 durch Umsetzung mit Aminen der Formel 4 austauschen.
Andererseits können Carbonsäuren der Formel 1 durch Umsetzung mit einer Base in eines ihrer physiologisch unbedenklichen Metall- bzw. Ammoniumsalze übergeführt werden. Als Salze kommen insbesondere die Natrium-, Kalium-, Magnesium-, Calcium- und Ammoniumsalze in Betracht, ferner substituierte Ammoniumsalze, wie z.B. die Dimethyl- und Diäthylammonium-, Monoäthanol-, Diäthanol- und Triäthanolammonium-, Cyclohexylammonium- und Dibenzyläthylendiammonium-Salze.
Umgekehrt können Verbindungen der Formel 1 aus ihren Säureadditionssalzen durch Behandlung mit starken Basen, wie Natrium- oder Kaliumhydroxid, Natrium- oder Kaliumcarbonat, bzw. gegebenenfalls aus ihren Metall- und Ammoniumsalzen durch Behandlung mit Säuren, vor allem Mineralsäuren wie Salz- oder Schwefelsäure, in Freiheit gesetzt werden.
Schliesslich kann eine Verbindung der Formel 1 zur Herstellung eines quaternären Ammoniumsalzes verwendet werden. Diese Herstellung erfolgt durch Behandlung der Verbindung der Formel 1 mit einem quaternierenden Mittel. Für diese Quaternierung sind beispielsweise Alkylhalogenide, wie Methylbromid oder Äthylchlorid oder auch Dialkylsulfate, wie z.B. Diäthyl- oder Dimethylsulfat, geeignet. Die Quaternierungsbedingungen sind die üblichen, wie sie in der Literatur beschrieben bzw. zitiert sind.
Falls die Verbindungen der Formel 1 ein Asymmetriezentrum enthalten, liegen sie gewöhnlich in racemischer Form vor. Weisen sie zwei oder mehrere Asymmetriezentren auf, dann fallen diese Verbindungen bei der Synthese im allgemeinen als Gemische von Racematen an, aus denen man in an sich bekannter Weise die einzelnen Racemate beispielsweise durch mehrmaliges Umkristallisieren aus geeigneten Lösungsmitteln isolieren und in reiner Form gewinnen kann.
Solche Racemate können nach einer Vielzahl bekannter Methoden, wie sie in der Literatur angegeben sind, in ihre optischen Antipoden getrennt werden.
So kann man einige racemische Gemische als Eutektika anstatt in Form von Mischkristallen fällen und auf diese Weise rasch trennen, wobei in diesen Fällen auch eine selektive Ausfällung möglich sein kann. Die Methode der chemischen Trennung ist jedoch zu bevorzugen. Danach werden aus dem racemischen Gemisch durch Umsetzung mit einem optisch aktiven Trennmittel Diastereomere gebildet. So kann man gegebenenfalls eine optisch aktive Base mit der Carboxylgruppe oder eine optisch aktive Säure mit der Aminogruppe einer substituierten Aminophenylessigsäure der Formel 1 umsetzen.
Zum Beispiel kann man Salz-Diastereomere der substituierten Aminophenylessigsäuren der Formel 1 ge pebenenfalls mit optisch aktiven Aminen, wie Chinin, Cinchonidin, Brucin, Cinchonin, Hydroxyhydrindamin, Morphin, 1-Phenyläthylamin, Phenyloxynaphthylmethylamin, Chinidin, 1 -Fenchylamin, Strychnin, basischen Aminosäuren, wie Lysin, Arginin, Aminosäureestern oder mit optisch aktiven Säuren, wie D- und L-Weinsäure, Dibenzoyl-D- und -L-weinsäure, Diacetyl-D- und -L-weinsäure, p-Camphersulfonsäure. D- und L-Mandelsäure, D- und L-Äpfelsäure oder D- und L-Milchsäure bilden. In ähnlicher Weise lassen sich Ester-Diastereomere der Aminophenylessigsäuren der Formel 1 (A = COOH) mit optisch aktiven Alkoholen, wie Borneol, Menthol, Octanol-2, herstellen.
Der Unterschied in der Löslichkeit der anfallenden Diastereomere erlaubt die selektive Kristallisation der einen Form und die Regeneration der jeweiligen optisch aktiven substituierten Aminophenylessigsäuren aus dem Gemisch. Die optischen Antipoden können auch biochemisch unter Anwendung selektiver, enzymatischer Reaktionen getrennt werden. So kann man die racemischen substituierten Aminophenylessigsäuren einer asymmetrischen Oxydase oder gegebenenfalls Decarboxylase aussetzen, die durch Oxydation oder Decarboxylierung eine Form zerstört, während die andere Form unverändert bleibt. Möglich ist die Verwendung einer Hydrolase bei einem Derivat des racemischen Gemischs zur bevorzugten Bildung einer Form der substituierten Aminophenylessigsäuren.
So kann man Ester oder Amide der Aminophenylessigsäuren der Formel 1 der Einwirkung einer Hydrolase aussetzen, welche das eine Enantiomorph selektiv verseift und das andere unverändert lässt.
Weiterhin ist es natürlich möglich, optisch aktive Verbindungen nach den oben beschriebenen Methoden zu erhalten, indem man Ausgangsstoffe verwendet, die bereits optisch aktiv sind.
Die Verbindungen der Formel 1 und/oder gegebenenfalls ihre physiologisch unbedenklichen Metall- und Ammonium- sowie Säureadditionssalze können im Gemisch mit üblichen Arzneimittelträgern in der Human- oder Veterinärmedizin eingesetzt werden. Als Trägersubstanzen kommen solche organischen oder anorganischen Stoffe in Frage, die für die parenterale, enterale oder topikale Applikation geeignet sind und die mit den neuen Verbindungen nicht in Reaktion treten, wie beispielsweise Wasser, pflanzliche Öle, Polyäthylenglykole, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, Vaseline, Cholesterin. Zur parenteralen Applikation dienen insbesondere Lösungen, vorzugsweise ölige oder wässrige Lösungen, sowie Suspensionen oder Emulsionen.
Für die enterale Applikation eignen sich ferner Tabletten oder Dragees, für die topikale Anwendung Salben oder Cremes, die gegebenenfalls sterilisiert oder mit Hilfsstoffen, wie Konservierungs-, Stabilisierungsoder Netzmitteln oder Salzen zur Beeinflussung des osmotischen Druckes oder mit Puffersubstanzen versetzt sind.
Die erfindungsgemässen Substanzen werden vorzugsweise in einer Dosierung von 1 bis 2000 mg pro Dosierungseinheit appliziert.
In den nachfolgenden Beispielen sind die Temperaturen in Grad Celsius angegeben. Beispiel I a) 12g 3-Brom-4-amino-phenylessigsäure und 12,5g 1,5-Dibrompentan werden mit 25 g l0%iger wässeriger Natronlauge versetzt und unter Rühren gekocht. Nach 4 Stunden werden nochmals 35g ölige wässerige Natronlauge zugesetzt. Nach weiterem 3-4stündigem Kochen extrahiert man mit Äther und arbeitet wie üblich auf (pH 5,4-5,6). b) 53,5 g 3-Chlor-4-piperidino-α-methyl-phenylessig- säure und 18,2g L-Ephedrin werden in 200ml Äther 2 Tage bei Raumtemperatur stehengelassen. Das ausgefallene Salz wird abgesaugt, dreimal aus Äthylacetat und danach zweimal aus Aceton umkristallisiert F. 125 bis 126 ; (a) -26,80.
6 g dieses Salzes werden in einem Gemisch von 150 ml Methanol und 150ml Äthanol gelöst. Die Lösung wird über einen schwach sauren Kationenaustauscher geleitet und mit Methanol/Äthanol (1: 1) eluiert. 2 g dieses Salzes werden in 100 ml Methanol/Äthano} (1: 1) unter Erwärmen gelöst, über einen schwach sauren Kationenaustauscher geleitet und mit Methanol/Äthanol (1: 1) eluiert. Die Eluate werden eingedampft. Man erhält 1,2g (+)-3-Chlor-4-piperidino-α-methyl-phenyles- sigsäuremit(a) +37,4 .
c) 26,7 g 3-Chlor-4-piperidino-α-methyl-phenylessig- säure werden in 100 ml Acetonitril mit 9,1 g L-Ephedrin 48 Stunden bei 0- 6 stehengelassen. Das ausgefallene Salz wird zweimal aus Äthylacetat umkristallisiert. F.
124 - 1260; (a) - 15,60. 5 g dieses Salzes werden in 250 ml Methanol/Äthanol (1:1) gelöst, über einen schwach sauren Kationenaustauscher geleitet und mit Methanol/Äthanol (1:1) eluiert. Die Eluate werden eingedampft. Der Rückstand [2 g Öl, (a) 37,20] wird in 40 ml Äther gelöst und mit einer Lösung von 0,73 g Cyclohexylamin in 10 ml Äther versetzt. Das ausgefallene Cyclohexylaminsalz [F. 197 - 1980; (a) + 8,10] wird abgesaugt, in 100 ml Methanol/Äthanol (1:1) gelöst, über einen schwach sauren Kationenaustauscher geleitet und mit Methanol/Äthanol (1:1) eluiert.
Die Eluate werden eingedampft. Man erhält 1,1 g ()-3-(:lhlor-4-piperidino- -x-methyl-phenylessigsäure, (a) -41,10.
d) 6,2 g 3-Brom-4-piperidino-y;methyl-phenylessig säure werden in 30 ml Acetanhydrid 10 Stunden gekocht.
Nach Abdestillieren der Essigsäure und des überschüssigen Acetanhydrids erhält man 3-Brom-4-piperidino-,a- -methyl-phenylessigsäureanhydrid.
e) Durch 12stündiges Erhitzen von 3-Chlor-4-piperi dino-a-methyl-phenylessigsäure mit überschüssigem Methyljodid bzw. Dimethylsulfat in Acetonitril auf 1000 im Rohr erhält man das Methojodid bzw. das Metho-methylsulfat der 3-Chlor-4-piperidino-α-methyl-phenylessig- saure.

70 g 3 -Nitro-4-amino-phenylessigsäuremethylester (F.
95 - 960; hergestellt durch Veresterung der Säure mit Methanol in Gegenwart von HCI) und 23 g 1,5-Dibrom- pentan werden 6 Stunden auf 90 - 950 erwärmt und nach dem Abkühlen mit 200 ml Äther versetzt. Das ausgefallene 4-Piperidino-3-nitro-phenylessigsäuremethylester-hydrobromid wird abgesaugt und das ätherische Filtrat eingedampft. Der ölige Rückstand wird mit einer Lösung von 12g KOH in 120ml absolutem Äthanol 90 Minuten gekocht, anschliessend wird der Alkohol abgedampft. Der Rückstand wird in 40 ml Wasser gelöst, die Lösung mit Äther gewaschen und wie üblich aufgearbeitet (pH 4). Man erhält 3-Nitro-4-piperidino-phenylessig- säure, F. 108 - 1100 (Benzol-Hexan).

The precipitated salt is filtered off with suction, recrystallized three times from ethyl acetate and then twice from acetone. F. 125 to 126; (a) -26.80.
6 g of this salt are dissolved in a mixture of 150 ml of methanol and 150 ml of ethanol. The solution is passed over a weakly acidic cation exchanger and eluted with methanol / ethanol (1: 1). The eluates are evaporated at 40. The oily residue [3.12 g; (a) +35.5] is dissolved in 50 ml of ether, filtered through charcoal and mixed with a solution of 1.15 g of cyclohexylamine in 10 ml of ether. The precipitated cyclohexylammonium-3-chloro-4-piperidino-α-methyl-phenyl acetate is filtered off with suction. 197-198 (methanol); (, a) 7.2.
2 g of this salt are dissolved in 100 ml of methanol / ethanol (1: 1) with heating, passed through a weakly acidic cation exchanger and eluted with methanol / ethanol (1: 1). The eluates are evaporated. 1.2 g of (+) - 3-chloro-4-piperidino-α-methyl-phenyl-acetic acid with (a) +37.4 are obtained.
c) 26.7 g of 3-chloro-4-piperidino-α-methyl-phenylacetic acid are left to stand in 100 ml of acetonitrile with 9.1 g of L-ephedrine at 0-6 for 48 hours. The precipitated salt is recrystallized twice from ethyl acetate. F.
124-1260; (a) - 15.60. 5 g of this salt are dissolved in 250 ml of methanol / ethanol (1: 1), passed through a weakly acidic cation exchanger and eluted with methanol / ethanol (1: 1). The eluates are evaporated. The residue [2 g of oil, (a) 37.20] is dissolved in 40 ml of ether and a solution of 0.73 g of cyclohexylamine in 10 ml of ether is added. The precipitated cyclohexylamine salt [F. 197-1980; (a) + 8.10] is filtered off with suction, dissolved in 100 ml of methanol / ethanol (1: 1), passed through a weakly acidic cation exchanger and eluted with methanol / ethanol (1: 1).
The eluates are evaporated. 1.1 g of () -3 - (: chloro-4-piperidino-x-methyl-phenylacetic acid, (a) -41.10 are obtained.
d) 6.2 g of 3-bromo-4-piperidino-y; methyl-phenylacetic acid are boiled in 30 ml of acetic anhydride for 10 hours.
After the acetic acid and the excess acetic anhydride have been distilled off, 3-bromo-4-piperidino-, α-methyl-phenylacetic anhydride is obtained.
e) By heating 3-chloro-4-piperidino-a-methyl-phenylacetic acid with excess methyl iodide or dimethyl sulfate in acetonitrile to 1000 in the tube for 12 hours, the methoiodide or metho-methyl sulfate of 3-chloro-4-piperidino is obtained -? -methyl-phenylacetic acid.
Example 2
70 g of methyl 3-nitro-4-aminophenylacetate (F.
95-960; produced by esterification of the acid with methanol in the presence of HCl) and 23 g of 1,5-dibromopentane are heated to 90-950 for 6 hours and, after cooling, 200 ml of ether are added. The precipitated 4-piperidino-3-nitro-phenylacetic acid methyl ester hydrobromide is filtered off with suction and the ethereal filtrate is evaporated. The oily residue is boiled for 90 minutes with a solution of 12 g of KOH in 120 ml of absolute ethanol, then the alcohol is evaporated off. The residue is dissolved in 40 ml of water, the solution is washed with ether and worked up as usual (pH 4). 3-Nitro-4-piperidino-phenyl acetic acid, mp 108-1100 (benzene-hexane) is obtained.

Example 3
26 g of 3-nitro-4-bromophenylacetic acid are boiled with 25.5 g of piperidine in 200 ml of ethyl alcohol for 24 hours, most of the alcohol is distilled off, the residue is taken up in 2N hydrochloric acid, filtered and the filtrate is worked up as usual (pH 4). This gives 3-nitro-4-piperidino-phenylacetic acid, mp 108-1100 (benzene / diisopropyl ether).

25.5 g of 3-chloro-4-amino-α-methyl-phenylacetic acid-n-butyl ester (boiling point 158-1600 / 0.1 mm; obtainable from 4-amino-3-chloro-α-methyl-phenylacetic acid and n-butanol in the presence of HCl) are u in 180 ml of n-butanol with 23 g of 1,5-dibromopentane. 28 g of finely powdered K2CO3 boiled for 15 hours while stirring. It is filtered off with suction and washed with n-butanol. The filtrate is evaporated, the residue is taken up in benzene and boiled with 15 ml of acetic anhydride for 90 minutes. The benzene solution is extracted with dilute hydrochloric acid and the acidic extract is worked up as usual (pH 8-10). 3-Chloro-4-piperidino - - methyl-phenylacetic acid-n-butyl ester, b.p. 160-1620 / 0.01 mm is obtained.
