Verfahren zur Herstellung von neuen ll-Amino-5,6-dihydro-6-morphanthridonen
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von neuen 11-Amino-5,6-dihydro- -6-morphanthridonen der allgemeinen Formel I

worin jedes der Symbole Ph1 und Ph2 für ein gegebenen- falls substituiertes 1,2-Phenylenradikal steht, A ein niederes Monoaza-alkylen oder -aralkylen, in welchen Re stel() die Heteroatome durch mindestens 2 Kohlenstoffatome getrennt sind und das N-Heteroatom des Restes A beliebig substituiert sein kann, bedeutet, und R3 für Wasserstoff oder Niederalkyl steht, ihren Salzen und N-Acylderivaten.
Die Phenylenreste Ph, und Ph2 sind substituiert oder können durch einen oder mehrere, gleiche oder verschiedene Substituenten substituiert-sein. Solche Substituenten sind z.B. Niederalkyl, z.B. Methyl, Äthyl, n- oder i-Propyl oder -Butyl, freie oder funktionell abgewandelte Hydroxy- oder Mercaptogruppen, z.B. Niederalkoxy oder Niederalkylmercaptogruppen, wie Methoxy, Äthoxy, n- oder i-Propoxy oder -Butoxy, Methyl- oder Äthylmercapto, Halogen, z.B. Fluor, Chlor oder Brom, Trifluormethyl, Nitro oder Amino, insbesondere Di-niederalkylamino, z.B. Dimethylamino oder Diäthylamino.
Der Ausdruck nieder bedeutet im Zusammenhang mit Kohlenwasserstoffresten solche Reste mit höchstens 7, vorzugsweise höchstens 4 Kohlenstoffatomen. Bevorzugte Reste Phl und Ph, sind 1,2-Phenylen, (Niederalkyl) -1,2-phenylen, (Niederalkoxy)-1,2-phenylen, (Niederalkyl mercapto)-1,2-phenylen, (Halogen)-1,2-phenylen, (Trifluormethyl)-1,2-phenylen, (Nitro)-1,2-phenylen und (Di -niederalkylamino)- 1 ,2-phenylen.
Ein Niederalkylrest R2 ist z.B. einer der oben genannten, aber auch n- oder i-Pentyl, n-Hexyl oder n Heptyl.
Das Symbol A steht vorzugsweise für Monoaza-niederalkylen, oder Monoaza-niederalkylen, worin das Stickstoffatom durch Niederalkyl, Hydroxy-niederalkyl, verestertes Hydroxy-niederalkyl (z.B. Halogen-niederalkyl oder Niederalkanoyloxy-niederalkyl), veräthertes Hydroxy-niederalkyl (z.B. Niederalkoxy-niederalkyl oder Niederalkoxy-niederalkoxy-niederalkyl), tert. Amino-niederalkyl (z.B.
Di-niederalkylamino-niederalkyl, Monoaza - niederalkylenamino - niederalkyl, Monooxa-niederal kylenamino-niederalkyl oder Monothia-niederalkylenamino-niederalkyl), (N-Niederalkyl-monoaza-niederalky lenamino) -niederalkyl, (N-Hydroxy-niederalkyl-monoaza -niederalkylenamino) - niederalkyl, (N - verestertes Hydroxyniederalkyl-monoaza-niederalkylenamino) -niederalkyl, (N-veräthertes Hydroxy-niederalkyl-monoaza-niederalkylenamino)-niederalkyl, Phenyl-niederalkyl, Niederalkoxycarbonyl oder Phenyl substituiert ist, z.B.
3-Aza pentylen-(1,S), oder für 3-Aza-pentylen-(1,5), worin das Stickstoffatom durch Methyl, Äthyl, Propyl, 2-Hydroxy äthyl, 3-Hydroxy-propyl, 2-Chlor-äthyl, 2-Acetoxy-äthyl, 2-Methoxy-äthyl, 3-Pivalyloxy-propyl, 4-Methoxy-butyl, 2-(4-Methyl-piperazino)-äthyl, 2-14-(2-Hydroxy-äthyl)-pi perazino]-äthyl, 2-Morpholino-äthyl, Benzyl, l- oder 2 -Phenyl-äthyl, Äthoxycarbonyl oder Phenyl substituiert sein kann; 3-Aza-hexylen-(1,5) oder 3-Aza-hexylen-(1,6), 3 -Methyl-, 3-Äthyl- oder 3 -Benzyl-3-aza-hexylen-(l ,5) oder -(1,6), oder 4-Aza-heptylen-(2,6).
Die aromatischen Teile, nämlich die Phenylgruppen der obigen Reste können unsubstituiert oder wie die Reste Ph1 und Ph2 substituiert sein.
Die Acylderivate weisen das Acylradikal auf dem Stickstoffatom in 5-Stellung auf. Die Acylgruppe dieser Acylderivate ist vorzugsweise Niederalkanoyl, wie Ace tyl, Propionyl, Butyryl oder Pivalyl, aber auch Niederalkenoyl, wie Arcyloyl oder Methacryloyl, monocyclisches Aroyl oder Aryl-niederalkanoyl oder Aryl-niederalkenoyl, wie Benzoyl, Phenylacetyl oder Cinnamoyl.
Diese Acylradikale können unsubstituiert oder, insbesondere im aromatischen Teil, wie die Reste Phl und Ph2 substituiert sein.
Die Verbindungen der vorliegenden Anmeldung zeigen wertvolle pharmakologische Eigenschaften. Ausser ihren krampflösenden und antiinflammatorischen Effekten, rufen die neuen Verbindungen in erster Linie eine Abnahme der Magensaftsekretion, insbesondere der Ma gensäuresekretion, hervor, wie diese in Tierversuchen, z.B. an Säugetieren, z.B. Ratten oder Hunden nachge- wiesen werden können. In den letzteren kann die Magen saftsekretion entweder durch Futter oder durch parenterale Verabreichung von Histamin hervorgerufen werden. Die neuen Verfahrensprodukte können daher in erster Linie für die Behandlung von Reizungen der Ma genschleimhaut und von Magengeschwüren eingesetzt werden.
Weiter können die neuen Verfahrensprodukte bei der Untersuchung des Mechanismus der Magensaft sekretion oder als Zwischenprodukte für die Herstellung von anderen wertvollen, insbesondere pharmakologisch aktiven Verbindungen eingesetzt werden.
Besonders wertvoll sind Verbindungen der Formel I, worin jedes der Symbole Phl und Phe 1,2-Phenylen, (Niederalkyl)-1,2-phenylen, (Niederalkoxy)-1,2-phenylen, (Niederalkylmercapto) -12-phenylen, (Halogen)- 1,2-phe- nylen, (Trifluormethyl)-1,2-phenylen, (Mtro)-1,2-pheny- len oder (Diniederalkylamino)-1,2-phenylen bedeutet, A für Monoaza-niederalkylen oder Monoaza-niederalkylen, worin das Stickstoffatom durch Niederalkyl, Hydroxy -niederalkyl, Halogen-niederalkyl, Niederalkanoyloxy -niederalkyl, Niederalkoxy-niederalkyl, Niederalkoxy-nie deralkoxy-niederalkyl, Monoaza-niederalkylenamino-nie- deralkyl, Monooxa-niederalkylenamino-niederalkyl,

und R3 Wasserstoff oder Niederalkyl bedeutet, ihre Salze und N-Acylderivate mit Mederalkan-, Niederalken-, Benzoe-, Phenyl-niederalkan- und Phenyl-niederalkensäuren, wobei in den oben genannten Verbindungen die Phenylreste unsubstituiert oder durch Niederalkyl, Niederalkoxy, Niederalkylmercapto, Halogen, Trifluormethyl, Nitro oder Di-niederalkylamino substituiert sind.
Besonders wertvoll sind auch die Verbindungen der Formel I, worin jedes der Symbole Phl und Phe 1,2-Phe- nylen, (Niederalkyl)-1,2-phenylen, (Niederalkoxy)-1,2- phenylen oder (Halogen)-192-phenylen bedeutet, A für Monoaza-niederalkylen, N-Niederalkyl-monoaza - niederalkylen, N-(Hydroxy-niederalkyl)-monoaza-niederalkylen oder N-(Niederalkanoyloxy-niederalkyl)-monoaza-nieder- alkylen, Monoaza-niederalkylen, dessen Stickstoffatom durch Halogen-niederalkyl, Niederalkoxy-niederalkyl, Mederalkoxy niederalkoxy-niederalkyl, Niederalkoxyvar- bonyl, Phenyl-niederalkyl,
Phenyl oder durch (N-Hydroxy-niederalkyl -monoaza-niederalkylenamino) - nieder- alkyl, (N - Halogen - niederalkyl - monoaza - niederalkylenamino)-niederalkyl, (N - Niederalkanoyloxy - niederalkyl -monoaza-niederalkylenamino)-niederalkyl oder (N-Me- deralkoxy-niederalkyl-monoaza-niederalkylenamino)-nie- deralkyl substituiert ist, wobei die Heteroatome durch mindestens zwei Kohlenstoffatome getrennt sind, und R3 Wasserstoff, Niederalkyl oder Niederalkanoyl bedeutet, und ihre Salze.
Besonders wertvolle Verbindungen sind diejenigen der Formel II

Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man eine Verbindung der allgemeinen Formel III

worin X1 je eine freie oder reaktionsfähige veresterte Hydroxygruppe bedeutet und jedes der Symbole Z1 und Z2 gegebenenfalls durch Aryl substituiertes Niederalkylen bedeutet, welches X1 vom Stickstoffatom durch mindestens 2 Kohlenstoffatome trennt, wobei die Summe der Kohlenstoffatome der Niederalkylenreste von Z1 und Z2 höchstens 7 ist, oder eine entsprechende Verbindung worin mindestens eine der Gruppen X1 zusammen mit einem Wasserstoffatom abgespalten ist,
und die andere Gruppe X1 eine reaktionsfähige veresterte Hydroxygruppe bedeutet, mit einer Verbindung der Formel R1H2 oder einem Alkalimetallsalz davon umsetzt, worin R1 eine gegebenenfalls substituierte Iminogruppe bedeutet, wobei Z1, Z und R1 zusammen den oben definierten Rest A ergeben.
Wenn erwünscht, kann eine erhaltene freie Verbindung in ihre Salze übergeführt, oder ein erhaltenes Salz in die entsprechende freie Verbindung umgewandelt, und/oder, wenn erwünscht, ein erhaltenes Gemisch von Isomeren oder Racematen in die einzelnen Isomeren oder Racemate aufgetrennt, und/oder, wenn erwünscht, erhaltene Racemate in die optischen Antipoden aufgespaltet, und/oder, wenn erwünscht, eine erhaltene Verbindung in eine andere erfindungsgemässe Verbindung umgewandelt werden.
In den Ausgangsstoffen bedeutet X1 vorzugsweise Halogen, wie Chlor, Brom oder Jod, aber auch Sulfonyloxy, z.B. Methan-, Äthan-, oder Benzolsulfonyloxy, p Toluol- oder p-Brom-benzol-sulfonyloxy. Ist in den Ausgangsstoffen die Gruppe X1 zusammen mit einem Wasserstoffatom abgespalten, so enthält die Gruppe Z1 und/ oder Z eine Doppelbindung.
Die oben genannte Reaktion wird nach an sich bekannten Methoden, in Gegenwart oder Abwesenheit von Verdünnungsmitteln, vorzugsweise in solchen, welche gegenüber der Reagenzien inert sind und diese lösen, Katalysatoren, Kondensationsmitteln und/oder in inerten Atmosphären, bei normalem oder erhöhtem Druck, unter Kühlung, bei Zimmertemperatur oder vorzugsweise bei erhöhten Temperaturen, durchgeführt.
Im obigen Verfahren kann der Aminoausgangsstoff im Überschuss verwendet werden, um die entstehende Säure zu eliminieren, aber er kann auch als Lösungsmittel dienen. Dieser Ausgangsstoff kann aber auch in einer äquivalenten Menge in Gegenwart von anderen Kondensationsmitteln, wie anorganischen oder organischen Basen, z.B. Alkalimetallcarbonaten oder -bicarbonaten oder tertiären Stickstoffbasen, z.B. Tri-niederalkylaminen, N,N-Dimethylanilin oder Pyridin, umgesetzt werden.
Eine verfahrensgemäss erhaltene Verbindung kann nach an sich bekannten Methoden in andere Verbindungen der vorliegenden Erfindung umgewandelt werden.
So kann z.B. eine Verbindung der allgemeinen Formel 1, worin R-Wasserstoff bedeutet, mit einem reaktionsfähigen Ester eines entsprechenden Alkohols umgesetzt, oder mit einem reaktionsfähigen funktionellen Derivat einer entsprechenden Säure, z.B. mit ihren Halogeniden oder Anhydriden, acyliert werden. Erhaltene Acylderivate können z.B. mit sauren oder basischen Hydrolysierungsmitteln hydrolysiert werden. Erhaltene Ester können hydrolysiert, umestert oder mit Aminen oder Alkoholaten umgesetzt, und erhaltene Alkohole verestert werden.
Je nach den Verfahrensbedingungen und Ausgangsstoffen kann der Endstoff in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form seiner Säureadditionssalze erhalten werden. So können beispielsweise basische, neutrale, saure oder gemischte Salze, gegebenenfalls auch Hemi-, Mono-, Sesqui- oder Polyhydrate davon, erhalten werden. Die Säureadditionssalze der neuen Verbindungen können in an sich bekannter Weise in die freie Verbindung übergeführt werden, zum Beispiel mit basischen Mitteln, wie Alkalien oder Ionenaustauscher. Andererseits kann die erhaltene freie Base mit organischen oder anorganischen Säuren Salze bilden.
Zur Herstellung von Säureadditionssalzen können insbesondere therapeutisch verwendbare Säuren verwendet werden, z.B. Halogenwasserstoffsäuren, Schwefelsäure, Phosphorsäuren, Salpetersäure, Perchlorsäure; aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxymalein- oder Brenztraubensäure; Phenylessig-, Benzoe-, p-Amino-benzoe-, Anthranil-, p-Hydroxy-benzoe-, Salicyl- oder p-Amino-salicylsäure, Embonsäure, Nikotinsäure, Methansulfon-, Äthansulfon-, Hydroxyäthansulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure; Methionin, Trytophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindung, wie z.B. die Pikrate, können auch zur Reinigung der erhaltenen freien Verbindungen dienen, indem man die freie Verbindung in Salze überführt, diese abtrennt und aus den Salzen wiederum die freie Verbindung freimacht.
Infolge der engen Beziehung zwischen der neuen Verbindung in freier Form und in Form ihrer Säureadditionssalze sind im vorausgegangenen und nachfolgend unter der freien Verbindung sinn- und zweckgemäss gegebenenfalls auch die entsprechenden Säureadditionssalze zu verstehen.
Je nach den verwendeten Ausgangsstoffen und Reaktionsbedingungen können die neuen Verbindungen in Form von optischen Antipoden, Isomerengemischen oder Racematgemischen erhalten werden, welche in einzelne Isomere oder Racemate nach an sich bekannten Methoden aufgespalten werden können. Die Verfahrensprodukte können auch in Form von Racematen vorliegen, welche nach an sich bekannten Methoden in die optischen Antipoden aufgetrennt werden können.
Die Erfindung betrifft auch diejenigen Ausführungsformen des Verfahrens, nach denen man einen Ausgangsstoff in Form eines unter den Reaktionsbedingungen erhältlichen rohen Reaktionsgemisches oder bei denen die Reaktionskomponenten gegebenenfalls in Form ihrer Salze verwendet. So können z.B. die oben genannten Amine in der Form ihrer Alkalimetallsalze, z.B. Natrium- oder Kaliumsalze, welche die Umsetzung mit Alkalimetallen oder ihren Hydriden ergibt, verwendet werden.
Beim Verfahren der vorliegenden Erfindung werden vorzugsweise solche Ausgangsstoffe verwendet, welche zu den eingangs als besonders wertvoll geschilderten Verbindungen führen.
Die Ausgangsstoffe sind bekannt, oder können nach an sich bekannten Methoden hergestellt werden. So kann z.B. der verwendete Ausgangsstoff durch Reduktion von entsprechenden 5,6-Dihydro-morphanthridin-6, 11 -dionen mit Natriumborhydrid und Veresterung der erhaltenen 11-Hydroxy-verbindung in üblicher Weise, z.B. mit Thionyl- oder Sulfonylhalogeniden, und Umsetzung der erhaltenen 11 -Halogenide mit entsprechenden Aminen der Formel

erhalten werden.
Die neuen Verbindungen können als Heilmittel, zum Beispiel in Form von pharmazeutischen Präparaten, verwendet werden, welche diese Verbindungen zusammen mit pharmazeutischen, organischen oder anorganischen, festen oder flüssigen Trägerstoffen, die für enterale, zum Beispiel orale, oder parenterale oder (als antiinflammatorische Mittel) topicale Gabe geeignet sind, enthalten.
Die Temperaturen sind in den folgenden Beispielen in Celsiusgraden angegeben.
Beispiel I
Ein Gemisch von 5 g 2-Chlor- 11 -(bis-2-hydroxyäthyl- amino)-5,6-dihydro-6-morphanthridon und 2,1 g Anilin wird mit Chlorwasserstoffsäure neutralisiert, in einem offenen Gefäss langsam auf 2250 erhitzt uund bei dieser Temperatur 4 Stunden gehalten. Nach Abkühlen wird das Reaktionsgemisch in verdünnter Chlorwasserstoffsäure aufgenommen, die Lösung mit Methylenchloridge- waschen, filtriert und das Filtrat mit wässrigem Natriumhydroxyd basisch gemacht. Das Gemisch wird mit Äther extrahiert, der Extrakt getrocknet, filtriert und eingedampft. Der Rückstand wird aus Methanol umkristallisiert. Man erhält das 2-Chlor-1 1(4-phenyl-piperazino)- -5,6-dihydro-6-morphanthridon der Formel

welches bei 248-2500 schmilzt.
Der Ausgangsstoff kann wie folgt hergestellt werden:
Ein Gemisch von 5 g 2,11 -Dichlor-5,6-dihydro-6 -morphanthridon und 10 ml Di-(2-hydroxy-äthyl)-amin wird auf dem Dampfbad 2 Stunden erhitzt. Das Reaktionsgemisch wird mit einem Luftstrom eingedampft und der Rückstand auf Eis-Wasser gegossen. Das Gemisch wird über Nacht bei Zimmertemperatur stehen gelassen, die wässrige Schicht abdekantiert, der Rückstand mit Wasser gewaschen und zuerst aus wässrigem Äthanol und dann aus Äthanol-Äther umkristallisiert. Man erhält das 2-Chlor- 11 -(bis-2-Hydroxy - äthylamino).5,6-dihydro- -6-morphanthridon, welches bei 172-1730 schmilzt.
In analoger Weise kann auch das 11 -(bis.2-Hydroxy- äthyl-amino)-5,6-dihydro-6-morphanthridon, welches bei 180-1850 schmilzt, erhalten werden.
In analoger Weise werden auch die folgenden Verbindungen erhalten: 2-Chlor-1 1 -(4-methyl-piperazino)-5,6.dihydro.6.morph.
anthridon, welches nach Umkristallisation aus Methanol bei 218.2200 schmilzt. Nach Abkühlen schmilzt es wieder bei 230-2320; 2-Chlor- 11 -2-hydroxy-äthyl) -piperazino-5,6-dihydro-6- -morphanthridon, F. 241-2430 (unter Zersetzung) nach Umkristallisation aus Äthanol-Äther; 2-Chlor-l 1 -(4-benzyl-piperazino).5,6.dihydro.6.morph.

Beispiel 2
Ein Gemisch von 2 g 2-Chlor-1 1.(4.methyl.piperazi.
no)-5,6-dihydro-6-morphanthridon in 50 ml Essigsäureanhydrid wird 3 Stunden unter Rückfluss gekocht und dann eingedampft. Der Rückstand wird mit Essigester trituriert und aus Essigester-Äther umkristallisiert. Man erhält das 5-Acetyl-2-chlor-11-(4-methyl-piperazino)-5,6- -dihydro-6-morphanthridon der Formel

welches bei 193-1950 schmilzt.
In analoger Weise wird auch das 5-Acetyl-11-(4-me- thyl - piperazino).5, 6.dihydro.6.morphanthridon, welches nach Umkristallisation aus Äther, bei 223-2250 schmilzt, hergestellt.
Beispiel 3
Herstellung von 160 000 Tabletten mit einem Gehalt von je 0,025 g der wirksamen Substanz:

Verfahren:
Der Wirkstoff, der Milchzucker, 2500 g der Maisstärke, der Pulverzucker und das Silicagel werden mit einem Sieb mit 1 mm Maschenweite in einen Mischer gesiebt und 20 Minuten langsam vermischt. Der Rest der Maisstärke wird in 1 Liter kaltem Wasser suspendiert und durch stufenweise Zugabe von 4 Liter kochendem Wasser in eine Paste umgewandelt. Die vermischten Pulver werden mit dieser Paste granuliert, wobei man gegebenenfalls eine weitere Wassermenge zusetzt. Die erhaltene feuchte Masse wird durch ein Sieb mit 4 mm Maschenweite getrieben und auf Platten bei 380 bis zu einem Feuchtigkeitsgehalt von 2-3% getrocknet.
Das Granulat wird in einer Schlagmühle (Messerschneiden vorwärts) bearbeitet, durch ein Sieb mit 1 mm Maschenweite gesiebt und mit Stearinsäure und Calciumstearat, welche man vorher mit einem Sieb mit 0,8 mm Maschenweite gesiebt hat, versetzt. Das Gemisch wird 20 Minuten gemischt und nachher zu Tabletten gepresst.
In analoger Weise können auch Präparate, welche andere beschriebene Produkte enthalten, hergestellt werden.
Beispiel 4
Ein Gemisch von 3,8 g 2-Chlor-1 1-(4.äthoxycarbonyl- piperazino)-5,6-dihydro-6-morphanthridon und 200 ml Toluol wird mit 1 g einer 56%igen Suspension von Natriumhydrid in Mineralöl versetzt und unter Rühren eine halbe Stunde unter Rückfluss gekocht. Man setzt nachher 50 ml Methyljodid zu und rührt das Gemisch unter Rückfluss weitere 5 Stunden. Nach Abkühlen wird das Gemisch mit Wasser und Äther versetzt, die organische Schicht abgetrennt, getrocknet, filtriert und eingedampft.
Der Rückstand wird in Äthanol aufgenommen, die Lösung mit Kohle entfärbt, filtriert und mit Wasser gesättigt. Der in der Kälte erhaltene Niederschlag wird abfiltriert und aus Äther umkristallisiert. Man erhält das 2-Chlor- 5. methyl-1 1.(4.äthoxycarbonyl.piperazino).5,6.

Verfahren:
Sämtliche Bestandteile werden durch ein Sieb mit 0,6 mm Maschenweite gesiebt. Der Wirkstoff, Milchzukker, Talk, Magnesiumstearat und die halbe Stärkenmenge wird in einem geeigneten Mischer vermischt. Der Rest der Stärke wird in 45 ml Wasser suspendiert und die Suspension wird zu einer siedenden Lösung von Polyäthylenglykol in 180 ml Wasser zugesetzt. Die erhaltene Paste wird zum Pulvergemisch zugesetzt und, gegebenenfalls unter Zugabe einer weiteren Wassermenge, granuliert. Das Granulat wird über Nacht bei 350 getrocknet, durch ein Sieb mit 1,2 mm Maschenweite getrieben und zu Tabletten gepresst.
Analog zu dem hier oder im Beispiel 10 beschriebenen Verfahren können auch pharmazeutische Präparate, welche als Wirkstoff pro Einheit ungefähr 25 bis 50 mg der in den Beispielen 1, 2, 4 bis 11, insbesondere Beispiel 7, genannten Verbindungen enthalten, hergestellt worden. Diese Präparateeinheiten können höchstens 3mal täglich verabreicht werden.
Die erfindungsgemässen Verbindungen vom in den Beispielen 8 bis 10 genannten Typus zeigen an Mäusen bei einer oralen Dosis zwischen ungefähr 10 und 50 mg/ kg/Tag auch eine depressive Wirkung auf das zentrale Nervensystem. Die entsprechenden pharmazeutischen Päparate können daher als Beruhigungsmittel verwendet werden.

Depending on the process conditions and starting materials, the end product can be obtained in free form or in the form of its acid addition salts, which is also included in the invention. For example, basic, neutral, acidic or mixed salts, optionally also hemi-, mono-, sesqui- or polyhydrates thereof, can be obtained. The acid addition salts of the new compounds can be converted into the free compound in a manner known per se, for example using basic agents such as alkalis or ion exchangers. On the other hand, the free base obtained can form salts with organic or inorganic acids.
In particular, therapeutically useful acids can be used for the preparation of acid addition salts, e.g. Hydrohalic acids, sulfuric acid, phosphoric acids, nitric acid, perchloric acid; aliphatic, alicyclic, aromatic or heterocyclic carboxylic or sulfonic acids such as formic, acetic, propionic, succinic, glycolic, lactic, malic, tartaric, lemon, ascorbic, maleic, hydroxymaleic or pyruvic acid ; Phenylacetic, benzoic, p-amino-benzoic, anthranil, p-hydroxy-benzoic, salicylic or p-amino-salicylic acid, emboxylic acid, nicotinic acid, methanesulphonic, ethanesulphonic, hydroxyethanesulphonic, ethylene sulphonic acid; Halobenzenesulfonic, toluenesulfonic, naphthalenesulfonic acids or sulfanilic acid; Methionine, trytophan, lysine or arginine.
These or other salts of the new compound, e.g. the picrates can also be used to purify the free compounds obtained by converting the free compound into salts, separating them and in turn liberating the free compound from the salts.
As a result of the close relationship between the new compound in free form and in the form of its acid addition salts, in the preceding and below the free compound is to be understood, appropriately and appropriately, also to mean the corresponding acid addition salts.
Depending on the starting materials used and the reaction conditions, the new compounds can be obtained in the form of optical antipodes, mixtures of isomers or mixtures of racemates, which can be split into individual isomers or racemates by methods known per se. The invention also relates to those embodiments of the process in which a starting material is used in the form of a crude reaction mixture obtainable under the reaction conditions or in which the reaction components are optionally used in the form of their salts. In the process of the present invention it is preferred to use those starting materials which lead to the compounds described at the beginning as being particularly valuable.
The starting materials are known or can be prepared by methods known per se. E.g. the starting material used by reducing the corresponding 5,6-dihydro-morphanthridine-6, 11 -diones with sodium borohydride and esterifying the 11-hydroxy compound obtained in the usual way, e.g. with thionyl or sulfonyl halides, and reaction of the 11 halides obtained with corresponding amines of the formula

The new compounds can be used as medicaments, for example in the form of pharmaceutical preparations, which contain these compounds together with pharmaceutical, organic or inorganic, solid or liquid excipients for enteral, for example oral, or parenteral or (as anti-inflammatory agents) topical administration are suitable.

Example I.

The raw material can be produced as follows:
A mixture of 5 g of 2,11-dichloro-5,6-dihydro-6-morphanthridone and 10 ml of di- (2-hydroxy-ethyl) -amine is heated on the steam bath for 2 hours. Procedure:
The active ingredient, the milk sugar, 2500 g of the corn starch, the powdered sugar and the silica gel are sieved into a mixer with a sieve with 1 mm mesh size and slowly mixed for 20 minutes. The rest of the corn starch is suspended in 1 liter of cold water and converted into a paste by gradually adding 4 liters of boiling water. The mixed powders are granulated with this paste, optionally adding a further amount of water. The moist mass obtained is forced through a sieve with a mesh size of 4 mm and dried on plates at 380 to a moisture content of 2-3%.
The granulate is processed in a hammer mill (knife cutting forward), sieved through a sieve with 1 mm mesh size and mixed with stearic acid and calcium stearate, which have previously been sieved with a sieve with 0.8 mm mesh size. The mixture is mixed for 20 minutes and then pressed into tablets.
Preparations which contain other products described can also be produced in an analogous manner.

A mixture of 3.8 g of 2-chloro-1 1- (4.ethoxycarbonyl-piperazino) -5,6-dihydro-6-morphanthridone and 200 ml of toluene is mixed with 1 g of a 56% suspension of sodium hydride in mineral oil and refluxed with stirring for half an hour. 50 ml of methyl iodide are then added and the mixture is stirred under reflux for a further 5 hours. After cooling, water and ether are added to the mixture, the organic layer is separated off, dried, filtered and evaporated.
The residue is taken up in ethanol, the solution is decolorized with charcoal, filtered and saturated with water. The precipitate obtained in the cold is filtered off and recrystallized from ether. The 2-chloro-5.methyl-1 1. (4.ethoxycarbonyl.piperazino) .5,6 is obtained.

Procedure:
All components are sieved through a sieve with a mesh size of 0.6 mm. The active ingredient, milk sugar, talc, magnesium stearate and half the amount of starch are mixed in a suitable mixer. The remainder of the starch is suspended in 45 ml of water and the suspension is added to a boiling solution of polyethylene glycol in 180 ml of water. The paste obtained is added to the powder mixture and granulated, optionally with the addition of a further amount of water. The granules are dried overnight at 350, driven through a sieve with a mesh size of 1.2 mm and pressed into tablets.
Analogous to the method described here or in example 10, pharmaceutical preparations containing as active ingredient per unit approximately 25 to 50 mg of the compounds mentioned in examples 1, 2, 4 to 11, in particular example 7, can also be produced. These preparation units can be administered a maximum of 3 times a day.
