Verfahren zur Herstellung von Alkanolaminderivaten Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Alkanolaminderivaten die ss-adrenergische Blockierungswirksamkeit besitzen und daher bei der Behandlung oder Prophylaxe von Herz krankheiten brauchbar sind.
Gewisse 1- Amino - 3 - phenoxy-2-propanolderivate sind bekannt, aber es war bisher nicht bekannt, dass Verbindungen dieses Typs ss-adrenergische Blockie rungswirksamkeit besitzen.
Erfindungsgemäss werden Alkanolaminderivate der Formel:

-verzweigten Aralkylrest mit 10 bis 15 Kohlen stoffatomen, R3 und R4, die gleich oder verschieden sein können, Wasserstoff oder Alkylreste mit bis zu 5 Kohlenstoffatomen und eines von A3 und A4 Was serstoff, das andere ein Halogenatom, oder die Hy- droxyl- oder Nitrogruppe, oder eine Alkyl-, Alkoxy-, Alkenyl-, Alkylthio- oder Halogenalkylgruppe jeweils mit bis zu 4 Kohlenstoffatomen, oder eine Acyl-, Aryl-, Aryloxy- (z. B. Alkylaryloxy-), Arylthio-, Arylsulfonyl-, Arylamino-, Aralkyl- oder Aralkoxygruppe jeweils mit bis zu 10 Kohlenstoffatomen, bzw. ihre Salze herge stellt.
Es versteht sich, dass die obige Definition der Alkanolaminderivate alle möglichen Stereoisomere der selben und Gemische davon umfasst.
Als geeignete Bedeutung von R2 sei z. B. der Iso- propyl-, sek.-Butyl-, tert.-Butyl-, 1-Methyloctyl-, 2-Hy- droxy -1,1- dimethyläthyl-, 1- Methyl-3-phenylpropyl-, Cyclopentyl und Allylrest erwähnt.
Als geeignete Bedeutung von R3 und R4, wenn sie Alkylreste bedeuten, sei z. B. der Methylrest genannt. Als geeignete Bedeutung des Substituenten A3 oder A4 seien beispielsweise die folgenden genannt: die Fluor-, Chlor-, Brom- und Jodatome, die Hydroxyl-, Nitro-, Methyl-, Äthyl-, Isopropyl-, tert.-Butyl-, Meth- oxy-, Äthoxy-, n-Butoxy-, Allyl-, Methylthio-, Ace- tyl-, Benzoyl-, Trifluormethyl-, Phenyl-, Phenoxy-, 4- Tolyloxy-, Phenylthio-, Phenylsulfonyl-, Anilino-, Ben zyl-, α,α-Dimethylbenzyl- und Benzyloxyreste.
Als besonders wertvolle Verbindungen, die ver fahrensgemäss hergestellt werden können, seien z. B. genannt: 1-(3-Tolyloxy)-, 1-(4-Tolyloxy)-, 1-(3-Chlorphenoxy)-, 1-(3-Fluorphenoxy)-, 1-(3-Methoxyphenoxy)-, 1-(3-Nitrophenoxy)-, 1-(3-Trifluormethylphenoxy)-, 1-(3-Phenoxyphenoxy)- und 1-(4-Anilinophenoxy)-3-isopropylamino-2-propanol, 1-(2-Hydroxy-1,1-dimethyläthylamino)-3-(3-tolyloxy)- 2-propanol und 1-tert.-Butylamino-3-(3-tolyloxy)-2-propanol und ihre Salze.
Als geeignete Salze der Alkanolamine seien die fol genden erwähnt: Säureadditionssalze, z. B. von anorga nischen Säuren abgeleitete Salze, beispielsweise Hy- drochloride, Hydrobromide, Phosphate oder Sulfate, oder von organischen Säuren abgeleitete Salze, beispiels weise Oxalate, Lactate, Tartrate, Acetate, Salicylate, Citrate, Benzoate, ss-Naphthoate, Adipate oder 1,1'- Methylen-bis-(2-hydroxy-3-naphthoate), oder von sau ren synthetischen Harzen, beispielsweise von sulfonier- ten Polystyrolharzen, z.
B. Zeo-Karb 225 ( Zeo- Karb ist eine eingetragene Marke), abgeleitete Salze. Verhältnismässig unlösliche Salze, beispielsweise die 1,1'-Methylen-bis-(2-hydroxy-3-naphthoate), haben den Vorteil, dass sie verlängerte Blutspiegel des Medika mentes liefern.
Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass man eine Verbindung der Formel:

Als eine geeignete Bedeutung von Z sei beispiels weise ein Chlor- oder Bromatom erwähnt. Die Um setzung kann zweckmässig durch Zuführung von Wärme beschleunigt oder vervollständigt werden, und sie kann in einem inerten Verdünnungsmittel oder Lösungsmittel ausgeführt werden.
In den folgenden Beispielen sind die Teile gewichts mässig angegeben.
<I>Beispiel 1</I> Ein Gemisch von 1,94 Teilen 1,2-Epoxy-3-(3'- äthoxyphenoxy)-propan und 25 Teilen Isopropylamin wird 2 Stunden lang unter Rückfluss erhitzt. Überschüs siges Isopropylamin wird durch Destillation unter ver mindertem Druck entfernt. Der Rückstand wird in 2n Salzsäure gelöst. Die Lösung wird zweimal mit 50 Tei len Äther gewaschen und dann mit 2n Natriumhydro xydlösung basisch gemacht. Das Gemisch wird zweimal mit 100 Teilen Äther extrahiert. Die vereinigten Äther extrakte werden mit 100 Teilen Wasser gewaschen, über wasserfreiem Magnesiumsulfat getrocknet und fil triert. Das Lösungsmittel wird durch Destillation ent fernt. Der Rückstand wird in 20 Teilen Äther gelöst, und ätherische Oxalsäure wird zugegeben, bis die Ausfällung beendet ist.
Das Gemisch wird filtriert, und der feste Rückstand wird aus Dimethylformamid kri stallisiert. Auf diese Weise wird 1-(3'-Äthoxy-phenoxy)-3-isopropylamino-2-propanol- oxalat vom Schmelzpunkt 155-156 C erhalten. <I>Beispiel 2</I> Ein Gemisch von 10,8 Teilen m-Kresol, 25 Teilen Epichlorhydrin, 4,8 Teilen Natriumhydroxyd und 100 Teilen Wasser wird 24 Stunden lang bei Umgebungs temperatur gerührt. Das Gemisch wird mit Eisessig neutralisiert und dann zweimal mit 200 Teilen Chloro form extrahiert. Die vereinigten Chloroformextrakte werden dreimal mit einem gleichen Volumen Wasser gewaschen und dann über wasserfreiem Magnesium sulfat getrocknet.
Das Gemisch wird filtriert, und das Lösungsmittel wird durch Destillation unter verminder tem Druck entfernt. Der Rückstand besteht aus 1,2- Epoxy-3-(3'-tolyloxy)-propan. Dieses wird zusammen mit 60 Teilen Isopropylamin eine Stunde lang am Rück fluss erhitzt, und das Gemisch wird dann 18 Stunden lang bei Umgebungstemperatur gehalten. Überschüs siges Isopropylamin wird durch Destillation entfernt. Der Rückstand wird in 500 Teilen Wasser suspendiert, und konzentrierte Salzsäure wird zugegeben, bis die Auflösung bewirkt worden ist. Die Lösung wird zweimal mit einem gleichen Volumen Äther gewaschen. Die wässrige Lösung wird mit 2n Natriumhydroxydlösung basisch gemacht, und das resultierende Gemisch wird filtriert.
Der feste Rückstand wird mit Wasser gewa schen, getrocknet und aus Cyclohexan kristallisiert. Auf diese Weise wird 1-Isopropylamino-3-(3'-tolyloxy)-2- propanol vom Schmelzpunkt 79 C erhalten.
In ähnlicher Weise wird durch Verwendung von 4- Benzyloxyphenol anstelle von m-Kresol 1-(4'-Benzyl- oxyphenyl)-3-isopropylamino-2-propanol vom Schmelz punkt 100-101 C (kristallisiert aus Cyclohexan) er halten.
<I>Beispiel 3</I> Ein Gemisch von 2,1 Teilen 1-Brom-3-(3'-tolyloxy)- 2-propanol und 15 Teilen Isopropylamin wird in einem zugeschmolzenen Rohr 6 Stunden lang auf 100 C er hitzt. Das Produkt wird mittels eines ähnlichen Ver fahrens wie des in Beispiel 1 beschriebenen isoliert, und es wird 1-Isopropylamino-3-(3'-tolyloxy)-2-propanol vom Schmelzpunkt 79 C (kristallisiert aus Cyclohe- xan) erhalten.
Das als Ausgangsmaterial verwendete 1-Brom-3- (3'-tolyloxy)-2-propanol kann folgendermassen erhalten werden: Ein Gemisch von 9 Teilen 3-Tolyloxyacetylchlorid in 150 Teilen wasserfreiem Äther wird tropfenweise während eines Zeitraumes von 30 Minuten zu einer gekühlten (-13 C) Lösung von 16,5 Teilen Diazome- than in wasserfreien Äther gegeben. Das Reaktions gemisch wird während der Zugabe und danach noch eine Stunde lang vorsichtig gerührt. Man lässt die Tem peratur der Lösung auf Umgebungstemperatur steigen, und die Lösung wird 18 Stunden lang bei dieser Tem peratur gehalten.
Die Lösung wird dann gerührt und auf -15 C abgekühlt, und 20 Teile 48 %ige Bromwasser stoffsäure werden tropfenweise zugegeben. Das Ge misch wird getrennt, und die ätherische Schicht wird mit einem gleichen Volumen von 5 %iger wässriger Natriumcarbonatlösung und dann zweimal mit einem gleichen Volumen Wasser gewaschen. Die ätherische Lösung wird getrocknet (wasserfreies Magnesiumsulfat), und das Lösungsmittel wird dann durch Destillation unter vermindertem Druck entfernt.
Der Rückstand ist 1-Brom-3-(3'-tolyloxy)-2-propa- non, dessen 2,4-Dinitrophenylhydrazon einen Schmelz punkt von 128-130 C aufweist (kristallisiert aus Ätha- nol). 2 Teile dieses Rückstandes werden in 25 Teilen Methanol gelöst, und die Lösung wird auf 0 C abge kühlt. 2 Teile Natriumborhydrid werden portionenweise zu der gekühlten Lösung gegeben, und das Gemisch wird 18 Stunden lang auf Umgebungstemperatur gehal ten. Das Gemisch wird in ein Gemisch von Eis und Salzsäure gegossen, und die resultierende saure Lösung wird dreimal unter Verwendung je eines gleichen Volu mens an Chloroform extrahiert.
Die vereinigten Chloro- formextrakte werden zweimal mit einem gleichen Vo lumen Wasser gewaschen, über wasserfreiem Magne- siumsulfat getrocknet und dann filtriert. Das Chloro form wird dann durch Destillation unter vermindertem Druck entfernt, und als Rückstand wird 1-Brom-3- (3'-tolyloxy)-2-propanol erhalten. <I>Beispiel 4</I> Ein Gemisch von 2 Teilen 3-Chlor-1-(3'-tolyloxy)- 2-butanol und 15 Teilen Isopropylamin wird in einem verschlossenen Gefäss 10 Stunden lang auf 100 C er hitzt.
Das Produkt wird analog Beispiel 1 isoliert, und es wird 3-Isopropylamino-1-(3'-tolyloxy)-2-butanol- monohydrat vom Schmelzpunkt 78-80 C (kristallisiert aus wässrigem Äthanol) erhalten.
Das als Ausgangsmaterial verwendete 3-Chlor-1- (3'-tolyloxy)-2-butanol kann folgendermassen erhalten werden: Eine Lösung von 10 Teilen 3-Tolyloxyacetylchlorid in 150 Teilen wasserfreiem Äther wird tropfenweise während eines Zeitraumes von 30 Minuten zu einer gut gekühlten (-15 C) und vorsichtig gerührten Lösung von 13,6 Teilen Diazoäthan in 300 Teilen wasserfreiem Äther gegeben. Nach der Zugabe wird das Reaktions gemisch eine Stunde lang bei -10 C gerührt. Dann lässt man die Temperatur der Lösung auf Umgebungs temperatur steigen, und die Lösung wird 10 Stunden lang bei Umgebungstemperatur gehalten.
Die Lösung wird dann auf -15 C abgekühlt, und wasserfreier Chlorwasserstoff wird durch die Lösung perlen gelas sen, bis die Entwicklung von Stickstoff aufhört. Die Lösung wird dann zweimal mit einem gleichen Volu men Wasser, einmal mit einem gleichen Volumen 5 %iger Natriumcarbonatlösung und wieder mit einem gleichen Volumen Wasser gewaschen. Die Lösung wird dann getrocknet (wasserfreies Magnesiumsulfat).
Das Lösungsmittel wird unter vermindertem Druck entfernt, wobei sich als Rückstand 3-Chlor-1-(3'-tolyloxy)-2-bu- tanon, dessen 2,4-Dinitrophenylhydrazon einen Schmelz punkt von 118-119 C (kristallisiert aus Äthanol) er gibt. 5 Teile dieses Rückstandes werden in 25 Teilen Äthanol gelöst, und die Lösung wird auf 0 C abgekühlt. Zu der Lösung werden 5 Teile Natriumborhydrid in Portionen zugegeben, und das resultierende Gemisch wird 4 Stunden lang auf Umgebungstemperatur ge halten. Das Gemisch wird in ein Gemisch von Eis und Salzsäure gegossen, und die resultierende Lösung wird zweimal mit gleichen Volumen Äther extrahiert.
Die vereinigten Extrakte werden mit einem gleichen Volu men Wasser gewaschen und getrocknet (wasserfreies Magnesiumsulfat). Das Lösungsmittel wird durch De stillation unter vermindertem Druck entfernt, und auf diese Weise wird als Rückstand 3-Chlor-1-(3'-tolyloxy)- 2-butanol erhalten.
<I>Beispiel 5</I> Die in Beispiel 4 beschriebene Verfahrensweise wird wiederholt mit der Ausnahme, dass das 3-Chlor-1- (3'-tolyloxy)-2-butanol durch 1-Chlor-3-(3-tolyloxy)-2- butanol ersetzt wird, und auf diese Weise wird 1-Isopropylamino-3-(3'-tolyloxy)-2-butanol- oxalathydrat vom Schmelzpunkt 98-108 C erhalten (kristallisiert aus einem Gemisch von Athylacetat und Äthanol). Diese Verbindung wird in der folgenden Weise in ihre 2 Diastereomere getrennt.
Die Verbindung wird zusammen mit 20 Teilen Äthylacetat 10 Minuten lang auf 80 C erhitzt, und das heisse Gemisch wird dann filtriert, wobei sowohl der feste Rückstand als auch das Filtrat zurückbehalten werden. Der feste Rückstand wird mehrere Male aus einem Gemisch von Äthylacetat und Äthanol umkri stallisiert, bis Material mit einem konstanten Schmelz punkt erhalten wird. Auf diese Weise wird das eine Diastereomere von 1-Isopropylamino-3-(3'-tolyloxy)-2- butanol-oxalat (Schmelzpunkt 151-152 C) erhalten. Das oben erwähnte Filtrat wird abgekühlt, und das re sultierende Gemisch wird filtriert.
Der feste Rückstand wird mehrere Male aus Äthylacetat kristallisiert, bis ein Material mit einem konstanten Schmelzpunkt er halten wird, und auf diese Weise wird das andere Diastereomere von 1-Isopropylamino-3-(3'-tolyloxy)-2- butanol-oxalat vom Schmelzpunkt 92-94 C erhalten.
Das als Ausgangsmaterial verwendete 1-Chlor-3- (3'-tolyloxy)-2-butanol kann mittels ähnlichen Verfah rensweisen wie die im Beispiel 4 beschriebenen aus gehen von α-(3-Tolyloxy)-propionylchlorid und Diazo- methan erhalten werden, und α-(3-Tolyloxy)-propio- nylchlorid seinerseits kann folgendermassen erhalten werden: Ein Gemisch von 10 Teilen α-(3-Tolyloxy)-pro- pionsäure und 6,6 Teilen Thionylchlorid wird 4 Stunden lang auf 60 C erhitzt. Das Gemisch wird abgekühlt, zusammen mit 20 Teilen Petroläther (Siedebereich 60 bis 80 C) gerührt und filtriert.
Das Filtrat wird unter vermindertem Druck eingedampft, und es wird als Rückstand α-(3-Tolyloxy)-propionylchlorid erhalten.
<I>Beispiel 6</I> Nach denselben Verfahrensweisen wie in den Bei spielen 1, 2 und 3, unter Verwendung der entsprechen den Ausgangsprodukte, werden die in folgenden Tabel len angeführten Verbindungen erhalten:

The mixture is filtered and the solid residue is crystallized from dimethylformamide. The aqueous solution is made basic with 2N sodium hydroxide solution and the resulting mixture is filtered.
The solid residue is washed with water, dried and crystallized from cyclohexane. 1-Isopropylamino-3- (3'-tolyloxy) -2-propanol with a melting point of 79 ° C. is obtained in this way.
Similarly, by using 4-benzyloxyphenol instead of m-cresol, 1- (4'-benzyloxyphenyl) -3-isopropylamino-2-propanol with a melting point of 100-101 ° C. (crystallized from cyclohexane) is obtained.
